Small fish abundance is usually high in heavily vegetated habitats in Yangtze lakes, China. Visual and swimming barriers created by dense macrophytes beds could reduce feeding efficiency and growth of small fishes. We tested the hypothesis that small fishes in habitats with dense macrophytes would show decreased feeding efficiency and reduced growth rates by comparing feeding efficiency (measured as the relative weight of fore-gut contents), total length, and condition factor of four small young-of-the-year fishes collected in the near-shore (heavily vegetated) and central (less vegetated) areas of Liangzi Lake. Feeding efficiency, total length, or condition factor were each significantly reduced in the near-shore area compared with the central area for Ctenogobius giurinus, Pseudorasbora parva and Carassius auratus auratus. This supports our hypothesis that vegetation abundance may mediate feeding efficiency and growth of small fishes. Although Hypseleotris swinhonis did not show significant decreases in feeding efficiency or growth in the near-shore area, there was not any reversed tendency, i.e. increased feeding rate or growth in the near-shore area compared to the central area.

Interferon (IFN) exerts its antiviral effect by inducing the expression of a number of IFN-stimulated genes (ISGs) to establish a host antiviral state. Earlier studies identified some important fish IFN system genes from IFN-induced CAB cells (crucian carp Carassius auratus L. embryonic blastulae cells) after treatment with UV-inactivated GCHV (grass carp hemorrhage virus). Herein, the cloning of 2 novel IFN-stimulated genes, termed Gig1 and Gig2, is described for the same cell system. The complete cDNA sequences of Gig1 and Gig2 contain 1244 bp encoding for a 194-amino-acid protein and 693 bp for a 158-amino-acid protein, respectively. A search of public databases revealed that these are 2 novel IFN-stimulated genes, since neither significant homologous genes nor conserved motifs were identified. Active GCHV, UV-inactivated GCHV and CAB IFN-containing supernatant (ICS) induced transcription of these genes and distinct kinetics were observed. An analysis of differences in expression between the 2 genes and the IFN signal factors CaSTAT1 and CaIRF7 indicated that GCHV infection activated different signal pathways for their up-regulation. Upon virus infection, the transcription of Gig1 but not of Gig2 is strongly suppressed by cycloheximide (CHX). In contrast, following treatment with CAB IFN-containing supernatant, CHX does not inhibit either gene transcription. The results suggest that GCHV infection can induce expression of both Gig1 and Gig2 via newly synthesized CAB IFN, most probably through the JAK-STAT signal pathway, and can also directly activate Gig2 transcription without ongoing protein synthesis.

The double-stranded-RNA-dependent protein kinase (PKR) is an important component in an antiviral defence pathway that is mediated by interferon (IFN) in vertebrates. Previously, some important IFN system genes had been identified from an IFN-producing CAB (crucian carp Carassius auratus blastulae embryonic) cells after treatment with UV-inactivated GCHV (grass carp haemorrhage virus). Here, a fish PKR-like gene, named CaPKR-like, is cloned and sequenced from the same virally infected CAB cells. It has 2192 base pairs in length with a largest open reading frame (ORF) encoding a protein of 513 amino acid residues. BLAST search reveals that the putative CaPKR-like protein is most homologous to human PKR and also has a high-level homology with all members of a family of eIF2alpha kinases. Structurally, CaPKR-like possesses a conserved C-terminal catalytic domain of eIF2alpha kinase family and the most similarity to mammalian PKRs. Within its N-terminus, there are no dsRNA-binding domains conserved in mammalian PKRs instead of two putative Z-DNA binding domains (Zalpha). Like mammalian PKRs, CaPKR-like had a very low level of constitutive expression in normal CAB cells but was up-regulated in response to active GCHV, UV-inactivated GCHV and CAB IFN, implying that the transcriptional activation of CaPKR-like by viral infection is mediated possibly by newly produced CAB IFN, which was further supported by using cycloheximide, a potent inhibitor of protein synthesis. The results together suggested that CaPKR-like was the first identified fish gene most similar to mammalian PKRs. (C) 2004 Elsevier Ltd. All rights reserved.

