A correlation between cancer and hypercoagulability has been described for more than a century. Patients with cancer are at increased risk for thrombotic complications and the clotting initiator protein, tissue factor (TF), is possibly involved in this process. Moreover, TF may promote angiogenesis and tumor growth. In addition to TF, thrombin seems to play a relevant role in tumor biology, mainly through activation of protease-activated receptor-1 (PAR-1). In the present study, we prospectively studied 39 lung adenocarcinoma patients in relation to the tumor expression levels of TF and PAR-1 and their correlation with thrombosis outcome and survival. Immunohistochemical analysis showed TF positivity in 22 patients (56%), most of them in advanced stages (III and IV). Expression of PAR-1 was found in 15 patients (39%), most of them also in advanced stages (III and IV). Remarkably, no correlation was observed between the expression of TF or PAR-1 and risk for thrombosis development. On the other hand, patients who were positive for TF or PAR-1 tended to have decreased long-term survival. We conclude that immunolocalization of either TF or PAR-1 in lung adenocarcinoma may predict a poor prognosis although lacking correlation with thrombosis outcome.

A bacterial strain (PAP04) isolated from cattle farm soil was shown to produce an extracellular, solvent-stable protease. Sequence analysis using 16S rRNA showed that this strain was highly homologous (99%) to Brevibacillus laterosporus. Growth conditions that optimize protease production in this strain were determined as maltose (carbon source), skim milk (nitrogen source), pH 7.0, 40°C temperature, and 48 h incubation. Overall, conditions were optimized to yield a 5.91-fold higher production of protease compared to standard conditions. Furthermore, the stability of the enzyme in organic solvents was assessed by incubation for 2 weeks in solutions containing 50% concentration of various organic solvents. The enzyme retained activity in all tested solvents except ethanol; however, the protease activity was stimulated in benzene (74%) followed by acetone (63%) and chloroform (54.8%). In addition, the plate assay and zymography results also confirmed the stability of the PAP04 protease in various organic solvents. The organic solvent stability of this protease at high (50%) concentrations of solvents makes it an alternative catalyst for peptide synthesis in non-aqueous media.

A disponibilidade do ferro é fator importante na dieta, pois a anemia ferropriva atinge grande parte da população mundial. A cisteína e a vitamina C são fatores que melhoram sua absorção. A pesquisa teve por objetivos avaliar a disponibilidade de ferro em carne, feijão comum (Phaseolus vulgaris) e a combinação de arroz e tomate ao feijão, bem como analisar se o ácido fítico, o ácido oxálico e os taninos das combinações contendo feijão influenciam na disponibilidade de ferro de forma significativa. Os alimentos foram preparados para consumo e as amostras obtidas com a combinação de carne, feijão, arroz e tomate foram: A1 - carne bovina; A2 - feijão comum; A3 - feijão comum e tomate; A4 - feijão comum e arroz; e A5 - feijão comum, arroz e tomate. Foram realizadas as análises de taninos, ácido fítico, ácido oxálico, vitamina C e teor e diálise de ferro in vitro. Os teores de taninos, ácido fítico e ácido oxálico foram baixos para influenciar a disponibilidade do ferro. A diálise de 5,73 a 13,61% indicando que a adição de cisteína e vitamina C em conjunto tem efeito maior do que quando presentes separadamente.

The common bean (Phaseolus vulgaris L.) is a staple food in the Brazilian diet and represents the major source of dietary protein and other micronutrients and minerals. Despite the considerable protein concentration in beans, the food is considered of low biological value when compared to animal proteins and other plant protein sources. To improve the availability of protein in beans, enzymatic treatments were performed in four cultivars (ON, OPNS, TAL and VC3). The approach was a completely randomized design with four replicates. We used a 4 × 3 factorial arrangement (four cultivars and three treatments: treatment 1-addition of commercial protease (Trypsin 250, Difco), treatment 2-addition of protease from Bacillus sp., and treatment 3:-control without enzyme addition). The enzyme: substrate ratio was 5% w/w (amount of enzyme per total protein in bean flour). The approach was a completely randomized design with four replicates. A 4 × 3 factorial arrangement (four cultivars and three treatments, the same as those mentioned above) was used. The concentration of total protein (g.100 g-1 of dry matter) in the samples ranged from 16.94 to 18.06%, while the concentration of total phenolics was between 0.78 and 1.12% (g Eq. tannic acid.100 g-1 dry matter). The in vitro protein digestibility of enzymatically untreated bean flour (control) ranged from 47.30 to 56.17% based on the digestibility of casein. Concentrations of P, K, Ca, Mg, and Zn observed in the four cultivars tested were within the average values available in the literature. Treatment 2 with protease from Bacillus sp. induced decreases in the levels of Cu and Mn. The average Fe content increased in all bean flour samples when treated with proteases, reaching a maximum increase of 102% in the TAL flour treated with protease from Bacillus sp. The digestibility of all beans tested was significantly increased (p < 0.05) after the enzyme treatment. The greatest change was observed in the OPNS cultivar treated with protease from Bacillus sp., which increased its digestibility from 54.4% (control treatment) to 81.6%.

