Mixed infections in cucurbits are frequently observed in natural conditions between viruses from the Potyvirus genus and Cucumber mosaic virus (CMV), which significantly decreases productivity. The objectives of the present study was to compare the host range of PRSV-W, WMV, and ZYMV isolates and evaluate the effects of mixed infections with CMV in zucchini plants (Cucurbita pepo L.). Host range studies comprising 23 plant species confirmed some similarities and biological differences among the isolates of PRSV-W, ZYMV, and WMV. RT-PCR confirmed the amplification of DNA fragments of the PRSV-W, WMV, and ZYMV coat protein gene (cp) and cytoplasm inclusion gene (ci). The virus interaction studies in zucchini Caserta plants indicated synergistic interactions, particularly among species from the Potyvirus genus, and some CMV interference with some virus combinations.

O feijão-de-metro é uma hortaliça amplamente cultivada nos municípios da região metropolitana de Belém. Diversas doenças podem comprometer a sua produtividade, dentre elas as viroses. Recentemente, foi detectado o Cucumber mosaic virus (CMV) em vagens de feijão-de-metro provenientes do município de Castanhal-PA. Este trabalho teve como objetivo identificar o subgrupo do CMV detectado em vagens de feijão-de-metro, por meio de RT-PCR, sequenciamento do ácido nucléico e análise utilizando o programa Blast, ClustalW e MEGA 7.0. Para isso, foi feita a extração de ácidos nucleicos total a partir de folhas de fumo inoculado com o isolado. Posteriormente, foi realizado o RT-PCR utilizando os primers específicos (CMV-CPR e CMV-CPF). A partir da análise da filogenia foi observado que o isolado formou um clado com os acessos do subgrupo IB de CMV.

O feijão-de-porco é uma leguminosa utilizada em recuperação e recobrimento do solo e controle de plantas invasoras, por possuir efeito alelopático sobre outras espécies de plantas. A sua produtividade pode ser afetada por doenças, entre elas destacam-se as causadas por vírus. Em Altamira-PA, no campo experimental da Embrapa, observou-se plantas de feijão-de-porco apresentando os sintomas característicos de viroses como o mosaico, nanismo e deformação foliar. Assim, o objetivo deste trabalho foi identificar a espécie de vírus em amostras de feijão-de-porco. Para isso, as amostras foliares foram avaliadas utilizando os testes ELISA e em sequida o de RT-PCR com primers específicos para CABMV (CABMV-F e CABMV-R). No teste de RT-PCR obteve-se a banda esperada de 221 pb correspondente à parte do gene da capa proteica do vírus CABMV. Este foi o primeiro relato de CABMV em feijão-de-porco no Estado do Pará.

RNA templates of 33 nucleotides containing the brome mosaic virus (BMV) core subgenomic promoter were used to determine the promoter elements recognized by the BMV RNA-dependent RNA polymerase (RdRp) to initiate RNA synthesis. Nucleotides at positions −17, −14, −13, and −11 relative to the subgenomic initiation site must be maintained for interaction with the RdRp. Changes to every other nucleotide at these four positions allow predictions for the base-specific functional groups required for RdRp recognition. RdRp contact of the nucleotide at position −17 was suggested with a template competition assay. Comparison of the BMV subgenomic promoter to those from other plant and animal alphaviruses shows a remarkable degree of conservation of the nucleotides required for BMV subgenomic RNA synthesis. We show that the RdRp of the plant-infecting BMV is capable of accurately, albeit inefficiently, initiating RNA synthesis from the subgenomic promoter of the animal-infecting Semliki Forest virus. The sequence-specific recognition of RNA by the BMV RdRp is analogous to the recognition of DNA promoters by DNA-dependent RNA polymerases.

The mouse mammary tumor virus (MMTV) promoter is regulated by steroid hormones through a hormone-responsive region that is organized in a positioned nucleosome. Hormone induction leads to a structural change of this nucleosome which makes its DNA more sensitive to cleavage by DNase I and enables simultaneous binding of all relevant transcription factors. In cells carrying either episomal or chromosomally integrated MMTV promoters, moderate acetylation of core histones, generated by treatment with low concentrations of the histone deacetylase inhibitors sodium butyrate or trichostatin A, enhances transcription from the MMTV promoter in the absence of hormone and potentiates transactivation by either glucocorticoids or progestins. At higher concentrations, histone deacetylase inhibitors reduce basal and hormone induced MMTV transcription. Inducing inhibitor concentrations lead to the same type of nucleosomal DNase I hypersensitivity as hormone treatment, suggesting that moderate acetylation of core histone activates the MMTV promoter by mechanisms involving chromatin remodeling similar to that generated by the inducing hormones.

We report the complete molecular characterization of the DNA-A and DNA-B of a Brazilian tomato isolate of Tomato severe rugose virus (ToSRV) and the experimental host range of the virus determined using white-fly transmission tests. Genome analysis showed that ToSRV has a close evolutionary relationship with Tomato rugose mosaic virus. Of 33 plants species inoculated with viruliferous Bemisia tabaci biotype B, 13 species were susceptible to ToSRV, nine asymptomatically. Therefore, ToSRV disease management strategy should include the control of infected weeds close to tomato fields.

The RT-PCR technique for the detection of apple stem grooving virus (ASGV), apple stem pitting virus (ASPV), apple chlorotic leaf spot virus (ACLSV), apple mosaic virus (ApMV) and pear blister canker viroid (PBCV) was evaluated for health control of fruit plants from nurseries. The technique was evaluated in purified RNA and crude extracts and also in phloem collected in autumn and from young spring shoots. The results obtained for phytoplasma detection with ribosomal and non-ribosomal primers are also presented.