UV-inactivated grass carp hemorrhage virus (GCHV) can induce high titer of interferon in cultured CAB (crucian carp (Carassius auratus L.) blastulae) cells, and thus defend host cells against the virus invasion. The mechanism is proposed that an antiviral state should be established in the host cells by activating expression of a set of antiviral-relevant genes. In this study, suppressive subtractive hybridization is applied to constructing a subtracted cDNA library with mRNAs isolated from UV-inactivated GCHV infected and mock-infected CAB cells. 272 differential cDNA fragments are identified by both PCR and dot blot from the subtractive cDNA library. Sequencing analysis reveals 69 genes, including 46 known gene homologues, and 23 unknown putative genes. The known genes include the genes involved in interferon signaling pathways, such as Stat1 and Jak1, the antiviral genes, such as Mx and Viperin, and a set of interferon-stimulated genes observed in mammalian cells. Most of the unknown putative genes contain AU-rich element in their sequences. Differential expressions of these genes are further confirmed by virtual Northern blot and RT-PCR. The data imply that UV-inactivated GCHV is not only able to induce production of interferon in the infected CAB cells, but also leads to the expression of a series of antiviral-relevant genes or immune-relevant genes, and therefore reveals that the signaling pathway of interferon system and antiviral mechanism in fish are similar to those in mammals.

Mature eggs of allotetraploid carp were activated by inactive sperm or crossed with normal sperms of common carp (Cyprinus carpio), crucian carp (Carassius auratus), Chinese blunt snout bream (Megalobrama amblycephala), Hemiculter leucisculus and Pseudorasbora parva. Chromosome counts showed that all offspring of these crosses presented a mode number of 200 chromosomes (4n = 200), and their morphological traits are much like maternal. Microsatelite marker and RAPD patterns between allotetraploid maternal and its offspring, reproduced from different paternal species, were identical. Cytological, morphological and molecular evidences suggested that allotetraploid carp female nucleus would not fuse with any male nucleus and its reproduction mode might be gynogenesis and therefore their offspring are retaining their tetraploidy and give origin to clonal individuals.

Small fish communities were compared between the vegetated and vegetation-free regions of the Liangzi Lake, a shallow lake along the middle reach of the Yangtze River, China. Fish were sampled using 10 x 10 m(2) block nets and poisoning. Three samples were taken from either the near shore area or lake centre of each region. A total of 19 fish species were collected; all species occurred in the vegetated region but only 12 occurred in the vegetation-free region. The dominant small fish were Carassius auratus auratus in the vegetated region and Ctenogobius giurinus in the vegetation-free region. Diversity, density and biomass of small fishes were significantly higher in the vegetated region than in the vegetation-free region in both near shore and lake centre areas. In the vegetated region, density and biomass of small fishes was significantly higher, while species diversity significantly lower in the near shore area than in the lake centre. In the vegetation-free region, density of small fishes was significantly higher in the near shore area than in the lake centre area, but species diversity or biomass was unaffected by location.

