O uso agrÃcola do lodo é uma alternativa viável para a reciclagem de nutrientes, embora haja certa preocupação com o aumento de espécies quÃmicas solúveis originadas da mineralização da fração orgânica. Assim, para conhecer a disponibilidade de nutrientes e o risco de contaminação do solo e da água, o estudo da solução do solo é uma ferramenta valiosa. Nesses estudos, o número de variáveis utilizadas ultrapassa facilmente uma dezena, e ferramentas de análise exploratória, como a análise de componentes principais (ACP), revelam a existência ou não de amostras anômalas, de relações entre as variáveis medidas e sua contribuição relativa e de relações ou agrupamentos entre amostras. O objetivo deste trabalho foi identificar variáveis relacionadas com a solução do solo e alteradas, em razão do uso do lodo de esgoto e da adubação mineral, de forma a complementar os procedimentos para monitoramento/avaliação de áreas que recebem esse tipo de resÃduo. A composição da solução do solo de uma área que recebeu lodo de esgoto por sete anos consecutivos foi estudada dos 22 aos 46 meses após a sua última aplicação. Com a solução do solo foi possÃvel, por meio da ACP, distinguir as áreas em função da adubação aplicada. Após quatro anos da interrupção das aplicações sucessivas de lodo de esgoto, não verificou-se concentração apreciável de metais na solução do solo, embora ainda seja possÃvel identificar traços da mineralização do resÃduo. As variáveis pH, Mn2+, COD, SO4(2-), NO3- e NH4+ na solução do solo podem ser utilizadas no monitoramento de áreas tratadas com lodo de esgoto.

RESUMEL'agriculture urbaine et périurbalne - nommée ci-après AU - est un thème fort de recherche transversale, au vu des nombreux enjeux économiques, sociaux et environnementaux. L'objectif de cette recherche était de contribuer à une meilleure connaissance des processus de transfert de polluants et du cycle des nutriments à l'échelle locale, afin de déterminer sous quelles conditions l'AU de Dakar peut être pratiquée sans porter atteinte à la santé et à l'environnement.Une approche basée sur l'étude des processus géochimiques dans ie sol jusqu'à la nappe a été choisie, à l'échelle de la parcelle cultivée et à une échelle un peu plus large de la zone périurbaine de Dakar pour déterminer les influences du type d'occupation du sol.L'évaluation des impacts de l'irrigation avec des eaux usées brutes et des eaux de nappe saumâtres sur la qualité des sols (chapitre 2) a montré que l'alcalinité et les teneurs en calcium élevées des eaux saumâtres induisent la précipitation de CaC03 dans l'horizon superficiel du sol. Na remplace consécutivement Ca sur le complexe argilo-humique du sol et les bicarbonates diminuent dans la solution du sol. Le carbone organique dissout (COD) augmente significativement dans la solution du sol et dans la nappe sous-jacente. Malgré l'alcalinité et les teneurs très élevées en calcium des eaux usées, il y a peu de précipitation de CaC03 dans l'horizon superficiel du sol et une faible augmentation du sodium échangeable ESP. La nitrification de l'ammonium des eaux usées (moy 190mg/L à Pikine) produit des protons, qui ne sont plus tamponnés par les bicarbonates exportés hors du profil. Il y a alors une nette baisse de pH des sols irrigués par des eaux usées non traitées. Les sols irrigués par des eaux usées et saumâtres stockent moins de C et Ν que les sols de référence.L'évaluation de l'influence de l'occupation des sols en zone périurbaine sur à la nappe phréatique peu profonde (chapitre 3) a permis de déterminer les traceurs représentatifs de l'occupation du sol, à savoir Br/CI, NO3/CI et δ180-Ν03 pour l'irrigation par des eaux usées, pH et δ15Ν-Ν03 pour l'irrigation par des eaux de nappe, et Rb+Cr et Κ pour les lixiviats de fosses septiques. Ce chapitre a mis en évidence des points importants de la dynamique de l'azote en zone périurbaine sous deux occupations du sol : (1) la dénitrification est un processus important dans l'agrosystème périurbain de Dakar en bas de dune, dans les gleysols où l'on trouve des conditions temporairement réduites et un substrat organique favorables aux microorganismes de la dénitrification. Les teneurs en nitrates sont presque nulles avec irrigation d'eau de nappe. (2) en bas de pente, mais avec irrigation