Renal cell carcinoma (RCC), also known as kidney cancer, renal adenocarcinoma or hypernephroma, and metastatic renal cell carcinoma is a global burden. This article aims to provide a brief overview of RCC. It outlines epidemiology and presentation; invesitgation and staging; treatments and prognosis. The article also includes a focus on currently available drug treatments, and serves as an introduction to the topic.

Müllerian inhibiting substance (MIS), a member of the transforming growth factor-beta superfamily, induces regression of the Müllerian duct in male embryos. In this report, we demonstrate MIS type II receptor expression in normal breast tissue and in human breast cancer cell lines, breast fibroadenoma, and ductal adenocarcinomas. MIS inhibited the growth of both estrogen receptor (ER)-positive T47D and ER-negative MDA-MB-231 breast cancer cell lines, suggesting a broader range of target tissues for MIS action. Inhibition of growth was manifested by an increase in the fraction of cells in the G(1) phase of the cell cycle and induction of apoptosis. Treatment of breast cancer cells with MIS activated the NFkappaB pathway and selectively up-regulated the immediate early gene IEX-1S, which, when overexpressed, inhibited breast cancer cell growth. Dominant negative IkappaBalpha expression ablated both MIS-mediated induction of IEX-1S and inhibition of growth, indicating that activation of the NFkappaB signaling pathway was required for these processes. These results identify the NFkappaB-mediated signaling pathway and a target gene for MIS action and suggest a putative role for the MIS ligand and its downstream interactors in the treatment of ER-positive as well as negative breast cancers.

Clear cell renal cell carcinoma (ccRCC), a tubular epithelial cell (TEC) malignancy, frequently secretes tumor necrosis factor (TNF). TNF signals via two distinct receptors (TNFRs). TNFR1, expressed in normal kidney primarily on endothelial cells, activates apoptotic signaling kinase 1 and nuclear factor-kappaB (NF-kappaB) and induces cell death, whereas TNFR2, inducibly expressed on endothelial cells and on TECs by injury, activates endothelial/epithelial tyrosine kinase (Etk), which trans-activates vascular endothelial growth factor receptor 2 (VEGFR2) to promote cell proliferation. We investigated TNFR expression in clinical samples and function in short-term organ cultures of ccRCC tissue treated with wild-type TNF or specific muteins selective for TNFR1 (R1-TNF) or TNFR2 (R2-TNF). There is a significant increase in TNFR2 but not TNFR1 expression on malignant TECs that correlates with increasing malignant grade. In ccRCC organ cultures, R1-TNF increases TNFR1, activates apoptotic signaling kinase and NF-kappaB, and promotes apoptosis in malignant TECs. R2-TNF increases TNFR2, activates NF-kappaB, Etk, and VEGFR2 and increases entry into the cell cycle. Wild-type TNF induces both sets of responses. R2-TNF actions are blocked by pretreatment with a VEGFR2 kinase inhibitor. We conclude that TNF, acting through TNFR2, is an autocrine growth factor for ccRCC acting via Etk-VEGFR2 cross-talk, insights that may provide a more effective therapeutic approach to this disease.

Semliki Forest virus (SFV) vectors have been efficiently used for rapid high level expression of several G protein-coupled receptors. Here we describe the use of SFV vectors to express the alpha 1b-adrenergic receptor (AR) alone or in the presence of the G protein alpha q and/or beta 2 and gamma 2 subunits. Infection of baby hamster kidney (BHK) cells with recombinant SFV-alpha 1b-AR particles resulted in high specific binding activity of the alpha 1b-AR (24 pmol receptor/mg protein). Time-course studies indicated that the highest level of receptor expression was obtained 30 hours post-infection. The stimulation of BHK cells, with epinephrine led to a 5-fold increase in inositol phosphate (IP) accumulation, confirming the functional coupling of the receptor to G protein-mediated activation of phospholipase C. The SFV expression system represents a rapid and reproducible system to study the pharmacological properties and interactions of G protein coupled receptors and of G protein subunits.

