A doença de Alzheimer (DA) é a forma mais comum de demência, representando cerca de 80% dos casos. A DA é caracterizada por um processo de declínio progressivo e irreversível das funções cognitivas e da memória, que se estende para a desorganização do comportamento. Atualmente, 46,8 milhões de pessoas em todo o mundo foram diagnosticadas com demência. Embora vários fatores tenham sido implicados na DA, sua etiologia ainda não é completamente conhecida. Do ponto de vista neuropatológico, é observado no cérebro de indivíduos com DA atrofia cortical difusa, presença de grande número de placas senis, emaranhados neurofibrilares, processo inflamatório e perda neuronal. A progressão dos sintomas está associada a mudanças estruturais nas sinapses colinérgicas em certas regiões do cérebro, que consequentemente, apresentam neurotransmissão colinérgica reduzida. Os vários eventos patológicos interligados contribuem para o avanço da doença e direcionam diversas pesquisas na busca por tratamentos multialvos com base no processo multifatorial de DA. Assim o presente trabalho descreve a síntese de derivados híbridos dual binding site de donepezila-tacrina (fármacos inibidores de acetilcolinesterase), com potencial para agir em dois alvos terapêuticos pela (i) inibição da acetilcolinesterase em ambos os sítios ativo e periférico, como demonstrado pelos estudos de modelagem molecular, e (ii) na agregação do peptídeo A? neurotóxico induzido pela acetilcolinesterase, na tentativa de interromper a progressão da doença. A estratégia sintética envolveu a condensação da 5,6-dimetóxiindanona com a unidade 4-piperidinil carbaldeído, a qual forneceu o intermediário 5,6- dimetóxindan-1-ona-4-piperidinil-metileno-1-[(4-cloroquinolin-2-il)metil], seguido de redução da dupla ligação, gerada na reação de condensação anterior, e substituição do átomo de cloro-quinolina por amino para obtenção do produto final, ou manutenção da função olefina, seguido de substituição do átomo de cloro-quinolina por azido ou amino, gerando cinco híbridos estruturalmente correlacionados. Os híbridos foram testados em ensaio de inibição de acetilcolinesterase e butirilcolinesterase pelo método de Ellman, e o híbrido insaturado, contendo a função amino-quinolina foi o mais ativo da série com IC50 na faixa de nanomolar (0,014 ?M). Futuramente, os intermediários da reação e produto final serão submetidos ao ensaio de inibição da agregação do peptídeo A? neurotóxico pelo método da tioflavina T. Neste trabalho, também são descritos os testes de predição in vitro para permeação pela barreira hematoencefálica, bem como sua absorção intestinal, pelo método PAMPA.

Atualmente a nossa sociedade depara-se com diversos problemas sociais, como o envelhecimento populacional, a mudança de padrões económicos e de bem-estar seguros, a dissolvência das estruturas familiares e o isolamento social dos idosos. Com o objetivo de minimizar esta tendência, pretendemos promover a prática intergeracional visando unir as pessoas em atividades propositadas, mutuamente benéficas, que promovam uma maior compreensão e respeito entre gerações, contribuindo para a construção de comunidades mais coesas. Participantes. Um grupo de 17 jovens alunos da turma C do 7º ano de escolaridade da Escola Básica e Secundária de Ourém e um grupo de 15 utentes da Universidade Sénior de Ourém. Método. Os participantes realizaram, em conjunto, ações em diversas áreas como o voluntariado, solidariedade, promoção da saúde e qualidade de vida em cooperação entre gerações. Com o apoio das várias entidades locais, estes tornaram-se mentores e dinamizadores de diversas ações, incentivando os seus pares numa perspetiva aglutinadora a envolverem-se nas diferentes iniciativas inerentes ao Projeto ELSS - Estreitar Laços com Solidariedade e Saúde. Resultados. Com este projeto desenvolveu-se uma comunidade ativa, envolvida num conjunto de boas práticas, tendo em vista o envelhecimento ativo e a prática de atividades intergeracionais, incentivando o envelhecimento saudável e pró-ativo. Através do estudo efetuado evidenciamos que após a sua implementação, o aumento da perceção da felicidade por parte dos seniores aumentou. Desenvolvemos uma comunidade sustentável através de diversas ações comunitárias e sociais com impacto sobre a população. As atividades intergeracionais que se concretizaram, foram significativas e tiveram um contributo importante na vida dos jovens e seniores. Conclusões. Os resultados obtidos foram muito satisfatórios; o projeto teve uma projeção e adesão surpreendentes, que evidenciam a importância da sua continuidade no mesmo local, ou com possível replicação em comunidades com caraterísticas idênticas.

