A revista Arquitectura foi criada em Portugal em 1927 e publicou‑se até 1939. Em 1948 foi comprada por um grupo de arquitectos denominado ICAT (Iniciativas Culturais Arte e Técnica) que, até 1957, a edita como um meio de divulgação da arquitectura moderna, sob a orientação decisiva de Francisco Keil do Amaral. O número 57/58, de Janeiro/Fevereiro de 1957, é o primeiro a ser editado por uma nova geração de arquitectos, que procurava exprimir as tendências de revisão do Movimento Moderno que começavam a dominar o debate internacional, e o número seguinte é o primeiro da 3.ª série. Na viragem dos anos 50 para os anos 60 as revistas Architectural Review e Casabella protagonizavam o debate arquitectónico na Europa. Neste artigo, a partir da leitura de alguns textos chave de Carlos Duarte e Nuno Portas publicados nos primeiros números da 3.ª série da revista portuguesa, é analisada a influência que tiveram as duas principais publicações internacionais sobre os editores de Arquitectura.

Analogues of Peptaibolin, a peptaibol with antibiotic activity, incorporating α,α-dialkylglycines (Deg, Dpg, and Ac6c) at selected positions were synthesised by MW-SPPS and fully characterized. A control analogue incorporating L-alanine was also prepared. The native peptide and the analogues were studied by fluorescence spectroscopy for their membrane permeating activity. Small unilamellar vesicles (SUVs) of egg phosphatidylcholine/ cholesterol (70:30) containing an encapsulated fluorescence probe (6-carboxyfluorescein) were used as membrane models. The assays of carboxyfluorescein release from SUVs upon peptide addition showed that Peptaibolin-Dpg and Peptaibolin-Ac6c are the most active peptides. These results indicate that the structure of the α,α-dialkylglycines is crucial for the membrane permeating ability of these Peptaibolin analogues.

Software reconfigurability became increasingly relevant to the architectural process due to the crescent dependency of modern societies on reliable and adaptable systems. Such systems are supposed to adapt themselves to surrounding environmental changes with minimal service disruption, if any. This paper introduces an engine that statically applies reconfigurations to (formal) models of software architectures. Reconfigurations are specified using a domain specific language— ReCooPLa—which targets the manipulation of software coordinationstructures,typicallyusedinservice-orientedarchitectures(soa).Theengine is responsible for the compilation of ReCooPLa instances and their application to the relevant coordination structures. The resulting configurations are amenable to formal analysis of qualitative and quantitative (probabilistic) properties.

In a reconfigurable system, the response to contextual or internal change may trigger reconfiguration events which, on their turn, activate scripts that change the system׳s architecture at runtime. To be safe, however, such reconfigurations are expected to obey the fundamental principles originally specified by its architect. This paper introduces an approach to ensure that such principles are observed along reconfigurations by verifying them against concrete specifications in a suitable logic. Architectures, reconfiguration scripts, and principles are specified in Archery, an architectural description language with formal semantics. Principles are encoded as constraints, which become formulas of a two-layer graded hybrid logic, where the upper layer restricts reconfigurations, and the lower layer constrains the resulting configurations. Constraints are verified by translating them into logic formulas, which are interpreted over models derived from Archery specifications of architectures and reconfigurations. Suitable notions of bisimulation and refinement, to which the architect may resort to compare configurations, are given, and their relationship with modal validity is discussed.

Las reducciones jesuíticas en Argentina reconocen generalmente un único aporte en la región guaraní. Pero lo cierto es que una cantidad importante de reducciones, equivalente en número con las anteriormente mencionadas, se desarrollaron en el interior del país, fundamentalmente en las regiones del Chaco, noroeste y sur argentino. Muchas de ellas reconocen hoy su continuidad en centros urbanos y otras tan sólo, y en el mejor de los casos, en vestigios arqueológicos. Se propone el análisis de este conjunto desde las primeras incursiones en "misiones volantes" en el siglo XVII hasta la expulsión de los jesuitas en 1767. También se abordarán las modalidades y procesos generadores de centros reduccionales en estas regiones y sus interrelaciones territoriales. Además se pretende analizar sus funciones y morfologías originales, su evolución, traslados y posibles transformaciones posteriores, en las etapas previas al impacto originado ante la ausencia de la Compañía de Jesús. Los resultados incluirán la interpretación de los procesos formativos, con su diversidad de casos, en escalas regionales y locales, la recopilación de cartografía regional y urbana, y la determinación de series tipológicas de formas de trazados y organizaciones de tejidos, tanto en las demarcaciones de origen como en sus remodelaciones y ensanches cuando así correspondiere.

Aberrant blood vessels enable tumor growth, provide a barrier to immune infiltration, and serve as a source of protumorigenic signals. Targeting tumor blood vessels for destruction, or tumor vascular disruption therapy, can therefore provide significant therapeutic benefit. Here, we describe the ability of chimeric antigen receptor (CAR)-bearing T cells to recognize human prostate-specific membrane antigen (hPSMA) on endothelial targets in vitro as well as in vivo. CAR T cells were generated using the anti-PSMA scFv, J591, and the intracellular signaling domains: CD3ζ, CD28, and/or CD137/4-1BB. We found that all anti-hPSMA CAR T cells recognized and eliminated PSMA(+) endothelial targets in vitro, regardless of the signaling domain. T cells bearing the third-generation anti-hPSMA CAR, P28BBζ, were able to recognize and kill primary human endothelial cells isolated from gynecologic cancers. In addition, the P28BBζ CAR T cells mediated regression of hPSMA-expressing vascular neoplasms in mice. Finally, in murine models of ovarian cancers populated by murine vessels expressing hPSMA, the P28BBζ CAR T cells were able to ablate PSMA(+) vessels, cause secondary depletion of tumor cells, and reduce tumor burden. Taken together, these results provide a strong rationale for the use of CAR T cells as agents of tumor vascular disruption, specifically those targeting PSMA.