Transitional-cell carcinoma of the renal pelvis or ureter is a relatively rare disease. Several risk factors are smoking, occupational carcinogens, analgesic abuse or Balkan nephropathy. The grade and stage of the disease have the most significant impact on the outcome. The treatment of renal pelvis and ureter tumours is open or laparoscopic surgery varying from conservative to more extensive surgical procedures, i.e. radical nephroureterectomy including removal of the contents of Gerota's fascia with ipsilateral ureter and a cuff of bladder at its distal extent. Most available data are from retrospective studies and surgery is the mainstay of treatment. Chemotherapy and/or radiation therapy are possible adjuvant or primary treatment for selected patients; however, prospective studies are needed to confirm their use.

The membrane organization of the alpha-subunit of purified (Na+ + K+)-ATPase ((Na+ + K+)-dependent adenosine triphosphate phosphorylase, EC 3.6.1.3) and of the microsomal enzyme of the kidney of the toad Bufo marinus was compared by using controlled trypsinolysis. With both enzyme preparations, digestions performed in the presence of Na+ yielded a 73 kDa fragment and in the presence of K+ a 56 kDa, a 40 kDa and small amounts of a 83 kDa fragment from the 96 kDa alpha-subunit. In contrast to mammalian preparations (Jørgensen, P.L. (1975) Biochim. Biophys. Acta 401, 399-415), trypsinolysis of the purified amphibian enzyme led to a biphasic loss of (Na+ + K+)-ATPase activity in the presence of both Na+ and K+. These data could be correlated with an early rapid cleavage of 3 kDa from the alpha-subunit in both ionic conditions and a slower degradation of the remaining 93 kDa polypeptide. On the other hand, in the microsomal enzyme, a 3 kDa shift of the alpha-subunit could only be produced in the presence of Na+. Our data indicate that (1) purification of the amphibian enzyme with detergent does not influence the overall topology of the alpha-subunit but produces a distinct structural alteration of its N-terminus and (2) the amphibian kidney enzyme responds to cations with similar conformational transitions as the mammalian kidney enzyme. In addition, anti alpha-serum used on digested enzyme samples revealed on immunoblots that the 40 kDa fragment was better recognized than the 56 kDa fragment. It is concluded that the NH2-terminal of the alpha-subunit contains more antigenic sites than the COOH-terminal domain in agreement with the results of Farley et al. (Farley, R.A., Ochoa, G.T. and Kudrow, A. (1986) Am. J. Physiol. 250, C896-C906).

