The objective of this study was to compare the BLUP selection method with different selection strategies in F-2:4 and assess the efficiency of this method on the early choice of the best common bean (Phaseolus vulgaris) lines. Fifty-one F-2:4 progenies were produced from a cross between the CVIII8511 x RP-26 lines. A randomized block design was used with 20 replications and one-plant field plots. Character data on plant architecture and grain yield were obtained and then the sum of the standardized variables was estimated for simultaneous selection of both traits. Analysis was carried out by mixed models (BLUP) and the least squares method to compare different selection strategies, like mass selection, stratified mass selection and between and within progeny selection. The progenies selected by BLUP were assessed in advanced generations, always selecting the greatest and smallest sum of the standardized variables. Analyses by the least squares method and BLUP procedure ranked the progenies in the same way. The coincidence of the individuals identified by BLUP and between and within progeny selection was high and of the greatest magnitude when BLUP was compared with mass selection. Although BLUP is the best estimator of genotypic value, its efficiency in the response to long term selection is not different from any of the other methods, because it is also unable to predict the future effect of the progenies x environments interaction. It was inferred that selection success will always depend on the most accurate possible progeny assessment and using alternatives to reduce the progenies x environments interaction effect.

The aim of this study was to classify some markers of common herbs used in Western medicine according to the Biopharmaceutical Classification System (BCS). The BCS is a scientific approach to classify drug substances based upon their intestinal permeability and their solubility, at the highest single dose used, within the physiologically relevant pH ranges. Known marker components of twelve herbs were chosen from the USP Dietary Supplement Compendium Monographs. Different BCS parameters such as intestinal permeability (P-eff) and solubility (C-s) were predicted using the ADMET Predictor, which is a software program to estimate biopharmaceutical relevant molecular descriptors. The dose number (D-0) was calculated when information from the literature was available to identify an upper dose for individual markers. In these cases the herbs were classified according to the traditional BCS parameters using Peff and Do. When no upper dose could be determined, then the amount of a marker that is just soluble in 250 mL of water was calculated. This value, M-x, defines when a marker is changing from highly soluble to poorly soluble according to BCS criteria. This biopharmaceutically relevant value can be a useful tool for marker selection. The present study showed that a provisional BCS classification of herbs is possible but some special considerations need to be included into the classification strategy. The BCS classification can be used to choose appropriate quality control tests for products containing these markers. A provisional BCS classification of twelve common herbs and their 35 marker compounds is presented.

Rationale and aim: This paper has the object to present the impact of nuts' and seeds' injuries withdrawing data from the Susy Safe registry, highlighting that as for other foreign bodies the main item efficiently and substantially susceptible to changes to decrease the accidents' rates is the education of adults and children, that can be shared with parents both from pediatricians and general practitioners. Indeed labeling and age related warnings have also a fundamental relevance in prevention. Methods: The present study draws its data from the Susy Safe registry. Details on injuries are entered in the Susy Safe Web-registry through a standardized case report form, that includes information regarding: children age and gender, features of the object, circumstances of injury (presence of parents and activity) and hospitalization's details (lasting, complications and removal details). Cases are prospectively collected using the Susy Safe system from 06/2005; moreover, also information regarding past consecutive cases available in each centre adhering to the project have been entered in the Susy Safe registry. Results: Nuts and seeds are one of the most common food item retrieved in foreign bodies injuries in children. In Susy Safe registry they represent the 38% in food group, and almost the 10% in general cases. Trachea, bronchi and lungs were the main location of FB's retrieval, showing an incidence of 68%. Hospitalization occurred in 83% of cases, showing the major frequency for foreign bodies located in trachea. This location was also the principal site of complications, with a frequency of 68%. There were no significant associations between these outcomes and the age class of the children. The most common complications seen (22.4%) was bronchitis, followed by pneumonia (19.7%). Adult presence was recorded as positive in 71.2% of cases, showing an association (p value 0.009) between the adult supervision and the hospitalization outcome. On the contrary there was a non significant association between adult presence and the occurrence of complications. In 80.7% of cases, the incident happened while the child was eating. Among those cases, 88.6% interested trachea, lungs and bronchi. Conclusions: Food-related aspiration injuries are common events for young children, particularly under 4 years of age, and may lead to severe complication. There is a need to study in more depth specific characteristics of foreign bodies associated with increased hazard, such as size, shape, hardness or firmness, lubricity, pliability and elasticity, in order to better identify risky foods, and more precisely described the pathogenetic pathway. Parents are not adequately conscious and aware toward this risk; therefore, the number and severity of the injuries could be reduced by educating parents and children. Information about food safety should be included in all visits to pediatricians in order to make parents able to understand, select, and identify key characteristics of hazardous foods and better control the hazard level of various foods. Abstract Background Prior to the selection of disinfectants for low, intermediate and high (sterilizing) levels, the decimal reduction time, D-value, for the most common and persistent bacteria identified at a health care facility should be determined. Methods The D-value was determined by inoculating 100 mL of disinfecting solution with 1 mL of a bacterial suspension (104 – 105 CFU/mL for vegetative and spore forms). At regular intervals, 1 mL aliquots of this mixture were transferred to 8 mL of growth media containing a neutralizing agent, and incubated at optimal conditions for the microorganism. Results The highest D-values for various bacteria were determined for the following solutions: (i) 0.1% sodium dichloroisocyanurate (pH 7.0) – E. coli and A. calcoaceticus (D = 5.9 min); (ii) sodium hypochlorite (pH 7.0) at 0.025% for B. stearothermophilus (D = 24 min), E. coli and E. cloacae (D = 7.5 min); at 0.05% for B. stearothermophilus (D = 9.4 min) and E. coli (D = 6.1 min) and 0.1% for B. stearothermophilus (D = 3.5 min) and B. subtilis (D = 3.2 min); (iii) 2.0% glutaraldehyde (pH 7.4) – B. stearothermophilus, B. subtilis (D = 25 min) and E. coli (D = 7.1 min); (iv) 0.5% formaldehyde (pH 6.5) – B. subtilis (D = 11.8 min), B. stearothermophilus (D = 10.9 min) and A. calcoaceticus (D = 5.2 min); (v) 2.0% chlorhexidine (pH 6.2) – B. stearothermophilus (D = 9.1 min), and at 0.4% for E. cloacae (D = 8.3 min); (vi) 1.0% Minncare® (peracetic acid and hydrogen peroxide, pH 2.3) – B. stearothermophilus (D = 9.1 min) and E. coli (D = 6.7 min). Conclusions The suspension studies were an indication of the disinfectant efficacy on a surface. The data in this study reflect the formulations used and may vary from product to product. The expected effectiveness from the studied formulations showed that the tested agents can be recommended for surface disinfection as stated in present guidelines and emphasizes the importance and need to develop routine and novel programs to evaluate product utility.

