Axisymmetric notched bars with notch roots of large and small radii were tested under large strain cyclic loading. The main attention is focused on the fracture behaviour of steels having cycles to failure within the range 1-100. Our study shows that a gradual transition from a static ductile nature to one of fatigue cleavage can be observed and characterized by the Coffin-Manson formula in a generalized form. Both the triaxial tensile stress within the central region of specimens and static damage caused by the first increasing load have effects on the final failure event. A generalized cyclic strain range parameter DELTAepsilon is proposed as a measure of the numerous factors affecting behaviour. Fractographs are presented to illustrate the behaviour reported in the paper.

Partial differential equations (PDEs) with multiscale coefficients are very difficult to solve due to the wide range of scales in the solutions. In the thesis, we propose some efficient numerical methods for both deterministic and stochastic PDEs based on the model reduction technique.

For the deterministic PDEs, the main purpose of our method is to derive an effective equation for the multiscale problem. An essential ingredient is to decompose the harmonic coordinate into a smooth part and a highly oscillatory part of which the magnitude is small. Such a decomposition plays a key role in our construction of the effective equation. We show that the solution to the effective equation is smooth, and could be resolved on a regular coarse mesh grid. Furthermore, we provide error analysis and show that the solution to the effective equation plus a correction term is close to the original multiscale solution.

For the stochastic PDEs, we propose the model reduction based data-driven stochastic method and multilevel Monte Carlo method. In the multiquery, setting and on the assumption that the ratio of the smallest scale and largest scale is not too small, we propose the multiscale data-driven stochastic method. We construct a data-driven stochastic basis and solve the coupled deterministic PDEs to obtain the solutions. For the tougher problems, we propose the multiscale multilevel Monte Carlo method. We apply the multilevel scheme to the effective equations and assemble the stiffness matrices efficiently on each coarse mesh grid. In both methods, the $\KL$ expansion plays an important role in extracting the main parts of some stochastic quantities.

For both the deterministic and stochastic PDEs, numerical results are presented to demonstrate the accuracy and robustness of the methods. We also show the computational time cost reduction in the numerical examples.

Wide field-of-view (FOV) microscopy is of high importance to biological research and clinical diagnosis where a high-throughput screening of samples is needed. This thesis presents the development of several novel wide FOV imaging technologies and demonstrates their capabilities in longitudinal imaging of living organisms, on the scale of viral plaques to live cells and tissues.

The ePetri Dish is a wide FOV on-chip bright-field microscope. Here we applied an ePetri platform for plaque analysis of murine norovirus 1 (MNV-1). The ePetri offers the ability to dynamically track plaques at the individual cell death event level over a wide FOV of 6 mm × 4 mm at 30 min intervals. A density-based clustering algorithm is used to analyze the spatial-temporal distribution of cell death events to identify plaques at their earliest stages. We also demonstrate the capabilities of the ePetri in viral titer count and dynamically monitoring plaque formation, growth, and the influence of antiviral drugs.

We developed another wide FOV imaging technique, the Talbot microscope, for the fluorescence imaging of live cells. The Talbot microscope takes advantage of the Talbot effect and can generate a focal spot array to scan the fluorescence samples directly on-chip. It has a resolution of 1.2 μm and a FOV of ~13 mm2. We further upgraded the Talbot microscope for the long-term time-lapse fluorescence imaging of live cell cultures, and analyzed the cells’ dynamic response to an anticancer drug.

We present two wide FOV endoscopes for tissue imaging, named the AnCam and the PanCam. The AnCam is based on the contact image sensor (CIS) technology, and can scan the whole anal canal within 10 seconds with a resolution of 89 μm, a maximum FOV of 100 mm × 120 mm, and a depth-of-field (DOF) of 0.65 mm. We also demonstrate the performance of the AnCam in whole anal canal imaging in both animal models and real patients. In addition to this, the PanCam is based on a smartphone platform integrated with a panoramic annular lens (PAL), and can capture a FOV of 18 mm × 120 mm in a single shot with a resolution of 100─140 μm. In this work we demonstrate the PanCam’s performance in imaging a stained tissue sample.

