O presente relatório resultou do estágio efetuado na DGPC, no âmbito da componente não-letiva do Mestrado em Arqueologia. O seu propósito centrou-se no Inventário e Georreferenciação das manifestações funerárias rupestres medievais, enquadradas na região do Centro de Portugal. Este trabalho possibilitou a atualização e introdução de inúmeras ocorrências, respeitantes a sepulturas e necrópoles escavadas na rocha, na base de dados da DGPC – o Endovélico. A concretização deste objetivo assentou essencialmente na consulta de bibliografia especializada, assim como de relatórios técnico-científicos. Ao constar do Endovélico, a informação inserida afigura-se como um contributo para o conhecimento e divulgação deste tipo de patrinio medieval, auxiliando, inevitavelmente, no que concerne à sua salvaguarda e proteção. O processamento e sistematização da vasta informação inventariada e georreferenciada possibilitou o desenvolvimento de uma análise, na perspetiva das sepulturas escavadas na rocha como fenómeno funerário alto-medieval. Esta permitiu a criação de uma imagem, ainda que incompleta, dos tipos de agrupamentos existentes e da relação entre as sepulturas e as zonas envolventes, com lugar numa parte do território de Viseu.

Cette thèse porte sur la capacité à détecter des compagnons de faible intensité en présence de bruit de tavelures dans le contexte de limagerie à haute gamme dynamique pour lastronomie spatiale. On s’intéressera plus particulièrement à limagerie spectrale différentielle (ISD) obtenue en utilisant un étalon Fabry-Pérot comme filtre accordable. Les performances d’un tel filtre accordable sont présentées dans le cadre du Tunable Filter Imager (TFI), instrument conçu pour le télescope spatial James Webb (JWST). La capacité de l’étalon à supprimer les tavelures avec ISD est démontrée expérimentalement grâce à un prototype de l’étalon installé sur un banc de laboratoire. Les aliorations de contraste varient en fonction de la séparation, s’étendant d’un facteur 10 pour les séparations supérieures à 11 lambda/D jusqu’à un facteur 60 à 5 lambda/D. Ces résultats sont cohérents avec une étude théorique qui utilise un modèle basé sur la propagation de Fresnel pour montrer que les performances de suppression de tavelures sont limitées par le banc optique et non pas par l’étalon. De plus, il est démontré qu’un filtre accordable est une optionduisante pour limagerie à haute gamme dynamique combinée à la technique ISD. Une seconde étude basée sur la propagation de Fresnel de linstrument TFI et du télescope, a permis de définir les performances de la technique ISD combinée avec un étalon pour lastronomie spatiale. Les résultats prévoient une alioration de contraste de l’ordre de 7 jusqu’à 100, selon la configuration de linstrument. Une comparaison entre ISD et la soustraction par rotation a également été simulée. Enfin, la dernière partie de ce chapitre porte sur les performances de la technique ISD dans le cadre de linstrument Near-Infrared Imager and Slitless Spectrograph (NIRISS), conçu pour remplacer TFI comme module scientifique à bord du Fine Guidance Sensor du JWST. Cent quatre objets localisés vers la région centrale de la nébuleuse d’Orion ont été caractérisés grâce à un spectrographe multi-objet, de basse résolution et multi-bande (0.85-2.4 um). Cette étude a relevé 7 nouvelles naines brunes et 4 nouveaux candidats de masse planétaire. Ces objets sont utiles pour déterminer la fonction de masse initiale sous-stellaire et pour évaluer les modèles atmosphériques et évolutifs futurs des jeunes objets stellaires et sous-stellaires. Combinant les magnitudes en bande H mesurées et les valeurs d’extinction, les objets classifiés sont utilisés pour créer un diagramme de Hertzsprung-Russell de cet amas stellaire. En accord avec des études antérieures, nos résultats montrent qu’il existe une seule époque de formation d’étoiles qui a débuté il y a environ 1 million dannées. La fonction de masse initiale qui en dérive est en accord avec des études antérieures portant sur dautres amas jeunes et sur le disque galactique.