Chlorophyll determination with a portable chlorophyll meter can indicate the period of highest N demand of plants and whether sidedressing is required or not. In this sense, defining the optimal timing of N application to common bean is fundamental to increase N use efficiency, increase yields and reduce the cost of fertilization. The objectives of this study were to evaluate the efficiency of N sufficiency index (NSI) calculated based on the relative chlorophyll index (RCI) in leaves, measured with a portable chlorophyll meter, as an indicator of time of N sidedressing fertilization and to verify which NSI (90 and 95 %) value is the most appropriate to indicate the moment of N fertilization of common bean cultivar Perola. The experiment was carried out in the rainy and dry growing seasons of the agricultural year 2009/10 on a dystroferric Red Nitosol, in Botucatu, São Paulo State, Brazil. The experiment was arranged in a randomized complete block design with five treatments, consisting of N managements (M1: 200 kg ha-1 N (40 kg at sowing + 80 kg 15 days after emergence (DAE) + 80 kg 30 DAE); M2: 100 kg ha-1 N (20 kg at sowing + 40 kg 15 DAE + 40 kg 30 DAE); M3: 20 kg ha-1 N at sowing + 30 kg ha-1 when chlorophyll meter readings indicated NSI < 95 %; M4: 20 kg ha-1 N at sowing + 30 kg ha-1 N when chlorophyll meter readings indicated NSI < 90 % and, M5: control (without N application)) and four replications. The variables RCI, aboveground dry matter, total leaf N concentration, production components, grain yield, relative yield, and N use efficiency were evaluated. The RCI correlated with leaf N concentrations. By monitoring the RCI with the chlorophyll meter, the period of N sidedressing of common bean could be defined, improving N use efficiency and avoiding unnecessary N supply to common bean. The NSI 90 % of the reference area was more efficient to define the moment of N sidedressing of common bean, to increase N use efficiency.

Descrizione, tema e obiettivi della ricerca La ricerca si propone lo studio delle possibili influenze che la teoria di Aldo Rossi ha avuto sulla pratica progettuale nella Penisola Iberica, intende quindi affrontare i caratteri fondamentali della teoria che sta alla base di un metodo progettuale ed in particolar modo porre l'attenzione alle nuove costruzioni quando queste si confrontano con le città storiche. Ha come oggetto principale lo studio dei documenti, saggi e scritti riguardanti il tema della costruzione all'interno delle città storiche. Dallo studio di testi selezionati di Aldo Rossi sulla città si vuole concentrare l'attenzione sull'influenza che tale teoria ha avuto nei progetti della Penisola Iberica, studiare come è stata recepita e trasmessa successivamente, attraverso gli scritti di autori spagnoli e come ha visto un suo concretizzarsi poi nei progetti di nuove costruzioni all'interno delle città storiche. Si intende restringere il campo su un periodo ed un luogo precisi, Spagna e Portogallo a partire dagli anni Settanta, tramite la lettura di un importante evento che ha ufficializzato il contatto dell'architetto italiano con la Penisola Iberica, quale il Seminario di Santiago de Compostela tenutosi nel 1976. Al Seminario parteciparono numerosi architetti che si confrontarono su di un progetto per la città di Santiago e furono invitati personaggi di fama internazionale a tenere lezioni introduttive sul tema di dibattito in merito al progetto e alla città storica. Il Seminario di Santiago si colloca in un periodo storico cruciale per la Penisola Iberica, nel 1974 cade il regime salazarista in Portogallo e nel 1975 cade il regime franchista in Spagna ed è quindi di rilevante importanza capire il legame tra l'architettura e la nuova situazione politica. Dallo studio degli interventi, dei progetti che furono prodotti durante il Seminario, della relazione tra questo evento ed il periodo storico in cui esso va contestualizzato, si intende giungere alla individuazione delle tracce della reale presenza di tale eredità. Presupposti metodologici. Percorso e strumenti di ricerca La ricerca può quindi essere articolata in distinte fasi corrispondenti per lo più ai capitoli in cui si articola la tesi: una prima fase con carattere prevalentemente storica, di ricerca del materiale per poter definire il contesto in cui si sviluppano poi le vicende oggetto della tesi; una seconda fase di impronta teorica, ossia di ricerca bibliografica del materiale e delle testimonianze che provvedono alla definizione della reale presenza di effetti scaturiti dai contatti tra Rossi e la Penisola Iberica, per andare a costruire una eredità ; una terza fase che entra nel merito della composizione attraverso lo studio e la verifica delle prime due parti, tramite l'analisi grafica applicata ad uno specifico esempio architettonico selezionato; una quarta fase dove il punto di vista viene ribaltato e si indaga l'influenza dei luoghi visitati e dei contatti intrattenuti con alcuni personaggi della Penisola Iberica sull'architettura di Rossi, ricercandone i riferimenti. La ricerca è stata condotta attraverso lo studio di alcuni eventi selezionati nel corso degli anni che si sono mostrati significativi per l'indagine, per la risonanza che hanno avuto sulla storia dell'architettura della Penisola. A questo scopo si sono utilizzati principalmente tre strumenti: lo studio dei documenti, le pubblicazioni e le riviste prodotte in Spagna, gli scritti di Aldo Rossi in merito, e la testimonianza diretta attraverso interviste di personaggi chiave. La ricerca ha prodotto un testo suddiviso per capitoli che rispetta l'organizzazione in fasi di lavoro. A seguito di determinate condizioni storiche e politiche, studiate nella ricerca a supporto della tesi espressa, nella Penisola Iberica si è verificato il diffondersi della necessità e del desiderio di guardare e prendere a riferimento l'architettura europea e in particolar modo quella italiana. Il periodo sul quale viene