O Bacilo de Calmette e Guérin (BCG) é utilizado em muitos países como profilaxia contra a tuberculose. Tem sido sugerido que esta vacina possa determinar, através de atividade imunomoduladora, proteção também contra outras patologias como alergia e doenças autoimunes. Entretanto, este assunto é ainda bastante controverso. A proposta desta investigação foi avaliar o efeito da imunização prévia com BCG no desenvolvimento do diabetes experimental do tipo I em camundongos C57BL/6. Para isto foram realizados três protocolos objetivando caracterizar a resposta imune desencadeada por BCG, padronizar o diabetes experimental tipo I e avaliar o efeito do BCG neste modelo de diabetes. A imunização desta cepa de camundongos com BCG determinou produção discreta de IFN- mas produção significativa de IL-10 após estímulo in vitro das culturas com hsp65 recombinante (rhsp65). Os ensaios de padronização indicaram que o protocolo de inoculação de streptozotocina adotado determinou um quadro de diabetes típico caracterizado por glicemia elevada e graus variáveis de insulite. A imunização com BCG antes da indução do diabetes não alterou de forma significativa o peso, os níveis glicêmicos e a intensidade do processo inflamatório. Os resultados obtidos sugerem que nas condições experimentais adotadas, o BCG não alterou o desenvolvimento do diabetes tipo I induzido por STZ em camundongos C57BL/6

Experimental models are necessary to elucidate pathophysiological mechanisms not yet understood in humans. To evaluate the repercussions of the diabetes, considering two methodologies, on the pregnancy of Wistar rats and on the development of their offspring. In the 1st induction, female offspring were distributed into two experimental groups: Group streptozotocin (STZ, n=67): received the β-cytotoxic agent (100mg STZ/kg body weight - sc) on the 1st day of the life; and Non-diabetic Group (ND, n=14): received the vehicle in a similar time period. In the adult life, the animals were mated. After a positive diagnosis of pregnancy (0), female rats from group STZ presenting with lower glycemia than 120 mg/dL received more 20 mg STZ/kg (ip) at day 7 of pregnancy (2nd induction). The female rats with glycemia higher than 120mg/dL were discarded because they reproduced results already found in the literature. In the mornings of days 0, 7, 14 and 21 of the pregnancy glycemia was determined. At day 21 of pregnancy (at term), the female rats were anesthetized and killed for maternal reproductive performance and fetal development analysis. The data were analyzed using Student-Newman-Keuls, Chi-square and Zero-inflated Poisson (ZIP) Tests (p<0.05). STZ rats presented with increased rates of pre (STZ=22.0%; ND=5.1%) and post-implantation losses (STZ=26.1%; ND=5.7%), reduced rates of fetuses with appropriate weight for gestational age (STZ=66%; ND=93%) and reduced degree of development (ossification sites). Conclusion: Mild diabetes led a negative impact on maternal reproductive performance and caused intrauterine growth restriction and impaired fetal development

The exercise-induced collapse (EIC) is a hereditary disease characterized by muscle weakness, impaired locomotion and collapse after intense exercise. This autossomic recessive disorder affects mainly Labrador Retriever presenting the mutation c.767G>T in the dynamin 1 (DNM1) gene. The objective of this study is to report the first case of exercise-induced collapse in Labrador Retriever in Brazil. The molecular test detected the specific genetic mutation and confirmed the clinical diagnosis in a Labrador Retriever with clinical history of weakness and collapse after exercise. It is important to include this disease as part of the differential diagnosis of neuromuscular diseases in Labrador Retriever and use the molecular test to guide matings.

The long-term effects of five different treatments of diabetes were evaluated in alloxan-induced diabetic rats. Seven experimental groups, with 50 rats each (GN--normal control; GD--untreated diabetic control; GI, GA, GIA--treated groups with insulin, acarbose, and insulin plus acarbose, respectively; GTIL, GTPD--treated groups with islet of Langerhans and pancreas transplantation) were studied. Clinical (body weight, water intake, food intake and urine output) and laboratory (blood and urinary glucose, and plasma insulin) parameters were analyzed at the beginning of the study, and after 1, 3, 6, 9 and 12 months of follow-up. Mortality was observed in all groups, except GN, during 12 months (GD = 50%; GI = 20%; GA = 26%; GIA = 18%; GTIL = 4%; GTPD = 20%). Rats from the GD, GI, and GIA groups died due to metabolic or hydrossaline disbalance, and/or pneumonia, diarrhoea, and cachexy. All deaths observed in GTIL and GTPD groups were in decorrence of technical failure at the immediate postoperative, until 72h. Animals from the GI, GA and GIA had significative improving of the clinical and laboratory parameters (p < 0,05) observed in diabetic rats, being the efficacy of theses treatments equal. However, rats from the GTIL and GTPD groups had better control of these parameters than GI, GA, and GIA groups. Transplanted rats had complete restoration, at the normal levels, of all analyzed variables (p < 0.01). Conventional treatments with insulin, acarbose, and insulin plus acarbose improved the severe diabetic state of the alloxan-diabetic rats, but pancreas and islet transplantation have a better performance for treatment of diabetes.