quotidienne par les eaux usées, l'apport continu d'ammonium inhibe probablement la dénitrification, mais favorise la volatilisation. (3) la nitrification de l'ammonium dans la nappe lors de la lixiviation des fosses septiques se distingue de la nitrification de l'ammonium dans la zone non saturée dans la zone d'agriculture périurbaine par la composition isotopique de l'oxygène de l'eau. Une comparaison des flux d'azote entre l'agrosystème et les quartiers périurbains de Dakar (chapitre 4) ont révélé que ces derniers étaient du même ordre de grandeur par unité de surface, à savoir 2-4 tonnes Ν /ha/an.L'évaluation des flux de pesticides dans l'agrosystème et des risques induits pour les eaux souterraines (chapitre 5) a révélé un fiux total de pesticides de 60kg/ha/an, totalisant 15 matières actives. Seules deux de ces matières actives sont autorisées par le comité des pesticides sahélien. Les pesticides les plus utilisés par les producteurs sont l'organochloré dicofol, les organophosphorés methamidophos, dimethoate et fenithrotion ainsi que le cabamate methomyl. Les flux les plus importants sont de 9 à 7 kg/ha/an (methomyl, methamidophos, ethoprophos et dicofol). Les pesticides qui présentent un risque élevé de contamination des eaux souterraines et qui devraient être prioritaires pour un suivi analytique sont : le carbofuran, le dimethoate, l'ethoprophos et le methomyl.En conclusion, une meilleure gestion de la fertilisation est nécessaire dans la zone d'AU de Dakar, afin de (1) réduire les pertes gazeuses qui contribuent à l'effet de serre, (2) de ralentir la minéralisation du carbone et de l'azote organiques pour créer un stock de C et Ν dans ces sols, (3) de limiter le lessivage dans la nappe et enfin, 4) d'augmenter l'efficacité d'utilisation de Ν par les plantes. Une optimisation de l'irrigation devrait limiter l'alcalinisation secondaire. Enfin, la mise en place d'une lutte intégrée ou biologique contre les ravageurs est indispensable afin de minimiser les risques pour les eaux souterraines et les mares permanentes.ABSTRACTUrban and periurban agriculture (UA) is an important issue in southern countries, because of its key role in their social and economical development and its environmental concern. The goal of this study was to contribute to a better understanding of pollutant transfer and nutrient cycling at the local scale, in order to implement the necessary improvements to guarantee the sustainability of this practice.An approach based on geochemical processes occurring in the vadose zone from the surface down to the groundwater level was chosen, at the scale of cultivated plots and at the regional scale of Dakar periurban areas, to determine the influence of land use.The assessment of irrigation with untreated domestic wastewater and brackish water on soil quality (chapter 2) showed: (1) that the high alkalinity and calcium contents of brackish water induce CaC03 precipitation in the top layer of the soil and therefore a replacement of Ca by Na on the clay- humic complexes, strongly marked during the dry season. Dissolved organic carbon (DOC) increased significantly in the soil solution and in the underlying groundwater. (2) in spite of the similarly high alkalinity and Ca contents of waste water, there is only little CaC03 precipitation and a low increase of the percentage of exchangeable sodium (ESP) in the soil top layer. The nitrification of the ammonium of wastewater (mean 190 mg/L in Pikine) produces protons, which are not any more buffered by bicarbonates exported out of the soil profile, which leads to a net decline of soil pH. Both soils irrigated with untreated wastewater and brackish water store less of C and Ν than soils irrigated with non saline groundwater.The assessment of the impact of land use on the shallow groundwater (chapter 3) allowed determining representative tracers of the land use. Low Br/CI ratio, high NO3/CI ratio and low δ1βΟ- nitrate indicated the influence of wastewater; high pH and high 515N-nitrates indicated the influence of brackish water together with high amendments of organic fertilizers; high Rb+Cr and Κ indicated the influence of poor sanitation facilities in periurban districts (septic tank leakage). This chapter also pointed out the following facts about the nitrogen