focalizzata l'attenzione ha inizio negli anni Sessanta, gli ultimi prima della caduta delle dittature, scenario dei primi viaggi di Aldo Rossi nella Penisola Iberica. Questi primi contatti pongono le basi per intense e significative relazioni future. Attraverso l'approfondimento e la studio dei materiali relativi all'oggetto della tesi, si è cercato di mettere in luce il contesto culturale, l'attenzione e l'interesse per l'apertura di un dibattito intorno all'architettura, non solo a livello nazionale, ma europeo. Ciò ha evidenziato il desiderio di innescare un meccanismo di discussione e scambio di idee, facendo leva sull'importanza dello sviluppo e ricerca di una base teorica comune che rende coerente i lavori prodotti nel panorama architettonico iberico, seppur ottenendo risultati che si differenziano gli uni dagli altri. E' emerso un forte interesse per il discorso teorico sull'architettura, trasmissibile e comunicabile, che diventa punto di partenza per un metodo progettuale. Ciò ha reso palese una condivisione di intenti e l'assunzione della teoria di Aldo Rossi, acquisita, diffusa e discussa, attraverso la pubblicazione dei suoi saggi, la conoscenza diretta con l'architetto e la sua architettura, conferenze, seminari, come base teorica su cui fondare il proprio sapere architettonico ed il processo metodologico progettuale da applicare di volta in volta negli interventi concreti. Si è giunti così alla definizione di determinati eventi che hanno permesso di entrare nel profondo della questione e di sondare la relazione tra Rossi e la Penisola Iberica, il materiale fornito dallo studio di tali episodi, quali il I SIAC, la diffusione della rivista "2C. Construccion de la Ciudad", la Coleccion Arquitectura y Critica di Gustavo Gili, hanno poi dato impulso per il reperimento di una rete di ulteriori riferimenti. E' stato possibile quindi individuare un gruppo di architetti spagnoli, che si identificano come allievi del maestro Rossi, impegnato per altro in quegli anni nella formazione di una Scuola e di un insegnamento, che non viene recepito tanto nelle forme, piuttosto nei contenuti. I punti su cui si fondano le connessioni tra l'analisi urbana e il progetto architettonico si centrano attorno due temi di base che riprendono la teoria esposta da Rossi nel saggio L'architettura della città : - relazione tra l'area-studio e la città nella sua globalità, - relazione tra la tipologia edificatoria e gli aspetti morfologici. La ricerca presentata ha visto nelle sue successive fasi di approfondimento, come si è detto, lo sviluppo parallelo di più tematiche. Nell'affrontare ciascuna fase è stato necessario, di volta in volta, operare una verifica delle tappe percorse precedentemente, per mantenere costante il filo del discorso col lavoro svolto e ritrovare, durante lo svolgimento stesso della ricerca, gli elementi di connessione tra i diversi episodi analizzati. Tale operazione ha messo in luce talvolta nodi della ricerca rimasti in sospeso che richiedevano un ulteriore approfondimento o talvolta solo una rivisitazione per renderne possibile un più proficuo collegamento con la rete di informazioni accumulate. La ricerca ha percorso strade diverse che corrono parallele, per quanto riguarda il periodo preso in analisi: - i testi sulla storia dell'architettura spagnola e la situazione contestuale agli anni Settanta - il materiale riguardante il I SIAC - le interviste ai partecipanti al I SIAC - le traduzioni di Gustavo Gili nella Coleccion Arquitectura y Critica - la rivista "2C. Construccion de la Ciudad" Esse hanno portato alla luce una notevole quantità di tematiche, attraverso le quali, queste strade vengono ad intrecciarsi e a coincidere, verificando l'una la veridicità dell'altra e rafforzandone il valore delle affermazioni. Esposizione sintetica dei principali contenuti esposti dalla ricerca Andiamo ora a vedere brevemente i contenuti dei singoli capitoli. Nel primo capitolo Anni Settanta. Periodo di transizione per la Penisola Iberica si è cercato di dare un contesto storico agli eventi studiati successivamente, andando ad evidenziare gli elementi chiave che permettono di rintracciare la presenza della predisposizione ad un cambiamento culturale. La fase di passaggio da una condizione di chiusura rispetto alle contaminazioni provenienti dall'esterno, che caratterizza Spagna e Portogallo negli anni Sessanta, lascia il posto ad un graduale abbandono della situazione di isolamento venutasi a creare intorno al Paese a causa del regime dittatoriale, fino a giungere all'apertura e all'interesse nei confronti degli apporti culturali esterni. E' in questo contesto che si gettano le basi per la realizzazione del I Seminario Internazionale di Architettura Contemporanea a Santiago de Compostela, del 1976, diretto da Aldo Rossi e organizzato da César Portela e Salvador Tarragó, di cui tratta il capitolo secondo. Questo è uno degli eventi rintracciati nella storia delle relazioni tra Rossi e la Penisola Iberica, attraverso il quale è stato possibile constatare la presenza di uno scambio culturale e l'importazione in Spagna delle teorie di Aldo Rossi. Organizzato all'indomani della caduta del franchismo, ne conserva una reminescenza formale. Il capitolo è organizzato in tre parti, la prima si occupa della ricostruzione dei momenti salienti del Seminario Proyecto y ciudad historica, dagli interventi di architetti di fama internazionale, quali lo stesso Aldo Rossi, Carlo Aymonino, James Stirling, Oswald Mathias Ungers e molti altri, che si confrontano sul tema delle città storiche, alle giornate seminariali dedicate all’elaborazione di un progetto per cinque aree individuate all’interno di Santiago de Compostela e quindi dell’applicazione alla pratica progettuale dell’inscindibile base teorica esposta. Segue la seconda parte dello stesso capitolo riguardante La selezione di interviste ai partecipanti al Seminario. Esso contiene la raccolta dei colloqui avuti con alcuni dei