Epidemiological studies show that elevated plasma levels of advanced glycation end products (AGEs) are associated with diabetes, kidney disease, and heart disease. Thus AGEs have been used as disease progression markers. However, the effects of variations in biological sample processing procedures on the level of AGEs in plasma/serum samples have not been investigated. The objective of this investigation was to assess the effect of variations in blood sample collection on measured Ne_(carboxy-methyl)lysine (CML), the best characterised AGE, and its homolog, Ne_(carboxyethyl)lysine (CEL). The investigation examined the effect on CML and CEL of different blood collection tubes, inclusion of a stabilising cocktail, effect of freeze thaw cycles, different storage times and temperatures, and effects of delaying centrifugation on a pooled sample from healthy volunteers. CML and CEL were measured in extracted samples by ultra_performance liquid chromatography-tandem mass spectrometry. Median CML and CEL ranged from 0.132 to 0.140 mM/M lys and from 0.053 to 0.060 mM/M lys, respectively. No significant difference was shown CML or CEL in plasma/serum samples. Therefore samples collected as part of epidemiological studies that do not undergo specific sample treatment at collection are suitable for measuring CML and CEL.

Cationic amphipathic α-helical peptides are intensively studied classes of host defence peptides (HDPs). Three peptides, peptide glycine-leucine-amide (PGLa-AM1), caerulein-precursor fragment (CPF-AM1) and magainin-AM1, originally isolated from norepinephrine-stimulated skin secretions of the African volcano frog Xenopus amieti (Pipidae), were studied for their antimicrobial and immunomodulatory activities against oral and respiratory pathogens. Minimal effective concentrations (MECs), determined by radial diffusion assay, were generally lower than minimal inhibitory concentrations (MICs) determined by microbroth dilution. PGLa-AM1 and CPF-AM1 were particularly active against Streptococcus mutans and all three peptides were effective against Fusobacterium nucleatum, whereas Enterococcus faecalis and Candida albicans proved to be relatively resistant micro-organisms. A type strain of Pseudomonas aeruginosa was shown to be more susceptible than the clinical isolate studied. PGLa-AM1 displayed the greatest propensity to bind lipopolysaccharide (LPS) from Escherichia coli, P. aeruginosa and Porphyromonas gingivalis. All three peptides showed less binding to P. gingivalis LPS than to LPS from the other species studied. Oral fibroblast viability was unaffected by 50. μM peptide treatments. Production of the pro-inflammatory cytokine IL-8 by oral fibroblasts was significantly increased following treatment with 1 or 10. μM magainin-AM1 but not following treatment with PGLa-AM1 or CPF-AM1. In conclusion, as well as possessing potent antimicrobial actions, the X. amieti peptides bound to LPS from three human pathogens and had no effect on oral fibroblast viability. CPF-AM1 and PGLa-AM1 show promise as templates for the design of novel analogues for the treatment of oral and dental diseases associated with bacteria or fungi.

Apesar dos hidratos de carbono representarem apenas 1% do peso corporal, são essencialmente importantes nas reações químicas que fornecem energia às células, principalmente ao cérebro. A glicose é o monossacarídeo com maior relevância para obtenção de energia. As outras oses provenientes dos hidratos de carbono vão-se integrar no metabolismo da glicose. Para manter a glicémia dentro do intervalo de valores de referência, há um sistema de regulação endócrino, do qual a insulina e o glucagon desempenham um papel predominante. A hipoglicémia define-se como um estado metabólico caracterizado por níveis de glicémia inferiores a 55 mg/dL, acompanhada de manifestações clínicas de intensidade e expressão variáveis, que refletem sintomas como a ansiedade, palpitações, tremores, défice cognitivo e coma. A glicose presente no organismo pode ser proveniente da dieta, ou da produção endógena. Deste modo, a hipoglicémia pode resultar de um consumo excessivo de glicose (exercício físico ou aumento de perdas externas) ou de um inadequado aporte de glicose (produção endógena insuficiente ou inanição). A hipoglicémia é uma complicação aguda, muito frequente, que surge como consequência do tratamento da diabetes com insulina e/ou sulfonilureias e, com muito menos frequência, no individuo não diabético. Neste, a hipoglicémia pode ser consequência primária de uma patologia, ou seja, por intervenção direta no metabolismo da glicose, ou consequência secundária de uma patologia, ou seja, por um mecanismo não direto. O estado de jejum ou pós-prandial do individuo quando surge a hipoglicémia também auxilia no diagnóstico diferencial. Para estabelecer o diagnóstico é necessário se verificar a tríade de Whipple: (1) sinais e sintomas compatíveis com hipoglicémia; (2) baixa concentração de glicémia; (3) melhoria dos sintomas após aumento da glicémia. O tratamento da hipoglicémia passa pela administração de hidratos de carbono, nomeadamente sacarose. O presente trabalho tem como objetivo, numa primeira parte, fazer a revisão, do ponto de vista bioquímico, dos mecanismos associados ao metabolismo dos glícidos e da regulação da glicémia que contribuem para a rápida correção da hipoglicémia. Numa segunda parte, pretende-se descreve-se as manifestações clinicas, as várias classificações de hipoglicémia e os mecanismos em cada caso/patologia. Numa terceira parte apresenta-se a abordagem diagnóstica na suspeita de hipoglicémia e qual a terapêutica mais adequada. Por último pretende-se evidenciar a relação da hipoglicémia versus hiperglicemia. Enquanto estados crónicos de hiperglicemia têm sido associados a disfunções de vários órgãos a longo prazo, episódios hipoglicémicos, apesar de pontuais, estão associados, a lesões neurológicas a curto prazo e até morte. Associações cientificas como The American Diabetes Association e The Endocrine Society têm vindo a disponibilizar informação para uma maior compreensão do episódio hipoglicémico, as suas implicações e estratégias para prevenção. Neste sentido, concluiu-se que é essencial identificar o mecanismo que conduziu à hipoglicémia de modo a prevenir a sua recorrência.