Os principais constituintes do ar, nitrogênio, oxigênio e argônio, estão cada vez mais presentes nas indústrias, onde são empregados nos processos químicos, para o transporte de alimentos e processamento de resíduos. As duas principais tecnologias para a separação dos componentes do ar são a adsorção e a destilação criogênica. Entretanto, para ambos os processos é necessário que os contaminantes do ar, como o gás carbônico, o vapor dágua e hidrocarbonetos, sejam removidos para evitar problemas operacionais e de segurança. Desta forma, o presente trabalho trata do estudo do processo de pré-purificação de ar utilizando adsorção. Neste sistema a corrente de ar flui alternadamente entre dois leitos adsorvedores para produzir ar purificado continuamente. Mais especificamente, o foco da dissertação corresponde à investigação do comportamento de unidades de pré-purificação tipo PSA (pressure swing adsorption), onde a etapa de dessorção é realizada pela redução da pressão. A análise da unidade de pré-purificação parte da modelagem dos leitos de adsorção através de um sistema de equações diferenciais parciais de balanço de massa na corrente gasosa e no leito. Neste modelo, a relação de equilíbrio relativa à adsorção é descrita pela isoterma de Dubinin-Astakhov estendida para misturas multicomponentes. Para a simulação do modelo, as derivadas espaciais são discretizadas via diferenças finitas e o sistema de equações diferenciais ordinárias resultante é resolvido por um solver apropriado (método das linhas). Para a simulação da unidade em operação, este modelo é acoplado a um algoritmo de convergência relativo às quatro etapas do ciclo de operação: adsorção, despressurização, purga e dessorção. O algoritmo em questão deve garantir que as condições finais da última etapa são equivalentes às condições iniciais da primeira etapa (estado estacionário cíclico). Desta forma, a simulação foi implementada na forma de um código computacional baseado no ambiente de programação Scilab (Scilab 5.3.0, 2010), que é um programa de distribuição gratuita. Os algoritmos de simulação de cada etapa individual e do ciclo completo são finalmente utilizados para analisar o comportamento da unidade de pré-purificação, verificando como o seu desempenho é afetado por alterações nas variáveis de projeto ou operacionais. Por exemplo, foi investigado o sistema de carregamento do leito que mostrou que a configuração ideal do leito é de 50% de alumina seguido de 50% de zeólita. Variáveis do processo foram também analisadas, a pressão de adsorção, a vazão de alimentação e o tempo do ciclo de adsorção, mostrando que o aumento da vazão de alimentação leva a perda da especificação que pode ser retomada reduzindo-se o tempo do ciclo de adsorção. Mostrou-se também que uma pressão de adsorção maior leva a uma maior remoção de contaminantes.

We consider the quanti fied constraint satisfaction problem (QCSP) which is to decide, given a structure and a first-order sentence (not assumed here to be in prenex form) built from conjunction and quanti fication, whether or not the sentence is true on the structure. We present a proof system for certifying the falsity of QCSP instances and develop its basic theory; for instance, we provide an algorithmic interpretation of its behavior. Our proof system places the established Q-resolution proof system in a broader context, and also allows us to derive QCSP tractability results.

EXTRACT (SEE PDF FOR FULL ABSTRACT): Catch of coho salmon off the coast of Washington and Oregon since 1925 appears to be related to large-scale events in the atmosphere, which in turn affect ocean currents and coastal upwelling intensities in the northeastern Pacific. At least two time scales of variations can be identified. The first is that of the El Nino/Southern Oscillation phenomenon giving rise to an irregular cycle of between 3 to 7 years. ... The second time scale of variation seems to have a periodicity of about 20 years, although this is based on a limited dataset. ... This paper endeavors to describe how, if real, these atmospheric/oceanic effects are integrated and might affect the salmon catch. The possibility must also be considered that the atmospheric events are symbiotically related to the oceanic events and, further, that both may be enmeshed in even longer-term variability of climate.
