The objective of this work was to determine the early physiological changes throughout shelf life of fresh broccoli (Brassica oleracea L. var. italica) cv. Piracicaba Precoce at 25ºC and relative humidity of 96% in the dark until complete senescence. Head inflorescences showed lack of turgidity and commercial value when weight loss reached up to 5%, coinciding with 48 hour after harvest. Chlorophyll content was stable until 24 hours after harvesting; afterwards, an intense degradation phase took place. At 72 hours, total head yellowing was observed when chlorophyll content dropped to 30% of its initial content. Peroxidase activity increased by 1.4 fold during the first six hours, dropping to its lowest level approximately 24 hours after harvesting. However, from this time on, an increment of activity was observed until 72 hours. At 24 hours after harvesting, respiration was reduced by 50%. At later stages of senescence, respiration of florets was stable, but in a lower level than that determined at harvest. Sharp reduction of starch and reducing sugars was observed within 24 hours after harvesting, followed by continuous period of decline in starch and non-reducing sugars.

O objetivo deste estudo foi obter calos e embriões somáticos de couve-comum (Brassica oleracea L., var. leucocephala) e caracterizá-los por meio de observações histológicas. Foram cultivadas, in vitro, explantes foliares, sob condições assépticas, em meio nutritivo MS, suplementado com várias combinações entre 2,4-D/BAP e 2,4-D/KIN. Na fase de indução de calos (fase I), houve o desenvolvimento de calos com características embriogênicas nos meios suplementados com 5,0 mg/L de 2,4-D; 1,0 mg/L de 2,4-D e 0,1 mg/L de BAP; 1,0 mg/L de 2,4-D e 1,0 mg/L de BAP e 2,0 mg/L de 2,4-D e 0,05 mg/L de BAP, que foram selecionados para o subcultivo em meio de regeneração (fase II). Nesta fase, foram utilizados meios de cultura suplementados com reduzidas concentrações de 2,4-D, BAP e AG3, ou totalmente desprovidos destes, em que os calos permaneceram por mais 30 dias em cultivo. Os calos subcultivados nos meios de regeneração com 0,1 mg/L de BAP (R3) e 0,01 e 0,001 mg/L de BAP e 2,4-D respectivamente (R8), provenientes dos meios para indução, acrescidos de 5,0 mg/L de 2,4-D (M16) e 1,0 e 1,0 mg/L de BAP e 2,4-D (M30), desenvolveram estruturas semelhantes a embriões. No entanto, a caracterização histológica revelou a superioridade da interação entre os meios M30 e R8, para a regeneração de calos e embriões somáticos.

The effects of combined pressure/temperature treatments (200, 400 and 600 MPa, at 20 and 40 °C) on the physical and nutritional properties of swede roots (Brassica napus var. napobrassica) were assessed. Changes induced by high pressure processing (HPP) on the original properties of swede samples were compared with those produced by thermal treatment (blanching). All studied treatments altered the physical properties of swede, resulting in a loss of hardness and water binding capacity. The strongest alteration of texture was observed after HPP at 400 MPa, while 600 MPa was the treatment that better preserved the texture properties of swede. Blanching caused less total colour changes (ΔE) than HPP. Antioxidant properties of swede were measured as total antioxidant capacity, ascorbic acid and total phenol content. All treatments caused a loss of antioxidant capacity, which was less pronounced after HPP at 600 MPa and 20 °C and blanching. Four glucosinolates were detected in swede roots, glucoraphanin, progoitrin, glucobrassicanapin and glucobrassicin. Glucobrassicanapin and glucobrassicin contents were reduced with all studied treatments. Progoitrin content was not affected by blanching and HPP at 200 MPa. HPP at higher pressure levels (400 and 600 MPa), though, induced an increase of progoitrin levels. The results indicated that blanching and HPP at 600 MPa and 20 °C were the treatments that better preserved the original quality properties of swede.