The SST convection relation over tropical ocean and its impact on the South Asian monsoon is the first part of this thesis. Understanding the complicated relation between SST and convection is important for better prediction of the variability of the Indian monsoon in subseasonal, seasonal, interannual, and longer time scales. Improved global data sets from satellite scatterometer observations of SST, precipitation and refined reanalysis of global wind fields have made it possible to do a comprehensive study of the SST convection relation. Interaction of the monsoon and Indian ocean has been discussed. A coupled feedback process between SST and the Active-Break cycle of the Asian summer monsoon is a central theme of the thesis. The relation between SST and convection is very important in the field of numerical modeling of tropical rainfall. It is well known that models generally do very well simulating rainfall in areas of tropical convergence zones but are found unable to do satisfactory simulation in the monsoon areas. Thus in this study we critically examined the different mechanisms of generation of deep convection over these two distinct regions.The study reported in chapter 3 has shown that SST - convection relation over the warm pool regions of Indian and west Pacific oceans (monsoon areas) is in such a way that convection increases with SST in the SST range 26-29 C and for SST higher than 29-30 C convection decreases with increase of SST (it is called Waliser type). It is found that convection is induced in areas with SST gradients in the warm pool areas of Indian and west Pacific oceans. Once deep convection is initiated in the south of the warmest region of warm pool, the deep tropospheric heating by the latent heat released in the convective clouds produces strong low level wind fields (Low level Jet - LLJ) on the equatorward side of the warm pool and both the convection and wind are found to grow through a positive feedback process. Thus SST through its gradient acts only as an initiator of convection. The central region of the warm pool has very small SST gradients and large values of convection are associated with the cyclonic vorticity of the LLJ in the atmospheric boundary layer. The conditionally unstable atmosphere in the tropics is favorable for the production of deep convective clouds.

Perennial plants are the main pollen and nectar sources for bees in the tropical areas where most of the annual flora are burned in dry seasons. Therefore perennial plants constitute the most reliable bio materials for determining and evaluating the beekeeping regions of the Republic of Benin. A silvo-melliferous region (S-MR) is a geographical area characterised by a particular set of homogenous melliferous plants that can produce timber. Using both the prevailing climatic and the agro-ecological conditions six S-MRs could be identified, i.e. the South region, the Common Central region, the Central West region, the Central North region, the Middle North region and the Extreme North region. At the country level, the melliferous plants were dominated by Vitellaria paradoxa which is common to all regions. The most diversified family was the Caesalpiniaceae (12 species) followed by the Combretaceae (10 species) and Combretum being the richest genus. The effect of dominance is particularly high in the South region where Elaeis guineensis alone represented 72.6% of the tree density and 140% of the total plant importance. The total melliferous plant density varied from 99.3 plants ha^(−1) in the Common Central region to 178.0 plants ha^(−1) in the Central West region. On the basis of nectar and pollen source, the best region for beekeeping is the CentralWest region with 46.7% of nectar producing trees, 9.4% of pollen producing trees and 40.6% of plants that issue both, this in opposition to the South region which was characterised by an unbalanced distribution of melliferous trees.

A completely effective vaccine for malaria (one of the major infectious diseases worldwide) is not yet available; different membrane proteins involved in parasite-host interactions have been proposed as candidates for designing it. It has been found that proteins encoded by the merozoite surface protein (msp)-7 multigene family are antibody targets in natural infection; the nucleotide diversity of three Pvmsp-7 genes was thus analyzed in a Colombian parasite population. By contrast with P. falciparum msp-7 loci and ancestral P. vivax msp-7 genes, specie-specific duplicates of the latter specie display high genetic variability, generated by single nucleotide polymorphisms, repeat regions, and recombination. At least three major allele types are present in Pvmsp-7C, Pvmsp-7H and Pvmsp-7I and positive selection seems to be operating on the central region of these msp-7 genes. Although this region has high genetic polymorphism, the C-terminus (Pfam domain ID: PF12948) is conserved and could be an important candidate when designing a subunit-based antimalarial vaccine.

