ENGLISH: The fishing power of the tuna purse-seine fleet of the eastern Pacific Ocean has increased since the early 1960's. Because the entire fleet seems to have adopted equipment and techniques to increase its efficiency in capturing tunas, traditional methods of adjusting catch rates to a reference vessel type of fixed efficiency to index tuna abundance from fishing success are inapplicable. Instead, a methodology for such adjustment based on a mathematical representation of purse seining activities is developed. Observed changes in efficiency in subprocesses of purse seining are then used to adjust catch rates when computing abundance histories for yellowfin and skipjack in large regions of the eastern Pacific Ocean. SPANISH: La eficacia de pesca de la flota de cerco atunera en el Océano Pacífico oriental ha aumentado desde el comienzo del decenio de 1960. Como toda la flota parece haber adoptado equipo y métodos para incrementar su eficaciaen capturar atunes, no se pueden aplicar los métodos tradicionales de ajustar los índices de captura a un tipo normalizado de barco (es decir de eficacia fija) para indicar la abundancia del atún según los resultados de pesca. En su lugar se ha desarrollado un método para realizar tal ajuste basado en una representación matemática de las actividades de las embarcaciones de cerco. Cuando se calcula la abundancia histórica del atún aleta amarilla y barrilete en grandes regiones del Océano Pacífico oriental, se usan entonces los cambios observados en la eficacia de los subprocesos cerqueros para ajustar los índices de captura. Neocarzinostatin chromophore 1 is the active component of the antitumor antibiotic neocarzinostatin (NCS). The chromophore reacts with thiols to form a highly strained cumulene-enyne species which rapidly rearranges to a biradical intermediate which can abstract hydrogen atoms from DNA, leading to strand cleavage. DNA damage is the proposed source of biological activity for NCS. The structure of the methyl thioglycolate monoadduct 2 of NCS chromophore, including the absolute stereochemistry, was determined by NMR studies. The presence of the cumulene-enyne intermediate and the rearrangement to a biradical were supported by data from low temperature NMR investigations. Also included are synthetic approaches to NCS chromophore model compounds based on intramolecular addition of an acetylide to an aldehyde.

Three mutants of Drosophila melanogaster have been isolated in which the free-running period of the circadian eclosion rhythm and the adult locomotor activity rhythm is affected. One mutant is arrhythmic, another has a short period of 19 hours, and the third has a long period of 28 hours. The mutants retain their phenotypes over the temperature range 18° to 25° C. All three mutants map near the tip of the X chromosome (distal to the centromere). By deficiency mapping, the short-period mutation has been localized to the 3B1-2 region. Complementation tests show that all three mutations affect the same functional gene.

Analysis of activity rhythms of individual mosaic flies indicates that the site of action of the short-period mutation is probably located in the head of the fly. A few activity patterns of split-head and mixed-head mosaics appear to possess both mutant and heterozygous components, suggesting that the fly head may contain two complete clocks capable of maintaining their periodicities independently.

The short-period mutation affects both the duration of the light-insensitive part of the oscillation and the degree to which the clock can be reset during the light-sensitive part of the oscillation.

Both the short-period and long-period mutant eclosion rhythms can be entrained to a period of 24 hours by a 12:12 light-dark cycle having a light intensity at least two orders of magnitude greater than that required to entrain the normal rhythm. The arrhythmic mutant does not entrain under these conditions. In the presence of a temperature cycle, however, the arrhythmic mutant does entrain, but its rhythm damps out when the temperature cycle is removed.

Evidence is presented that Pittendrigh's two-oscillator model for the clock in D. pseudoobscura applies to D. melanogaster as well. The three clock mutations primarily affect the light- sensitive driving oscillator. The arrhythmic mutation appears to have eliminated the driving oscillator while leaving the temperature-sensitive driven oscillator relatively intact.