dynamics : (1) denitrification is a key-process in the Dakar UA agrosystem in the gleysols irrigated with groundwater. The underlying groundwater is almost nitrate free. (2) in the Gleysols irrigated with waste water, ammonium inhibits denitrification but facilitate ammoniac volatilization. A comparison of nitrogen balance between the UA agrosystem and the periurban districts of Dakar (chapter 4) revealed similar flows per surface unit, namely 2-4 tons Ν / ha / year.The evaluation of pesticides use in the UA agrosystem and the risk assessment for the groundwater (chapter 5) revealed a total flow of pesticides of 60kg / ha / year, totalizing 15 active substances. Only two of these are authorized by the Sahelian Pesticides Committee. The most used pesticides are dicofol (organochlorinated), methamidophos, dimethoate and fenithrotion (organophosphate) as well as methomyl. (carbamate). The most important flows vary between 9 to 7 kg / ha / year. Pesticides with a high risk of groundwater contamination - according to SIRIS and EPRIP 2 indicators - are: carbofuran, dimethoate, ethoprophos and methomyl. These substances should be established as a priority for an analytical follow-up in the different environmental compartments.In conclusion, a better management of the fertilization is necessary in the Dakar UA, (1) to reduce the gaseous losses which contribute to greenhouse emissions (2) to slow down the mineralization of the organic carbon and the nitrogen, in order to enhance the C and Ν stock in these soils, (3) to limit the nitrate leaching in the groundwater and finally, 4) to increase the N-use efficiency of plants. An optimization of the irrigation scheme should limit the secondary sodisation if coupled with an increase the stable organic matter of the soil. An integrated or biologic crop pest strategy is urgently needed to minimize risks with respect to ground and surface water (ponds used for fishing).RESUME LARGE PUBLICL'agriculture mondiale connaît actuellement une crise majeure, affectée par les changements climatiques, la sécurité alimentaire et les dégradations de l'environnement. Elle n'a plus le rôle unique de produire, mais devient un élément essentiel de la protection des ressources naturelles et du paysage. Les politiques agricoles basées sur les marchés mondiaux devront se réorienter vers une agriculture locale basée sur le développement durable.La production alimentaire située dans l'enceinte des villes, nommée agriculture urbaine ou périurbaine (AU ci-après) joue un rôle important dans le contexte actuel d'accroissement de la population et de la pauvreté urbaines. L'AU concerne en effet la majorité des mégapoies du monde, fait vivre plus de 200 millions de personnes dans les pays du Sud, fournit jusqu'à 80% de la demande urbaine en certains produits frais, fait barrière à l'extension urbaine et permet un recyclage de certains déchets urbains. L'AU a pour particularité d'être à cheval entre des politiques rurales et urbaines, d'où un délaissement ce cette activité au secteur informel. Ce qui a développé de nombreuses stratégies à risques, comme à Dakar, où les petits producteurs périurbains irriguent quotidiennement avec des eaux usées domestiques par manque d'accès à une eau de bonne qualité et pour raccourcir les cycles de production. L'extrême précarité foncière des acteurs de l'AU de Dakar les empêchent d'investir à long terme et induit des pratiques inadéquates d'irrigation, d'usage de pesticides et de fertilisation de ces sols sableux.L'objectif de cette recherche était de contribuer à une meilleure connaissance des processus de transfert de polluants et du cycle des nutriments à l'échelle des parcelles cultivées par des eaux usées et des eaux saumâtres, afin de déterminer sous quelles conditions l'AU de Dakar peut être pratiquée et surtout maintenue sans porter atteinte à la santé et à l'environnement. Pour cela, une approche basée sur l'étude des processus géochimiques dans le sol jusqu'à la nappe a été choisie, à l'échelle de la parcelle cultivée et à une échelle un peu plus large de la zone périurbaine de Dakar pour déterminer les influences du type d'occupation du sol.Les résultats principaux de cette étude ont montré que (1) il y a un processus de salinisation anthropique des sols (sodisation) lors d'irrigation avec des eaux de nappe saumâtres, un processus accentué en saison sèche et lors d'années à pluviométrie déficitaire. Bien que les eaux usées soient aussi salines que les eaux de nappe, la salinisation des sols irrigués' par des eaux usées est limitée par l'ammonium présent dans les eaux usées (moy 190mg NH4/L à Pikine) qui produit de l'acidité lors de la transformation en nitrates dans le sol (nitrification). (2) les sols irrigués par des eaux usées (EU) stockent moins de C et Ν que les sois de référence, ce qui montrent bien que l'azote des eaux usées n'est pas disponible pour les plantes, mais est lessivé dans la nappe (100 à 450 mg/L N03 sous irrigation par EU, alors que la limite de OMS est de 50mg/L). (3) l'utilisation des isotopes stables des nitrates et des éléments traces, notamment le bore et le brome, ont permis de distinguer l'influence de l'irrigation par des eaux usées, de l'irrigation par des eaux de nappe et des lixiviats de fosses septiques sur les propriétés de la nappe. (4) Le processus de la dénitrification (atténuation naturelle des concentrations en nitrates de la nappe par biotransformation en azote gazeux) est important dans les zones basses de l'agrosystème périurbain de Dakar, sous irrigation par eaux naturelles (ΝΟ3 < 50mg/L). Tandis que sous habitat sans assainissement adéquat, les nitrates atteignent 300 à 700 mg/L. (5) Le flux total de pesticides dans l'AU est énorme (60kg/ha/an) totalisant 15 pesticides, dont deux seulement sont autorisés. Les pesticides les plus utilisés sont des insecticides organophosphorés et organochlorés classés extrêmement dangereux à dangereux par l'OMS, appliqués à des doses de 2 à 9 kg/ha/an. Les pesticides qui ont montré un risque élevé de contamination des eaux souterraines avec les indicateurs SIRIS et EPRIP2 sont : le carbofuran, le dimethoate, l'ethoprophos et le methomyl.En conclusion, nous recommandons la reconstitution d'un horizon superficiel des sols riche en matière organique stable et structuré par production locale de compost. Cette mesure réduira les pertes gazeuses contribuant à l'effet de serre, augmentera le stock de Ν dans ces sols, alors utilisable par les plantes et permettra de diminuer l'irrigation car la capacité de rétention de l'eau dans le sol sera accru, ce qui limitera le lessivage des nitrates dans la nappe et l'alcalinisation secondaire. Enfin, la mise en place d'une lutte intégrée ou biologique contre les ravageurs est indispensable afin de minimiser les risques pour les eaux souterraines et lesmares permanentes.

ABSTRACT Soil solution samplers may have the same working principle, but they differ in relation to chemical and physical characteristics, cost and handling, and these aspects exert influence on the chemical composition of the soil solution obtained. This study was carried out to evaluate, over time, the chemical composition of solutions extracted by Suolo Acqua, with the hydrophilic membrane (HM) as a standard, using soils with contrasting characteristics, and to determine the relationship between electrical conductivity (EC) and concentration of ions and pH of soil solution samples. This study was carried out under laboratory conditions, using three soils samples with different clay and organic matter (OM) contents. Soil solution contents of F−, Cl−, NO−3, Br−, SO42−, Na+, NH4+, K+, Mg2+, Ca2+, were analyzed, as well as inorganic, organic, and total C contents, pH, and EC, in four successive sampling times. Soil solution chemical composition extracted by the Suolo Acqua sampler is similar to that collected by the HM, but the Suolo Acqua extracted more Na+ and soluble organic C than the HM solution. Solution EC, cation and anion concentrations, and soluble C levels are higher in the soil with greater clay and OM contents (Latossolo and Cambissolo in this case). Soil solution composition varied over time, with considerable changes in pH, EC, and nutrient concentrations, especially associated with soil OM. Thus, single and isolated sampling of the soil solution must be avoided, otherwise composition of the soil solution may not be correctly evaluated. Soil solution EC was regulated by pH, as well as the sum of cation and anion concentrations, and the C contents determined in the soil liquid phase.

