We assessed relationships between phytoplankton standing stock, measured as chlorophyll a (Chl a), primary production (PP), and heterotrophic picoplankton production (HPP), in the epipelagic zone (0-100 m) as well as in the mesopelagic zone (100-1,000 m) in the polar frontal zone of the Atlantic sector of the Southern Ocean in austral summer (late December to January) and fall (March to early May). Integrated epipelagic HPP was positively correlated to integrated PP in summer (data for fall are not available) but not to integrated Chl a. However, integrated mesopelagic HPP was positively correlated to Chl a in summer as well as fall. The mesopelagic fraction of HPP as a percentage of total HPP was also positively correlated to Chl a, whereas the epipelagic fraction of HPP was negatively correlated to it. These results indicate that with increasing phytoplankton standing stock, constituted mainly of highly silicified diatoms, the focus of its consumption by heterotrophic picoplankton shifts from epipelagic to mesopelagic waters. With a growth efficiency of 30%, our HPP data indicate that in both the epipelagic and mesopelagic zone heterotrophic picoplankton consume 20% of PP. Mesopelagic heterotrophic picoplankton consumed around 80% of the sinking flux, measured from depletion of 234Th, which is a lower fraction than that reported from the central and subarctic Pacific. Our analysis indicates that it is important to include mesopelagic HPP in comprehensive assessments of the microbial consumption of PP, phytoplankton biomass, and particulate organic matter in cold oceanic systems with high rates of export production.

The insertion of the blood retrotransposon into the untranslated region of exon 7 of the sn-glycerol-3-phosphate dehydrogenase-encoding gene (Gpdh) in Drosophila melanogaster induces a GPDH isozyme-GPDH-4-and alters the pattern of expression of the three normal isozymes-GPDH-1 to GPDH-3. The process of transcript terminus formation inside the retrotransposon insertion reduces the level of the Gpdh transcript that contains exon 8 and increases the level of the transcript that contains exons 1-7. The induced GPDH-4 isozyme is a translation product of the three transcripts that contain fragments of the blood retrotransposon. The mechanism of mutagenesis by the blood insertion is postulated to involve the pause or termination of transcription within the blood sequence, which in turn is caused by the interference of a DNA-binding protein with the RNA polymerase. Thus, we show the formation of a new functional GPDH protein by the insertion of a transposable element and discuss the evolutionary significance of this phenomenon.

alpha-Amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptors that lack the glutamate receptor GluR2 subunit are Ca(2+)-permeable and exhibit inwardly rectifying current responses to kainate and AMPA. A proportion of cultured rat hippocampal neurons show similar Ca(2+)-permeable inwardly rectifying AMPA receptor currents. Inward rectification in these neurons was lost with intracellular dialysis and was not present in excised outside-out patches but was maintained in perforated-patch whole-cell recordings, suggesting that a diffusible cytoplasmic factor may be responsible for rectification. Inclusion of the naturally occurring polyamines spermine and spermidine in the recording pipette prevented loss of rectification in both whole-cell and excised-patch recordings; Mg2+ and putrescine were without effect. Inward rectification of Ca(2+)-permeable AMPA receptors may reflect voltage-dependent channel block by intracellular polyamines.
