BackgroundThere are no published reports on studies comparing itraconazole (ITC), sulfamethoxazole-trimethoprim (cotrimoxazole, CMX), and ITC followed by CMX (ITC/CMX) in the treatment of paracoccidiodomycosis. This study aimed to compare the efficacy, effectiveness, safety and time to clinical and serologic cure in paracoccidioidomycosis patients treated with ITC or CMX, the antifungal agents most widely used.MethodologyA quasi-experimental study was performed in 177 patients with a confirmed or probable diagnosis of paracoccidioidomycosis. Treatment was divided into two stages: 1) initial, which was continued until clinical cure was achieved and the erythrocyte sedimentation rate decreased to normal values; 2) complementary, which was continued until serologic cure was achieved. Medians were compared via the Mann-Whitney test, and frequencies were compared via the chi-squared test. The assessment of variables as a function of time was performed using Kaplan-Meier curves and Cox regression. The significance level was established as p <= 0.05.Principal FindingsNo difference was found in the efficacy and effectiveness of the initial treatment of 47 individuals given ITC and 130 individuals given CMX; however, the time to clinical cure was shorter in the former compared with the latter group (105 vs. 159 days; p = 0.001), specifically in patients with the chronic form. Efficacy and effectiveness of the three regimens were similar in the complementary treatment; however, the time to serologic cure was shorter when ITC (161 days) or CMX (495 days) was used compared with ITC/CMX (881 days) [p = 0.02]. The independent predictors of a shorter time to serologic cure were treatment with ITC [risk ratio = 6.61 (2.01-21.75)] or with CMX [risk ratio = 5.11 (1.91-13.67)]). The prevalence of side effects was lower with ITC (6.4%) than with CMX (20.0%; p = 0.03).ConclusionsSince ITC induced earlier clinical cure and was better tolerated than CMX, such triazole should be considered the first-choice for PCM treatment.

The main objective of this article is to suggest a conceptual taxonomy of thegenerically called International Technical Cooperation (ITC) and Foreign Financial Aid (FFA) phenomena, which will have the pretension to ease their comprehension and their practical consequences for the Brazilian society and for the national State. Insofar, wheninduced by a dynamical structural change in the international society, some Brazilianfederated sectors, specially, municipalities, by the exercise of so-called paradiplomacy or federated diplomacy, are developing public management instruments to acquire resources through ITC and FFA experiences, without making a reflexive thought about their benefits and their consequences.

The incipient but quickly expansion action on the Information and Communications Technologies (ICT) in Africa it is now just having different impact on these societies. One of these relates bear on how users are identified with these tools. Just like that we find individuals identify as bloggers, twitter followers or cyber activist. This contribution analyzes the Senegal’s fact where a successful use of social nets and web 2.0 tools experience (at least in repercussion) as social and political involvement while presidential elections in 2012 is tied to come back an identity: Cyber activist. Senegalese circumstance shows us how this identity has a personal and assertiveness dimension as well collective aspects of belonging to a community. One as much as the other, show us personal traits in contrast to previous beliefs, basically because it fuse and confuse virtual and reality. Due to dynamics from expanding technology, this identity is youthful and urban, but not only. This situation creates new dynamics at least in this affected group. For this reason, besides knowing emergence and evolution of this fact, it raises some of the involvement in social and political involvement from groups traditionally “invisible”. Beyond the new social behavior there are new changes in the rules of the game in order to start new social revolution.

ELECTROCHEMICAL AND CALORIMETRIC INVESTIGATION OF INTERACTION OF NOVEL BISCATIONIC ANTICANCER AGENTS WITH DNA. Biscationic amidines bind in the DNA minor groove and present biological activity against a range of infectious diseases. Two new biscationic compounds (bis-alpha,omega-S-thioureido, amino and sulfide analogues) were synthesized in good yields and fully characterized, and their interaction with DNA was also investigated. Isothermal titration calorimetry (ITC) was used to measure the thermodynamic properties of binding interactions between DNA and these ligands. A double stranded calf thymus DNA immobilized on an electrode surface was used to study the possible DNA-interacting abilities of these compounds towards dsDNA in situ. A remarkable interaction of these compounds with DNA was demonstrated and their potential application as anticancer agents was furthered.