The construction of adenovirus vectors for cloning and foreign gene expression requires packaging cell lines that can complement missing viral functions caused by sequence deletions and/or replacement with foreign DNA sequences. In this study, packaging cell lines were designed to provide in trans the missing bovine adenovirus functions, so that recombinant viruses could be generated. Fetal bovine kidney and lUng cells, acquired at the trimester term from a pregnant cow, were tranfected with both digested wild type BAV2 genomic DNA and pCMV-EI. The plasmid pCMV-EI was specifically constructed to express El of BAV2 under the control of the cytomegalovirus enhancer/promoter (CMV). Selection for "true" transformants by continuous passaging showed no success in isolating immortalised cells, since the cells underwent crisis resulting in complete cell death. Moreover, selection for G418 resistance, using the same cells, also did not result in the isolation of an immortalised cell line and the same culture-collapse event was observed. The lack of success in establishing an immortalised cell line from fetal tissue prompted us to transfect a pre-established cell line. We began by transfecting MDBK (Mardin-Dardy bovine kidney) cells with pCMV-El-neo, which contain the bacterial selectable marker neo gene. A series of MDBK-derived cell lines, that constitutively express bovine adenoviral (BAV) early region 1 (El), were then isolated. Cells selected for resistance to the drug G418 were isolated collectively for full characterisation to assess their suitability as packaging cell lines. Individual colonies were isolated by limiting dilution and further tested for El expression and efficiency of DNA uptake. Two cell lines, L-23 and L-24, out of 48 generated foci tested positive for £1 expression using Northern Blot analysis. DNA uptake studies, using both lipofectamine and calcium phosphate methods, were performed to compare these cells, their parental MDBK cells, 8 and the unrelated human 293 cells as a benchmark. The results revealed that the new MDBKderived clones were no more efficient than MDBK cells in the transient expression of transfected DNA and that they were inferior to 293 cells, when using lacZ as the reporter gene. In view of the inherently poor transfection efficiency of MDBK cells and their derivatives, a number of other bovine cells were investigated for their potential as packaging cells. The cell line CCL40 was chosen for its high efficiency in DNA uptake and subsequently transfected with the plasmid vector pCMV El-neo. By selection with the drug G418, two cell lines were isolated, ProCell 1 and ProCell 2. These cell lines were tested for El expression, permissivity to BAV2 and DNA uptake efficiency, revealing a DNA uptake efficiency of 37 % , comparable to that of CCL40. Attempts to rescue BAV2 mutants carrying the lacZ gene in place of £1 or £3 were carried out by co-transfecting wild type viral DNA with either the plasmid pdlElE-Z (which contains BAV2 sequences from 0% to 40.4% with the lacZ gene in place of the £1 region from 1.1% to 8.25%) or with the plasmid pdlE3-5-Z (which contains BAV2 sequences from 64.8% to 100% with the lacZ gene in place of the E3 region from 75.8% to 81.4%). These cotransfections did not result in the generation of a viral mutant. The lack of mutant generation was thought to be caused by the relative inefficiency ofDNA uptake. Consequently, cosBAV2, a cosmid vector carrying the BAV2 genome, was modified to carry the neo reporter gene in place of the £3 region from 75.8% to 81.4%. The use of a single cosmid vector earring the whole genome would eliminate the need for homologous recombination in order to generate a viral vector. Unfortunately, the transfection of cosBAV2- neo also did not result in the generation of a viral mutant. This may have been caused by the size of the £3 deletion, where excess sequences that are essential to the virus' survival might have been deleted. As an extension to this study, the spontaneous E3 deletion, accidently discovered in our viral stock, could be used as site of foreign gene insertion.

Human adenoviruses (Ads), members of the family adenoviridae, are medium-sized DNA viruses which have been used as valuable research tools for the study of RNA processing, oncogenic transformation, and for the development of viral vectors for use in gene delivery and immunization technology. The left 12% of the linear Ad genollle codes for products which are necessary for the efficient replication of the virus, as well as being responsible for the forlllation of tumors in animallllodels. The establishlllent of the 293 cell line, by immortalization of human embryonic kidney cells with th~ E1 region of Ad type S (AdS), has facilitated extensive manipulation of the Ads and the development of recombinant Ad vectors. The study of bovine adenoviruses (BAVs), which cause mild respiratory and gastrointestinal infections in cattle has, on the other hand, been limited primarily to that of infectivity, immunology and clinicallllanifestations. As a result, any potential as gene delivery vehicles has not yet been realized. Continued research into the molecular biolo~gy of BAVs and the development of recolllbinant vectors would benefit from the development of a cell line analogous to that of the 293 cells. In an attelllpt to establish such a cell line, the recombinant plaslllid pKC-neo was constructed, containing the left 0-19.7% of the BAV type 3 (BAV3) genome, and the selectable marker for resistance to the aminoglycoside G418, a neomycin derivative. The plasmid construct was then used to transfect both the Madin-Darby bovine kidney (MDBK) -iicell line and primary bovine lung cells, after which G418-resistant foci were selected for analysis. Two cell lines, E61 (MDBK) and E24 (primary lung), were subsequently selected and analysed for DNA content, revealing the presence of the pKC-neo sequences in their respective genomes. In addition, BAV3 RNA transcripts were detected in the E61 cells. Although the presence of E1 products has yet to be confirmed in both cell lines, the E24 cells exhibit a phenotype characteristic of partial transformation by E1. The apparent immortalization of the primary lung cells will permit exploitation of their ability to take up exogenous DNA at high efficiency.