Oxalic and oxamic acids are the ultimate and more persistent by-products of the degradation of N-aromatics by electrochemical advanced oxidation processes (EAOPs). In this paper, the kinetics and oxidative paths of these acids have been studied for several EAOPs using a boron-doped diamond (BDD) anode and a stainless steel or an air-diffusion cathode. Anodic oxidation (AO-BDD) in the presence of Fe2+ (AO-BDD-Fe2+) and under UVA irradiation (AO-BDD-Fe2+-UVA), along with electro-Fenton (EF-BDD), was tested. The oxidation of both acids and their iron complexes on BDD was clarified by cyclic voltammetry. AO-BDD allowed the overall mineralization of oxalic acid, but oxamic acid was removed much more slowly. Each acid underwent a similar decay in AO-BDD-Fe2+ and EFBDD, as expected if its iron complexes were not attacked by hydroxyl radicals in the bulk. The faster and total mineralization of both acids was achieved in AO-BDD-Fe2+-UVA due to the high photoactivity of their Fe(III) complexes that were continuously regenerated by oxidation of their Fe(II) complexes. Oxamic acid always released a larger proportion of NH4 + than NO3- ion, as well as volatile NOx species. Both acids were independently oxidized at the anode in AO-BDD, but in AO-BDD-Fe2+-UVA oxamic acid was more slowlydegraded as its content decreased, without significant effect on oxalic acid decay. The increase in current density enhanced the oxidation power of the latter method, with loss of efficiency. High Fe2+ contents inhibited the oxidation of Fe(II) complexes by the competitive oxidation of Fe2+ to Fe3+. Low current densities and Fe2+ contents are preferable to remove more efficiently these acids by the most potent AO-BDD-Fe2+-UVA method.

Thermal analysis, powder diffraction, and Raman scattering as a function of the temperature were carried out on K2BeF4. Moreover, the crystal structure was determined at 293 K from powder diffraction. The compound shows a transition from Pna21 to Pnam space group at 921 K with a transition enthalpy of 5 kJ/mol. The transition is assumed to be first order because the compound shows metastability. Structurally and spectroscopically the transition is similar to those observed in (NH4)2SO4, which suggests that the low-temperature phase is ferroelectric. In order to confirm it, the spontaneous polarization has been computed using an ionic model.

An in situ energy budget of the hydropolyp Eudendrium racemosum (Cavolini, 1785) is presented. Ingestion and respiration rates and ammonium excretion were studied over two 24 h cycles, with two-hour sample intervals. The species ingested as much as 25.9% of its own biomass per day (minimum rate). Respiration was 1.62 ml O2 g-1 d w h-1 while excretion was 13.6 mM NH4 g-1dw h-1. We estimated that the species increased its biomass at a rate of 9.6% per day (Growth + Reproduction). This value is higher than those previously reported for other cnidarians. We can assume that the capacity of E. racemosum to survive - albeit for a limited period of the year - in the highly-competitive shallow-water communities is based on its high growth rate.