personaggi che presero parte al Seminario e attraverso le loro parole si è cercato di approfondire la materia, in particolar modo andando ad evidenziare l’ambiente culturale in cui nacque l’idea del Seminario, il ruolo avuto nella diffusione della teoria di Aldo Rossi in Spagna e la ripercussione che ebbe nella pratica costruttiva. Le diverse interviste, seppur rivolte a persone che oggi vivono in contesti distanti e che in seguito a questa esperienza collettiva hanno intrapreso strade diverse, hanno fatto emergere aspetti comuni, tale unanimità ha dato ancor più importanza al valore di testimonianza offerta. L’elemento che risulta più evidente è il lascito teorico, di molto prevalente rispetto a quello progettuale che si è andato mescolando di volta in volta con la tradizione e l’esperienza dei cosiddetti allievi di Aldo Rossi. Negli stessi anni comincia a farsi strada l’importanza del confronto e del dibattito circa i temi architettonici e nel capitolo La fortuna critica della teoria di Aldo Rossi nella Penisola Iberica è stato affrontato proprio questo rinnovato interesse per la teoria che in quegli anni si stava diffondendo. Si è portato avanti lo studio delle pubblicazioni di Gustavo Gili nella Coleccion Arquitectura y Critica che, a partire dalla fine degli anni Sessanta, pubblica e traduce in lingua spagnola i più importanti saggi di architettura, tra i quali La arquitectura de la ciudad di Aldo Rossi, nel 1971, e Comlejidad y contradiccion en arquitectura di Robert Venturi nel 1972. Entrambi fondamentali per il modo di affrontare determinate tematiche di cui sempre più in quegli anni si stava interessando la cultura architettonica iberica, diventando così ¬ testi di riferimento anche nelle scuole. Le tracce dell’influenza di Rossi sulla Penisola Iberica si sono poi ricercate nella rivista “2C. Construccion de la Ciudad” individuata come strumento di espressione di una teoria condivisa. Con la nascita nel 1972 a Barcellona di questa rivista viene portato avanti l’impegno di promuovere la Tendenza, facendo riferimento all’opera e alle idee di Rossi ed altri architetti europei, mirando inoltre al recupero di un ruolo privilegiato dell’architettura catalana. A questo proposito sono emersi due fondamentali aspetti che hanno legittimato l’indagine e lo studio di questa fonte: - la diffusione della cultura architettonica, il controllo ideologico e di informazione operato dal lavoro compiuto dalla rivista; - la documentazione circa i criteri di scelta della redazione a proposito del materiale pubblicato. E’ infatti attraverso le pubblicazioni di “2C. Construccion de la Ciudad” che è stato possibile il ritrovamento delle notizie sulla mostra Arquitectura y razionalismo. Aldo Rossi + 21 arquitectos españoles, che accomuna in un’unica esposizione le opere del maestro e di ventuno giovani allievi che hanno recepito e condiviso la teoria espressa ne “L’architettura della città”. Tale mostra viene poi riproposta nella Sezione Internazionale di Architettura della XV Triennale di Milano, la quale dedica un Padiglione col titolo Barcelona, tres epocas tres propuestas. Dalla disamina dei progetti presentati è emerso un interessante caso di confronto tra le Viviendas para gitanos di César Portela e la Casa Bay di Borgo Ticino di Aldo Rossi, di cui si è occupato l’ultimo paragrafo di questo capitolo. Nel corso degli studi è poi emerso un interessante risvolto della ricerca che, capovolgendone l’oggetto stesso, ne ha approfondito gli aspetti cercando di scavare più in profondità nell’analisi della reciproca influenza tra la cultura iberica e Aldo Rossi, questa parte, sviscerata nell’ultimo capitolo, La Penisola Iberica nel “magazzino della memoria” di Aldo Rossi, ha preso il posto di quello che inizialmente doveva presentarsi come il risvolto progettuale della tesi. Era previsto infatti, al termine dello studio dell’influenza di Aldo Rossi sulla Penisola Iberica, un capitolo che concentrava l’attenzione sulla produzione progettuale. A seguito dell’emergere di un’influenza di carattere prettamente teorica, che ha sicuramente modificato la pratica dal punto di vista delle scelte architettoniche, senza però rendersi esplicita dal punto di vista formale, si è preferito, anche per la difficoltà di individuare un solo esempio rappresentativo di quanto espresso, sostituire quest’ultima parte con lo studio dell’altra faccia della medaglia, ossia l’importanza che a sua volta ha avuto la cultura iberica nella formazione della collezione dei riferimenti di Aldo Rossi. L’articolarsi della tesi in fasi distinte, strettamente connesse tra loro da un filo conduttore, ha reso necessari successivi aggiustamenti nel percorso intrapreso, dettati dall’emergere durante la ricerca di nuovi elementi di indagine. Si è pertanto resa esplicita la ricercata eredità di Aldo Rossi, configurandosi però prevalentemente come un’influenza teorica che ha preso le sfumature del contesto e dell’esperienza personale di chi se ne è fatto ricevente, diventandone così un continuatore attraverso il proprio percorso autonomo o collettivo intrapreso in seguito. Come suggerisce José Charters Monteiro, l’eredità di Rossi può essere letta attraverso tre aspetti su cui si basa la sua lezione: la biografia, la teoria dell’architettura, l’opera. In particolar modo per quanto riguarda la Penisola Iberica si può parlare dell’individuazione di un insegnamento riferito alla seconda categoria, i suoi libri di testo, le sue partecipazioni, le traduzioni. Questo è un lascito che rende possibile la continuazione di un dibattito in merito ai temi della teoria dell’architettura, della sue finalità e delle concrete applicazioni nelle opere, che ha permesso il verificarsi di una apertura mentale che mette in relazione l’architettura con altre discipline umanistiche e scientifiche, dalla politica, alla sociologia, comprendendo l’arte, le città la morfologia, la topografia, mediate e messe in relazione proprio attraverso l’architettura.