Although photodynamic therapy have been used as a useful tool over the past 30 years in oncology, few clinical trials have been conducted in dentistry. Photodynamic therapy (PDT) uses non - toxic photosensitizers and selective which are administered in target cells followed by local application of visible light, producing reactive oxygen species capable of causing cell death by apoptosis or necrosis, injured the local vasculature, and exert important effects on the im mune system. New generations of photosensitizing agents, such as nanoparticulate phthalocyanines, has shown excellent results in antitumor and antibacterial activity . In this context, the present work constitutes the first clinical protocol of local appli cation of nanoemulsion chloro - aluminum phthalocyanine (AlClFc) followed by irradiation in human gingiva, and analyzed descriptively and comparatively , by means of immunohistochemistry , the expression of RANK , RANKL , OPG and VEGF in a split - mouth model . Eight healthy volunteers with clinical indication for extraction were included in the study . Seven days before the extraction, was injected in the gingiva of participants, 5 μ M of nanoemulsion AlClFc followed by irra diation with diode laser (660nm , 7 J/cm2 ), the contralateral side was used as control. Tissue specimens were removed seven days after the TFD is performed. Tissues sample were divided into two groups (test and con trol groups) for histological and immunohistochemical analysis. Patients were monitored at days, 0, 7, 14 and 30 to assess adverse effects of the therapy. Vascular alterations were seen in gingival samples that received PDT. Areas of edema and vascular con gestion, and intense vascularization were viewed . Additionally, dystrophic calcification in subepithelial region were observed in the test group. The results showed a similar pattern of immunostaining scores of RANK, RANKL and VEGF between the test and co ntrol groups, with no statistically significant difference (p = 0.317, p = 0.777, p = 0 .814, respectively). RANK and RANKL exhibited weak or absent immunostaining in most specimens analyzed. There was n o immunostaining for OPG. VEGF showed moderate to stro ng immunostaining in specimens from the test group. In addition, the clinical study showed that therapy was well tolerated by all patients. Adverse effects were short - time and completely reversible. Taken together, the results presented in this study showe d that PDT mediated by nanoemulsion containing AlClPc is safe for clinical application in gingival tissue and suggests that a strong immunostaining for VEGF after therapy .

In view of the evident increase in the population group of people over 60 years old, the aging process has raised important discussions on social, political and economical planning that recommend that the increase in longevity must be followed by autonomy and well-being. Faced to these demands, the literature reports that physical activity is a key strategy to promote better living conditions for the elderly. Studies about personenvironment are making efforts to understand the socio physical environment role to encourage old people to make activities in many different environments. Among these, the urban public open space has been reported by many researches as a promising alternative in order to adopt a less sedentary life style. Based on these considerations, this study proposed to investigate the relationship between the social environment support elements and social interaction of seniors in public squares in the city of Natal, RN, using a multimethodological approach that included a experts panel, environmental characteristics analysis of 252 squares, case studies containing collective and individual interviews with elderly users and the spatial representation of their social interactions. The association between physical activities with the socialization practiced by them was identified as the main motivation for their presence in these places. The results indicate that the relationship between the socio-environmental characteristics (concept developed in this study) and the socio-spatial behaviors that include the spatial appropriation, spatial arrangements and interpersonal distances, helps to promote social interactions in carrying out activities, which, according to the assumptions Active Ageing Program, enable the maintenance of an active way of life, with important implications for health, wellness and quality of life in old age

The conventional control schemes applied to Shunt Active Power Filters (SAPF) are Harmonic extractor-based strategies (HEBSs) because their effectiveness depends on how quickly and accurately the harmonic components of the nonlinear loads are identified. The SAPF can be also implemented without the use of the load harmonic extractors. In this case, the harmonic compensating term is obtained from the system active power balance. These systems can be considered as balanced-energy-based schemes (BEBSs) and their performance depends on how fast the system reaches the equilibrium state. In this case, the phase currents of the power grid are indirectly regulated by double sequence controllers with two degrees of freedom, where the internal model principle is employed to avoid reference frame transformation. Additionally the DSC controller presents robustness when the SAPF is operating under unbalanced conditions. Furthermore, SAPF implemented without harmonic detection schemes compensate simultaneously harmonic distortion and reactive power of the load. Their compensation capabilities, however, are limited by the SAPF power converter rating. Such a restriction can be minimized if the level of the reactive power correction is managed. In this work an estimation scheme for determining the filter currents is introduced to manage the compensation of reactive power. Experimental results are shown for demonstrating the performance of the proposed SAPF system.