Transthyretin (TTR) is a carrier protein involved in human amyloidosis. The development of small molecules that may act as TTR amyloid inhibitors is a promising strategy to treat these pathologies. Here we selected and characterized the interaction of flavonoids with the wild type and the V30M amyloidogenic mutant TTR. TTR acid aggregation was evaluated in vitro in the presence of the different flavonoids. The best TTR aggregation inhibitors were studied by Isothermal Titration Calorimetry (ITC) in order to reveal their thermodynamic signature of binding to TTRwt. Crystal structures of TTRwt in complex with the top binders were also obtained, enabling us to in depth inspect TTR interactions with these flavonoids. The results indicate that changing the number and position of hydroxyl groups attached to the flavonoid core strongly influence flavonoid recognition by TTR, either by changing ligand affinity or its mechanism of interaction with the two sites of TTR. We also compared the results obtained for ITRwt with the V30M mutant structure in the apo form, allowing us to pinpoint structural features that may facilitate or hamper ligand binding to the V30M mutant. Our data show that the TTRwt binding site is labile and, in particular, the central region of the cavity is sensible for the small differences in the ligands tested and can be influenced by the Met30 amyloidogenic mutation, therefore playing important roles in flavonoid binding affinity, mechanism and mutant protein ligand binding specificities. Protective immunity against Plasmodium falciparum may be obtained after repeated exposure to infection. Several studies indicate that immunity against the blood stages of the P. Falciparum infection is mainly antibody mediated. Protective antibodies may act either on their own, mediate antibody-dependent phagocytosis and/or cell-mediated neutralization of parasites. This thesis describes several aspects of humoral immune responses to P. falciparum infection in individuals of different age groups, different genetic background and with different degrees of malaria exposure. Several target antigens for antibody-mediated inhibition of parasite growth or invasion have been identified. One such antigen is Pf332, which appears on the surface of parasitized erythrocytes at late trophozoite and schizont stage. This surface exposure makes the antigen a possible target for opsonizing antibodies. We optimized an in vitro assay for studying cellmediated parasite neutralization in the presence of Pf332-reactive antibodies. Our data demonstrate that, Pf332 specific antibodies are able to inhibit parasite growth on their own and in cooperation with human monocytes. The P. falciparum parasites have evolved several mechanisms to evade the host neutralizing immune responses. In this thesis, we show that freshly isolated P. falciparum parasites from children living in a malaria endemic area of Burkina Faso were less sensitive for growth inhibition in vitro by autologous immunoglobulins (Ig) compared with heterologous ones. Analyses of two consecutive isolates taken 14 days apart, with regard to genotypes and sensitivity to growth inhibition in vitro, did not give any clear-cut indications on possible mechanisms leading to a reduced inhibitory activity in autologous parasite/antibody combinations. The frequent presence of persisting parasite clones in asymptomatic children indicates that the parasite possesses as yet undefined mechanisms to evade neutralizing immune responses. Transmission reducing measures such insecticide treated nets (ITNs) have been shown to be effective in reducing morbidity and mortality from malaria. However, concerns have been raised that ITNs usage could affect the acquisition of malaria immunity. We studied the effect of the use of insecticide treated curtains (ITC) on anti-malarial immune responses of children living in villages with ITC since birth. The use of ITC did neither affect the levels of parasite neutralizing immune responses nor the multiplicity of infection. These results indicate that the use of ITC does not interfere with the acquisition of anti-malarial immunity in children living in a malaria hyperendemic area. There is substantial evidence that the African Fulani tribe is markedly less susceptible to malaria infection compared to other sympatrically living ethnic tribes. We investigated the isotypic humoral responses against P. falciparum asexual blood stages in different ethnic groups living in sympatry in two countries exhibiting different malaria transmission intensities, Burkina Faso and Mali. We observed higher levels of the total malaria-specific-IgG and its cytophilic subclasses in individuals of the Fulani tribe as compared to non-Fulani individuals. Fulani individuals also showed higher levels of antibodies to measles antigen, indicating that the intertribal differences are not specific for malaria and might reflect a generally activated immune system in the Fulani.

