Breast cancer is a leading contributor to the burden of disease in Australia. Fortunately, the recent introduction of diverse therapeutic strategies have improved the survival outcome for many women. Despite this, the clinical management of breast cancer remains problematic as not all approaches are sufficiently sophisticated to take into account the heterogeneity of this disease and are unable to predict disease progression, in particular, metastasis. As such, women with good prognostic outcomes are exposed to the side effects of therapies without added benefit. Furthermore, women with aggressive disease for whom these advanced treatments would deliver benefit cannot be distinguished and opportunities for more intensive or novel treatment are lost. This study is designed to identify novel factors associated with disease progression, and the potential to inform disease prognosis. Frequently overlooked, yet common mediators of disease are the interactions that take place between the insulin-like growth factor (IGF) system and the extracellular matrix (ECM). Our laboratory has previously demonstrated that multiprotein insulin-like growth factor-I (IGF-I): insulin-like growth factor binding protein (IGFBP): vitronectin (VN) complexes stimulate migration of breast cancer cells in vitro, via the cooperative involvement of the insulin-like growth factor type I receptor (IGF-IR) and VN-binding integrins. However, the effects of IGF and ECM protein interactions on the dissemination and progression of breast cancer in vivo are unknown. It was hypothesised that interactions between proteins required for IGF induced signalling events and those within the ECM contribute to breast cancer metastasis and are prognostic and predictive indicators of patient outcome. To address this hypothesis, semiquantitative immunohistochemistry (IHC) analyses were performed to compare the extracellular and subcellular distribution of IGF and ECM induced signalling proteins between matched normal, primary cancer, and metastatic cancer among archival formalin-fixed paraffin-embedded (FFPE) breast tissue samples collected from women attending the Princess Alexandra Hospital, Brisbane. Multivariate Cox proportional hazards (PH) regression survival models in conjunction with a modified „purposeful selection of covariates. method were applied to determine the prognostic potential of these proteins. This study provides the first in-depth, compartmentalised analysis of the distribution of IGF and ECM induced signalling proteins. As protein function and protein localisation are closely correlated, these findings provide novel insights into IGF signalling and ECM protein function during breast cancer development and progression. Distinct IGF signalling and ECM protein immunoreactivity was observed in the stroma and/or in subcellular locations in normal breast, primary cancer and metastatic cancer tissues. Analysis of the presence and location of stratifin (SFN) suggested a causal relationship in ECM remodelling events during breast cancer development and progression. The results of this study have also suggested that fibronectin (FN) and ¥â1 integrin are important for the formation of invadopodia and epithelial-to-mesenchymal transition (EMT) events. Our data also highlighted the importance of the temporal and spatial distribution of IGF induced signalling proteins in breast cancer metastasis; in particular, SFN, enhancer-of-split and hairy-related protein 2 (SHARP-2), total-akt/protein kinase B 1 (Total-AKT1), phosphorylated-akt/protein kinase B (P-AKT), extracellular signal-related kinase-1 and extracellular signal-related kinase-2 (ERK1/2) and phosphorylated-extracellular signal-related kinase-1 and extracellular signal-related kinase-2 (P-ERK1/2). Multivariate survival models were created from the immunohistochemical data. These models were found to fit well with these data with very high statistical confidence. Numerous prognostic confounding effects and effect modifications were identified among elements of the ECM and IGF signalling cascade and corroborate the survival models. This finding provides further evidence for the prognostic potential of IGF and ECM induced signalling proteins. In addition, the adjusted measures of associations obtained in this study have strengthened the validity and utility of the resulting models. The findings from this study provide insights into the biological interactions that occur during the development of breast tissue and contribute to disease progression. Importantly, these multivariate survival models could provide important prognostic and predictive indicators that assist the clinical management of breast disease, namely in the early identification of cancers with a propensity to metastasise, and/or recur following adjuvant therapy. The outcomes of this study further inform the development of new therapeutics to aid patient recovery. The findings from this study have widespread clinical application in the diagnosis of disease and prognosis of disease progression, and inform the most appropriate clinical management of individuals with breast cancer.