This work aimed to promote the synthesis, characterization and propose a plausible molecular structure for coordination compounds involving furosemide (4-Chloro-2-(2- furylmethylamino)-5-sulfamoyl-benzoic acid) with the metal ions Ni+2, Zn+2 and Co+2. The compounds were obtained in methanoic medium by evaporation of the solvent after the synthesis procedure. For characterization of coordination compounds determining the levels of metals by EDTA complexometry, infrared spectroscopy (FTIR), solubility of compounds in various solvents, thermogravimetry (TG), differential scanning calorimetry (DSC), differential thermal analysis were made (DTA), determination of the carbon , hydrogen and nitrogen (CHN). The results of infrared spectroscopy in the region suggest that the organic ligand is coordinated in a bidentate fashion to the metal ions, the metal center interactions to occur by the coordination of the nitrogen atom of the amino group and the oxygen atom of the carboxylic acid of the structure of furosemide. With the results of the levels of metal, elemental analysis (CHN) and thermal analysis has been possible to propose the structure of the ligand. The values of the molar conductivity of the complex in acetonitrile behavior suggest the non acetonitrile electrolyte solution. With the solubility tests it was found that the compounds have high solubility in methanol and acetonitrile, as are partially insoluble in water. From the results of thermal analysis (TG, DSC, DTA), it was possible to obtain the thermal behavior of the compounds as stages of dehydration, thermal stability, decomposition and the energies involved.

This Thesis main objective is to implement a supporting architecture to Autonomic Hardware systems, capable of manage the hardware running in reconfigurable devices. The proposed architecture implements manipulation, generation and communication functionalities, using the Context Oriented Active Repository approach. The solution consists in a Hardware-Software based architecture called "Autonomic Hardware Manager (AHM)" that contains an Active Repository of Hardware Components. Using the repository the architecture will be able to manage the connected systems at run time allowing the implementation of autonomic features such as self-management, self-optimization, self-description and self-configuration. The proposed architecture also contains a meta-model that allows the representation of the Operating Context for hardware systems. This meta-model will be used as basis to the context sensing modules, that are needed in the Active Repository architecture. In order to demonstrate the proposed architecture functionalities, experiments were proposed and implemented in order to proof the Thesis hypothesis and achieved objectives. Three experiments were planned and implemented: the Hardware Reconfigurable Filter, that consists of an application that implements Digital Filters using reconfigurable hardware; the Autonomic Image Segmentation Filter, that shows the project and implementation of an image processing autonomic application; finally, the Autonomic Autopilot application that consist of an auto pilot to unmanned aerial vehicles. In this work, the applications architectures were organized in modules, according their functionalities. Some modules were implemented using HDL and synthesized in hardware. Other modules were implemented kept in software. After that, applications were integrated to the AHM to allow their adaptation to different Operating Context, making them autonomic.

The time perception is critical for environmental adaptation in humans and other species. The temporal processing, has evolved through different neural systems, each responsible for processing different time scales. Among the most studied scales is that spans the arrangement of seconds to minutes. Evidence suggests that the dorsolateral prefrontal (DLPFC) cortex has relationship with the time perception scale of seconds. However, it is unclear whether the deficit of time perception in patients with brain injuries or even "reversible lesions" caused by transcranial magnetic stimulation (TMS) in this region, whether by disruption of other cognitive processes (such as attention and working memory) or the time perception itself. Studies also link the region of DLPFC in emotional regulation and specifically the judgment and emotional anticipation. Given this, our objective was to study the role of the dorsolateral prefrontal cortex in the time perception intervals of active and emotionally neutral stimuli, from the effects of cortical modulation by transcranial direct current stimulation (tDCS), through the cortical excitation (anodic current), inhibition (cathode current) and control (sham) using the ranges of 4 and 8 seconds. Our results showed that there is an underestimation when the picture was presented by 8 seconds, with the anodic current in the right DLPFC, there is an underestimation and with cathodic current in the left DLPFC, there is an overestimation of the time reproduction with neutral ones. The cathodic current over the left DLPFC leads to an inverse effect of neutral ones, an underestimation of time with negative pictures. Positive or negative pictures improved estimates for 8 second and positive pictures inhibited the effect of tDCS in DLPFC in estimating time to 4 seconds. With this work, we conclude that the DLPFC plays a key role in the o time perception and largely corresponds to the stages of memory and decision on the internal clock model. The left hemisphere participates in the perception of time in both active and emotionally neutral contexts, and we can conclude that the ETCC and an effective method to study the cortical functions in the time perception in terms of cause and effect.