Protease and α-amylase production by a thermophilic Bacillus sp. SMIA-2 cultivated in liquid cultures containing 0.25% (w/v) starch as a carbon source reached a maximum at 18 hours (47 U.mg-1 Protein) and 36 hours (325 U.mg-1 Protein), respectively. Culture medium supplementation with whey protein concentrate (0.1%, w/v) and corn steep liquor (0.3%, w/v) not only improved the production of both enzymes but also enabled them to be produced simultaneously. Under these conditions, α-amylase and protease production reached a maximum in 18 hours with levels of 401 U.mg-1 protein and 78 U.mg-1 protein, respectively. The compatibility of the enzymes produced with commercial laundry detergent was investigated. In the presence of Campeiro® detergent, α-amylase activity increased while protease activity decreased by about 27%. These enzymes improved the cleaning power of Campeiro® detergent since they were able to remove egg yolk and tomato sauce stains when used in this detergent.

Abstract This study evaluated the chemical and volatile composition of jujube wines fermented with Saccharomyces cerevisiae A1.25 with and without pulp contact and protease treatment during fermentation. Yeast cell population, total reducing sugar and methanol contents had significant differences between nonextracted and extracted wine. The nonextracted wines had significantly higher concentrations of ethyl 9-hexadecenoate, ethyl palmitate and ethyl oleate than the extracted wines. Pulp contact also could enhance phenylethyl alcohol, furfuryl alcohol, ethyl palmitat and ethyl oleate. Furthermore, protease treatment can accelerate the release of fusel oils. The first principal component separated the wine from the extracted juice without protease from other samples based on the higher concentrations of medium-chain fatty acids and medium-chain ethyl esters. Sensory evaluation showed pulp contact and protease could improve the intensity and complexity of wine aroma due to the increase of the assimilable nitrogen.