Virus-like particle-based vaccines for high-risk human papillomaviruses (HPVs) appear to have great promise; however, cell culture-derived vaccines will probably be very expensive. The optimization of expression of different codon-optimized versions of the HPV-16 L1 capsid protein gene in plants has been explored by means of transient expression from a novel suite of Agrobacterium tumefaciens binary expression vectors, which allow targeting of recombinant protein to the cytoplasm, endoplasmic reticulum (ER) or chloroplasts. A gene resynthesized to reflect human codon usage expresses better than the native gene, which expresses better than a plant-optimized gene. Moreover, chloroplast localization allows significantly higher levels of accumulation of L1 protein than does cytoplasmic localization, whilst ER retention was least successful. High levels of L1 (>17% total soluble protein) could be produced via transient expression: the protein assembled into higher-order structures visible by electron microscopy, and a concentrated extract was highly immunogenic in mice after subcutaneous injection and elicited high-titre neutralizing antibodies. Transgenic tobacco plants expressing a human codon-optimized gene linked to a chloroplast-targeting signal expressed L1 at levels up to 11% of the total soluble protein. These are the highest levels of HPV L1 expression reported for plants: these results, and the excellent immunogenicity of the product, significantly improve the prospects of making a conventional HPV vaccine by this means. © 2007 SGM.

This study investigated the effects of high-intensity interval training (HIIT) vs. work-matched moderate-intensity continuous exercise (MOD) on metabolism and counterregulatory stress hormones. In a randomized and counterbalanced order, 10 well-trained male cyclists and triathletes completed a HIIT session [81.6 ± 3.7% maximum oxygen consumption (V̇o2 max); 72.0 ± 3.2% peak power output; 792 ± 95 kJ] and a MOD session (66.7 ± 3.5% V̇o2 max; 48.5 ± 3.1% peak power output; 797 ± 95 kJ). Blood samples were collected before, immediately after, and 1 and 2 h postexercise. Carbohydrate oxidation was higher (P = 0.037; 20%), whereas fat oxidation was lower (P = 0.037; −47%) during HIIT vs. MOD. Immediately after exercise, plasma glucose (P = 0.024; 20%) and lactate (P < 0.01; 5.4×) were higher in HIIT vs. MOD, whereas total serum free fatty acid concentration was not significantly different (P = 0.33). Targeted gas chromatography-mass spectromtery metabolomics analysis identified and quantified 49 metabolites in plasma, among which 11 changed after both HIIT and MOD, 13 changed only after HIIT, and 5 changed only after MOD. Notable changes included substantial increases in tricarboxylic acid intermediates and monounsaturated fatty acids after HIIT and marked decreases in amino acids during recovery from both trials. Plasma adrenocorticotrophic hormone (P = 0.019), cortisol (P < 0.01), and growth hormone (P < 0.01) were all higher immediately after HIIT. Plasma norepinephrine (P = 0.11) and interleukin-6 (P = 0.20) immediately after exercise were not significantly different between trials. Plasma insulin decreased during recovery from both HIIT and MOD (P < 0.01). These data indicate distinct differences in specific metabolites and counterregulatory hormones following HIIT vs. MOD and highlight the value of targeted metabolomic analysis to provide more detailed insights into the metabolic demands of exercise.

Several genetic variants are thought to influence white matter (WM) integrity, measured with diffusion tensor imaging (DTI). Voxel based methods can test genetic associations, but heavy multiple comparisons corrections are required to adjust for searching the whole brain and for all genetic variants analyzed. Thus, genetic associations are hard to detect even in large studies. Using a recently developed multi-SNP analysis, we examined the joint predictive power of a group of 18 cholesterol-related single nucleotide polymorphisms (SNPs) on WM integrity, measured by fractional anisotropy. To boost power, we limited the analysis to brain voxels that showed significant associations with total serum cholesterol levels. From this space, we identified two genes with effects that replicated in individual voxel-wise analyses of the whole brain. Multivariate analyses of genetic variants on a reduced anatomical search space may help to identify SNPs with strongest effects on the brain from a broad panel of genes.

In vitro methyiation of Escherichia coli transfer ribonucleic acid by cell free extracts of Mycobacterium smegmatis leads exclusively to the formation of 1-methyl adenine [Vani, B. R., Ramakrishnan, T., Taya, Y., Noguchi, S., Yamaiuzumi, Z. and Nishimura, S.(1978) J. Bact., 137,1085]. We have studied the effect of this modification on aminoacylation of Escherichia coli tRNA by mycobacterial enzymes. Aminoacylation with total algal protein hydrolysate as well as several individual aminoacids like methionine, valine, tyrosine, aspartic acid and lysine were monitored. In all the cases methyiation had a positive effect on the extent of aminoacylation by mycobacterial enzymes. Decreased aminoacylation in vitro was observed when hypomethylated transfer RNA from ethionine treated cells was used as the substrate for aminoacylation.