Species in Liangzi Lake were clustered into four trophic groups: Hemiramphus kurumeus and Hemiculter bleekeri bleekeri fed predominantly on terrestrial insects; Carassius auratus auratus and Abbottina rivularis on non-animal food; Hypseleotris swinhonis, Ctenogobius giurinus, Pseudorasbora parva and Toxabramis swinhonis on cladocerans or copepods; Culterichthys erythropterus on decapod shrimps. Gut length, mouth width, mouth height, gill raker length and gill raker spacing, varied widely among species. With the exception of three species pairs (H. swinhonis, C. glurinus; C. erythropterus, H. kurumeus; T. swinhonis, H. bleekeri bleekeri), principal components analysis of morphological variables revealed over-dispersion of species. Canonical correspondence analysis of dietary and morphological data revealed five significant dietary-morphological correlations. The first three roots explained > 85% of the total variance. The first root reflected mainly the relationship of gut length to non-animal feud, with an increase in gut length associated with an increase in non-animal food. The second root was influenced strongly by the relationship of the gill raker spacing to consumption of copepods, with an increase in gill raker spacing associated positively with copepods in the diet. The third root was influenced by the relationship of mouth gape to consumption of fish and decapod shrimps, with an increase in mouth gape associated with more fish and decapod shrimps in the diet. These significant dietary-morphological relationships supported the eco-morphological hypotheses that fish morphology influence food use, and morphological variation is important in determining ecological segregation of co-existing fish species. Two growth trials using a range of ration sizes from starvation to maximum feeding suggested that linear relationships existed between specific growth rate and ration size for Nile tilapia and givel carp, Continuous measurement of activity showed that activity level, in terms of distance swum per day, was not affected significantly by ration size in both Nile tilapia and gibel carp. Like other transgenic animals, transgenic fishes produced by microinjection are transgenic mosaics. In order to produce homogenous transgenic fish, the transgenic blastula or gastrula cells were dissociated from Carassius auratus, Pengze var, and Cyprinus carpio, Huanghe var., and the nuclei were transferred into the mature eggs of the same species via microinjection or electro-fusion. Five nuclear-transferred Carassius auratus, Pengze var. and one Cyprinus carpio, Huanghe var. were obtained and the existence of the transgene was detected. The possibility of generating homogenous strain of transgenic fish by nuclear transplantation with transgenic early-embryonic cells is discussed.

The SR-protein kinase activity was analyzed and the cytological changes were observed during oocyte maturation in bisexual transparent color crucian carp ( Carassius auratus color variety). The results revealed that the SR-protein kinase activity was sensitive to the artificially induced spawning hormones, and the change of oscillatory activity was similar to that of the maturation-promoting factor (MPF) kinase that regulates meiotic cell cycle in fish.

The spatial pattern of the small fish community was studied seasonally in 1996 in the Biandantang Lake. Based on plant cover, the lake was divided into five habitats, arranged in the order by plant structure complexity from complex to simple: Vallisneria spiralis habitat (V habitat), Vallisneria spiralis-Myriophyllum spicatum habitat (V-M habitat), Myriophyllum spicatum habitat (M habitat), Nelunbo nucefera habitat (N habitat), and no vegetation habitat (NV habitat). A modified popnet was used for quantitative sampling of small fishes. A total of 16 fish species were collected; Hypseleotris swinhonis, Ctenogobius giurinus, Pseudorasbora parva, Carassius auratus and Paracheilognathus imberis were the five numerically dominant species. In both summer and autumn, the total density of small fishes was about 10 ind m(-2). Generally, Ctenogobius giurinus, a sedatory, benthic fish, was distributed more or less evenly among the five habitats, while the other four species had lower densities in the N habitat and NV habitat, which had the simplest structures. The distribution of the small fish species showed seasonal variations. In winter, most species concentrated in the V habitat, which had the most complex structure. In spring, the fish had low densities in the N and NV habitat, and were more or less evenly distributed in the other habitats. In summer, the fish had a low density in the NV habitat, and were evenly distributed in the other habitats. In autumn, the fish had higher densities in the V-M and M habitats than in the others. Generally, spatial overlaps between the dominant species were higher in winter than in the other seasons. It was suggested that the variations in the importance of predation risk and resource competition in habitat choice determined the seasonal changes of spatial patterns in the small fishes in the Biandantang Lake.