Introducción: Las deficiencias de micronutrientes continúan siendo un problema de salud pública en la población infantil, dentro de las ellas se ha encontrado a la deficiencia de zinc causa importante de morbi-mortalidad en los países en desarrollo, la nutrición adecuada de zinc es esencial para un crecimiento adecuado, inmunocompetencia y desarrollo neuroconductual; se dispone de información insuficiente sobre el estado de zinc en la población preescolar lo cual dificulta la expansión de las intervenciones para el control de su deficiencia. Colombia presenta una deficiencia de este micronutriente, considerándose a nivel mundial como un problema de salud pública moderado a severo. Una evaluación sobre la prevalencia y factores determinantes asociados puede proporcionar datos sobre el riesgo de deficiencia de zinc en una población, considerando factores demográficos, sociales y nutricionales que podan predisponer a la población preescolar colombiana a sufrir este déficit. Metodología: Estudio observacional de corte transversal que incluyó 4275 niños entre 1 y 4 años, utilizando datos de la Encuesta Nacional de Situación Nutricional (ENSIN-2010). Se realizaron análisis bivariados y multivariados para determinar factores asociados positiva y negativamente con deficiencia de zinc. Resultados: El 49,1% de los niños encuestados cursaban con deficiencia de zinc. Los factores de riesgo asociados a deficiencia de zinc encontrados fueron menor edad, peso y talla bajos, vivir en región Atlántica, región Central, Territorios Nacionales, vivienda en área de población dispersa, pertenencia a etnia afrocolombiana, pertenencia a etnia indígena, estar afiliado a régimen subsidiado, no estar afiliado a ninn régimen de salud, madre sin educación, no asistencia a programa de alimentación dirigido y el grado severo de inseguridad Conclusiones: El déficit de zinc en los niños entre 1 y 4 años de edad es multifactorial, siendo un reflejo probable de la situación de inequidad de la población colombiana, en especial, la más pobre y vulnerable. En aquest treball es caracteriza per primera vegada la capacitat de coordinació metàl·lica d'una metal·lotinna (MT) de planta i es proposa un model de plegament per a les MTs de planta en general. Els resultat mostren que aquestes protnes poden tenir un paper molt important en la regulació de l'estat redox de les cèl·lules, probablement a través de la coordinació a Cu. Les MTs de plantan protnes molt desconegudes. Es postula que participen en l'homeòstasi del Cu i en la protecció contra l'estrès oxidatiu, però es desconeix la capacitat de coordinació metàl·lica i el plegament. En aquest treball s'han estudiat una metal·lotionna d'alzina surera, QsMT, aïllada d'una llibreria de cDNA de fel·lema. Els objectius concrets han estat: (1) estudiar l'expressió de QsMT i la resposta a l'estrès oxidatiu; (2) determinar la capacitat de coordinació metàl·lica i la funcionalitat in vivo; (3) fer una aproximació al plegament de les MTs de planta. L'expressió del gen s'ha estudiat mitjançant hibridació in situ en plàntules i en embrions d'alzina surera. QsMT s'expressa majoritàriament enl·lules amb fort estrès oxidatiu, associat a lantesi de polifenols (suberització i lignificació) i a la senescència. També s'expressa enl·lules meristemàtiques, cèl·lules en divisió molt activa on la funció de les MTs podria estar relacionada amb el manteniment de l'estat redox. L'aplicació d'estrès oxidatiu exogen (H2O2 i paraquat) incrementa fortament l'expressió de QsMT en teixits amb expressió constitutiva, confirmant la regulació de l'expressió del gen per estrès oxidatiu. Per l'estudi de les propietats de coordinació metàl·lica es va expressar QsMT enl·lules d'E. coli en medi de cultiu suplementat amb Cu, Zn o Cd. Es van aïllar els agregats metàl·lics corresponents i es van analitzar mitjançant tècniques espectroscòpiques i espectromètriques (ICP-OES, ESI-MS i CD). Els resultats mostren que QsMT coordina de forma estable Cu (8 ions metàl·lics/molècula), Zn (4 ions de Zn/molècula) i Cd (6 ions de Cd/molècula), i adopta una estructura especialment quiral en coordinació a Cu. L'elevada capacitat quelant de la protna i la quiralitat de l'estructura indiquen que QsMT possiblement té preferència metàl·lica pel Cu i per tant una funció relacionada amb aquest metall in vivo. Estudis de complementació en llevat demostren que QsMT coordina Cu de forma funcional in vivo. En coordinació a Cd QsMT presenta una peculiaritat no observada fins ara en altres MTs: la participació d'ions sulfur en la formació de l'agregat metàl·lic incrementant la capacitat de coordinació metàl·lica (6 ions metàl·lics divalents de Cd enlloc de 4 ions de Zn). A més QsMT coordina Cd de forma funcional en llevat, i per tant la seva funció també podria estar relacionada amb la destoxicació de Cd en la planta. QsMT s'ha utilitzat com a model per fer una aproximació al plegament de les MTs de planta. Amb aquest objectiu vam dissenyar tres pèptids mutants derivats de QsMT: N25 corresponent a la zona rica en cistna en posició amino-terminal, C18 corresponent a la zona rica en cistna en posició carboxil-terminal, i N25-C18 corresponent a les dues zones riques en cistna enllaçades per 4 glicines substituint la zona central de 39 aminoàcids. Es van expressar i estudiar aquests pèptids per les mateixes tècniques utilitzades en l'estudi de QsMT. Els resultats indiquen que QsMT es plega formant un sol agregat metàl·lic per la interacció de les dues zones riques en cistna. En aquest model la zona central d'enllaç, típica de les MTs de planta, no participa en la coordinació metàl·lica però és imprescindible per a la funció de la protna. El paper de la zona central podria variar en funció del metall que coordina, participant en el plegament i estructura de la protna quan coordina Zn i Cd i en la seva regulació i estabilització quan coordina Cu.