In this work, a sliding mode controller with fuzzy inference is applied for active vibration control in an SMA two-bar truss. In order to obtain a simpler controller, a polynomial model is used in the control law, while a more sophisticated version, which presents close agreement with experimental data, is applied to describe the SMA behavior of the structural elements. This system has a rich dynamic response and can easily reach a chaotic behavior even at moderate loads and frequencies. Therefore, this approach has the advantage of not only obtaining a simpler control law, but also allows its robustness be evidenced. Numerical simulations are carried out in order to demonstrate the control system performance.

Photodynamic therapy (PDT) consists of a non-toxic photosensitizing agent (FS) administration followed by a laser source resulting in a sequence of photochemical and photobiological processes that generate reactive oxygen species (ROS) that damaging cells. The present work evaluated the effects of PDT nanoemulsion-aluminum chloride phthalocyanine (AlClFc) mediated on malondialdehyde (MDA), glutathione (GSH), superoxide dismutase (SOD) and glutathione peroxidase (GPx) levels, which represent indicators involved in oxidative stress and antioxidant defenses. For this purpose, this study used 120 female rats of the Rattus norvegicus species, Wistar race, divided into 5 groups: Healthy (H), with periodontal disease (PD), with periodontal disease and treatment with FS (F), with periodontal disease and treatment with the laser (L); and periodontal disease and treatment with PDT (FL). An experimental model for represent periodontal disease (PD) was induced by ligature (split-mouth). Seven days later the induction of PD, the treatments were instituted according to the groups. In the group treated with PDT was applied 40μl FS (5μM) followed by laser irradiation diode InGaAlP (660nm, 100J / cm2). The rats were sacrificed on the 7th and 28th day after treatment and tissue specimens were removed and subjected to histological, immunohistochemical methods and enzymatic colorimetric measurements with detection by UV / VIS spectroscopy. Inflammatory changes, connective tissue disorganization and alveolar bone loss were displaying in groups with PD induced. The enzyme dosages showed that MDA levels were higher in PD induced groups, with no statistically significant differences (p> 0.05). High levels of GSH were found in groups L (p = 0.028) and FL (p = 0.028) compared with PD group, with statistically significant differences. Immunohistochemistry for SOD showed higher immunostaining in L and FL groups, compared to the PD group without statistically significant differences (p> 0.05). GPx showed lower immunoreactivity in the DP group when compared to the other groups and statistically significant differences were observed between the DPxL groups (p <0.05). TFD administered in this experiment did not induce elevation of MDA levels significantly increased the GSH levels and showed intense immunostaining pada SOD and GPx, showing that this therapy does not accentuated lipid peroxidation, however, it was able to induce effects on the antioxidant defenses processes. The LBI therapy appeared to show photomodulatory promoting effects reduction of the MDA levels, increasing GSH levels and with intense immunostaining for SOD and GPx, demonstrating that laser therapy induced antioxidant effects.

Um dos fenómenos mais significativos da sociedade contemporânea, com tendência a acentuar-se, é o aumento da população idosa, que tem vindo a constituir um sério desafio, não só na área da saúde mas também no campo social, cultural, económico e político, ao qual ninguém pode ficar indiferente. Com o projeto que concebemos e implementámos pretendemos dar o nosso contributo na área da animação sociocultural, não apenas como agentes de mudança mas, sobretudo no desenvolvimento de ferramentas para enriquecer e formar os idosos promovendo o seu envelhecimento digno, saudável e ativo. Neste âmbito, pretendemos verificar que o contacto entre os idosos e as imagens de obras de arte lhes faculta a aquisição de novas aprendizagens, infundindo ânimo e participação crítica, favorecendo o crescimento não só pessoal mas também grupal. Seguindo uma metodologia de investigação-ação, desenvolvemos uma sequência de atividades, em contexto de lar, envolvendo idosos com idades entre os 59 e os 88 anos. A avaliação do projeto permitiu-nos constatar que os idosos que convivem com o mundo das artes (imagens de pinturas de obras de arte) aprendem não só a “olhar” mas sim a refletir e a apreciar, estimulando assim a sua capacidade cognitiva. De igual modo, através da promoção do diálogo, mantêm a sua vida social ativa.