Myzus persicae (Sulzer) was reared continuously for over thirty years (until it died out in December 2008) on a totally defined synthetic artificial diet, the procedure for which is described. Development time was extended on diet compared with rearing on Brussels sprout plants (Brassica oleracea L. var. gemmifera L.), and generation time was further increased by an added pre-reproductive period of 4 days. Fecundity was reduced by about two-thirds, and mean relative growth rate in weight (MRGR) was only 60% in comparison with plant-reared aphids. Applying 2 kg/cm(2) pressure to a 10% sucrose solution extended the adult longevity of Aphis fabae Scopoli by less than I day. In contrast, a short experience of half-strength diet Caused a sharp rise in honeydew excretion by A. fabae for several hours, and alternating full-strength diet with diluted diets (including water) Caused a greater weight increase. The poor performance of aphids on diet thus seems to have a behavioural rather than a mechanical explanation. The diet, designed to give optimal performance of the aphids, has proved not to be useful for nutritional studies, as any change is deleterious. Areas of aphid research where the diet has been useful, however, are studies on repellents/attractants/toxins, role of symbionts, maintenance of genotype collections, work on parasitoid behaviour in relation to plant chemistry, and collection of aphid saliva.

Postharvest senescence in broccoli (Brassica oleracea L. var Italica) florets results in phenotypic changes similar to those seen in developmental leaf senescence. To compare these two processes in more detail, we investigated molecular and biochemical changes in broccoli florets stored at two different temperatures after harvest. We found that storage at cooler temperatures delayed the symptoms of senescence at both the biochemical and gene expression levels. Changes in key biochemical components (lipids, protein, and chlorophyll) and in gene expression patterns occurred in the harvested tissue well before any visible signs of senescence were detected. Using previously identified senescence-enhanced genes and also newly isolated, differentially expressed genes, we found that the majority of these showed a similar enhancement of expression in postharvest broccoli as in developmental leaf senescence. At the biochemical level, a rapid loss of membrane fatty acids was detected after harvest, when stored at room temperature. However, there was no corresponding increase in levels of lipid peroxidation products. This, together with an increased expression of protective antioxidant genes, indicated that, in the initial stages of postharvest senescence, an orderly dismantling of the cellular constituents occurs, using the available lipid as an energy source. Postharvest changes in broccoli florets, therefore, show many similarities to the processes of developmental leaf senescence.

Broccoli seeds were coated in a conical-cylindrical spouted bed with an aqueous suspension of hydroxy ethyl cellulose aiming to improve the seeds coating technique using a fluid-dynamic process. An experimental design was applied to investigate the effects of the operating variables: gas temperature, atomizing air pressure and suspension flow rate on the germination of the seeds and on the process efficiency. Results indicated that the operating variables affect both the coating process efficiency and the germination ability. However, the analysis didn t identify differences between the germination potential of coated and uncoated seeds. Coated seeds absorbed up to 10 percent less moisture than the uncoated ones, when the environment temperature and humidity were controlled over a period of time.

The effects of irrigation with reclaimed wastewater (RWW) were compared with well water (WW) on citrus (Citrus paradisi Macfad. X Citrus aurantium L) nutrition. The deviation from the optimum percentage (DOP) index of macro- and micro-nutrients were used to evaluate the nutritional status: optimal (DOP = 0), deficiency (DOP < 0) or excess (DOP > 0). After 11 years of RWW irrigation the influence on nutrient concentration in plants decreased in the order: B > Zn > Mn = Ca > Cu > Mg > P > K. Reclaimed wastewater irritation positively affected citrus nutrition as it rendered the concentration of macronutrients, i.e. P, Ca, and K. closer to their optimum levels (Sigma DOP(macro) = 7). However micro-nutrients tended to be excessive in plants (EDOP(micro) = 753) due to imbalanced supply of these elements in the RWW, particularly, for B and Cu. Citrus groves with long-term RWW irrigation may exercised caution in monitoring concentrations of B and Cu to avoid plant toxicity and soil quality degradation. (C) 2011 Elsevier B.V. A execução adequada do programa de adubação na cultura do repolho pode ser obtida pela sincronização da demanda da planta com o suprimento de N durante o ciclo da cultura. Assim, o objetivo deste trabalho foi avaliar a evolução de indicadores nutricionais, a intensidade da cor verde (índice SPAD), medida na quarta folha completamente expandida; o teor de nitrogênio total e o teor de nitrato e as características agronômicas, comprimento do caule, número de folhas e massas de matéria fresca e seca de folhas, caules e raízes da planta, para determinar o teor de nitrogênio no repolho e determinar o valor crítico de cada característica em diferentes épocas de avaliação. Aos 50, 64 e 78 dias após a semeadura, foram determinados: a intensidade da cor verde (índice SPAD), medida na quarta folha; o comprimento do caule, o número de folhas, as massas de matéria fresca e seca e o teor de N foliar. Foram avaliadas cinco doses de N (0; 75; 150; 300 e 450 kg ha-1). Os tratamentos foram arranjados no delineamento experimental de blocos ao acaso, com quatro repetições. A colheita final das cabeças de repolho ocorreu aos 103 dias após a semeadura. Os valores ótimos das variáveis ao longo do tempo dependeram das doses de N e da época de avaliação. O valor ótimo do índice SPAD foi de 60,8; 58,4 e 53,5 aos 50; 64 e 78 dias após a semeadura. O valor ótimo do teor de nitrogênio total foi de 62,9; 60,3 e 59,0 g kg-1 nas folhas, de 48,6; 40,6 e 40,3 g kg-1 no caule e de 39,9; 36,9 e 20,1 g kg-1.nas raízes aos 50,64 e 78 dias após a semeadura.