We examined the effects of riparian vegetation removal on algal dynamics and stream nutrient retention efficiency by comparing NH4-N and PO4-P uptake lengths from a logged and an unlogged reach in Riera Major, a forested Mediterranean stream in northeastern Spain. From June to September 1995, we executed 6 short-term additions of N (as NH4Cl) and P (as Na2HPO4) in a 200-m section to measure nutrient uptake lengths. The study site included 2 clearly differentiated reaches in terms of canopy cover by riparian trees: the first 100 m were completely logged (i.e., the logged reach) and the remaining 100 m were left intact (i.e., the shaded reach). Trees were removed from the banks of the logged reach in the winter previous to our sampling. In the shaded reach, riparian vegetation was dominated by alders (Alnus glutinosa). The study was conducted during summer and fall months when differences in light availability between the 2 reaches were greatest because of forest canopy conditions. Algal biomass and % of stream surface covered by algae were higher in the logged than in the shaded reach, indicating that logging had a stimulatory effect on algae in the stream. Overall, nutrient retention efficiency was higher (i.e., shorter uptake lengths) in the logged than in the shaded reach, especially for PO4-P. Despite a greater increase in PO4-P retention efficiency relative to that of NH4-N following logging, retention efficiency for NH4-N was higher than for PO4-P in both study reaches. The PO4-P mass-transfer coefficient was correlated with primary production in both study reaches, indicating that algal activity plays an important role in controlling PO4-P dynamics in this stream. In contrast, the NH4-N mass-transfer coefficient showed a positive relation-ship only with % of algal coverage in the logged reach, and was not correlated with any algal-related parameter in the shaded reach. The lack of correlation with algal production suggests that mechanisms other than algal activity (i.e., microbial heterotrophic processes or abiotic mechanisms) may also influence NH4-N retention in this stream. Overall, this study shows that logging disturbances in small shaded streams may alter in-stream ecological features that lead to changes in stream nutrient retention efficiency. Moreover, it emphasizes that alteration of the tight linkage between the stream channel and the adjacent riparian zone may directly and indirectly impact biogeochemical processes with implications for stream ecosystem functioning.

Paspalum vaginatum Schwartz plants were grown under greenhouse conditions in a continuous-flow hydroponic culture, containing NO3- or NH4+or NH4NO3 as nitrogen source. After 30 days, the size of aerial biomass and root system decreased significantly when plants were supplied with NH4+as exclusive nitrogen source. Compared to NO3- treatment, reducing and non-reducing sugars were decreasing together with a significant increase in amino acids content. NH4+-nutrition caused tillers to grow toward an orthogravitropic position (average angle of 68° with respect to the horizontal), and with NO3--nutrition, tillers tended to become diagravitropic (average angle of 23°). With NH4NO3 all the parameters measured had values in between those of the other two sources. Thus, the morphologic differences among plants growing in NO3- or NH4+ nutrition confirm the hypothesis that nitrogen source determines the growth habit of tillers in P. vaginatum by modulating the endogenous levels of reducing-non-reducing sugars.

O presente estudo, desenvolvido em laboratório no perÃodo de outubro de 1995 a janeiro de 1996, objetivou avaliar a influência da calagem e fósforo (P) sobre a mineralização de nitrogênio (N) e enxofre (S) em sete solos, com ampla variabilidade nas caracterÃsticas quÃmicas e fÃsicas. Os solos (200 g), numa etapa anterior à aplicação de P (KH2PO4 p.a.), foram pré-incubados por sete dias com CaCO3p.a.Aseguir,foramincubadosnovamente,pormais70dias.OsteoresdeN (NO3- + NH4+) e S (SO4(2-)) mineralizados em condições aeróbias foram avaliados a cada catorze dias, e notou-se neles uma dinâmica diferenciada, com o N mineral apresentando maior variação do que o sulfato. As quantidades de N mineralizado não se mostraram dependentes da acidez do solo, porém notou-se uma aceleração na mineralização do N com a calagem do Glei Húmico e do Latossolo Vermelho-Amarelo coletado no Rio de Janeiro. Ao contrário, a mineralização de S no Glei Pouco Húmico e no Latossolo Vermelho-Amarelo coletado no Rio de Janeiro foi influenciada pela correção da acidez do solo, que promoveu maior disponibilidade de sulfato nesses solos. Os teores de N e S mineralizados não foram influenciados pelos nÃveis de P disponÃvel. A maior prevalência de nitrato e sulfato em alguns dos solos calcariados implica maior cuidado no estabelecimento da época de aplicação de corretivo.

