High bone mass (HBM) can be an incidental clinical finding; however, monogenic HBM disorders (eg, LRP5 or SOST mutations) are rare. We aimed to determine to what extent HBM is explained by mutations in known HBM genes. A total of 258 unrelated HBM cases were identified from a review of 335,115 DXA scans from 13 UK centers. Cases were assessed clinically and underwent sequencing of known anabolic HBM loci: LRP5 (exons 2, 3, 4), LRP4 (exons 25, 26), SOST (exons 1, 2, and the van Buchem's disease [VBD] 52-kb intronic deletion 3'). Family members were assessed for HBM segregation with identified variants. Three-dimensional protein models were constructed for identified variants. Two novel missense LRP5 HBM mutations ([c.518C>T; p.Thr173Met], [c.796C>T; p.Arg266Cys]) were identified, plus three previously reported missense LRP5 mutations ([c.593A>G; p.Asn198Ser], [c.724G>A; p.Ala242Thr], [c.266A>G; p.Gln89Arg]), associated with HBM in 11 adults from seven families. Individuals with LRP5 HBM ( approximately prevalence 5/100,000) displayed a variable phenotype of skeletal dysplasia with increased trabecular BMD and cortical thickness on HRpQCT, and gynoid fat mass accumulation on DXA, compared with both non-LRP5 HBM and controls. One mostly asymptomatic woman carried a novel heterozygous nonsense SOST mutation (c.530C>A; p.Ser177X) predicted to prematurely truncate sclerostin. Protein modeling suggests the severity of the LRP5-HBM phenotype corresponds to the degree of protein disruption and the consequent effect on SOST-LRP5 binding. We predict p.Asn198Ser and p.Ala242Thr directly disrupt SOST binding; both correspond to severe HBM phenotypes (BMD Z-scores +3.1 to +12.2, inability to float). Less disruptive structural alterations predicted from p.Arg266Cys, p.Thr173Met, and p.Gln89Arg were associated with less severe phenotypes (Z-scores +2.4 to +6.2, ability to float). In conclusion, although mutations in known HBM loci may be asymptomatic, they only account for a very small proportion ( approximately 3%) of HBM individuals, suggesting the great majority are explained by either unknown monogenic causes or polygenic inheritance.

A detailed study is presented of the expected performance of the ATLAS detector. The reconstruction of tracks, leptons, photons, missing energy and jets is investigated, together with the performance of b-tagging and the trigger. The physics potential for a variety of interesting physics processes, within the Standard Model and beyond, is examined. The study comprises a series of notes based on simulations of the detector and physics processes, with particular emphasis given to the data expected from the first years of operation of the LHC at CERN.

Tässä tutkimuksessa tutkittiin kanadalaisen kirjailijan L. M. Montgomeryn (1874 1942) elämää vuosina 1921 1927. Kyseisinä vuosina Montgomery eli presbyteeripapin vaimona maaseutuseurakunnissa, ensin Leaskdalessa ja sitten Norvalissa Kanadan Ontariossa. Tutkimus keskittyi tarkastelemaan kirjailijan elämää hallinnutta ristiriitaa, joka muodostui uskonnollisten ihanteiden ja yksilöllisen kokemuksen välisistä eroavaisuuksista. Primäärilähteinä olivat Montgomeryn toimitetut ja julkaistut päiväkirjat The Selected Journals of L. M. Montgomery 1 5 (1985 2004), joista käytettiin etenkin tutkimusvuosiin keskittyvää kolmatta osaa, Montgomeryn neljä romaania Emily of New Moon (1923), Emily Climbs (1925), Blue Castle (1926) ja Emily's Quest (1927) sekä Montgomeryn toimitettu ja julkaistu kirjeenvaihto After Green Gables: L. M. Montgomery's Letters to Ephraim Weber , 1916 1941 (2006) ja My Dear Mr. M.: Letters to G. B. MacMillan (1980). Lisäksi lähteinä oli Leaskdalen seurakuntalaisten kokoama muistelmateos Montgomerysta. Kirjallisuutta käytettiin kirkkohistorian, kirjallisuudentutkimuksen ja naistutkimuksen alalta. Suositusta kirjailijasta on tehty useampia elämäkertoja ja niitä hyödynnettiin tutkimuksessa. Tutkimus jakautui kahteen päälukuun, jotka toimivat toistensa vastaparina. Ensimmäinen tutkimusluku keskittyi tarkastelemaan uskonnollisia ja viktoriaanisia ihanteita, joiden mukaisesti Montgomery oli saanut kasvatuksen. Hän pyrki täyttämään näitä ihanteita myös pappilan emäntänä. Toinen tutkimusluku tarkasteli kirjailijan elämää, uskonnollisia ajatuksia ja avioliittoa julkisivun takana, jolloin kävi selvästi ilmi kirjailijan kahtiajakautunut elämä. Montgomery sai presbyteerisen ja viktoriaanisen kasvatuksen ja oppi jo lapsena huomioimaan sen, mitä muut ihmiset ajattelevat hänestä. Hän eli pienissä maaseutuyhteisöissä, joissa juoruilu oli yksi kontrollin väline. Papin vaimolla oli velvollisuuksia seurakunnassa ja pappilan hoidossa, ja Montgomery hoiti ne tarmokkaasti ja velvollisuudentuntoisesti. Kirjailijan ajattelussa on yhtymäkohtia transsendentalismiin, teosofiaan ja darwinismiin. Hän oli innokas psykologisen kirjallisuuden lukija ja sovelsi oppejaan käytäntöön. Hän uskoi unien välittävän viestejä. Kirkkokunta oli hänelle kuitenkin merkittävä identiteetin rakentaja. Papin vaimon roolin ja yksityisesti kriittisen ajattelijan roolin yhteensovittaminen aiheutti Montogmerylle ongelmia ja väsymystä. Hän kätki uskonnolliset ajatuksensa ja yksinäisyyden aiheuttaman kivun päiväkirjaansa, jonka hän toivoi julkaistavan viimeistään kuolemansa jälkeen. Kaikenlainen kirjoittaminen oli hänelle pakoa todellisuudesta ja auttoi häntä jaksamaan. Otsikon sitatti kuvaa Montgomeryn suhdetta kristinuskoon: hän käytti kristillisiä ilmauksia, mutta muokkasi niiden sisältöä omaa uskoaan vastaavaksi. Taivas ja helvetti kuvasivat usein hänen henkistä olotilaansa sanojen uskonnollisen merkityksen sijasta.