O objetivo deste trabalho foi avaliar a influência de diferentes doses de nitrogênio no crescimento e produtividade do pak choi. O experimento foi conduzido, em cultivo protegido, de outubro a novembro de 2007, em Marechal Cândido Rondon, PR. O delineamento experimental foi de blocos casualizados, com cinco tratamentos (0, 60, 105, 150 e 195 kg ha-1 de N) e quatro repetições. O nitrogênio foi aplicado em três diferentes épocas (transplantio, 7 e 14 dias após o transplantio). Foram realizadas cinco coletas das plantas, semanalmente, avaliando-se a produção de massa da matéria seca e área foliar, para obtenção das taxas de crescimento absoluto e relativo, taxa assimilatória líquida, razão de área foliar e área foliar específica. Na colheita, foram avaliados altura da planta, diâmetro e matéria fresca da parte aérea e produtividade. Incrementos na adubação nitrogenada até a dose de 195 kg ha-1 proporcionam incrementos no crescimento e produtividade do pak choi.

RESUMO Estudos realizados com Brassica sp. demonstram que o estresse salino afeta a qualidade fisiológica e o vigor das sementes. Assim, a busca por substâncias que minimizem os efeitos provocados pelo estresse salino durante a germinação são de grande importância. Objetivou-se com este trabalho avaliar a ação de substâncias liberadoras de óxido nítrico durante a germinação de sementes de repolho cultivado sob condições de salinidade. As sementes foram pré-embebidas em soluções de nitrato de potássio (KNO3) e nitroprus siato de sódio (SNP) nas concentrações de zero (controle), 0,01, 0,10, 1,00 e 10,00 mmol L-1, por 30 minutos. Posteriormente, distribuídas sobre papel germitest em placas de Petri, umedecido com solução salina de NaCl nos potenciais de zero, -0,6 e -1,2 MPa. Foram avaliados a germinação, primeira contagem de germinação, índice de velocidade de germinação, tempo médio de germinação, comprimento de parte aérea, comprimento de raiz e massa seca das plântulas. Não houve germinação no potencial osmótico de -1,2 MPa. O potencial de -0,6 MPa reduz a germinação e o desenvolvimento das plântulas de repolho. Os resultados sugerem que houve ação positiva do KNO3 e do SNP na concentração de 0,01 mmol L-1, revertendo o estresse provocado pela salinidade (-0,6 MPa de NaCl).

The author carried out a study on the behavior of cabbage for a period of two years (1952 and 1953). From the results obtained it was concluded that, cabbage, Brassica oleraceae, var. capitata, developed better during the cooler season of the year, that is, from April to June and, the yield decresed from the beginning to the end of the sowing time, that is, from April to Ocotber.

O presente trabalho apresenta um método de réplica de superfície para o estudo da estrutura das fôlhas de vegetais ao microscípio eletrônico. O método permitiu a observação do mesmo tipo de estrutura observada por outros autores na superfície das fôlhas de couve (Brassica oleracea). O método permitiu observar que nas superfícies das fôlhas de café (Coffea arabica) a ultraestrutura característica e pràticamente desprovida do tipo de ornamentação encontrado nas fôlhas de couve.