O objetivo deste trabalho foi determinar as melhores condições de crescimento e esporulação de dois isolados de Verticillium lecanii (Zimm.) Viégas, cultivados em vários meios de cultura, fontes de C, fontes orgânicas e inorgânicas de N e relações C:N. Após 20 dias de cultivo, BDA e Meio Completo mostraram ser os melhores meios de cultura, e a lactose e o amido foram as fontes de C que mais favoreceram o crescimento dos isolados JAB 02 e JAB 45, respectivamente; baixa esporulação foi apresentada por JAB 02, enquanto JAB 45 obteve boa esporulação em glicose e maltose. JAB 02 desenvolveu melhor quando a fonte orgânica de N era casitona; JAB 45 obteve bom desenvolvimento em presença de casitona e peptona, mas houve pouca esporulação em presença de triptona e caseÃna hidrolisada. As fontes inorgânicas de N que favoreceram o melhor desenvolvimento dos isolados foram (NH4)2HPO4 e NaNO3, porém, JAB 02 apresentou baixa esporulação. Entre as relações C:N analisadas, JAB 02 obteve melhor crescimento na relação 60:1 e JAB 45 nas relações 5:1, 10:1 e 20:1; baixa esporulação foi obtida por JAB 02 nas relações testadas, mas não se verificou efeito da relação C:N do meio sobre a esporulação dos isolados.

The objective of this work was to study the effects of fire on net N mineralization and soil microbial biomass in burned and unburned cerrado stricto sensu sites. The study was carried out from April 1998 to April 2000. The pH values were significantly higher in the burned site while soil moisture content was significantly higher in the unburned site (P<0.05). The soil C/N ratio was 22/1 and the available NO3-N ranged between 1.5 and 2.8 mg kg-¹ dry weight. However, the NH4-N concentration ranged between 3 and 34 mg kg-1 dry weight in the burned site and between 3 and 22 mg kg-1 dry weight in the unburned site. The NH4-N increased after fire, but no significant changes were observed for NO3-N (P<0.05). The NO3-N accumulation occurred in short periods during the rainy season. The rates of net N mineralization increased during the rainy season while reductions in soil microbial biomass were observed at both sites. This suggested that the peak in microbial activities occurred with the first rain events, with an initial net immobilization followed by net mineralization. Both sites presented the same pattern for mineralization/immobilization, however, the amount of inorganic-N cycled annually in unburned site was 14.7 kg ha-1 per year while the burned site presented only 3.8 kg ha-¹ of inorganic-N, one year after the burning.

Bactérias fixadoras de nitrogênio podem contribuir para o crescimento vegetal pela produção de auxinas. Os objetivos deste trabalho foram quantificar a produção de hormônios de crescimento por estirpes de bactérias fixadoras de nitrogênio e avaliar o efeito da inoculação destas estirpes em plântulas de milho e trigo. Todas as estirpes avaliadas começaram a produzir indóis no final da fase logarÃtmica. Houve efeito da adição de diferentes concentrações de triptofano ao meio de cultivo no aumento da produção de indóis até o nÃvel de 200 µM. Nas estirpes de Azospirillum, as formas de N: NH4Cl, (NH4)2SO4, NH4H2PO4 e NH4NO3 estimularam a produção de indóis em nÃveis baixos. A adição de KNO3, NaNO3 e KNO2 inibiu a produção de indóis em todas as bactérias testadas e no crescimento de células de Azospirillum. Os efeitos da inoculação foram também comparados com concentrações crescentes do ácido 3-indolacético, KNO3 e triptofano. Em condições axênicas, a elevada produção de indóis reduz o comprimento das raÃzes e colmos na presença de triptofano, especialmente quando submetidas à inoculação de Azospirillum.

