The analytical vectorial structure of HGB is investigated in the far field based on the vector plane wave spectrum and the method of stationary phase. The energy flux distributions of HGB in the far-field, which is composed of TE term and TM term, are demonstrated. The physics pictures of HGB is illustrated from the vectorial structure, which is important to understand the theoretical aspects of both scalar and vector HGB propagation. Diversas aplicações industriais relevantes envolvem os processos de adsorção, citando como exemplos a purificação de produtos, separação de substâncias, controle de poluição e umidade entre outros. O interesse crescente pelos processos de purificação de biomoléculas deve-se principalmente ao desenvolvimento da biotecnologia e à demanda das indústrias farmacêutica e química por produtos com alto grau de pureza. O leito móvel simulado (LMS) é um processo cromatográfico contínuo que tem sido aplicado para simular o movimento do leito de adsorvente, de forma contracorrente ao movimento do líquido, através da troca periódica das posições das correntes de entrada e saída, sendo operado de forma contínua, sem prejuízo da pureza das correntes de saída. Esta consiste no extrato, rico no componente mais fortemente adsorvido, e no rafinado, rico no componente mais fracamente adsorvido, sendo o processo particularmente adequado a separações binárias. O objetivo desta tese é estudar e avaliar diferentes abordagens utilizando métodos estocásticos de otimização para o problema inverso dos fenômenos envolvidos no processo de separação em LMS. Foram utilizados modelos discretos com diferentes abordagens de transferência de massa, com a vantagem da utilização de um grande número de pratos teóricos em uma coluna de comprimento moderado, neste processo a separação cresce à medida que os solutos fluem através do leito, isto é, ao maior número de vezes que as moléculas interagem entre a fase móvel e a fase estacionária alcançando assim o equilíbrio. A modelagem e a simulação verificadas nestas abordagens permitiram a avaliação e a identificação das principais características de uma unidade de separação do LMS. A aplicação em estudo refere-se à simulação de processos de separação do Baclofen e da Cetamina. Estes compostos foram escolhidos por estarem bem caracterizados na literatura, estando disponíveis em estudos de cinética e de equilíbrio de adsorção nos resultados experimentais. De posse de resultados experimentais avaliou-se o comportamento do problema direto e inverso de uma unidade de separação LMS visando comparar os resultados obtidos com os experimentais, sempre se baseando em critérios de eficiência de separação entre as fases móvel e estacionária. Os métodos estudados foram o GA (Genetic Algorithm) e o PCA (Particle Collision Algorithm) e também foi feita uma hibridização entre o GA e o PCA. Como resultado desta tese analisouse e comparou-se os métodos de otimização em diferentes aspectos relacionados com o mecanismo cinético de transferência de massa por adsorção e dessorção entre as fases sólidas do adsorvente.