cell of origin and triggering events for leukaemia are mostly unknown. Here we show that the bone marrow contains a progenitor that expresses renin throughout development and possesses a B-lymphocyte pedigree. This cell requires RBP-J to differentiate. Deletion of RBP-J in these renin-expressing progenitors enriches the precursor B-cell gene programme and constrains lymphocyte differentiation, facilitated by H3K4me3 activating marks in genes that control the pre-B stage. Mutant cells undergo neoplastic transformation, and mice develop a highly penetrant B-cell leukaemia with multi-organ infiltration and early death. These reninexpressing cells appear uniquely vulnerable as other conditional models of RBP-J deletion do not result in leukaemia. The discovery of these unique renin progenitors in the bone marrow and the model of leukaemia described herein may enhance our understanding of normal and neoplastic haematopoiesis.

Le syndrome reproducteur et respiratoire porcin (SRRP) est une des maladies les plus dévastatrices économiquement pour l'industrie mondiale du porc. L'agent étiologique du SRRP est le virus du SRRP (VSRRP) lequel est connu pour avoir une spécificité d'hôte très restreinte et pour sa transmission par voie aerosol. Les antigènes et les ARN du VSRRP ont été trouvés dans des cellules épithéliales du tractus respiratoire de porcs infectés par le virus. L’interaction entre les macrophages alvéolaires porcins (PAMs) et le VSRRP a été démontrée comme jouant un rôle important dans l’infection causée par le virus. Malgré cela, l’interaction prenant place entre les cellules épithéliales du tractus respiratoire porcin et le virus ne devrait pas être négligée. Jusqu’à présent, la réplication du VSRRP in vitro dans des cellules épithéliales du tractus respiratoire porcin n’a pas été conduite avec succès et les tentatives pour le faire ont échoué. Une nouvelle lignée de cellules épithéliales de poumon de porc (SJPL) est maintenant disponible et sera utilisée dans cette étude afin de déterminer si elle est permissive à la réplication du VSRRP et si elle peut être un modèle approprié pour l’étude de la pathogénèse virale du VSRRP. L’expérimentation a démontré que cette nouvelle lignée cellulaire était permissive à l’infection et à la réplication du VSRRP. Afin de corroborer ces résultats, la cinétique de réplication du virus à été effectuée avec les cellules MARC-145 et SJPL. Aucune différence significative dans la production virale totale n’a été trouvée entre les deux lignées cellulaires. Les cellules SJPL ont permis la réplication de plusieurs souches Nord-Américaines du VSRRP, quoiqu’elles sont légèrement moins efficaces que les cellules MARC-145 pour l’isolement du virus. De plus, les cellules SJPL sont phénotypiquement différentes des cellules MARC-145. Plus précisément, les cellules SJPL sont plus sensibles à l’activation par le VSRRP des pro-caspases 3/7 et plusieurs inducteurs apoptotiques. Elles ont également montré de 8 à 16 fois plus de sensibilité à l’effet antiviral causé par l’IFN-α sur la réplication du virus contrairement aux cellules MARC-145. Ces résultats démontrent que les cellules SJPL pourraient représenter un substitut intéressant aux cellules MARC-145 pour la production d’antigènes pour un vaccin anti-VSRRP. Également, dû à leurs origines (poumon de l’hôte naturel), elles pourraient s’avérer être un modèle in vitro plus approprié pour l’étude de la pathogénèse du VSRRP.