The enteric nervous system regulates autonomously from the central nervous system all the reflex pathways that control blood flow, motility, water and electrolyte transport and acid secretion. The ability of the gut to function in isolation is one of the most intriguing phenomenons in neurogastroenterology. This requires coding of sensory stimuli by cells in the gut wall. Enteric neurons are prominent candidates to relay mechanosensitivity. Surprisingly, the identity of mechanosensitive neurons in the enteric nervous system as well as the appropriate stimulus modality is unknown despite the evidence that enteric neurons respond to sustained distension. Objectives: The aim of our study was to record from mechanosensitive neurons using physiological stimulus modalities. Identification of sensory neurons is of central importance to understand sensory transmission under normal conditions and in gut diseases associated with sensorimotor dysfunctions, such as Irritable Bowel Syndrome. Only then it will be possible to identify novel targets that help to normalise sensory functions. Methods: We used guinea-pig ileum myenteric plexus preparations and recorded responses of all neurons in a given ganglion with a fast neuroimaging technique based on voltage sensitive dyes. To evoke a mechanical response we used two different kinds of stimuli: firstly we applied a local mechanical distortion of the ganglion surface with von Frey hair. Secondarily we mimic the ganglia deformation during physiological movements of myenteric ganglia in a freely contracting ileal preparation. We were able to reliably and reproducibly mimic this distortion by intraganglionic injections of small volumes of oxygenated and buffered Krebs solution using stimulus parameters that correspond to single contractions. We also performed in every ganglion tested, electrical stimulations to evoke fast excitatory postsynaptic potentials. Immunohistochemistry reactions were done with antibodies against Calbindin and NeuN, considered markers for sensory neurons. Results: Recordings were performed in 46 ganglia from 31 guinea pigs. In every ganglion tested we found from 1 to 21 (from 3% to 62%) responding cells with a median value of 7 (24% of the total number of neurons). The response consisted of an almost instantaneous spike discharge that showed adaptation. The median value of the action potential frequency in the responding neurons was 2.0 Hz, with a recording time of 1255 ms. The spike discharge lasted for 302 ± 231 ms and occurred only during the initial deformation phase. During sustained deformation no spike discharge was observed. The response was reproducible and was a direct activation of the enteric neurons since it remained after synaptic blockade with hexamethonium or ω-conotoxin and after long time perfusion with capsaicin. Muscle tone appears not to be required for activation of mechanosensory neurons. Mechanosensory neurons showed a response to mechanical stimulation related to the stimulus strength. All mechanosensory neurons received fast synaptic inputs. There was no correlation between mechanosensitivity and Calbindin-IR and NeuN-IR (44% of mechanosensitive neurones Calb-IR-/NeuN-IR-). Conclusions: We identified mechanosensitive neurons in the myenteric plexus of the guinea pig ileum which responded to brief deformation. These cells appear to be rapidly accommodating neurons which respond to dynamic change. All mechanosensitive neurons received fast synaptic input suggesting that their activity can be highly modulated by other neurons and hence there is a low stimulus fidelity which allows adjusting the gain in a sensory network. Mechanosensitivity appears to be a common feature of many enteric neurons belonging to different functional classes. This supports the existence of multifunctional enteric neurons which may fulfil sensory, integrative and motor functions.

Longstanding taxonomic ambiguity and uncertainty exist in the identification of the common (M. mustelus) and blackspotted (M. punctulatus) smooth-hound in the Adriatic Sea. The lack of a clear and accurate method of morphological identification, leading to frequent misidentification, prevents the collation of species-specific landings and survey data for these fishes and hampers the delineation of the distribution ranges and stock boundaries of the species. In this context, adequate species-specific conservation and management strategies can not be applied without risks of population declining and local extinction. In this thesis work I investigated the molecular ecology of the two smooth-hound sharks which are abundant in the demersal trawl surveys carried out in the NC Adriatic Sea to monitor and assess the fishery resources. Ecological and evolutionary relationships were assessed by two molecular tests: a DNA barcoding analysis to improve species identification (and consequently the knowledge of their spatial ecology and taxonomy) and a hybridization assay based on the nuclear codominant marker ITS2 to evaluate reproductive interactions (hybridization or gene introgression). The smooth-hound sharks (N=208) were collected during the MEDITS 2008 and 2010 campaigns along the Italian and Croatian coasts of the Adriatic Sea, in the Sicilian Channel and in the Algerian fisheries. Since the identification based on morphological characters is not strongly reliable, I performed a molecular identification of the specimens producing for each one the cytochrome oxidase subunit 1 (COI) gene sequence (ca. 640 bp long) and compared them with reference sequences from different databases (GenBank and BOLD). From these molecular ID data I inferred the distribution of the two target species in the NC Adriatic Sea. In almost the totality of the MEDITS hauls I found no evidence of species sympatry. The data collected during the MEDITS survey showed an almost different distribution of M. mustelus (confined along the Italian coasts) and M. punctulatus (confined along the Croatian coasts); just one sample (Gulf of Venice, where probably the ranges of the species overlap) was found to have catches of both the species. Despite these data results suggested no interaction occurred between my two target species at least during the summertime (the period in which MEDITS survey is carried out), I still wanted to know if there were inter-species reproductive interactions so I developed a simple molecular genetic method to detect hybridization. This method is based on DNA sequence polymorphism among species in the nuclear ribosomal Internal Transcribed Spacer 2 locus (ITS2). Its application to the 208 specimens collected raised important questions regarding the ecology of this two species in the Adriatic Sea. In fact results showed signs of hybridization and/or gene introgression in two sharks collected during the trawl survey of 2008 and one collected during the 2010 one along the Italian and Croatian coasts. In the case that it will be confirmed the hybrid nature of these individuals, a spatiotemporal overlapping of the mating behaviour and ecology must occur. At the spatial level, the northern part of the Adriatic Sea (an area where the two species occur with high frequency of immature individuals) could likely play the role of a common nursery area for both species.