Background We present new findings on liver steatosis detected in a group of 20 morbidly obese patients who were reassessed shortly after bariatric surgery (BS) by assaying hepatic markers in their serum.Methods We assayed aspartate aminotransferase (AST), alanine aminotransferase, lactate dehydrogenase, alkaline phosphatase, gamma-glutamyl transferase (gamma-GT), cholinesterase, cholesterol, total protein, and albumin, and measured the weight and the body mass index (BMI) of patients, before and one and three months after surgery.Results There were significant reductions in BMI following surgery and also falls in transaminases and gamma-GT activities three months after BS. No changes occurred in other parameters between periods, except that cholesterol was above reference values before BS and fell to normal levels three months after BS.Conclusions We suggest that before undergoing surgery, the patients suffered from slight steatosis, while after BS the reduction in AST and gamma-GT indicated that this condition was corrected within three months. Moreover, these enzymes may be useful markers for excess fat in the liver.

The values for the main hepatic enzymes included in the profiles of screen clinical biochemistry, alanine-aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (FA), lactate desidrogenase (LDH) and gamaglutamiltransferase (GGT), in samples of serum of broiler chickens in industrial system, clinically healthy, starting from the seventh day of life, until the slaughter (42 days) in weekly intervals were determined. Significant variations were not observed in the analyses in relation to the age of the birds for none of the appraised enzymes.

Clinical pathology has been used in veterinary medicine for many years to evaluate metabolic changes that may indicate any sign of disease in animals. Even though the use of blood parameters is routine in companion animals, it is not commonly used for avian clinical evaluation either wild or production birds. Among others, the lack of information of avian blood reference values has limited the use of this tool in avian medicine. The blood parameters in avian medicine are helpful to evaluate aspects related to animal health for wild and also production birds. Thus, this review paper presents some ways to use clinical pathology parameters to evaluate and to monitor the avian health.

Introduction: Calculated indexes from laboratory measurements and the determination of plasma markers are commonly used in cardiovascular risk assessment. Objective: To use triglycerides/high-density lipoprotein cholesterol (HDL-C) ratio, estimate of low-density lipoprotein (LDL) particle size and high sensitive C-reactive protein (hs-CRP) levels to the clinical and laboratorial assessment of coronary risk among patients with coronary disease. Material and methods: Sixty patients were chosen accordingly, 36 female (F) (65 ± 9 years of age) and 24 male (M) (55 ± 10 years of age) diagnosed with stable ischemic cardiopathy at Ana Neri Hospital, Salvador, Bahia, Brazil. The investigation was carried out at the Clinical Biochemistry Laboratory of the College of Pharmacy at Federal University of Bahia (UFBA). Blood samples were collected after a 12-hour fast to determine hs-CRP, lipid profile and risk indicators. All data were assessed through Syncron LX®20 Beckman Coulter. The inferential analysis results were considered significant when p ≤ 5%. Data analysis was carried out with GraphPad Prism® (4.03 version) software. Results: Data analysis showed a positive correlation between hs-CRP and triglycerides/HDL-C ratio only among women (r = 0.4; p = 0.0442), n = 36. The analysis of other parameters did not show any significance. Conclusion: The triglycerides/HDL-C ratio and hs-CRP may be used as indicators of increase in cardiovascular risk among women with coronary artery disease (CAD).