Once petroleum is na exhaustible source of energy, alternative fuels are having more prominence. A much discussed option for replacing fossil fuels is the use of biofuels derived from oils or fats, especially biodiesel. The biodiesel preparation is through a reaction named transesterification, a reaction of triglycerides with a short chain alcohol with a catalyst, producing a mixture of fatty acid esters and glycerol. According to ANP (National Petroleum Agency) specifications, biodiesel can have contaminants due to the catalyst or oil used on its synthesis, such as phosphorus, wich can damage the catalytic converter and cause significant increase in the particles emission. This project aims to develop na alternative method using chemically modified electrodes with iron nanoparticles for determination of phosphorus in biodiesel. For the formation of the iron nanoparticles film on the surface of a glassy carbon electrode, was used a iron sulfate solution. The film was formed after 10 successive cycles, with a scanning speed of 50 mV s-1 and a potential range of -0,9 to -1,25 V. To reduce possible oxides on the surface and activate the electrode, it has been subjected to a cathodic polarization with a potential of -1,25 V for 15 minutes in a sodium hydroxide solution. In cyclic voltammograms obtained in the study of the speed of scanning, there is an increase in the intensity of the anodic and cathodic current peaks. The cathodic peak current varied linearly with the square root of scan rate, showing that the electrode is controlled by diffusion. After successive additions of phosphate there is a linear variation in the current peak in the concentration range of 1,0 x 10-7 a 1,0 x 10-6 mol L-1. To determine if the concentration of phosphorus in real sample, the method of adding standard was used by adding aliquots of phosphate ions in the solution containing soy biodiesel extracted with ....

OBJETIVOS: Este estudo visa a analisar os efeitos, a longo prazo, de cinco diferentes tratamentos sobre o controle metabólico de ratos diabéticos aloxânicos. MÉTODOS: Foram analisados 7 grupos experimentais, com 50 ratos cada um, sendo: GN o grupo controle normal; GD o grupo controle diabético, sem tratamento; GI, GA e GIA os grupos tratados, respectivamente, com insulina, acarbose e associação insulina + acarbose; GTIL o grupo tratado com transplante de ilhotas de Langerhans; e o GTPD o grupo tratado com transplante pancreatoduodenal heterotópico. Parâmetros clínicos (peso, ingestão hídrica, ingestão alimentar e diurese) e laboratoriais (glicemia, glicose urinária e insulina plasmática) foram avaliados em todos os animais, no início do experimento, e após 1, 3, 6, 9 e 12 meses de seguimento. RESULTADOS: À exceção do GN, mortalidade foi observada em todos os grupos experimentais no seguimento de 12 meses (GD= 50%; GI= 20%; GA= 26%; GIA= 18%; GTIL= 4%; GTPD= 20%). em GD, GI, GA e GIA os óbitos ocorreram por distúrbios metabólicos ou hidroeletrolíticos e/ou pneumonia, diarréia e caquexia; em GTIL e GTPD todos os óbitos ocorreram por falhas técnicas no pós-operatório até 72h. Animais dos grupos GI, GA e GIA tiveram melhora significativa (p < 0,05) de todos os parâmetros clínicos e laboratoriais observados em ratos diabéticos, sem diferença de efetividade entre os tratamentos. Porém, os resultados observados nestes grupos, biologicamente não foram comparáveis aos observados em GTIL e GTPD, onde observou-se correção completa, aos níveis normais, de todas as variáveis analisadas (p<0,01). CONCLUSÕES: Os tratamentos convencionais com insulina, acarbose e insulina + acarbose melhoraram o estado diabético grave dos ratos tratados, contudo, a eficácia dos tratamentos foi significativamente inferior à oferecida pelo GTIL e GTPD.

Several nutritional interventions are performed in order to reduce the impact of EO induced by exercise. Some studies suggest that mate tea (CM) have compounds, which can act as antioxidants reducing EO. The aim of this study was to evaluate the effect of CM induced under the EO after a single bout of aerobic exercise. Methods: We used male Wistar rats (50 days and 200g) distributed in four experimental groups: control (CON); mate tea (CM), Swimming (N); Swimming+mate tea (N + CM) . The CM and N+CM groups received CM diluted in distilled water at 96 ° C for five days, oropharyngeal route (50 mg / kg body weight, 0.5 mL). Both groups were adapted to the aquatic environment prior to experimental day, after a single swimming session, with 5% of body weight attached to the tail, until the animals reached exhaustion. Immediately after the state of exhaustion, peripheral blood was collected for further analysis. Results: The animals of the group N + CM showed improved resistance swimming compared to group C (p < 0.0001). No increase in lipid oxidative damage and production of lactate in group N + CM , compared to group N , may be attributed to the significant increase in plasma uric acid concentrations demonstrated in this study was observed . Conclusion: Therefore, the results indicate that consumption of CM may be natural strategy for improving aerobic exercise endurance and reduce the impact of EO induced by aerobic exercise.