A incorporação de fósforo (P) foi avaliada através da técnica de marcador enzimático fluorescente a fim de determinar a atividade da fosfatase alcalina (PA) em dois clones de Phaeodactylum tricornutum (Bohlin), Ub3 e Ub7, isolados de Ubatuba (SP), em Tetraselmis aff. chui (Butcher) e Prorocentrum minimum (Pavillard) J. Schiller, isoladas da Baía de Guanabara (RJ) e na Comunidade Natural da Baía de Guanabara (RJ). A fosfatase alcalina (PA) é uma enzima extracelular associada à membrana que catalisa a hidrólise de compostos orgânicos de fósforo em resposta à limitação de fosfato. Sua análise, a partir do marcador ELF-97, proporciona uma avaliação individual e, portanto, determina as condições nutricionais de fósforo inorgânico em células fitoplanctônicas. Os clones de P. tricornutum apresentaram diferenças no desenvolvimento quando incubados no tratamento P-repleto. O clone Ub7 de P. tricornutum apresentou a maior atividade enzimática quando comparado às demais espécies testadas, em condições P-repletas. Enquanto P. minimum apresentou a maior atividade da fosfatase alcalina em condições P-limitadas. Entre as espécies T. aff. chui e P. minimum, a maior atividade enzimática ocorreu durante a fase estacionária de desenvolvimento, entretanto diferenças foram observadas somente nas menores concentrações de fosfato. P. tricornutum, T. aff. chui e P. minimum, ao longo dos experimentos, utilizaram duas estratégias para incorporação de fosfato, aumentando a atividade da fosfatase alcalina, assim como alterando o biovolume ou a máxima dimensão linear para manter a relação S/V estável. Em P. tricornutum os sítios da atividade enzimática ocorreram na membrana celular, em T. aff. chui encontrados intracelularmente, enquanto em P. minimum observados tanto nas membranas, quanto no interior das células. No experimento realizado com a comunidade natural, houve predomínio das diatomáceas entre todos os grupos e tratamentos; as espécies foram agrupadas nas estratégias adaptativas C e R e classificadas principalmente como R-estrategistas. Os dinoflagelados da Ordem Prorocentrales utilizaram a incorporação do fósforo orgânico como estratégia para obtenção de fósforo em condições limitantes. Entretanto, as diatomáceas apresentaram tal estratégia de forma mais variável. Quanto às prasinofíceas, embora Tetraselmis sp. tenha apresentado baixa atividade enzimática nos experimentos unialgais, as concentrações de fosfato ao longo do experimento não resultaram na utilização de P orgânico para o grupo. Os resultados destacaram as diferenças intra e interespecíficas na atividade da fosfatase alcalina, e, consequentemente, na incorporação de fósforo orgânico, uma vez que as espécies testadas regularam a atividade enzimática de acordo com as diferentes concentrações externas de fosfato.

In this paper, we report on the flexoelastic and viscoelastic ratios for a number of bimesogens compounds with the same generic structure. Values are obtained indirectly by measuring the flexoelectro-optic response in the chiral nematic phase. By varying the molecular structure we alter the bend angle, transverse dipole moment, and length of the molecule. First, to examine the influence of the bend angle we use a homologous series whereby the only alteration in the molecular structure is the number of methylene units in the aliphatic spacer, n . Results show that the flexoelastic ratio, e/K , and the effective flexoelectric coefficient, e , both exhibit an odd-even effect with values for n=odd being greater than that for n=even . This is understood in terms of an increase in the bend angle of the molecule and an increase in the transverse dipole moment. Second, in order to investigate the impact of the dipole moment, we have altered the mesogenic units so as to vary the longitudinal dipole moment and used different linkages in the aliphatic spacer in an attempt to alter the transverse dipole moment. Qualitatively, the results demonstrate that the odd-spaced bimesogen with larger transverse dipole moments exhibit larger flexoelastic ratios.

The growth of three microalgae species, viz., Nannochloropsis oculata, Tetraselmis chui and Chaetoceros muelleri which are commonly used in aquaculture, was investigated using three different inorganic nutrient media: (i) Modified Guillard's f/2 medium (ii) Rix Mix medium and (iii) BFRI medium. Each microalgae species was cultured for 24 days in small- scale with initial inoculation density of 17xl04 cell /ml in the three media with triplicates. N. oculata cultured in modified Guillard's f/2 medium showed superior growth with a mean peak density of 221 ±4.24 x 104 cell/ ml, to Rix Mix medium (141 ± 10.54xl04 cell/ml) and BFRI medium (47±4.94 x 104 cell/ml) on the 16th day of culture at stationary phase. Considering the increase in cell density for 20 days of culture in Rix Mix medium, C. muelleriwas significantly (P<0.05) highest than in other two media. N. oculata cultured in BFRI medium resulted in the poorest growth with a mean peak increase in density of 84±9.19 x 104 cell/ml in 12 days of culture. However, with an increase in cell density, growth of T. chui (182 ± 6.26 x 104 cell/ml) was significantly (P<0.05) higher in BFRI medium than in modified Guillard's f/2 medium. The results of the present study suggest that N. oculata and C. muelleri can be grown very well in both the modified Guillard's f/2 medium and Rix Mix medium. Better growth of T. chui can be obtained while culturing either in BFRI and Rix Mix medium. These three nutrient media used in the present study may be useful for microalgae species culture for establishing green-water culture for suitable target zooplankton, and fish and crustacean larvae in marine and brackishwater hatcheries.