Le dogme voulant que les récepteurs couplés aux protéines G (GPCRs) activent des voies de signalisation seulement lorsqu’ils sont localisés à la membrane plasmatique, a récemment été remis en question. Des données récentes indiquent que certains GPCRs peuvent également induire une réponse intracellulaire à partir des compartiments intracellulaires dont le noyau. Les récepteurs activés par la protéase (PAR) sont des membres de la famille GPCR. Les PARs sont activés par le clivage de la partie N–terminale du récepteur ce qui permet au ligand attaché sur le récepteur de se lier à sa poche réceptrice. Quatre PARs ont été décrits : PAR1, PAR2, PAR3 et PAR4. PAR2 peut susciter des effets mitogéniques et participer aux processus comme l’angiogenèse et l'inflammation. Alors que beaucoup d'effets intracellulaires de PAR2 peuvent être expliqués lorsqu’il est localisé à la membrane plasmatique, une fonction intracrine de PAR2 a aussi été proposée. Pourtant les mécanismes par lesquels PAR2 peut provoquer l’expression de gènes ciblés sont toujours inconnus. Le but de notre étude était de vérifier l’existence d’une population nucléaire de PAR2. Nous avons également émis l’hypothèse que les voies activées par l’activation de PAR2 dépendent de sa localization cellulaire. En utilisant des techniques de microscopie confocale et de "Western Blot" nous avons démontré la présence d’une population nucléaire de PAR2. À la suite de la stimulation de PAR2, nous avons observé une augmentation de la translocation du récepteur de la membrane plasmatique au noyau. En utilisant la technique de "RT – PCR", nous avons observé des rôles différents de PAR2 à la surface de la cellule et du noyau dans l’initiation de l’expression des gènes. Afin d’identifier les mécanismes responsables de la translocation nucléaire de PAR2, nous avons évalué l’implication des membres de la famille de "Sorting Nexins (SNX)" dans la translocation nucléaire de PAR2. "Sorting Nexins" est un groupe de protéines avec des fonctions de transport bien établies. SNX1 et SNX2 ont été identifiés comme responsables du transfert de PAR1 vers les lysosomes. SNX11 n'a pas encore été étudié et nous avons émis l’hypothèse qu'il pourrait être un autre membre de la famille des SNX impliqué dans la signalisation de PAR2. Pour ce faire, nous avons développé des "knockdowns" stables pour SNX1, SNX2 et SNX11 dans les cellules HEK293. En utilisant les essais d’immunofluorescence, "Western Blot" et de cytométrie en flux, nous avons déterminé que tous les trois membres du groupe SNX sont des partenaires d'interaction de PAR2. Toutefois, seul SNX11 se co-localise avec son partenaire au noyau et est responsable de sa translocation nucléaire. Les expériences de "RT - PCR" sur les lignées de cellule de SNXs "knockdowns" ont démontré que la fonction de PAR2 nucléaire dépend surtout de SNX11; néanmoins SNX1 et SNX2 peuvent aussi l’influencer, suggérant qu'ils font aussi partie du réseau signalétique de PAR2. En conclusion, PAR2 est déplacé de la membrane plasmatique à la membrane nucléaire après sa stimulation avec un agoniste. La translocation nucléaire de PAR2 par un mécanisme impliquant SNX11, initie des effets intracellulaires différents de sa signalisation membranaire. Connue pour son rôle dans la cascade de coagulation, la thrombine, une protéase à sérine, peut également agir par l’intermédiaire de PAR1, un récepteur activé par protéase et couplé aux protéines G liant le GTP (GPCR). La thrombine se lie et clive l’extrémité N-terminale du PAR1 entre l’Arg41 et la Ser42, exposant une nouvelle extrémité terminale qui agit elle-même comme un ligand. La thrombine et une séquence peptidique de cinq acides aminés, composée des résidus Ser42 à Arg46, nommée PAR1-AP, déclenchent dans diverses cellules de mammifères une réponse intracellulaire comportant une composante calcique. Dans cette étude, le système d’expression par baculovirus dans les cellules Sf9 d'insecte nous a permis d'exprimer le récepteur PAR1 du rat à la surface de ces cellules et de réaliser son couplage fonctionnel à leur signalisation intracellulaire (modèle rPAR1-Sf9). La composante calcique de celle-ci, en réponse au PAR1-AP, a ensuite été étudiée en détail à l’aide de la sonde fluorescente Fura-2 et de plusieurs inhibiteurs agissant sur les canaux calciques ou d'autres éléments de la cascade de signalisation du calcium intracellulaire. Lorsque le milieu extracellulaire contient du calcium (Ca2+), la thrombine ou PAR1-AP déclenchent un signal calcique qui consiste en une augmentation rapide de [Ca2+]i suivi d’un plateau relativement soutenu, puis d'un retour lent vers le niveau de base initial. En l'absence de Ca2+ dans le milieu extracellulaire, l'augmentation initiale rapide de [Ca2+]i est suivie d'un retour rapide vers le [Ca2+]i de base. À l’aide d’inhibiteurs de canaux calciques, tels que le lanthane, la nifédipine et le D-600, l'entrée du calcium du milieu extracellulaire dans les cellules est inhibée, abolissant le plateau soutenu de [Ca2+]i. L’inhibition de la pompe Ca2+-ATPase par la thapsigargine supprime la réponse au PAR1-AP après épuisement des sites de stockage de Ca2+intracellulaire. Le TMB-8 agit de façon discordante quant à l’inhibition de la libération de Ca2+ des sites de stockage intracellulaires. La réponse à PAR1-AP n’est pas affectée par le D-609, un inhibiteur de la phospholipase β. L’inhibition de la protéine kinase C (PKC) par le bisindolylmaléimide induit des oscillations en présence de Ca2+ extracellulaire et atténue fortement le signal calcique en absence de Ca2+ extracellulaire. En présence de Ca2+ extracellulaire, l’activation de la PKC par le PBDu tronque le flux de [Ca2+]i tandis que la réponse calcique est abolie en absence de Ca2+ dans le milieu extracellulaire. Le H-89, un inhibiteur de la protéine kinase A (PKA), cause une prolongation de la durée du plateau de [Ca2+]i dans un milieu riche en calcium et la suppression de la réponse à PAR1-AP lorsque le milieu extracellulaire est dépourvu de Ca2+. Les résultats obtenus nous permettent de conclure que la PKC et possiblement la PKA jouent un rôle critique dans la mobilisation du Ca2+ induite par le PAR1-AP dans le modèle rPAR1-Sf9.