[ES] El empleo de la simulación software aplicada a la enseñanza en entornos académicos es una realidad desde hace algún tiempo. sin embargo, en lo que a formación y entrenamiento del personal clínico y estudiantes de medicina respecta, es un concepto innovador. En este artículo se presenta una breve descripción del estado de desarrollo e implantación actual de estas nuevas tecnologías para la formación médica. se expone el trabajo realizado entre el centro de tecnología Médica de la universidad de las Palmas de Gran canaria (ctMulPGc) y el Instituto tecnológico de canarias (Itc) para el desarrollo de un entorno de simulación virtual genérico para la formación y entrenamiento médico en técnicas mínimamente invasivas. también se describe la experiencia piloto realizada con los alumnos de la Facultad de ciencias de la salud de la universidad de las Palmas de Gran canaria.

[EN] The correct determination of the energy generated or absorbed in the sample cell of an Isothermal Titration Calorimeter (ITC) requires a thorough analysis of the calorimetric signal. This means the identification and quantification of any thermal effect inherent to the working method. In this work, it is carried out a review on several thermal effects, studied by us in previous work, and which appear when an ITC is used for measuring the heats of mixing of liquids in a continuous mode. These effects are due to: (i) the difference between the temperature of the injected liquid and the temperature of the mixture during the mixing process, (ii) the increase of the liquid volume located in the mixing cell and (iii) the stirring velocity. Besides, methods for the identification and quantification of the mentioned effects are suggested.

[ES] El Instituto Tecnológico de Canarias (ITC), como coordinador del Proyecto OMARCOST, junto a los socios Instituto Español de Oceanografía (IEO) y Gestión del Medio Rural de Canarias (GMR Canarias) han editado esta sencilla obra divulgativa que resume los principales resultados obtenidos en el marco de este Proyecto de Cooperación española-marroquí cofinanciado por el Programa de Cooperación Transfronteriza POCTEFEX. El libro resume más de dos años de investigaciones; concretamente explica los estudios realizados, las herramientas y metodologías novedosas aplicadas y la transferencia de conocimiento y divulgación realizada. Todo ello, ha beneficiado directamente en la implementación nacional y regional de las políticas europea y nacional en materia de aguas y protección del medio marino.

Although nickel is a toxic metal for living organisms in its soluble form, its importance in many biological processes recently emerged. In this view, the investigation of the nickel-dependent enzymes urease and [NiFe]-hydrogenase, especially the mechanism of nickel insertion into their active sites, represent two intriguing case studies to understand other analogous systems and therefore to lead to a comprehension of the nickel trafficking inside the cell. Moreover, these two enzymes have been demonstrated to ensure survival and colonization of the human pathogen H. pylori, the only known microorganism able to proliferate in the gastric niche. The right nickel delivering into the urease active site requires the presence of at least four accessory proteins, UreD, UreE, UreF and UreG. Similarly, analogous process is principally mediated by HypA and HypB proteins in the [NiFe]-hydrogenase system. Indeed, HpHypA and HpHypB also have been proposed to act in the activation of the urease enzyme from H. pylori, probably mobilizing nickel ions from HpHypA to the HpUreE-HpUreG complex. A complete comprehension of the interaction mechanism between the accessory proteins and the crosstalk between urease and hydrogenase accessory systems requires the determination of the role of each protein chaperone that strictly depends on their structural and biochemical properties. The availability of HpUreE, HpUreG and HpHypA proteins in a pure form is a pre-requisite to perform all the subsequent protein characterizations, thus their purification was the first aim of this work. Subsequently, the structural and biochemical properties of HpUreE were investigated using multi-angle and quasi-elastic light scattering, as well as NMR and circular dichroism spectroscopy. The thermodynamic parameters of Ni2+ and Zn2+ binding to HpUreE were principally established using isothermal titration calorimetry and the importance of key histidine residues in the process of binding metal ions was studied using site-directed mutagenesis. The molecular details of the HpUreE-HpUreG and HpUreE-HpHypA protein-protein assemblies were also elucidated. The interaction between HpUreE and HpUreG was investigated using ITC and NMR spectroscopy, and the influence of Ni2+ and Zn2+ metal ions on the stabilization of this association was established using native gel electrophoresis, light scattering and thermal denaturation scanning followed by CD spectroscopy. Preliminary HpUreE-HpHypA interaction studies were conducted using ITC. Finally, the possible structural architectures of the two protein-protein assemblies were rationalized using homology modeling and docking computational approaches. All the obtained data were interpreted in order to achieve a more exhaustive picture of the urease activation process, and the correlation with the accessory system of the hydrogenase enzyme, considering the specific role and activity of the involved protein players. A possible function for Zn2+ in the chaperone network involved in Ni2+ trafficking and urease activation is also envisaged.