Biomicrocapsules mean microscopic living organisms which carry important nutrients, very essential for the growth and development of aquatic organisms as well as other animals. Among these biomicrocapsules, Chlorella ellipsoidea, an important green microalga (Chlorophyceae) which contains 40-45% crude protein, 12-16% crude lipid, 14-15% minerals, colour pigments, vitamins and carotene. The microalga, C. ellipsoidea was cultured in four different dilutions of supernatant of digested sugar mill effluent (DSME) i.e. 25, 50, 75 and 100% DSME and Bold basal medium (BBM) as control in laboratory condition. Maximum cell growth and chlorophyll a content of C. ellipsoidea were obtained on l0th day of culture in supernatant of 50% diluted DSME followed by those of this biomicrocapsule grown in BBM, and 75, 25 and 100% DSME at stationary phase. Cell number had highly (p<0.01) direct correlation with chlorophyll a (r = 0.889) of C. ellipsoidea, and optical density (r = 0.926) of media. Chlorophyll a was also highly (p<0.01) and directly correlated with optical density (r= 0.877) of media. The specific growth rates (µ/day) of cell and chlorophyll a of C. ellipsoidea grown in supernatant of 50% DSME were significantly (p<0.01) varied from those of C. ellipsoidea cultured in BBM followed by other DSME. Total biomass of C. ellipsoidea cultured in supernatant of 50% DSME was found significantly (p<0.01) higher than that of this microalga cultured in BBM, and supernatant of 25, 75 and 100% DSME. Similar trend was also observed in the case of optical density. The physico-chemical properties of media were varied with the growth of cell of this microalga. It was recorded that cell number, chlorophyll a of biomicrocapsule, and optical density of media were highly (p<0.01) and directly correlated with pH, hardness and alkalinity, and inversely correlated with nitrate-N. Crude protein and crude lipid of C. ellipsoidea grown in supernatant of 50% DSME were significantly (p<0.01) higher than those of C. ellipsoidea cultured in other DSME and BBM. Due to best growth performance exhibited by this microalga grown in supernatant of 50% DSME, it may be used to grow in supernatant of 50% DSME to get more essential nutrients than that cultured in supernatant of other DSME media.

Abstract A theoretical model is developed for the sound scattered when a sound wave is incident on a cambered aerofoil at non-zero angle of attack. The model is based on the linearization of the Euler equations about a steady subsonic flow, and is an adaptation of previous work which considered incident vortical disturbances. Only high-frequency sound waves are considered. The aerofoil thickness, camber and angle of attack are restricted such that the steady flow past the aerofoil is a small perturbation to a uniform flow. The singular perturbation analysis identifies asymptotic regions around the aerofoil; local 'inner' regions, which scale on the incident wavelength, at the leading and trailing edges of the aerofoil; Fresnel regions emanating from the leading and trailing edges of the aerofoil due to the coalescence of singularities and points of stationary phase; a wake transition region downstream of the aerofoil leading and trailing edge; and an outer region far from the aerofoil and wake. An acoustic boundary layer on the aerofoil surface and within the transition region accounts for the effects of curvature. The final result is a uniformly-valid solution for the far-field sound; the effects of angle of attack, camber and thickness are investigated. © 2013 Cambridge University Press.