Le récepteur de la vasopressine de type 2 (V2R) joue un rôle crucial dans l’homéostasie hydrique. Exprimé principalement au niveau du rein, son activation par l’hormone antidiurétique arginine-vasopressine (AVP) favorise la réabsorption d’eau, participant ainsi à diminuer la diurèse. Plus de 200 mutations dans le gène du V2R ont été associées au diabète néphrogénique insipide congénital (DINc), une maladie causée par une perte de fonction du récepteur. À l’opposé, trois mutations découvertes récemment induisent un gain de fonction du V2R, et sont la cause du syndrome néphrogénique de l’anti-diurèse inappropriée (NSIAD). Les travaux de cette thèse visent à mieux comprendre les bases moléculaires responsables de la perte ou du gain de fonction des récepteurs mutants associés à ces deux maladies. Dans plus de 50% des cas, les mutations faux-sens affectent négativement l’adoption d’une conformation native par le V2R, provoquant la reconnaissance et la rétention intracellulaire des mutants par le système de contrôle de qualité du réticulum endoplasmique. Nos résultats ont démontré que l’interaction entre les récepteurs mutants et le chaperon moléculaire calnexine est dépendante de N-glycosylation et que sa durée varie en fonction de la mutation. De plus, l’importance de cette modification co-traductionnelle et des interactions lectines-sucres dans le processus de maturation d’un mutant donné s’est avérée une caractéristique intrinsèque, puisque l’absence de N-glycosylation n’a pas affecté le mutant Y128S (phénotype léger) tandis que la maturation du mutant W164S (phénotype sévère) a été totalement abolie. Nos résultats suggèrent aussi que l’action des chaperons pharmacologiques (CP), molécules favorisant la maturation des mutants du V2R, peut survenir à différentes étapes au cours du processus de maturation, selon le mutant réchappé. Ces différences entre muta nts suggèrent des processus biosynthétiques ‘personnalisés’ dictés par la nature de la mutation impliquée et pourraient expliquer la différence de sévérité des manifestations cliniques chez les patients porteurs de ces mutations. Bien qu’une récupération de fonction ait été obtenue pour les mutants Y128S et W164S par un traitement au CP, il n’en est pas de même pour toutes les mutations occasionnant un défaut conformationnel. C’est ce que nous avons démontré pour le mutant V88M, affligé de deux défauts, soit une faible efficacité de maturation combinée à une basse affinité pour l’AVP. Dans ce cas, et malgré une augmentation du nombre de récepteurs mutants la surface cellulaire, la diminution de l’affinité apparente du récepteur mutant pour l’AVP a été exacerbée par la présence résiduelle de CP à son site de liaison, rendant impossible l’activation du récepteur aux concentrations physiologiques d’AVP. Les mutants R137C et R137L ont une activité constitutive élevée et mènent au NSIAD tandis que la substitution de cette même arginine par une histidine (R137H) mène au DINc. Ces trois mutants se sont avéré partager plusieurs caractéristiques, dont une efficacité de maturation réduite et une désensibilisation spontanée élevée. La seule différence iden tifiée entre ces mutants est leur niveau d’activité constitutive. Le CP utilisé dans nos études possède aussi la propriété d’agoniste inverse, mais n’a pourtant pas diminué l’activité constitutive des mutants R137C/L, suggérant une conformation active ‘figée’. Seul l’effet chaperon a été observé, entraînant la hausse de récepteurs à la surface cellulaire, qui se traduit par une augmentation de la production de second messager. Nous avons par contre suggéré l’utilisation d’AVP puisqu’il favorise l’endocytose des récepteurs R137/L sans promouvoir leur activation, diminuant ainsi le nombre de récepteurs actifs à la surface cellulaire. Nous avons identifié la première mutation occasionnant un gain de fonction du V2R qui n’implique pas l’arginine 137. Le mutant F229V a une activité constitutive élevée et, contrairement aux R137C et R137L, il n’est pas sujet à une désensibilisation spontanée accrue. L’observation que des agonistes inverses sont aptes à inhiber l’activité constitutive de ce nouveau mutant est une découverte importante puisque l’insuccès obtenu avec les mutations précédentes suggérait que ces molécules n’étaient pas utiles pour le traitement du NSIAD. Considérés globalement, ces travaux illustrent le caractère particulier des formes mutantes du V2R et l’importance de bien cerner les conséquences fonctionnelles des mutations afin d’apporter aux patients atteints de DINc ou NSIAD une thérapie personnalisée, et de développer de nouveaux agents thérapeutiques adaptés aux besoins.