Purpose of this research is to deepen the study on the section in architecture. The survey aims as important elements in the project Teatro Domestico by Aldo Rossi built for the XVII Triennale di Milano in 1986 and, through the implementation on several topics of architecture, verify the timeliness and fertility in the new compositional exercises. Through the study of certain areas of the Rossi’s theory we tried to find a common thread for the reading of the theater project. The theater is the place of the ephemeral and the artificial, which is why his destiny is the end and the fatal loss. The design and construction of theater setting has always had a double meaning between the value of civil architecture and testing of new technologies available. Rossi's experience in this area are clear examples of the inseparable relationship between the representation of architecture as art and design of architecture as a model of reality. In the Teatro Domestico, the distinction between representation and the real world is constantly canceled and returned through the reversal of the meaning and through the skip of scale. At present, studies conducted on the work of Rossi concern the report that the architectural composition is the theory of form, focusing compositional development of a manufacturing process between the typological analysis and form invention. The research, through the analysis of some projects few designs, will try to analyze this issue through the rules of composition both graphical and concrete construction, hoping to decipher the mechanism underlying the invention. The almost total lack of published material on the project Teatro Domestico and the opportunity to visit the archives that preserve the drawings, has allowed the author of this study to deepen the internal issues in the project, thus placing this search as a first step toward possible further analysis on the works of Rossi linked to performance world. The final aim is therefore to produce material that can best describe the work of Rossi. Through the reading of the material published by the same author and the vision of unpublished material preserved in the archives, it was possible to develop new material and increasing knowledge about the work, otherwise difficult to analyze. The research is divided into two groups. The first, taking into account the close relationship most frequently mentioned by Rossi himself between archeology and architectural composition, stresses the importance of tipo such as urban composition reading system as well as open tool of invention. Resuming Ezio Bonfanti’s essay on the work of the architect we wanted to investigate how the paratactic method is applied to the early work conceived and, subsequently as the process reaches a complexity accentuated, while keeping stable the basic terms. Following a brief introduction related to the concept of the section and the different interpretations that over time the term had, we tried to identify with this facility a methodology for reading Rossi’s projects. The result is a constant typological interpretation of the term, not only related to the composition in plant but also through the elevation plans. The section is therefore intended as the overturning of such elevation is marked on the same plane of the terms used, there is a different approach, but a similarity of characters. The identification of architectural phonemes allows comparison with other arts. The research goes in the direction of language trying to identify the relationship between representation and construction, between the ephemeral and the real world. In this sense it will highlight the similarities between the graphic material produced by Ross and some important examples of contemporary author. The comparison between the composition system with the surrealist world of painting and literature will facilitate the understanding and identification of possible rules applied by Rossi. The second part of the research is characterized by a focus on the intent of the project chosen. Teatro Domestico embodies a number of elements that seem to conclude (assuming an end point but also to start) a curriculum author. With it, the experiments carried out on the theater started with the project for the Teatrino Scientifico (1978) through the project for the Teatro del Mondo (1979), into a Laic Tabernacle representative collective and private memory of the city. Starting from a reading of the draft, through the collection of published material, we’ve made an analysis on the explicit themes of the work, finding the conceptual references. Following the taking view of the original materials not published kept at Aldo Rossi's Archive Collection of the Canadian Center for Architecture in Montréal, will be implemented through the existing techniques for digital representation, a virtual reconstruction of the project, adding little to the material, a new element for future studies. The reconstruction is part of a larger research studies where the current technologies of composition and representation in architecture stand side by side with research on the method of composition of this architect. The results achieved are in addition to experiences in the past dealt with the reconstruction of some of the lost works of Aldo Rossi. A partial objective is to reactivate a discourse around this work is considered non-principal, among others born in the prolific activities. Reassessment of development projects which would bring the level of ephemeral works most frequented by giving them the value earned. In conclusion, the research aims to open a new field of interest on the part not only as a technical instrument of representation of an idea but as an actual mechanism through which composition is formed and the idea is developed.