Understanding source-sink dynamics of game birds is essential to harvest and habitat management but acquiring this information is often logistically and financially challenging using traditional methods of population surveys and banding studies. This is especially true for species such as the American Black Duck (Anas rubripes), which have low breeding densities and extensive breeding ranges that necessitate extensive surveys and banding programs across eastern North America. Despite this effort, the contribution of birds fledged from various landscapes and habitat types within specific breeding ranges to regional harvest is largely unknown but remains an important consideration in adaptive harvest management and targeted habitat conservation strategies. We investigated if stable isotope (δD, δ13C, δ15N) could augment our present understanding of connectivity between breeding and harvest areas and so provide information relevant to the two main management strategies for black ducks, harvest and habitat management. We obtained specimens from 200 hatch-year Black Duck wings submitted to the Canadian Wildlife Service Species Composition Survey. Samples were obtained from birds harvested in Western, Central, and Eastern breeding/harvest subregions to provide a sample representative of the range and harvest rate of birds harvested in Canada. We sampled only hatch-year birds to provide an unambiguous and direct link between production and harvest areas. Marine origins were assigned to 12%, 7%, and 5% of birds harvested in the Eastern, Central, and Western subregions, respectively. In contrast, 32%, 9%, and 5% of birds were assigned, respectively, to agricultural origins. All remaining birds were assigned to nonagricultural origins. We portrayed probability of origin using a combination of Bayesian statistical and GIS methods. Placement of most eastern birds was western Nova Scotia, eastern New Brunswick, Prince Edward Island, and southern Newfoundland. Agricultural birds from the Central region were consistent with the Saguenay region of Québec and the eastern claybelt with nonagricultural birds originating in the boreal. Western nonagricultural birds were associated with broad boreal origins from southern James Bay to Lake of the Woods and east to Cochrane, Ontario. Our work shows that the geographic origins, landscape, and habitat associations of hatch-year Black Ducks can be inferred using this technique and we recommend that a broad-scale isotopic study using a large sample of Canadian and US harvested birds be implemented to provide a continental perspective of source-sink population dynamics.

Background: MHC Class I molecules present antigenic peptides to cytotoxic T cells, which forms an integral part of the adaptive immune response. Peptides are bound within a groove formed by the MHC heavy chain. Previous approaches to MHC Class I-peptide binding prediction have largely concentrated on the peptide anchor residues located at the P2 and C-terminus positions. Results: A large dataset comprising MHC-peptide structural complexes was created by remodelling pre-determined x-ray crystallographic structures. Static energetic analysis, following energy minimisation, was performed on the dataset in order to characterise interactions between bound peptides and the MHC Class I molecule, partitioning the interactions within the groove into van der Waals, electrostatic and total non-bonded energy contributions. Conclusion: The QSAR techniques of Genetic Function Approximation (GFA) and Genetic Partial Least Squares (G/PLS) algorithms were used to identify key interactions between the two molecules by comparing the calculated energy values with experimentally-determined BL50 data. Although the peptide termini binding interactions help ensure the stability of the MHC Class I-peptide complex, the central region of the peptide is also important in defining the specificity of the interaction. As thermodynamic studies indicate that peptide association and dissociation may be driven entropically, it may be necessary to incorporate entropic contributions into future calculations.

There is growing evidence that a number of oral Treponema species, in particular Treponema denticola, are associated with the progression of human periodontal disease. The major sheath (or surface) protein (Msp) of T. denticola is implicated in adhesion of bacteria to host cells and tissue proteins and is likely to be an important virulence factor. However, the binding regions of the Msp are not known. We have purified from Escherichia coli recombinant Msp (rMsp) polypeptides corresponding to the following: full-length Msp (rMsp) minus 13 N-terminal amino acid (aa) residues, an amino-terminal fragment (rN-Msp, 189 aa residues), a 57-aa residue segment from the central region (rV-Msp), and a C-terminal fragment (rC-Msp, 272 aa residues). rMsp (530 aa residues) bound to immobilized fibronectin, keratin, laminin, collagen type 1, fibrinogen, hyaluronic acid, and heparin. The N- and V-region polypeptides, but not rC-Msp, also bound to these substrates. Binding of rMsp to fibronectin was targeted to the N-terminal heparin I/fibrin I domain. Antibodies to the N-region or V-region polypeptides, but not antibodies to the rC-Msp fragment, blocked adhesion of T. denticola ATCC 35405 cells to a range of host protein molecules. These results suggest that the N-terminal half of Msp carries epitopes that are surface exposed and that are involved in mediating adhesion. Binding of rMsp onto the cell surface of low-level fibronectin-binding Treponema isolates conferred a 10-fold increase in fibronectin binding. This confirms that Msp functions autonomously as an adhesin and raises the possibility that phenotypic complementation of virulence functions might occur within mixed populations of Treponema species.