Chromosome behavior in meiosis was studied by air-drying, C-banding and surface-spreading methods in female intersexes of artificial triploid transparent-colored crucian carp (Carassius auratus). Chromosome pairing and contraction were obviously asynchronous. The preferential pairing of two homologous chromosomes was the major pattern of chromosome pairing, and a few triple pairing, repeated pairing, telomer or centromere associating and multiple pairing were also observed in the pachytene cells. The metaphase I cells were mainly composed of univalents, bivalents and trivalents, as well as few of other multivalents, such as tetravalents, pentavalents, hexavalents and heptavalents, were also found in some metaphase I cells. The chromosome elements including uni-, bi-, tri- and other multivalents varied considerably among the metaphase I cells, and the associating patterns of multivalents were also diverse. Some 6 n and 12 n cells, in which premeiotic endomitosis occurred once or twice, were found at the prophase and first metaphase of meiosis, and the pairing and associating patterns were basically similar to that of the triploid cells.

Silver nanoparticles (AgNP) have been produced and applied in a variety of products ranging from personal care products to food package containers, clothing and medicine utilities. The antimicrobial function of AgNP makes it very useful to be applied for such purposes. Silver (Ag) is a non-essential metal for organisms, and it has been historically present in the environment at low concentrations. Those concentrations of silver increased in the last century due to the use of Ag in the photographic industry and lately are expected to increase due to the use of AgNPs in consumer products. The presence of AgNP in the aquatic environment may pose a risk for aquatic species, and the effects can vary from lethal to sublethal effects. Moreover, the contact of aquatic organisms with AgNP may not cause immediately the death of individuals but it can be accumulated inside the animals and consequently transferred within the food chain. Considering this, the objective of this work was to study the transfer of silver nanoparticles in comparison to silver ions, which was used as silver nitrate, within an aquatic food chain model. To achieve this goal, this study was divided into four steps: the toxicity assessment of AgNP and AgNO3 to aquatic test-species, the bioaccumulation assessment of AgNP and AgNO3 by Pseudokirchneriella subcapitata and Daphnia magna under different exposure scenarios, and finally the evaluation of the trophic transfer of Ag through an experimental design that included the goldfish Carassius auratus in a model trophic chain in which all the species were exposed to the worse-case scenario. We observed that the bioconcentration of Ag by P. subcapitata is mainly driven by ionic silver, and that algae cannot internalize these AgNPs, but it does internalizes dissolved Ag. Daphnia magna was exposed to AgNP and AgNO3 through different exposure routes: water, food and both water and food. The worse-case scenario for Daphnia Ag bioaccumulation was by the joint exposure of contaminated water and food, showing that Ag body burdens were higher for AgNPs than for AgNO3. Finally, by exposing C. auratus for 10 days through contaminated water and food (supplied as D. magna), with another 7 days of depuration phase, it was concluded that the 10 days of exposure were not enough for fish to reach a plateau on Ag internal concentration, and neither the 7 days of elimination were sufficient to cause total depuration of the accumulated Ag. Moreover, a higher concentration of Ag was found in the intestine of fish when compared with other organs, and the elimination rate constant of AgNP in the intestine was very low. Although a potential for trophic transfer of AgNP cannot be suggested based in the data acquired in this study, there is still a potential environmental risk for aquatic species.