Atopic dermatitis (AD) or atopic eczema is characterised by a superficial skin inflammation with an overall Th2 cell dominance and impaired function of the epidermal barrier. Patients also are at an increased risk for asthma and allergic rhinitis. Treatment with tacrolimus ointment inhibits T cell activation and blocks the production of several inflammatory cytokines in the skin, without suppressing collagen synthesis. The aims of this thesis were to determine: (1) long-term efficacy, safety, and effects on cell-mediated immunity and serum IgE levels in patients with moderate-to-severe AD treated for 1 year with tacrolimus ointment or a corticosteroid regimen, (2) the 10-year outcome of eczema, respiratory symptoms, and serum IgE levels in AD patients initially treated long-term with tacrolimus ointment, and (3) pharmacokinetics and long-term safety and efficacy of 0.03% tacrolimus ointment in infants under age 2 with AD. Cell-mediated immunity, reflecting Th1 cell reactivity, was measured by recall antigens and was at baseline lower in patients with AD compared to healthy controls. Treatment with either 0.1% tacrolimus ointment or a corticosteroid regimen for one year enhanced recall antigen reactivity. Transepidermal water loss (TEWL), an indicator of skin barrier function, decreased at months 6 and 12 in both tacrolimus- and corticosteroid-treated patients; TEWL for the head and neck was significantly lower in tacrolimus-treated patients. Patients in the 10-year open follow-up study showed a decrease in affected body surface area from a baseline 19.0% to a 10-year 1.6% and those with bronchial hyper-responsiveness at baseline showed an increase in the provocative dose of inhaled histamine producing a 15% decrease in FEV1, indicating less hyper-responsiveness. Respiratory symptoms (asthma and rhinitis) reported by the patient decreased in those with active symptoms at baseline. A good treatment response after one year of tacrolimus treatment predicted a good treatment response throughout the 10-year follow-up and a decrease in total serum IgE levels at the 10-year follow-up visit. The 2-week pharmacokinetic and the long-term study with 0.03% tacrolimus ointment showed good and continuous improvement of AD in the infants. Tacrolimus blood levels were throughout the study low and treatment well tolerated. This thesis underlines the importance of effective long-term topical treatment of AD. When the active skin inflammation decreases, cell-mediated immunity of the skin improves and a secondary marker for Th2 cell reactivity, total serum IgE, decreases. Respiratory symptoms seem to improve when the eczema area decreases. All these effects can be attributed to improvement of skin barrier function. One potential method to prevent a progression from AD to asthma and allergic rhinitis may be avoidance of early sensitisation through the skin, so early treatment of AD in infants is crucial. Long-term treatment with 0.03% tacrolimus ointment was effective and safe in infants over age 3 months.

The growth responses and yield of Heterotis niloticus on artificial diets of varying protein levels were studied in a bid to assess the implication of feeding Heterotis in intensive fish production venture for a rearing period of 84 days. One hundred and twenty juvenile H.niloticus were fed for 12 weeks on 28%, 31%, 34% and 37% dietary crude protein levels. The fish were reared in 4 concrete tanks stocked at the rate of 10 fish per M super(2) 100,00/hectare). Consequently, the weight gain, food conversion ratio, serum protein and albumin-globulin ratio were determined to assess the growth and state of health of the fish. The yield was appraised through economic considerations of cost of production of fish and diets (feed). The varying crude protein levels significantly influenced mean weight gain, percentage weight gain and food conversion ratio however, the 37% crude protein in diet produced the best growth. The serum protein was highest in fish raised on 31% crude protein diet while the highest value was recorded for albumin-globulin ratio on diet containing 34% crude protein. The yield from treatments 1, 2, 3, and 4 were 114.38 of/84 days 571.9kg/g hect);146.79g/84 days 733.95kg/hect), respectively. However, treatment 3 recorded the highest value for profit index