The generation of flow and current vortices in the dayside auroral ionosphere has been predicted for two processes ocurring at the dayside magnetopause. The first of these mechanisms is time-dependent magnetic reconnection, in “flux transfer events” (FTEs); the second is the action of solar wind dynamic pressure changes. The ionospheric flow signature of an FTE should be a twin vortex, with the mean flow velocity in the central region of the pattern equal to the velocity of the pattern as a whole. On the other hand, a pulse of enhanced or reduced dynamic pressure is also expected to produce a twin vortex, but with the central plasma flow being generally different in speed from, and almost orthogonal to, the motion of the whole pattern. In this paper, we make use of this distinction to discuss recent observations of vortical flow patterns in the dayside auroral ionosphere in terms of one or other of the proposed mechanisms. We conclude that some of the observations reported are consistent only with the predicted signature of FTEs. We then evaluate the dimensions of the open flux tubes required to explain some recent simultaneous radar and auroral observations and infer that they are typically 300 km in north–south extent but up to 2000 km in longitudinal extent (i.e., roughly 5 hours of MLT). Hence these observations suggest that recent theories of FTEs which invoke time-varying reconnection at an elongated neutral line may be correct. We also present some simultaneous observations of the interplanetary magnetic field (IMF) and solar wind dynamic pressure (observed using the IMP8 satellite) and the ionospheric flow (observed using the EISCAT radar) which are also only consistent with the FTE model. We estimate that for continuously southward IMF ( ≈ 5 nT) these FTEs contribute about 30 kV to the mean total transpolar voltage (∼30%).

We present a first overview of flows in the high latitude ionosphere observed at 15 s resolution using the U.K.-Polar EISCAT experiment. Data are described from experiments conducted on two days, 27 October 1984 and 29 August 1985, which together span the local times between about 0200 and 2130MLT and cover five different regions of ionospheric flow. With increasing local time, these are: the dawn auroral zone flow cell, the dayside region of low background flows equatorward of the flow cells, the dusk auroral zone flow cell, the boundary region between the dusk auroral zone and the polar cap, and the evening polar cap. Flows in both the equatorward and poleward portions of the auroral zone cells appear to be relatively smooth, while in the central region of high speed flow considerable variations are generally present. These have the form of irregular fluctuations on a wide range of time scales in the early morning dawn cell, and impulsive wave-like variations with periods of a few minutes in the afternoon dusk cell. In the dayside region between the flow cells, the ionosphere is often essentially stagnant for long intervals, but low amplitude ULF waves with a period of about 5 min can also occur and persist for many cycles. These conditions are punctuated at one to two hour intervals by sudden ‘flow burst’ events with impulsively generated damped wave trains. Initial burst flows are generally directed poleward and can peak at line-of-sight speeds in excess of 1 km s^{−1} after perhaps 45 s. Flows in the polar cap are reasonably smooth on time scales of a few minutes and show no evidence for the presence of ULF waves. Under most, but not all, of the above conditions, the beam-swinging algorithm used to determine background vector flows should produce meaningful results. Comparison of these flow data with simultaneous plasma and magnetic field measurements in the solar wind, made by the AMPTE IRM and UKS spacecraft, emphasizes the strong control exerted on high latitude flows by the north-south component of the IMF.

Artisanal and small-scale mining (ASM) is often characterised as rush-type activity undertaken by people looking to ‘get-rich quick’. An alternative view posits ASM as poverty-driven activity which provides a valuable source of employment to a rural population where agriculture has failed to provide an adequate income. Against this dichotomy, this article critically reviews the short-lived, but high profile, gold rush which occurred on the beaches of Elmina, a fishing town in Ghana’s Central Region. At its peak, more than 1000 people, including local fishermen and career miners from the Western Region and Eastern Region of the country, were mining on the beaches. Drawing on interviews with miners and local business owners, the analysis explores the dynamics of the rush and its implications for understanding the ASM sector in Ghana. The discussion highlights the challenges associated with managing resources in dynamic and heterogeneous contexts, reconciling separate and conflicting sectoral priorities and the need to go beyond simplified and dualistic representations of the sector.