En el marc d'un projecte més ampli sobre la comunitat de peixos de la conca lacustre de Banyoles, s'ha estudiat la distribució espacial de les diferents espècies així com l'ús que aquestes fan de l'hàbitat. El poblament piscícola de l'estany de Banyoles és el resultat d'un llarg historial d'introducció d'espècies exòtiques i extinció de les poblacions autòctones locals. S'ha revisat la seva composició actual detectant un total de 18 espècies (4 autòctones i 14 introduïdes) de les quals només 13 presenten una població estable. L'estudi de l'hàbitat s'ha centrat a l'Estany, l'element principal del sistema lacustre, analitzant per separat la zona litoral i la zona limnètica. En el primer cas s'han realitzat les captures d'individus mitjançant pesca elèctrica des d'una embarcació adaptada per aquesta pràctica. La totalitat del litoral ha estat dividida en trams de característiques homogènies on s'han obtingut les captures per unitat d'esforç per cada espècie. El mostreig s'ha desenvolupat entre l'estiu de 1997 i la primavera del 2000 realitzant un total de 10 campanyes de pesca. Les espècies més abundants al litoral són la perca americana (Micropterus salmoides) i el peix sol (Lepomis gibbosus), essent també presents la perca (Perca fluviatilis), carpa (Cyprinus carpio) i el gardí (Scardinius erythrophthalmus). S'han capturat altres espècies com ara anguila (Anguilla anguilla), bagra (Squalius cephalus), sandra (Sander lucioperca), carpí (Carassius auratus) i madrilleta vera (Rutilus rutilus), però són molt menys abundants en nombre. S'ha examinat, per cadascuna de les espècies, si existeix selecció de l'hàbitat i en cas afirmatiu, quin és el preferent en base a la classificació del litoral en sis tipus de vegetació predominant. Les espècies més abundants, perca americana i peix sol, ocupen tots els hàbitats disponibles però amb una densitat diferent. La perca mostra també una clara selecció de l'hàbitat a favor de les zones molt estructurades amb abundant presència de jonca litoral. Carpa i gardí seleccionen els ambients més fondos amb major presència de matèria orgànica d'origen vegetal procedent del bogar. En general els individus ocupen les zones amb una densitat de vegetació intermèdia, majoritàriament zones de jonca a l'estiu i zones amb mansega a l'hivern, on troben refugi i els recursos tròfics necessaris. La perca americana, a més, presenta una elevada fidelitat a un mateix punt del litoral al llarg de la seva vida. La zona limnètica ha estat prospectada mensualment mitjançant ecosondació, realitzant transectes perpendiculars a l'eix principal de l'Estany, cobrint la seva totalitat. La composició d'espècies s'ha obtingut a partir de les captures fetes amb xarxes (tresmalls) amb periodicitat estacional. L'anàlisi geoestadística de la densitat de peixos ha permès descriure l'estructura espacial d'aquesta a partir dels variogrames, així com la seva variabilitat tant espacial com temporal, i obtenir els mapes de densitat. A l'hivern, la densitat de peixos a la zona limnètica assoleix els seus valors mínims i els individus es troben formant agregats dispersos, pels diferents estrats de fondària. A partir de la primavera la densitat augmenta, pel reclutament i la major freqüència d'individus que abandonen el litoral; la densitat esdevé més homogènia a les primer capes de fondària. A l'estiu la densitat és màxima i l'estrat més homogeni coincideix amb la posició de la termoclina. Aquest estructura varia en disminuir la temperatura i barrejar-se la columna d'aigua, tornant a la situació hivernal. La perca i la madrilleta vera són les espècies predominants en aquest ambient, juntament amb la carpa. La seva distribució no és homogènia i respon a les característiques limnològiques de les diferents cubetes de l'Estany. Una particularitat d'aquest, relacionada amb el seu origen càrstic, es la formació d'una ploma hidrotermal que afecta la distribució dels peixos, probablement en augmentar la terbolesa. S'ha integrat l'ús de l'hàbitat de les espècies que ocupen tant la zona limnètica com la litoral a partir del seguiment d'individus, concretament de perca i bagra. S'ha utilitzat un sistema automàtic de posicionament que estima la localització dels individus marcats amb transmissor de telemetria acústica. L'anàlisi dels desplaçaments mostra un rang superior per la bagra en comparació amb la perca. Ambdues espècies mostren una orientació en els seus desplaçaments. La perca ocupa el litoral a la nit i es desplaça a la zona limnètica de dia, amb un ritme d'activitat marcat per dos màxims coincidint amb la sortida i posta de sol; en canvi la bagra mostra una major activitat nocturna amb zones de repòs properes al litoral. S'ha estimat igualment els dominis vital de cada individu marcat.
