Procedures for routine analysis of soil phosphorus (P) have been used for assessment of P status, distribution and P losses from cultivated mineral soils. No similar studies have been carried out on wetland peat soils. The objective was to compare extraction efficiency of ammonium lactate (PAL), sodium bicarbonate (P-Olsen), and double calcium lactate (P-DCaL) and P distribution in the soil profile of wetland peat soils. For this purpose, 34 samples of the 0-30, 30-60 and 60-90 cm layers were collected from peat soils in Germany, Israel, Poland, Slovenia, Sweden and the United Kingdom and analysed for P. Mean soil pH (CaCl2, 0.01 M) was 5.84, 5.51 and 5.47 in the 0-30, 30-60 and 60-90 cm layers, respectively. The P-DCaL was consistently about half the magnitude of either P-AL or P-Olsen. The efficiency of P extraction increased in the order P-DCaL < P-AL &LE; P-Olsen, with corresponding means (mg kg(-1)) for all soils (34 samples) of 15.32, 33.49 and 34.27 in 0-30 cm; 8.87, 17.30 and 21.46 in 30-60 cm; and 5.69, 14.00 and 21.40 in 60-90 cm. The means decreased with depth. When examining soils for each country separately, P-Olsen was relatively evenly distributed in the German, UK and Slovenian soils. P-Olsen was linearly correlated (r = 0.594, P = 0.0002) with pH, whereas the three P tests (except P-Olsen vs P-DCaL) significantly correlated with each other (P = 0.017850.0001). The strongest correlation (r = 0.617, P = 0.0001) was recorded for P-AL vs P-DCaL) and the two methods were inter-convertible using a regression equation: P-AL = -22.593 + 5.353 pH + 1.423 P-DCaL, R-2 = 0.550.

Single-cell analysis is essential for understanding the processes of cell differentiation and metabolic specialisation in rare cell types. The amount of single proteins in single cells can be as low as one copy per cell and is for most proteins in the attomole range or below; usually considered as insufficient for proteomic analysis. The development of modern mass spectrometers possessing increased sensitivity and mass accuracy in combination with nano-LC-MS/MS now enables the analysis of single-cell contents. In Arabidopsis thaliana, we have successfully identified nine unique proteins in a single-cell sample and 56 proteins from a pool of 15 single-cell samples from glucosinolate-rich S-cells by nanoLC-MS/MS proteomic analysis, thus establishing the proof-of-concept for true single-cell proteomic analysis. Dehydrin (ERD14_ARATH), two myrosinases (BGL37_ARATH and BGL38_ARATH), annexin (ANXD1_ARATH), vegetative storage proteins (VSP1_ARATH and VSP2_ARATH) and four proteins belonging to the S-adenosyl-l-methionine cycle (METE_ARATH, SAHH1_ARATH, METK4_ARATH and METK1/3_ARATH) with associated adenosine kinase (ADK1_ARATH), were amongst the proteins identified in these single-S-cell samples. Comparison of the functional groups of proteins identified in S-cells with epidermal/cortical cells and whole tissue provided a unique insight into the metabolism of S-cells. We conclude that S-cells are metabolically active and contain the machinery for de novo biosynthesis of methionine, a precursor for the most abundant glucosinolate glucoraphanine in these cells. Moreover, since abundant TGG2 and TGG1 peptides were consistently found in single-S-cell samples, previously shown to have high amounts of glucosinolates, we suggest that both myrosinases and glucosinolates can be localised in the same cells, but in separate subcellular compartments. The complex membrane structure of S-cells was reflected by the presence of a number of proteins involved in membrane maintenance and cellular organisation.

This article examines the role of advertisement and promotion in the successful development of nationwide building societies in interwar Britain and the rapid overall growth of the building society movement. Major building societies are shown to have used extensive advertising to compensate for their initial lack of established national brands, promote home-ownership, and make savers aware of the attractive earnings and high security of building society savings. During a period when most building societies had very limited branch networks, extensive advertising increased the public profile of the major societies and thus assisted their rapid expansion via lower-cost modes such as agency networks.

The phytopathogen Xylella fastidiosa produces long type IV pili and short type I pili involved in motility and adhesion. In this work, we have investigated the role of sigma factor sigma(54) (RpoN) in the regulation of fimbrial biogenesis in X. fastidiosa. An rpoN null mutant was constructed from the non-pathogenic citrus strain J1a12, and microarray analyses of global gene expression comparing the wild type and rpoN mutant strains showed few genes exhibiting differential expression. In particular, gene pilA1 (XF2542), which encodes the structural pilin protein of type IV pili, showed decreased expression in the rpoN mutant, whereas two-fold higher expression of an operon encoding proteins of type I pili was detected, as confirmed by quantitative RT-PCR (qRT-PCR) analysis. The transcriptional start site of pilA1 was determined by primer extension, downstream of a sigma(54)-dependent promoter. Microarray and qRT-PCR data demonstrated that expression of only one of the five pilA paralogues, pilA1, was significantly reduced in the rpoN mutant. The rpoN mutant made more biofilm than the wild type strain and presented a cell-cell aggregative phenotype. These results indicate that sigma(54) differentially regulates genes involved in type IV and type I fimbrial biogenesis, and is involved in biofilm formation in X. fastidiosa.