O objetivo deste trabalho foi avaliar o efeito do alumÃnio sobre a capacidade das cultivares de arroz Fernandes (tolerante) e Maravilha (sensÃvel) de modificar o pH de soluções nutritivas com diferentes proporções de NO3-/NH4+ . Após exposição das plantas ao Al (0 mM e 1,5 mM), durante 15 dias, foram determinados: quantidade de prótons exsudados, alongamento, produção de matéria seca e teor de Al nas raÃzes e folhas. O alongamento e produção de matéria seca, de raÃzes e folhas, se reduziram em presença de Al, independentemente da proporção NO3-/NH4+ , e essas reduções se intensificaram com o aumento na concentração de NH4+. Os teores de Al aumentaram nas raÃzes e folhas, com aumento na concentração de NH4+ na solução nutritiva. Houve forte alcalinização da solução nutritiva com nitrogênio exclusivamente na forma de NO3-, que se reduziu em presença de Al. Em soluções nutritivas com NH4+, ao contrário, observou-se acidificação, que se intensificou com aumento na concentração de NH4+. A cultivar tolerante sempre consumiu mais prótons da solução nutritiva com N exclusivamente na forma de NO3-, e menos prótons daquela que continha NH4+, independentemente da proporção NO3-/NH4+ . Essa cultivar foi, portanto, capaz de ajustar seu balanço de prótons e modificou o pH para valores que favoreciam menor absorção e maior tolerância ao alumÃnio.

O objetivo deste trabalho foi avaliar a sensibilidade de plantas transgênicas de citrumelo Swingle com elevada produção de prolina, ao herbicida glufosinato de amônio. As plantas utilizadas apresentavam a inserção do gene mutante da enzima delta1-pirrolina-5-carboxilato sintetase (P5CS), responsável pela biossÃntese de prolina. A expressão do gene p5cs em plantas transgênicas causou aumento nas quantidades de prolina em tecidos foliares, em até cinco vezes, quando comparadas à s plantas-controle tratadas com 200 µM de glufosinato de amônio. As plantas transgênicas acumularam maior quantidade de NH4+ nas folhas, em relação à s plantas não-transgênicas. Os danos causados pelo herbicida foram avaliados in vitro, utilizando-se discos foliares cultivados em meio MS com diferentes concentrações de glufosinato de amônio. Observou-se maior clorose em discos foliares das plantas transgênicas, o que comprova a maior suscetibilidade de plantas de citrumelo Swingle com alta produção de prolina ao herbicida.

O objetivo deste trabalho foi ativar, extrair e caracterizar parcialmente a fitase em sementes germinadas de girassol (Helianthus annuus L.), hÃbrido M734, e avaliar o efeito da fitase no farelo de girassol. Sementes foram colocadas para germinar por oito dias em câmara a 25°C. A fitase foi extraÃda com CaCl2 2%, depois do quinto dia de germinação, e fracionada com (NH4)2SO4 até 80% de saturação. O extrato bruto foi caracterizado parcialmente e aplicado em farelo de girassol desengordurado para avaliar a hidrólise do fitato. Com a germinação, houve aumento na atividade da fitase e redução no teor de fitato. A maior atividade da fitase foi observada do quinto ao oitavo dia de germinação. A fitase das sementes germinadas aos cinco dias apresentou atividade ótima em pH de 5,2 e temperatura de 55ºC. A enzima se manteve estável, quando pré-aquecida por 10 minutos a 50ºC, com Vmáx de 1,87 U g-1 de amostra e Km de 0,29 mM, indicando alta especificidade pelo fitato. Quando aplicada no farelo de girassol desengordurado, depois de oito horas de incubação, a fitase hidrolisou 92% do fitato. A germinação de sementes de girassol estimula a atividade da fitase, o que facilita sua extração para a produção de alimentos livres de fitatos.
