A synthesis of artificial sweetener dulcin starting from nitrobenzene was elaborated for undergraduate organic laboratory course. Paracetamol and phenacetin, both physiologically active analgesic compounds, were also prepared as intermediates. Besides a large scope of discussion subjects related with organic synthesis, interesting lectures about analgesics and sweeteners may also be performed in this project. The advantage of this project is the adaptability according to the conditions offered by the course, i.e., convenience and/or availability of time and reagents.

The oxidation of alcohols to obtain ketones, aldehydes or carboxylic acids is a fundamental transformation in organic synthesis and many reagents are known for these conversions. However, there is still a demand for mild and selective reagents for the oxidation of alcohols in the presence of other functional groups. As an alternative, the nitroxyl radical TEMPO (2,2,6,6-tetramethylpiperidine-N-oxyl) has been demonstrated to be a useful reagent for the transformation of alcohols. The oxidation of alcohols using TEMPO is often efficient, fast, selective, made in mild conditions and can tolerate sensitive functional groups. In this article we report different methodologies using TEMPO in the oxidation of alcohols.

The Lycopodium alkaloids are a structurally diverse group of natural products isolated from Lycopodium with important biological effects for the potential treatment of cancer and severe neurodegenerative diseases. To date, full biological studies have been hampered by lack of material from natural sources. Total synthesis represents a possible solution to meet this demand as well as the most effective way to design new compounds to determine structural activity relationships and obtain more potent compounds. The aim of this chapter is to summarise the work carried out in this field so far by presenting an overview of the synthetic strategies used to access each of the four key Lycopodium alkaloid types. Particular emphasis has been placed on methods that rapidly construct each nucleus utilizing tandem reactions.

The application of biocatalysis is a promising field related to new technologies for organic synthesis. The development of immobilization techniques is very important due to the multiple or repetitive use of a single batch of enzymes and the ability to stop the reaction rapidly, at any stage, by removing the enzymes. In most cases, after immobilization, enzymes and microorganisms maintain or even increase their activity and stability. This work presents an overview of the common methods for lipase immobilization in polymers and applications of these systems to obtain compounds of synthetic interest.

Solid-phase organic synthesis (SPOS) has been considered the main strategy for the construction of combinatorial libraries, because its simplicity leads to faster synthetic procedures. In addition to that, a series of reports in the specialized literature show great advantages in the use of microwave activation, when compared to classical heating, for instance: shorter reaction times, in some cases from several hours to a few minutes, increase of selectivity and product yields, energy economy and reduction and/or elimination of solvent. This review describes the use of microwave ovens/reactors in solid phase organic synthesis, describing the advantages, equipment and reactions using both techniques.

Thiazoles comprise an important class of heterocyclic compounds present in many potent biologically active molecules. This heterocyclic ring also shows various applications in organic synthesis to obtain new compounds. This paper presents different methodologies for the preparation of thiazole ring systems, as well as their synthetic applications.

The present contribution describes three different modern experiments for possible adoption in undergraduate organic chemistry laboratories. These are: 1. electrocatalytic hydrogenation of benzaldehyde to benzyl alcohol; 2. identification of three volatile components, obtained from pineapple fruit, by mass spectrometry and 3. microwave mediated fast synthesis of N-(p-chlorophenyl)phthalamic acid from phthalic anhydride and p-chloroaniline under solvent-free conditions. The experiments can be executed in a short period of time, putting the undergraduate student in contact with a variety of topics in organic chemistry and several techniques of analysis, showing multidisciplinarity in organic chemistry.

The Prins cyclization reaction has significantly advanced in the last years as demonstrated by a number of applications described in the literature. The objective of this report is to introduce this powerful synthetic methodology to the undergraduate and graduated student, since it is rarely presented in an organic synthesis formal course. This reaction is, in some cases, the methodology of choice for the preparation of natural products or drugs that present the tetrahydropyrane moiety in their structures. In this report we show some aspects of this reaction, including mechanism, scope and limitations.

The field of chiral catalysis has experienced explosive growth over the last two decades. By now, many of the classical reactions in organic synthesis can be carried out efficiently in asymmetric manner. As one of the fundamental and powerful C-C bond-forming reactions, enantioselective catalytic allylation (ECA) and crotylation (ECC) of aldehydes has attracted considerable attention. In this article, we present an overview about the importance of chiral Lewis acids and bases in catalytic enantioselective addition of allyl- and crotyl metals to aldehydes and the application of this methodology in the total synthesis of natural and non-natural products.

alpha-Bromoacetophenones are important in organic synthesis. They have been widely used as precursors of several natural products. Several methods of bromination aiming at their synthesis have been described, however they furnish a mixture of starting material 10, mono 8 and dibromide 11 products. We developed a novel, simple and efficient synthesis of these compounds with applications in the synthesis of benzoylbenzofurans 9, compounds with important pharmacological properties, such as the ability of dilating the coronary artery and analgesic action. Such compounds have also been used as key intermediates to obtain quinone systems.

Atropisomerism is a special kind of stereoisomeric relationship that arises from the freezing of a certain conformation of an organic molecule, associated with a high rotational barrier about a single covalent bond. Atropisomerism has been originally described in orto-functionalyzed biphenyl derivatives, but a lot of other organic functionalities can present this structural phenomenon, characterized by the presence of chiral properties in compounds that don't present classical stereogenic centers. Atropisomeric compounds, intermediates and catalysts have well-know importance in organic synthesis, but the influence of the axial chirality in substances able to modulate biological systems is still not very exploited in drug design and development. In this context, the present account describes the importance of this structural property in the medicinal chemistry of different classes of bioactive compounds or therapeutic agents, emphasizing how atropisomerism could affect the molecular recognition of a ligand or a prototype by the target bioreceptor.

The design and use of a novel apparatus for a variant of vacuum distillation is described. Relative to a conventional device, the apparatus/technique described permits superior recovery of multigram quantities of moderately volatile liquids from vacuum distillations.

D-mannitol is used in several fields including food, pharmaceuticals, cosmetics and textiles being an important carbohydrate found widespread in nature. Due to its chirality, it is largely used in organic synthesis with several applications, such as ligands, polymers, chiral pool, for preparation of small chiral building blocks, key intermediates in total synthesis. In this context, the aim of this review is to highlights recent applications of D-mannitol, especially in total synthesis.

The present work discusses the appearance of the concepts of valence and molecular structure, and describes the appropriation and evolution of the concept of molecule in the period following the publication of Avogadro's Hypothesis. The point of reference is the development of what became known as Organic Chemistry, which encompassed Pharmacy, Physiological Chemistry, Animal and Plant Chemistry, Chemistry of Dyestuffs, Agricultural Chemistry, and the fledgling Organic Synthesis industry in the early 19th century. The theories formulated in these areas and the quest for accurate atomic weights led to those concepts of valence and molecular structure and to a precise differentiation between atom and molecule.