The effects of temperature and light on the growth and geosmin production of Lyngbya kuetzingii were determined. Of the three temperatures tested, 10, 25 and 35A degrees C, the maximal geosmin concentration and geosmin productivity were yielded at 10A degrees C, while the highest chl a production was observed at 25A degrees C. In the studies on light intensity, the maximal geosmin concentration and geosmin productivity were observed at 10 mu mol m(-2) s(-1), while the highest chl a production was at 20 mu mol m(-2) s(-1). It was suggested that more geosmin was synthesized with lower chl a demand. Meanwhile, the relative amounts of extra- and intracellular geosmin were investigated. Under optimum growth conditions (20 mu mol m(-2) s(-1), 25A degrees C; BG-11 medium), the amounts of extracellular geosmin increased as the growth progressed and reached the maximum in the stationary phase, while the intracellular geosmin reached its maximum value in the late exponential phase, and then began to decline. However, under the low temperature (10A degrees C) or light (10 mu mol m(-2) s(-1)) conditions, more intracellular geosmin was synthesized and mainly accumulated in the cells. The proportions of extracellular geosmin were high, to 33.33 and 32.27%, respectively, during the stationary phase at 35A degrees C and 20 mu mol m(-2) s(-1). It was indicated that low temperature or light could stimulate geosmin production and favor the accumulation of geosmin in cells, while more intracellular geosmin may be released into the medium at higher temperatures or optimum light intensity.

The growth and toxin content of the dinoflagellate Alexandrium tamarense ATHK was markedly affected by culture methods. In early growth phase at lower cell density static or mild agitation methods were beneficial to growth, but continuous agitation or aeration, to some extent, had an adverse effect on cell growth. Static culture in 2 L Erlenmeyer flasks had the highest growth rate (0.38 d(-1)) but smaller cell size compared with other culture conditions. Cells grown under aerated conditions possessed low nitrogen and phosphorus cell yields, namely high N and P cell-quota. At day 18, cells grown in continuous agitated and 1 h aerated culture entered the late stationary phase and their cellular toxin contents were higher (0.67 and 0.54 pg cell(-1)) compared with cells grown by other culture methods (0.27-0.49 pg cell(-1)). The highest cell density and cellular toxin content were 17190 cells mL(-1) and 1.26 pg cell(-1) respectively in an airlift photobioreactor with two-step culture. The results indicate that A. tamarense could be grown successfully in airlift photobioreactor by a two-step culture method, which involved cultivating the cells statically for 4 days and then aerating the medium. This provides an efficient way to enhance cell and toxin yield of A. tamarense.

The effects of salt stress on carbohydrate metabolism in Microcoleus vaginatus Gom., a cyanobacterium isolated from desert algal crusts, were investigated in the present study. Extracellular total carbohydrates and exopolysaccharides (EPS) in the culture medium produced by M. vaginatus increased significantly during the growth phase and reached a maximum during the stationary phase. The production of extracellular carbohydrates also significantly increased under higher salt concentrations, which was attributed to an increase in low molecular weight carbohydrates. In the presence of NaCl, the production of cellular total carbohydrates decreased and photosynthetic activity was impaired, whereas cellular reducing sugars, water-soluble sugars and sucrose content and sucrose phosphate synthase activity increased, reaching a maximum in the presence of 200 mmol/L NaCl. These parameters were restored to original levels when the algae were transferred to a non-saline medium. Sodium and K+ concentrations of stressed cells decreased significantly and H+-ATPase activity increased after the addition of exogenous sucrose or EPS. The results suggest that EPS and sucrose are synthesized to maintain the cellular osmotic equilibrium between the intra- and extracellular environment, thus protecting algal cells from osmotic damage, which was attributed to the selective exclusion of cellular Na+ and K+ by H+-ATPase.