In dieser Arbeit wurden isotherme Schnitte der ternären Systeme Ti-Fe-Sb, Zr-Fe-Sb und Nb-Fe-Sb bei 800 bzw. 600 °C untersucht. Die Bildung von vier von den Binärbereichen getrennten ternären Verbindungen im System Ti-Fe-Sb, drei im System Zr-Fe-Sb und einer Verbindung im System Nb-Fe-Sb wurde festgestellt bzw. bestätigt. In den ersten zwei Systemen ist die Bildung von festen Lösungen auf der Basis von binären sowie ternären Phasen stark ausgeprägt. Es wurde die Abhängigkeit des Strukturtyps der Laves-Phasen M(Fe???Sb?)??? (M = Ti, Zr, Nb) von der Elektronenkonzentration und den Atomradien der Komponenten gezeigt. 18 isotype Verbindungen M?Me’???X??? (M = Zr, Hf; M’ = Fe, Co, Ni; X = Sn, Sb, Bi) des geordneten Fe?P-Strukturtyps wurden synthetisiert. Die Untersuchungen der Transporteigenschaften dieser Verbindungen belegen deren metallischen Charakter. Es wurde die Bildung der neuen equiatomen Verbindungen in den Systemen Zr-Cu-Sn und Hf-Cu-Sn der Strukturtypen TiNiSi bzw. LiGaGe und der Verbindung HfFe???Sb des TiNiSi-Strukturtyps festgestellt. Die Transporteigenschaften der Reihe von festen Lösungen V???Ti?FeSb wurden untersucht. Es wurde gezeigt, dass die größte Erhöhung des Seebeck-Koeffizienten bei der kleinen Konzentration der vierten Komponente erreicht wird. Der höchste Wert des Seebeck-Koeffizienten (370 ?V/K bei 380 K) wurde für die Zusammensetzung V????Ti????FeSb festgestellt. Die Serie der quaternären Phasen Sc???Nb???NiSn, ZrNiIn???Sb???, HfNiIn???Sb???, ZrCo???Cu???Sn und HfCo???Cu???Sn. zeigt die Möglichkeit der Phasenbildung der Strukturtypen AlLiSi, LiGaGe bzw. TiNiSi auch im Fall der Abwesenheit einer oder beider ternärer Randverbindungen. Für die Verbindung Sc???Nb???NiSn wurden Halbleitereigenschaften festgestellt. Insgesamt wurde die Kristallstruktur der 25 neuen, zum ersten Mal synthetisierten ternären und quaternären Verbindungen bestimmt. In dieser Arbeit werden Quantum-Hydrodynamische (QHD) Modelle betrachtet, die ihren Einsatz besonders in der Modellierung von Halbleiterbauteilen finden. Das QHD Modell besteht aus den Erhaltungsgleichungen für die Teilchendichte, das Momentum und die Energiedichte, inklusive der Quanten-Korrekturen durch das Bohmsche Potential. Zu Beginn wird eine Übersicht über die bekannten Ergebnisse der QHD Modelle unter Vernachlässigung von Kollisionseffekten gegeben, die aus ein­em Schrödinger-System für den gemischten-Zustand oder aus der Wigner-Glei­chung hergeleitet werden können. Nach der Reformulierung der eindimensionalen QHD Gleichungen mit linearem Potential als stationäre Schrö­din­ger-Gleichung werden die semianalytischen Fassungen der QHD Gleichungen für die Gleichspannungs-Kurve betrachtet. Weiterhin werden die viskosen Stabilisierungen des QHD Modells be­rück­sich­tigt, sowie die von Gardner vorgeschlagene numerische Viskosität für das {sf upwind} Finite-Differenzen Schema berechnet. Im Weiteren wird das viskose QHD Modell aus der Wigner-Glei­chung mit Fokker-Planck Kollisions-Ope­ra­tor hergeleitet. Dieses Modell enthält die physikalische Viskosität, die durch den Kollision-Operator eingeführt wird. Die Existenz der Lösungen (mit strikt positiver Teilchendichte) für das isotherme, stationäre, eindimensionale, viskose Modell für allgemeine Daten und nichthomogene Randbedingungen wird gezeigt. Die dafür notwendigen Abschätzungen hängen von der Viskosität ab und erlauben daher den Grenzübergang zum nicht-viskosen Fall nicht. Numerische Simulationen der Resonanz-Tunneldiode modelliert mit dem nichtisothermen, stationären, eindimensionalen, viskosen QHD Modell zeigen den Einfluss der Viskosität auf die Lösung. Unter Verwendung des von Degond und Ringhofer entwickelten Quanten-Entropie-Minimierungs-Verfahren werden die allgemeinen QHD-Gleichungen aus der Wigner-Boltzmann-Gleichung mit dem BGK-Kollisions-Operator hergeleitet. Die Herleitung basiert auf der vorsichtige Entwicklung des Quanten-Max­well­ians in Potenzen der skalierten Plankschen Konstante. Das so erhaltene Modell enthält auch vertex-Terme und dispersive Terme für die Ge­schwin­dig­keit. Dadurch bleibt die Gleichspannungs-Kurve für die Re­so­nanz-Tunnel­diode unter Verwendung des allgemeinen QHD Modells in einer Dimension numerisch erhalten. Die Ergebnisse zeigen, dass der dispersive Ge­schwin­dig­keits-Term die Lösung des Systems stabilisiert.