La rapamycine est un immunosuppresseur utilisé pour traiter plusieurs types de maladies dont le cancer du rein. Son fonctionnement par l’inhibition de la voie de Tor mène à des changements dans des processus physiologiques, incluant le cycle cellulaire. Chez Saccharomyces cerevisiae, la rapamycine conduit à une altération rapide et globale de l’expression génique, déclenchant un remodelage de la chromatine. Nous proposons que les modifications des histones peuvent jouer un rôle crucial dans le remodelage de la chromatine en réponse à la rapamycine. Notre objectif principal est d’identifier d’une banque de mutants d’histone les variantes qui vont échouer à répondre à la rapamycine dans une tentative de réaliser une caractérisation des modifications d’histone critiques pour la réponse à cette drogue. Ainsi, nous avons réalisé un criblage d’une banque de mutants d’histone et identifié plusieurs mutants d‘histone dont la résistance à la rapamycine a été altérée. Nous avons caractérisé une de ces variantes d’histone, à savoir H2B, qui porte une substitution de l’alanine en arginine en position 95 (H2B-R95A) et démontré que ce mutant est extrêmement résistant à la rapamycine, et non à d’autres drogues. Des immunoprécipitations ont démontré que H2B-R95A est défectueux pour former un complexe avec Spt16, un facteur essentiel pour la dissociation de H2A et H2B de la chromatine, permetant la réplication et la transcription par les ADN et ARN polymérases, respectivement. Des expériences de ChIP-Chip et de micropuce ont démontré que l’arginine 95 de H2B est requise pour recruter Spt16 afin de permettre l’expression d’une multitude de gènes, dont certains font partie de la voie des phéromones. Des évidences seront présentées pour la première fois démontrant que la rapamycine peut activer la voie des phéromones et qu’une défectuosité dans cette voie cause la résistante à cette drogue.

Background Airborne transmitted pathogens, such as porcine reproductive and respiratory syndrome virus (PRRSV), need to interact with host cells of the respiratory tract in order to be able to enter and disseminate in the host organism. Pulmonary alveolar macrophages (PAM) and MA104 derived monkey kidney MARC-145 cells are known to be permissive to PRRSV infection and replication and are the most studied cells in the literature. More recently, new cell lines developed to study PRRSV have been genetically modified to make them permissive to the virus. The SJPL cell line origin was initially reported to be epithelial cells of the respiratory tract of swine. Thus, the goal of this study was to determine if SJPL cells could support PRRSV infection and replication in vitro. Results The SJPL cell growth was significantly slower than MARC-145 cell growth. The SJPL cells were found to express the CD151 protein but not the CD163 and neither the sialoadhesin PRRSV receptors. During the course of the present study, the SJPL cells have been reported to be of monkey origin. Nevertheless, SJPL cells were found to be permissive to PRRSV infection and replication even if the development of the cytopathic effect was delayed compared to PRRSV-infected MARC-145 cells. Following PRRSV replication, the amount of infectious viral particles produced in SJPL and MARC-145 infected cells was similar. The SJPL cells allowed the replication of several PRRSV North American strains and were almost efficient as MARC-145 cells for virus isolation. Interestingly, PRRSV is 8 to 16 times more sensitive to IFNα antiviral effect in SJPL cell in comparison to that in MARC-145 cells. PRRSV induced an increase in IFNβ mRNA and no up regulation of IFNα mRNA in both infected cell types. In addition, PRRSV induced an up regulation of IFNγ and TNF-α mRNAs only in infected MARC-145 cells. Conclusions In conclusion, the SJPL cells are permissive to PRRSV. In addition, they are phenotypically different from MARC-145 cells and are an additional tool that could be used to study PRRSV pathogenesis mechanisms in vitro.