Microcystis aeruginosa Kutz. 7820 was cultured at 350 and 700 muL.L-1 CO2 to assess the impacts of doubled atmospheric CO2 concentration on this bloom-forming cyanobacterium. Doubling Of CO2 concentration in the airflow enhanced its growth by 52%-77%, with pH values decreased and dissolved inorganic carbon (DIC) increased in the medium. Photosynthetic efficiencies and dark respiratory rates expressed per unit chl a tended to increase with the doubling of CO2. However, saturating irradiances for photosynthesis and light-saturated photosynthetic rates normalized to cell number tended to decrease with the increase of DIC in the medium. Doubling of CO2 concentration in the airflow had less effect on DIC-saturated photosynthetic rates and apparent photosynthetic affinities for DIC. In the exponential phase, CO2 and HCO3- levels in the medium were higher than those required to saturate photosynthesis. Cultures with surface aeration were DIC limited in the stationary phase. The rate of CO2 dissolution into the liquid increased proportionally when CO2 in air was raised from 350 to 700 muL.L-1, thus increasing the availability of DIC in the medium and enhancing the rate of photosynthesis. Doubled CO2 could enhance CO2 dissolution, lower pH values, and influence the ionization fractions of various DIC species even when the photosynthesis was not DIC limited. Consequently, HCO3- concentrations in cultures were significantly higher than in controls, and the photosynthetic energy cost for the operation of CO2 concentrating mechanism might decrease.

A column method was developed to conveniently and reliably determine the soil organic partition coefficients (K-oc) of three insecticides (methiocarb, azinphos-methyl, fenthion), four fungicides (triadimenol, fuberidazole, tebuconazole, pencycuron), and one herbicide (atrazine), in which real soil acted as a stationary phase and the water solution of pesticide as an eluent. The processes of sorption equilibrium were directly shown through a breakthrough curve(BTC). The log K-oc values are 1.69, 1.95, 2.25, 2.55, 2.69, 2.67, 3.10, and 3.33 for atrazine, triadimenol, methiocarb, fuberidazole, azinphos-methyl, tebuconazole, fenthion and pencycuron, respectively.

It is reported that when a light beam travels through a slab of left-handed medium in the air, the lateral shift of the transmitted beam can be negative as well as positive. The necessary condition for the lateral shift to be positive is given. The validity of the stationary-phase approach is demonstrated by numerical simulations for a Gaussian-shaped beam. A restriction to the slab's thickness is provided that is necessary for the beam to retain its profile in the traveling. It is shown that the lateral shift of the reflected beam is equal to that of the transmitted beam in the symmetric configuration.

A novel monolithic stationary phase having long alkyl chain ligands was introduced and evaluated in pressurized-capillary electrochromatography of small neutral and charged compounds. The monolithic column was prepared by the in situ copolymerization of ethylene dimethacrylate, 1-hexadecene, allyl alcohol and 2-acrylamido-2-methyl-1-propanesulfonic acid in a quaternary porogenic solvent mixture consisting of 1,4-butanediol, cyclohexanol, dodecanol and water.

To avoid the limitation of the widely used prediction methods of soil organic carbon partition coefficients (K-OC) from hydrophobic parameters, e.g., the n-octanol/water partition coefficients (K-OW) and the reversed phase high performance liquid chromatographic (RP-HPLC) retention factors, the soil column liquid chromatographic (SCLC) method was developed for K-OC prediction. The real soils were used as the packing materials of RP-HPLC columns, and the correlations between the retention factors of organic compounds on soil columns (k(soil)) and K-OC measured by batch equilibrium method were studied. Good correlations were achieved between k(soil) and K-OC for three types of soils with different properties. All the square of the correlation coefficients (R-2) of the linear regression between log k(soi) and log K-OC were higher than 0.89 with standard deviations of less than 0.21. In addition, the prediction of K-OC from K-OW and the RP-HPLC retention factors on cyanopropyl (CN) stationary phase (k(CN)) was comparatively evaluated for the three types of soils. The results show that the prediction of K-OC from k(CN) and K-OW is only applicable to some specific types of soils. The results obtained in the present study proved that the SCLC method is appropriate for the K-OC prediction for different types of soils, however the applicability of using hydrophobic parameters to predict K-OC largely depends on the properties of soil concerned.