Este texto tem como objetivo analisar o papel e a atuação da Assessoria Econômica no segundo governo Vargas a partir de entrevistas que alguns de seus membros concederam ao Centro de Pesquisa e Documentação de História Contemporânea do Brasil (CPDOC), da Fundação Getulio Vargas, ao longo da década de 1980. Ao avaliar a posteriori a curta, porém intensa, experiência que tiveram como “braço imediato” do presidente da República,2 os depoimentos de Rômulo de Almeida, Cleantho Paiva Leite e Ignacio Rangel tornam-se fontes particularmente interessantes para quem está interessado em entender os meandros do processo de industrialização, em particular o papel que nele desempenharam as chamadas “elites técnicas”.

This paper analyzes the Brazilian Supreme Court (STF) Decision (ADPF n. 54) in which the court, by majority of votes, stated that the termination of the pregnancy of an anencephalic fetus does not constitute a crime of abortion. The paper analyzes the arguments used by the judges in their opinions to show that they agree on the conclusion, but strongly disagree on their reasons, with the result that the court remains deeply divided on the legal concept of life. The paper then points to the fact that the possibility of different majority positions regarding the decision and the arguments that justify it is not accidental, but the result of the court’s structure, which does not guarantee the collective rationality of the court’s decisions.

Brachiaria decumbens, vulgarmente conhecida por capim-braquiária, é considerada uma importante planta daninha, muito agressiva e de difícil controle em áreas onde foi introduzida como forrageira e posteriormente tornou-se lavoura. Com o objetivo de estudar a produção de massa seca, a distribuição e o acúmulo de macronutrientes em plantas de capim-braquiária, foi conduzido o presente trabalho em casa de vegetação. As plantas foram cultivadas em vasos preenchidos com areia de rio lavada e peneiradas e irrigadas diariamente com solução nutritiva completa de Hoagland & Arnon a 50% da concentração original. A primeira avaliação foi realizada aos 20 dias após a emergência (DAE), e as seguintes, em intervalos de 14 dias. Foi determinada a massa seca das diferentes partes da planta. O material foi moído e analisado quanto aos teores de macronutrientes. Os resultados indicaram que a planta apresentou crescimento durante toda a fase experimental. O maior acúmulo ocorreu aos 160 DAE, sendo de 38,27 g planta-1 de massa seca. Aos 160 DAE, cerca de 37,37% da massa seca estava alocada nas raízes, 37,54% nos colmos + bainhas e 25,09% nas folhas. O acúmulo total dos macronutrientes foi crescente até os 146 dias após a emergência. Aos 76 DAE (período de maior competição dessa espécie com a maioria das culturas anuais), uma planta de capim-braquiária acumulou 1,32 grama de massa seca; 40,8 mg de K; 18,2 mg de N; 6,3 mg de Ca; 5,2 mg de Mg; 2,68 mg de P; e 2,4 mg de S.

O objetivo deste trabalho foi determinar parâmetros genéticos relacionados a características morfológicas e suas correlações genéticas com a produção de leite, em vacas da raça Gir. Utilizaram-se 3.805 registros provenientes de 2.142 vacas. O modelo utilizado na análise de características morfológicas continha os efeitos fixos de rebanho, ano e estação de classificação, estádio da lactação e idade da vaca à classificação, além da identificação do classificador. Quanto à produção de leite, foram incluídos no modelo os efeitos fixos de rebanho, ano e estação de parição e idade da vaca ao parto. Os parâmetros genéticos foram obtidos por meio do aplicativo REMLF90. As estimativas de herdabilidade variaram de 0,09 a 0,54. A variabilidade genética aditiva da maioria das características é suficiente para que ganhos genéticos anuais significativos possam ser alcançados com o processo de seleção. As correlações genéticas entre as características morfológicas variaram de baixas a altas e, entre elas e a produção de leite, de baixas a moderadas. Altas correlações genéticas entre algumas características morfológicas implicam a possibilidade de exclusão de algumas delas do programa de melhoramento genético da raça Gir, no Brasil. As correlações genéticas entre produção de leite e algumas características morfológicas indicam que estas podem ser utilizadas na formação de índices de seleção.