O aumento da longevidade evidenciou nas mulheres os efeitos deletérios da deficiência ovariana como, a obesidade central e as doenças neurodegenerativas. Os hábitos alimentares modernos favorecem o consumo excessivo de gorduras, que contribuem para efeitos adversos sobre a saúde, incluindo alteração da composição corporal e da função nervosa. Assim, objetivamos estudar como a dieta hiperlipídica (DH), contendo óleo de soja, associada à perda da função gonadal influenciam o comportamento e o desenvolvimento corporal de ratas adultas. Ratas Wistar adultas foram ovariectomizadas (OVX) ou pseudo-operadas (C) e após 7d passaram a receber dieta contendo 4%(normo) ou 19%(hiperlípidica) de óleo de soja: C4 (n=29), OVX4 (n=30), C19 (n=30), OVX19 (n=30). Foram avaliados a massa e o comprimento corporais e a ingestão alimentar, e aspectos comportamentais de ansiedade, através de teste no Labirinto em Cruz Elevado (LCE); de busca pela novidade, por testes no Campo Vazado (CV); e aprendizagem/memória avaliada no labirinto aquático radial de 8 braços (LAROB). Ao final dos testes, os animais foram anestesiados, exsanguinados e o tecido adiposo intra-abdominal foi coletado e pesado. O soro foi utilizado para dosagens de triglicerídeo (TG), VLDL-c, colesterol (COL), HDL-c e estradiol. A ingestão alimentar média e o comprimento corporal não diferiram entre os grupos, mas o grupo OVX19 mostrou ganho significativo (13,6%) de massa corporal mais precocemente que os demais. O mesmo padrão foi observado quanto à massa de tecido adiposo, que foi 86% maior no OVX19. Os TG, VLDL e COL aumentaram cerca de 28% no OVX19, porém sem alteração de HDL-c (33,762,2 mg/dl). A DH e a ovariectomia se mostraram ansiogênicos e quando associados, a DH parece reverter o efeito ansiogênico da castração. Padrão similar de resposta foi observado na resposta ao teste de busca por novidade. No teste de aprendizagem/memória o OVX4 apresentou maior tempo de latência no primeiro dia, e todos responderam de forma semelhante nos demais dias. Conclui-se que a deficiência de estradiol acompanhada da ingestão de dieta hiperlipídica mantém o desempenho cognitivo, o comportamento emocional e a motivação, embora prejudique a adiposidade e o metabolismo lipídico.

O Aspergillus fumigatus é o principal agente etiológico da aspergilose invasiva, uma infecção fúngica oportunista que acomete, principalmente, pacientes de Unidades Hematológicas, como aqueles com neutropenia profunda e prolongada. Após a filamentação este fungo angioinvasivo é capaz de ativar e causar danos em células endoteliais de veia umbilical humana (HUVEC) que passam a expressar um fenótipo pró-trombótico. A ativação destas células, dependente de contato célulacélula, é mediada por TNF-α e caracterizada pela expressão de moléculas próinflamatórias, como citocinas, quimiocinas e moléculas de adesão. Recentemente, nosso grupo comparou a ativação endotelial de HUVECs desafiadas com cepas selvagens e uma cepa mutante para o gene UGM1. Nestes experimentos a cepa mutante Δugm1, que apresenta um fenótipo de maior produção de galactosaminogalactana (GAG) na parede celular, mostrou um fenótipo hiperadesivo e uma capacidade maior de ativar células endoteliais. Entretanto, os receptores e as vias de sinalização envolvidos nesta ativação permanecem desconhecidos. Assim, o objetivo deste trabalho foi verificar as proteínas envolvidas nestes processos através do estudo das proteínas diferencialmente expressas nas HUVECs após a interação com A. fumigatus, usando a técnica proteômica 2D-DIGE. Brevemente, as HUVECs foram infectadas com tubos germinativos da cepa selvagem (AF293) e da cepa Δugm1 de A. fumigatus. Em seguida, as proteínas foram marcadas com diferentes fluorocromos e separadas por eletroforese bidimensional. A análise quantitativa foi realizada utilizando o software DeCyder. Foram identificadas por MS/MS cinco proteínas diferencialmente expressas, incluindo a galectina-1 e a anexina A2, ambas mais expressas após a interação, sendo a primeira ~25% mais expressa após a interação com a mutante Δugm1. Este trabalho propõe que a galectina-1 poderia ser o receptor endotelial para polímeros de galactose presentes na parede celular do A. fumigatus, e que a Anexina A2 poderia estar envolvida na sinalização intracelular em resposta a este patógeno. No entanto, experimentos complementares, em curso, são necessários para comprovar esta hipótese.