Oxidativer Stress ist seit über 25 Jahren als ein Charakteristikum vieler pathologischer Prozesse bekannt. Helmut Sies beschrieb bereits in den 1980er Jahren oxidativen Stress als Störung in der prooxidativ – antioxidativen Balance zugunsten der prooxidativen Seite, wodurch es potentiell zu Schäden in verschiedenen Geweben kommt. Oxidativer Stress tritt sowohl bei neurodegenerativen Erkrankungen wie Morbus Alzheimer, Morbus Parkinson und zerebraler Ischämie, bei peripheren Erkrankungen wie Arteriosklerose, als auch beim Alterungsprozess per se auf und wird als Ursache oder zumindest als ein krankheitsfördernder Faktor diskutiert. Die in in vitro-Experimenten als vielversprechend antioxidativ getesteten Substanzen (meist phenolhaltig) ergaben in mehreren klinischen Studien keinen signifikanten Vorteil. Um die Ursachen dieser Ergebnisse näher zu analysieren, wurde in der vorliegenden Arbeit auf Basis des cytoprotektiven Phenothiazins, einem aromatischen trizyklischen Amin, der Einfluss von verschiedenen Substituenten im Hinblick auf Lipophilie, Radikalstabilisierung und Löslichkeit des Moleküls chemisch vorhergesagt. Anhand dieser in silicio Struktur-Wirkungs-Beziehung wurden anschließend neue Modellsubstanzen synthetisiert, welche sich systematisch in den drei zuvor genannten Parametern unterschieden. Dies wurde durch Substitution von unterschiedlich langen Fettsäureketten, von löslichkeitsbeeinflussenden funktionellen Gruppen, oder durch Anellierung zusätzlicher aromatischer Ringe erreicht. In den folgenden Versuchen zu antioxidativer Kapazität, zellulärem Überleben, Lipidperoxidation und Proteinoxidation zeigte sich, dass mit gesteigerter Stabilität der korrespondierenden Radikale und mit wachsender Lipophilie die antioxidativ cytoprotektive Aktivität der neuen Derivate bis zu einer gewissen Grenze (logP ≈ 7) signifikant zunahm; über diesen Wert hinaus sank die Effektivität wieder ab. Benzanellierte Phenothiazine entwickelten mit EC50-Werten von ungefähr 8-10 nM die höchste mittlere effektive Wirkkonzentration in oxidativ geschädigten, klonalen hippocampalen Neuronen (HT-22 Zellen). Dies entspricht einer etwa 20-fachen Verbesserung gegenüber α-Tocopherol, welches bisher als bestes natürliches lipophiles Antioxidans angesehen wurde. Im Vergleich zu Phenothiazin erreichen die neuen Antioxidantien immerhin eine höhere Effektivität um den Faktor 4. Folglich sind es sowohl Aspekte der Löslichkeit und der Distribution, welche die Potenz der gegenwärtigen Antioxidantien limitieren als auch Aspekte der Radikalstabilisierung, die Einfluss auf die primäre Wirksamkeit nehmen. Dieses Wissen sollte beim zukünftigen Design neuer, antioxidativ potenter Moleküle im Hinblick auf ihren langfristigen Einsatz bei neurodegenerativen Erkrankungen von Nutzen sein.

Based on an ethnographic case study in the border cities of Frankfurt (Oder), Germany and Słubice, Poland, this article explores the construction and maintenance of ethnic difference within the transnational economic and social spaces created by the European Union's common market. Through an examination of three domains of cross-border citizenship practice - shopping and consumption, housing and work - this article argues that even as the European Union deploys policies aimed at creating de-territorialised and supranational forms of identity and citizenship, economic asymmetries and hierarchies of value embedded within these policies grant rights differentially in ways that continue to be linked to ethnicity and nationality.

Background Chronic pancreatitis (CP) is an inflammatory disease that in some patients leads to exocrine and endocrine dysfunction. In industrialized countries the most common aetiology is chronic alcohol abuse. Descriptions of associated genetic alterations in alcoholic CP are rare. However, a common PNPLA3 variant (p.I148M) is associated with the development of alcoholic liver cirrhosis (ALC). Since, alcoholic CP and ALC share the same aetiology PNPLA3 variant (p.I148M) possibly influences the development of alcoholic CP. Methods Using melting curve analysis we genotyped the variant in 1510 patients with pancreatitis or liver disease (961 German and Dutch alcoholic CP patients, 414 German patients with idiopathic or hereditary CP, and 135 patients with ALC). In addition, we included in total 2781 healthy controls in the study. Results The previously published overrepresentation of GG-genotype was replicated in our cohort of ALC (p-value <0.0001, OR 2.3, 95% CI 1.6–3.3). Distributions of genotype and allele frequencies of the p.I148M variant were comparable in patients with alcoholic CP, idiopathic and hereditary CP and in healthy controls. Conclusions The absence of an association of PNPLA3 p.I148M with alcoholic CP seems not to point to a common pathway in the development of alcoholic CP and alcoholic liver cirrhosis.

BACKGROUND: Thirty-eight of the 73 consecutive acute ruptures of the anterior cruciate ligament (ACL) proven by a magnetic resonance imaging scan (MRI) in skeletally mature patients (16-55 years old) were classified as suitable for primarily conservative treatment. Patient selection was performed using a preoperative screening protocol based on the structural damage, clinical symptoms, compliance, sportive activity, and the consent of a well-informed patient. METHODS: In 12 of the 38 treated patients, the conservative protocol showed a good to very good outcome, 2 patients had persistent giving-way and were considering ACL reconstruction, 14 patients had a secondary ACL reconstruction in our clinic (average 5.3 months after injury), 9 patients were operated on in other hospitals (average 13.3 months after injury), and 1 patient was lost to follow-up. RESULTS: All patients with successful conservative treatment were able to perform low-risk pivoting sports and two patients are practicing high-risk pivoting sports. The average International Knee Documentation Committee (IKDC) score was 92.5 (82.8-98.9); the subjective overall knee function was 93% (60% to 100%). Of the 12 patients with good and very good results, 6 continued playing the same sports at an unreduced intensity, 4 patients reduced their activities slightly, and 1 patient played more sports than before. CONCLUSIONS: Although the authors performed a preoperative screening to select patients suitable for conservative treatment, almost two-thirds of the primarily conservatively treated ACL ruptures needed an operative reconstruction in the long term. In one-third of the patients, conservative treatment led to a good or very good result. At the endpoint of the study only 12 (16%) of a total of 73 patients with acute injuries of the ACL had successful conservative treatment. Therefore, patients must be comprehensively instructed about the treatment program and the chances of success of conservative ACL treatment.