Two-wheelers (TW) constitute a major proportion of urban traffic in developing countries and therefore their effect on the saturation flow at signalized intersections could be substantial. This paper attempts to study and analyze the effect of two-wheelers on the saturation flow of signalized intersections by collecting data at a few signalized intersections in Bangalore, India. A strong correlation is observed between the measured saturation flow and the proportion of two-wheeler traffic, which suggest that two-wheelers have significant impact and should be considered in the capacity analysis of signalized intersections. In this paper, the effect of two-wheelers on saturation flow rate is incorporated in a previous model by calibrating and introducing a new adjustment factor for two-wheelers. Results show that saturation flow measured using the modified HCM equation is closer to observed saturation flow values.

Chemokines help to establish cerebral inflammation after ischemia, which comprises a major component of secondary brain injury. The CXCR4 chemokine receptor system induces neural stem cell migration, and hence has been implicated in brain repair. We show that CXCR1 and interleukin-8 also stimulate chemotaxis in murine neural stem cells from the MHP36 cell line. The presence of CXCR1 was confirmed by reverse transcriptase PCR and immunohistochemistry. Interleukin-8 evoked intracellular calcium currents, upregulated doublecortin (a protein expressed by migrating neuroblasts), and elicited positive chemotaxis in vitro. Therefore, effectors of the early innate immune response may also influence brain repair mechanisms.

BACKGROUND: GABA(A) receptors are members of the Cys-loop family of neurotransmitter receptors, proteins which are responsible for fast synaptic transmission, and are the site of action of wide range of drugs. Recent work has shown that Cys-loop receptors are present on immune cells, but their physiological roles and the effects of drugs that modify their function in the innate immune system are currently unclear. We are interested in how and why anaesthetics increase infections in intensive care patients; a serious problem as more than 50% of patients with severe sepsis will die. As many anaesthetics act via GABA(A) receptors, the aim of this study was to determine if these receptors are present on immune cells, and could play a role in immunocompromising patients. PRINCIPAL FINDINGS: We demonstrate, using RT-PCR, that monocytes express GABA(A) receptors constructed of α1, α4, β2, γ1 and/or δ subunits. Whole cell patch clamp electrophysiological studies show that GABA can activate these receptors, resulting in the opening of a chloride-selective channel; activation is inhibited by the GABA(A) receptor antagonists bicuculline and picrotoxin, but not enhanced by the positive modulator diazepam. The anaesthetic drugs propofol and thiopental, which can act via GABA(A) receptors, impaired monocyte function in classic immunological chemotaxis and phagocytosis assays, an effect reversed by bicuculline and picrotoxin. SIGNIFICANCE: Our results show that functional GABA(A) receptors are present on monocytes with properties similar to CNS GABA(A) receptors. The functional data provide a possible explanation as to why chronic propofol and thiopental administration can increase the risk of infection in critically ill patients: their action on GABA(A) receptors inhibits normal monocyte behaviour. The data also suggest a potential solution: monocyte GABA(A) receptors are insensitive to diazepam, thus the use of benzodiazepines as an alternative anesthetising agent may be advantageous where infection is a life threatening problem.

A fifth-order theory for solving the problem of interaction between Stokes waves and exponential profile currents is proposed. The calculated flow fields are compared with measurements. Then the errors caused by the linear superposition method and approximate theory are discussed. It is found that the total wave-current field consists of pure wave, pure current and interaction components. The shear current not only directly changes the flow field, but also indirectly does sx, by changing the wave parameters due to wave-current interaction. The present theory can predict the wave kinematics on shear currents satisfactorily. The linear superposition method may give rise to more than 40% loading error in extreme conditions. When the apparent wave period is used and the Wheeler stretching method is adopted to extrapolate the current, application of the approximate theory is the best.