A protagonista do presente trabalho, a Utopia, a arte do pensar-contra, foi apresentada e definida, nas sendas da Filosofia da Esperança de Ernst Bloch, como uma consciência antecipadora que não se conforma com o está-aí das coisas, com a realidade fática; e como um logos, linguagem-ação que cria furos no tempo saltando para-adiante, para o topos-outro. Negativa e Esperançosa, ela representa a verdade-de-fora: não é o irreal, pois existe. E a existência do topos de fora, o topos-outro, se justifica pelo fato de que a vida e o mundo não são sistemas fechados, porque seus horizontes estão em aberto: atravessados por possibilidades, ainda-não-são. Contra o que é estático, o que é fatal e fático, se posiciona o sonho utópico, abrindo espaços no fluxo do mesmo. Ao fazê-lo, cria duas frentes reciprocamente reais: o aqui-e-agora de quem sonha e o aqui-e-agora do sonho, o u-topos. Assim, tanto seu caráter de projeção ao porvir quanto, na sua base, o descontentamento com o atual, revelam seu comprometimento com o presente. Negando e afirmando a história, transformou-se em conteúdo e, sobretudo, forma, de Morus a Fourrier, de Marx a Orwell. E é por comprometer-se com o futuro, o presente e o passado, que, nos tempos sombrios do início do século XX, ela subverte a si mesma e faz vir ao mundo sua versão pessimista: a Distopia. Articulando e fazendo dialogarem as obras distópicas de Orwell, Aldous Huxley e Jerome K. Jerome com os pensamentos de Adorno, Marcuse, Horkheimer, Hannah Arendt, Karl Kraus e Walter Benjamin, tentamos encaminhar a pergunta originária da nossa pesquisa: é possível uma utopia pessimista? Será este pessimismo, ainda, uma Utopia?

Esta tese de doutorado parte da perspectiva inicial de que a gramaticalização se restringe a tratado sobre itens lexicais ou discursivos que se tornam itens gramaticais (o que a enquadraria dentro da Teoria da Variação, inserta esta dentro da Pesquisa Sociolinguística), mas segue em direção a um salto epistemológico que remodele aquela perspectiva, ampliando-a a patamar do qual ela pode ser observada como teoria autônoma, investigativa de fenômenos limítrofes e nem sempre discretos entre linguagem e língua, discurso e texto, descrição e prescrição, oralidade e escrita, léxico e gramática. Desse modo, propugna-se pela visão epistemológica do tema, conduzido, até aqui, de modo puramente ontológico, circunscrito a um (e apenas um) dos muitos espectros que se podem alcançar com a aludida ampliação àquele que vem sendo perquirido como tratado, porém que, segundo se pretende demonstrar, pode e deve ser expandido à malha de uma teoria geral, qual seja a Teoria Geral da Gramaticalização: trata-se, aqui, de seu objetivo geral. Para esse propósito, vale-se a tese de filósofos da linguagem que atuaram sobre essa faculdade ou capacidade humana de forma direta ou indireta desde os seus primórdios ocidentais (como Sócrates, Platão e Aristóteles), passando pelos pensadores mais incisivamente preocupados com os aspectos cognitivos e interativos da linguagem e da língua (como Hegel, Husserl, Saussure, Sapir, Bloomfield, Wittgenstein, Derrida, Chomsky, Labov, Charaudeau, Maingueneau, Ducrot, Coseriu), além de ser necessária a incursão à Gramaticografia mais estrita (como a empreendida por Dionísio da Trácia, Varrão, Arnault e Lancelot, Nebrija, Jerônimo Soares Barbosa, Eduardo Carlos Pereira, Said Ali, Bechara), e, naturalmente, a contribuição filosófica dos pesquisadores sobre a gramaticalização (como Meillet, Vendryès, Bréal, Kurilowicz, Traugott, Heine, Hopper, Lehmann). Uma vez que se tenha mostrado ser verossímil aceitar-se a gramaticalização como teoria autônoma, esta tese pretende legar-lhe o papel instrumental de metodologia auxiliar a muitas entre as que ora se empreendem quando se trata de pesquisas em campos cuja ocupação é a linguagem e a língua: trata-se, aqui, de seu objetivo específico. Para essa duplicidade de metas ou objetivos, será necessário compreender conceitos, categorias e protótipos oriundos da Filosofia da Ciência (Epistemologia), do contraste entre ciências da linguagem e outros ramos do saber, da imersão em Gramaticologia e Gramaticografia (e, em alguns aspectos, em Gramatização e Gramatologia) referentes à Língua Portuguesa, da defesa, enfim, de que o ensino da Gramática Formal (ou Normativa) do idioma privilegia a acepção reflexiva e ativa (plena) dos usos ou atos a que a linguagem só pode chegar por meio do domínio da língua em toda a sua tessitura epistemológica, que gera comunicação e expressividade, raciocínio e emotividade, indo da concretude do discurso ou da oralidade à abstração da entidade pouco ou nada material, que, por sua vez, é mais nitidamente representada pela escrita, seu estágio por assim dizer de forma ainda mais pura, conquanto não excludente da substancialidade com que dialoga de modo incessante no seu constante e dialético passado-futuro ou diversidade-homogeneidade (tese e antítese) de onde emerge o seu presente ou a sua unidade (síntese)