Research and professional practices have the joint aim of re-structuring the preconceived notions of reality. They both want to gain the understanding about social reality. Social workers use their professional competence in order to grasp the reality of their clients, while researchers’ pursuit is to open the secrecies of the research material. Development and research are now so intertwined and inherent in almost all professional practices that making distinctions between practising, developing and researching has become difficult and in many aspects irrelevant. Moving towards research-based practices is possible and it is easily applied within the framework of the qualitative research approach (Dominelli 2005, 235; Humphries 2005, 280). Social work can be understood as acts and speech acts crisscrossing between social workers and clients. When trying to catch the verbal and non-verbal hints of each others’ behaviour, the actors have to do a lot of interpretations in a more or less uncertain mental landscape. Our point of departure is the idea that the study of social work practices requires tools which effectively reveal the internal complexity of social work (see, for example, Adams & Dominelli & Payne 2005, 294 – 295). The boom of qualitative research methodologies in recent decades is associated with much profound the rupture in humanities, which is called the linguistic turn (Rorty 1967). The idea that language is not transparently mediating our perceptions and thoughts about reality, but on the contrary it constitutes it was new and even confusing to many social scientists. Nowadays we have got used to read research reports which have applied different branches of discursive analyses or narratologic or semiotic approaches. Although differences are sophisticated between those orientations they share the idea of the predominance of language. Despite the lively research work of today’s social work and the research-minded atmosphere of social work practice, semiotics has rarely applied in social work research. However, social work as a communicative practice concerns symbols, metaphors and all kinds of the representative structures of language. Those items are at the core of semiotics, the science of signs, and the science which examines people using signs in their mutual interaction and their endeavours to make the sense of the world they live in, their semiosis. When thinking of the practice of social work and doing the research of it, a number of interpretational levels ought to be passed before reaching the research phase in social work. First of all, social workers have to interpret their clients’ situations, which will be recorded in the files. In some very rare cases those past situations will be reflected in discussions or perhaps interviews or put under the scrutiny of some researcher in the future. Each and every new observation adds its own flavour to the mixture of meanings. Social workers have combined their observations with previous experience and professional knowledge, furthermore, the situation on hand also influences the reactions. In addition, the interpretations made by social workers over the course of their daily working routines are never limited to being part of the personal process of the social worker, but are also always inherently cultural. The work aiming at social change is defined by the presence of an initial situation, a specific goal, and the means and ways of achieving it, which are – or which should be – agreed upon by the social worker and the client in situation which is unique and at the same time socially-driven. Because of the inherent plot-based nature of social work, the practices related to it can be analysed as stories (see Dominelli 2005, 234), given, of course, that they are signifying and told by someone. The research of the practices is concentrating on impressions, perceptions, judgements, accounts, documents etc. All these multifarious elements can be scrutinized as textual corpora, but not whatever textual material. In semiotic analysis, the material studied is characterised as verbal or textual and loaded with meanings. We present a contribution of research methodology, semiotic analysis, which has to our mind at least implicitly references to the social work practices. Our examples of semiotic interpretation have been picked up from our dissertations (Laine 2005; Saurama 2002). The data are official documents from the archives of a child welfare agency and transcriptions of the interviews of shelter employees. These data can be defined as stories told by the social workers of what they have seen and felt. The official documents present only fragmentations and they are often written in passive form. (Saurama 2002, 70.) The interviews carried out in the shelters can be described as stories where the narrators are more familiar and known. The material is characterised by the interaction between the interviewer and interviewee. The levels of the story and the telling of the story become apparent when interviews or documents are examined with the use of semiotic tools. The roots of semiotic interpretation can be found in three different branches; the American pragmatism, Saussurean linguistics in Paris and the so called formalism in Moscow and Tartu; however in this paper we are engaged with the so called Parisian School of semiology which prominent figure was A. J. Greimas. The Finnish sociologists Pekka Sulkunen and Jukka Törrönen (1997a; 1997b) have further developed the ideas of Greimas in their studies on socio-semiotics, and we lean on their ideas. In semiotics social reality is conceived as a relationship between subjects, observations, and interpretations and it is seen mediated by natural language which is the most common sign system among human beings (Mounin 1985; de Saussure 2006; Sebeok 1986). Signification is an act of associating an abstract context (signified) to some physical instrument (signifier). These two elements together form the basic concept, the “sign”, which never constitutes any kind of meaning alone. The meaning will be comprised in a distinction process where signs are being related to other signs. In this chain of signs, the meaning becomes diverged from reality. (Greimas 1980, 28; Potter 1996, 70; de Saussure 2006, 46-48.) One interpretative tool is to think of speech as a surface under which deep structures – i.e. values and norms – exist (Greimas & Courtes 1982; Greimas 1987). To our mind semiotics is very much about playing with two different levels of text: the syntagmatic surface which is more or less faithful to the grammar, and the paradigmatic, semantic structure of values and norms hidden in the deeper meanings of interpretations. Semiotic analysis deals precisely with the level of meaning which exists under the surface, but the only way to reach those meanings is through the textual level, the written or spoken text. That is why the tools are needed. In our studies, we have used the semiotic square and the actant analysis. The former is based on the distinctions and the categorisations of meanings, and the latter on opening the plotting of narratives in order to reach the value structures.

Much has happened in the past fifty years, and the broadcasting system and in fact the entire media landscape have changed in many significant ways. Yet, the debate on the role of public service media and the involvement of the state in them still perseveres. It has indeed been reinvigorated due to the tectonic shifts in media production, distribution, access and consumption caused by digital technologies in general, and the Internet in particular. The gist of the debates has however curiously remained almost the same and is still focused on a set of economic arguments that call for state intervention in public media, and not unimportantly, on the various political interpretations of these economic arguments. In Europe, the debate has another essential core too, as Public Service Broadcasting (PSB) has been traditionally entrusted to serve some higher goals intrinsically related to key democratic and cultural processes. Accordingly, PSB in Western Europe has developed as the core media institution at the national level and has become deeply embedded in many facets of the nation’s economic, political, social and cultural life. Against the backdrop of PSB’s history, its vital tasks in society, as well as the dramatic changes brought about by the digitally networked environment, the question on the future of PSB and its transition into Public Service Media (PSM) is very interesting, to say the least, and highly challenging at the same time. The book by Karen Donders, Public Service Media and Policy in Europe (Palgrave, 2012), makes an essential contribution to these complex debates, and more importantly, adds some new value to an otherwise saturated discourse.