Assistimos a um acelerado processo de envelhecimento demográfico. A população está cada vez mais envelhecida, surgindo a necessidade de promover e incentivar o Envelhecimento Ativo. Este é contínuo, difere de pessoa para pessoa, podendo ser compreendido e influenciado por diversos fatores. As alterações familiares levam a uma reorganização na própria visão do indivíduo ao longo da sua trajetória de vida. O objetivo deste estudo é compreender de que modo as trajetórias familiares influenciam o envelhecimento ativo. A metodologia utilizada foi qualitativa e a recolha de dados obteve-se através de entrevistas semiestruturadas. Foram selecionados 4 sujeitos, 2 homens e 2 mulheres, entre os 70 e os 77 anos, residentes em meio rural e urbano. A análise incidiu sobre as trajetórias familiares ao longo da infância, juventude, idade adulta e pós-reforma. Os resultados constatam que, na infância, a família foi um suporte sólido. Apesar da educação rígida típica da época, os progenitores incentivaram a escolaridade e autonomia dos sujeitos. Na juventude, o fenómeno da emigração esteve presente, tendo sido uma etapa marcada pela falta de liberdade, pela diferença de género e entrada no mundo laboral. Na fase adulta, todos namoraram, primando o respeito, casaram e constituíram família. Nesta fase, também marcada por algumas perdas, todos eles se mantiveram ativos profissionalmente e ao nível do lazer. A entrada na reforma foi vivida com naturalidade, sem implicações familiares, continuando ativos. As conclusões revelam que a família foi importante em todo o ciclo vital, destacando-se a relação mantida com os seus ascendentes, descendentes e cônjuges.

Os indicadores do envelhecimento demográfico sugerem desafios de impacto no ajustamento da sociedade portuguesa ao crescente aumento da população idosa e, com este, vai aumentando igualmente a preocupação com a qualidade de vida desta população, e com o envelhecimento ativo. A responsabilidade de ajudar o idoso a envelhecer com qualidade é de todos nós e por isso é urgente olhar os novos tempos do presente como tempos em que dar um sentido à velhice deverá fazer parte dos desafios da vida de cada um de nós, e em especial dos técnicos de intervenção social, no sentido de ajudar o idoso a encontrar o seu sentido de vida no presente, como força geradora e motivadora da construção do seu futuro. Fundamenta-se assim a escolha deste estudo científico intitulado “Vidas de idosos. Reviver o passado para construir um futuro mais ativo”. Com o objetivo de perceber se é ou não possível os técnicos de intervenção em envelhecimento ativo conciliarem o conhecimento da história de vida de um idoso, com o projeto de vida fundamental para a sua qualidade de vida, (re)desenhando o seu projeto de vida, no diálogo com ele mesmo, conciliando os interesses do passado com as suas perspetivas presentes e futuras, foram estudados três idosos independentes, autónomos, em que todos aderiam às atividades propostas pela instituição, em duas instituições distintas. Em função destes objetivos e numa tentativa de entender o significado que os idosos dão às suas vidas ou a aspetos circunscritos dela, optei por uma abordagem qualitativa que permite recolher e refletir os aspetos enraizados dos indivíduos, os seus hábitos quotidianos e nos dão acesso ao seu mundo subjetivo, em contraste com a despersonalização dos estudos estatísticos. Nesta perspetiva, recorri à metodologia etnográfica, e dentro desta à metodologia das histórias de vida, através da realização de diversas entrevistas profundas aos três sujeitos estudados, que foram sistematizadas em sinopses de cuja análise de conteúdo foram retirados os dados que permitiram chegar às conclusões deste estudo científico. Só em espaços de participação onde os técnicos de intervenção sejam mediadores, com um olhar holístico sobre o idoso, e onde é prioritário respeitar as diferenças, é possível ajuda-lo a (re)desenhar o seu projeto de vida e a construir um futuro com mais significado para si, por isso mais positivo e com mais qualidade de vida.