Blooms of the brevetoxin-producing dinoflagellate Karenia brevis have been linked to high mortality of bottlenose dolphins Tursiops truncatus on Florida’s Gulf of Mexico coast. A clear understanding of trophic transfer of brevetoxin from its algal source up the food web to top predators is needed to assess exposure of affected dolphin populations. Prey fish constitute a means of accumulating and transferring brevetoxins and are potential vectors of brevetoxin to dolphins frequently exposed to K. brevis blooms. Here we report results of brevetoxin analyses of the primary fish species consumed by long-term resident bottlenose dolphins inhabiting Sarasota Bay, Florida. Fish collected during K. brevis blooms in 2003 to 2006 were analyzed by competitive enzyme-linked immunosorbent assay (ELISA) and had brevetoxin concentrations ranging from 4 to 10844 ng PbTx-3 eq g–1 tissue. Receptor binding assay (RBA) and liquid chromatography–mass spectrometry (LC-MS) analysis confirmed toxicity and the presence of parent brevetoxins and known metabolites. Fish collected in the absence of K. brevis blooms tested positive for brevetoxin by ELISA and RBA, with concentrations up to 1500 ng PbTx-3 eq g–1 tissue. These findings implicate prey fish exposed to K. brevis blooms as brevetoxin vectors for their dolphin predators and provide a critical analysis of persistent brevetoxin loads in the food web of dolphins repeatedly exposed to Florida red tides.

Mayan cichlids (Cichlasoma urophthalmus) were collected monthly from March 1996 to October 1997 with hook-and-line gear at Taylor River, Florida, an area within the Crocodile Sanctuary of Everglades National Park, where human activities such as fishing are prohibited. Fish were aged by examining thin-sectioned otoliths, and past size-at-age information was generated by using back-calculation techniques. Marginal increment analysis showed that opaque growth zones were annuli deposited between January and May. The size of age-1 fish was estimated to be 33–66 mm standard length (mean=45.5 mm) and was supported by monthly length-frequency data of young-of-year fish collected with drop traps over a seven-year period. Mayan cichlids up to seven years old were observed. Male cichlids grew slower but achieved a larger size than females. Growth was asymptotic and was modeled by the von Bertalanffy growth equation Lt=263.6(1–exp[–0.166(t–0.001)]) for males (r2=0.82, n=581) and Lt=215.6 (1–exp[–0.197(t–0.058)]) for females (r2= 0.77, n=639). Separate estimates of total annual mortality were relatively consistent (0.44–0.60) and indicated moderate mortality at higher age classes, even in the absence of fishing mortality. Our data indicated that Mayan cichlids grow slower and live longer in Florida than previously reported from native Mexican habitats. Because the growth of Mayan cichlids in Florida periodically slowed and thus produced visible annuli, it may be possible to age introduced populations of other subtropical and tropical cichlids in a similar way.

EXTRACT (SEE PDF FOR FULL ABSTRACT): The 1988 summer drought over much of the United States is described in terms of hemispheric mid-tropospheric flow patterns, temperature and precipitation anomalies, and sea surface temperature patterns. This drought was similar to earlier Great Plains droughts, although spatially more extensive than most. Three attempts to predict this drought from antecedent spring were moderately successful, though no one anticipated its severity and extent. ... A modified barotropic model iterating from a mean summer estimate of seasonal forcing from the May mid-tropospheric height pattern was reasonably successful in forecasting the drought. Sea surface temperature indications show that cold water (La Niña) along the equator subsequent to the 1987 El Niño, while contributory, cannot be considered a principal cause of the drought, since earlier cold water episodes did not produce drought, and other drought episodes occurred in the absence of cold equatorial waters.