Il cemento è il materiale da edilizia più comune al mondo e le problematiche ambientali associate all’utilizzo ed al ciclo produttivo sono numerose (rilascio di gas serra, scelta delle materie prime, risparmio energetico) e fortemente impattanti. Tuttavia, nonostante la sua diffusione sia globale e le proprietà siano note da millenni vi sono ancora alcuni processi che non sono stati correttamente spiegati. In particolare non esiste una teoria unica e consolidata che descriva efficacemente la microstruttura delle fasi minerali del cemento che si sviluppano successivamente all'idratazione, e come esso di conseguenza interagisca con principi attivi, organici o inorganici, di cui è noto l’effetto accelerante sulla cinetica di indurimento. Questa tesi si pone l'obiettivo di condurre uno studio dell'idratazione del clinker finemente macinato (CFM - principale componente di molti cementi) focalizzato sull’interazione tra elementi chimici presenti nella matrice cementizia (Calcio, Ferro e Alluminio) ed additivi acceleranti (Trietanolammina, Triisopropanolammina). Il fenomeno è stato studiato tramite osservazione dell'andamento del rilascio dei suddetti ioni su cementi in via di idratazione e tramite utilizzo di tecniche di microscopia elettronica e di diffrazione di raggi X. L'analisi del rilascio di ioni, effettuata mediante spettroscopia ad assorbimento atomico, è stata condotta al fine di approfondire l'azione di trietanolammina e triisopropanolammina sulle dinamiche di idratazione dei CFM nelle fasi precoci di idratazione, al fine di confrontare queste dinamiche con processi che avvengono in fasi successive e che coinvolgono la formazione delle principali fasi amorfe e cristalline responsabili delle proprietà del materiale. Da indagini di diffrattometria e microscopia sono inoltre state estrapolate interessanti considerazioni circa le cinetiche che portano alla formazione delle fasi cristalline tipiche dell'idratazione del cemento (portlandite, ettringite) in presenza e assenza di additivi. Nel corso dello studio sono stati sintetizzati alcuni composti alternativi agli additivi commerciali, sviluppati a partire da building blocks provenienti da fonti rinnovabili, portando quindi l'attenzione sulla sostenibilità del processo di progettazione delle molecole, anche alla luce della degradazione in ambiente delle sostanze sintetizzate. Tali molecole sono attualmente in fase di test presso i laboratori di Italcementi (ITC Group).