The thesis entitled “Alkaline Protease Production by Marine Fungus Engyodontium BTMFS 10”.Proteases are the single class of enzymes, which occupy a pivotal position with respect to their application in both physiological and commercial filed. Protease in the industrial market is expected to increase further in the coming year. The current trend is to use microbial enzymes since they provide a greater diversity of catalytic activities and can be produced more economically. Main objective of theses studies are the optimization of various physicochemical factors in the solid state fermentation for the production of alkaline protease enzyme, characterization of the enzyme, evaluation of the enzyme for various industrial application. Protease inhibitors are one of the most important tools of nature for regulating the proteolytic activity of their target proteases. They are synthesized in biological systems and they play a critical role in controlling a number of diverse physiological functions. The current investigation focused on the isolation, purification and characterization of a novel protease inhibitor from Moringa oleifera. The results obtained during the course of study opens new perspectives for the utilization of protease inhibitor from Moringa oleifera for various pharmaceutical, agricultural and food industries. The biological and physicochemical properties exhibited by the novel protease inhibitor from Moringa oleifera clearly testify its suitability for the development as a drug for application in pharmaceutical industries such as anticoagulant agent or biocontrol agent in agriculture and even as a food preservant. There is a scope for further research on the structure elucidation and protein engineering towards a wide range of further applications. Detailed structure/function analysis of these proteins is important to facilitate their use in genetic engineering for various applications.

This thesis presents a detailed account of a cost - effective approach towards enhanced production of alkaline protease at profitable levels using different fermentation designs employing cheap agro-industrial residues. It involves the optimisation of process parameters for the production of a thermostable alkaline protease by Vibrio sp. V26 under solid state, submerged and biphasic fermentations, production of the enzyme using cell immobilisation technology and the application of the crude enzyme on the deproteinisation of crustacean waste.The present investigation suggests an economic move towards Improved production of alkaline protease at gainful altitudes employing different fermentation designs utilising inexpensive agro-industrial residues. Moreover, the use of agro-industrial and other solid waste substrates for fermentation helps to provide a substitute in conserving the already dwindling global energy resources. Another alternative for accomplishing economically feasible production is by the use of immobilisation technique. This method avoids the wasteful expense of continually growing microorganisms. The high protease producing potential of the organism under study ascertains their exploitation in the utilisation and management of wastes. However, strain improvement studies for the production of high yielding variants using mutagens or by gene transfer are required before recommending them to Industries.Industries, all over the world, have made several attempts to exploit the microbial diversity of this planet. For sustainable development, it is essential to discover, develop and defend this natural prosperity. The Industrial development of any country is critically dependent on the intellectual and financial investment in this area. The need of the hour is to harness the beneficial uses of microbes for maximum utilisation of natural resources and technological yields. Owing to the multitude of applications in a variety of industrial sectors, there has always been an increasing demand for novel producers and resources of alkaline proteases as well as for innovative methods of production at a commercial altitude. This investigation forms a humble endeavour towards this perspective and bequeaths hope and inspiration for inventions to follow.

The thesis presents a detailed account of the alkaline protease produced by Vibrio sp.(V26) a mangrove isolate,and the application of this enzyme in different fields.The protease producer strain was identified on the basis of biochemical characteristice,putative virulence traits and 16S rRNA gene sequencing.The purification and characterization of the protease has been carried out. Along with this, an attempt has been made to identifiy the protease gene. The physical parameters as well as the media components influencing protease production were optimized using Response Surfce Methodology(RSM).The scale up of the application of the protease from Vibrio sp.(V26) in the dissociation of cells in animal cell culture,in the recovery of silver from used X-ray films as well as an ingredient in commercial detergents were investigated.

Protease inhibitors are found abundantly in numerous plants, animals and microorganisms, owing their significance to their application in the study of enzyme structures, reaction mechanisms and also their utilization in pharmacology and agriculture. They are (synthetic/natural) substances that act directly on proteases to lower the catalytic rate. Although most of these inhibitory proteins are directed against serine proteases, some target cysteine, aspartyl or metalloproteases (Bode and Huber, 1992). Protease inhibitors are essential for regulating the activity of their corresponding proteases and play key regulatory roles in many biological processes. Applications of protease inhibitors are intimately connected to the proteases they inhibit; an overview of proteases with the modes of regulation of their proteolytic activity is discussed