The desire to design more efficient transport aircraft has led to many different attempts to minimize drag. One approach is the use of three-dimensional shock control bumps, which have gained popularity in the research community as simple, efficient and robust devices capable of reducing the wave drag of transonic wings. This paper presents a computational study of the performance of three-dimensional bumps, relating key bump design variables to the overall wing aerodynamic performance. An efficient parameterization scheme allows three-dimensional bumps to be directly compared to two-dimensional designs, indicating that two-dimensional bumps are capable of greater design point aerodynamic performance in the transonic regime. An advantage of three-dimensional bumps lies in the production of streamwise vortices, such that, while two-dimensional bumps are capable of superior performance near the design point, three-dimensional bumps are capable of breakingup regions of separated flow at high Mach numbers, suggesting improvement in terms of buffet margin. A range of bump designs are developed that exhibit a tradeoff between design point aerodynamic efficiency and improvementinbuffet margin, indicating the potential for bespoke designs to be generated for different sections of a wing based on its flow characteristics. The aerodynamic design of turbomachinery presents the design optimisation community with a number of exquisite challenges. Chief among these are the size of the design space and the extent of discontinuity therein. This discontinuity can serve to limit the full exploitation of high-fidelity computational fluid dynamics (CFD): such codes require detailed geometric information often available only sometime after the basic configuration of the machine has been set by other means. The premise of this paper is that it should be possible to produce higher performing designs in less time by exploiting multi-fidelity techniques to effectively harness CFD earlier in the design process, specifically by facilitating its participation in configuration selection. The adopted strategy of local multi-fidelity correction, generated on demand, combined with a global search algorithm via an adaptive trust region is first tested on a modest, smooth external aerodynamic problem. Speed-up of an order of magnitude is demonstrated, comparable to established techniques applied to smooth problems. A number of enhancements aimed principally at effectively evaluating a wide range of configurations quickly is then applied to the basic strategy, and the emerging technique is tested on a generic aeroengine core compression system. A similar order of magnitude speed-up is achieved on this relatively large and highly discontinuous problem. A five-fold increase in the number of configurations assessed with CFD is observed. As the technique places constraints neither on the underlying physical modelling of the constituent analysis codes nor on first-order agreement between those codes, it has potential applicability to a range of multidisciplinary design challenges. © 2012 by Jerome Jarrett and Tiziano Ghisu.

Dynamics of dewetting and phase separation in ultrathin films (thickness is ca. one radius of gyration, approximate to 1 R-g) of poly(methyl methacrylate) (PMMA) and poly(styrene-ran-acrylonitrile) (SAN) blends on Si substrate has been studied by in situ atomic force microscopy (AFM). In the miscible region, a "spinodal-like" dewetting driven by a composition fluctuation recently predicted by Wensink and Jerome (Langmuir 2002, 18, 413) occurs. In the two-phase region, the dewetting of the whole film is followed by phase separation in the droplets, coupling with the wetting of the substrate by the PMMA extracted by the strong attractive interaction between them.

The SIEGE (Smoking Induced Epithelial Gene Expression) database is a clinical resource for compiling and analyzing gene expression data from epithelial cells of the human intra-thoracic airway. This database supports a translational research study whose goal is to profile the changes in airway gene expression that are induced by cigarette smoke. RNA is isolated from airway epithelium obtained at bronchoscopy from current-, former- and never-smoker subjects, and hybridized to Affymetrix HG-U133A Genechips, which measure the level of expression of ~22 500 human transcripts. The microarray data generated along with relevant patient information is uploaded to SIEGE by study administrators using the database's web interface, found at http://pulm.bumc.bu.edu/siegeDB. PERL-coded scripts integrated with SIEGE perform various quality control functions including the processing, filtering and formatting of stored data. The R statistical package is used to import database expression values and execute a number of statistical analyses including t-tests, correlation coefficients and hierarchical clustering. Values from all statistical analyses can be queried through CGI-based tools and web forms found on the �Search� section of the database website. Query results are embedded with graphical capabilities as well as with links to other databases containing valuable gene resources, including Entrez Gene, GO, Biocarta, GeneCards, dbSNP and the NCBI Map Viewer.