A elaboração de um programa de treinamento físico depende do controle de diferentes parâmetros bioquímicos, que se relacionam com fatores como a intensidade do exercício, imunidade e com o estado redox. Além disso, estudos recentes também começaram a apontar a relevância da função executiva como componente determinante para o alcance de um alto nível de desempenho esportivo. Por outro lado, poucos estudos foram realizados em atletas até o momento utilizando estes marcadores na saliva juntamente com os testes de função cognitiva. O objetivo desse trabalho foi estudar a capacidade discriminatória das análises bioquímicas realizadas em saliva para avaliação do desempenho físico e sua relação com a função cognitiva em atletas de futebol. Trinta e dois atletas foram submetidos ao Bangsbo Sprint Test (BST) para avaliação da capacidade física e 48 horas depois ao Teste de Stroop (TSt) e Torre de Hanoi (ToH) para avaliação da função executiva. Os níveis de lactato na saliva aumentaram quando comparados aos valores Pré-BST (6,9 vezes; p<0,05). A proteína total salivar seguiu o mesmo padrão com aumento observado após o BST (+34%; p<0,05). As concentrações de imunoglobulina-A salivar (IgA-s) não mostraram diferença significativa após o BST. Os níveis de GSH e TBARs na saliva não mostraram diferença significativa, enquanto que a concentração de ácido úrico diminuiu após o BST (-26%; p<0,05). Interessantemente, a superóxido dismutase (SOD) salivar aumentou (3,6 vezes; p<0,05), enquanto que os níveis de catalase (CAT) na saliva não alteraram significativamente. Não houve correlação de nenhum dos parâmetros analisados com o desempenho no TSt, entretanto atletas localizados no percentil superior (P90) de cortisol na saliva (11,2 ng/ mL à 32,7 ng/ mL) apresentaram tempos mais longos para a resolução do ToH. A eletroforese 2D mostrou que 215 spots só apareceram no momento Pré-BST, 63 spots aumentaram e 108 diminuíram a sua expressão após o BST. Concluindo, a saliva é sensível às modificações induzidas pelo BST. A manutenção dos níveis salivares de TBARs após o BST parece ocorrer em função da diminuição dos níveis de ácido úrico, componente este que possui uma expressiva ação antioxidante. Neste sentido, o aumento de SOD pós-exercício parece agir como uma segunda linha de defesa antioxidante contra a produção de ROS induzidas pelo BST. Além disso, os resultados dos testes de função executiva indicam que níveis elevados de cortisol salivar possuem um efeito deletério no tempo de resolução da ToH, que se refere à memória de trabalho, o planejamento e solução de problemas. No entanto o TSt, que envolve a atenção seletiva e a velocidade de processamento de informações parecem não ser afetados pelos níveis de cortisol em repouso. E finalmente, a eletroforese 2D mostrou que o BST induziu a expressão diferencial de proteínas, visto que não surgiram proteínas novas após o teste, e dezenas de proteínas foram up-reguladas e down-reguladas após o BST. Estes dados sugerem que após uma análise proteômica, estas proteínas possam ser candidatas a marcadores de desempenho físico e/ou cognitivo em futuros estudos.

Porphyrin metabolic disruption from exposure to xenobiotic contaminants such as heavy metals, dioxins, and aromatic hydrocarbons can elicit overproduction of porphyrins. Measurement of porphyrin levels, when used in conjunction with other diagnostic assays, can help elucidate an organism’s physiological condition and provide evidence for exposure to certain toxicants. A sensitive microplate fluorometric assay has been optimized for detecting total porphyrin levels in detergent solubilized protein extracts from symbiotic, dinoflagellate containing cnidarian tissues. The denaturing buffer used in this modified assay contains a number of potentially interfering components (e.g., sodium dodecyl sulfate (SDS), dithiothreitol (DTT), protease inhibitors, and chlorophyll from the symbiotic zooxanthellae), which required examination and validation. Examination of buffer components were validated for use in this porphyrin assay; while the use of a specific spectrofluorometric filter (excitation 400 ± 15 nm; emission 600 ± 20 nm) minimized chlorophyll interference. The detection limit for this assay is 10 fmol of total porphyrin per μg of total soluble protein and linearity is maintained up to 5000 fmol. The ability to measure total porphyrins in a SDS protein extract now allows a single extract to be used in multiple assays. This is an advantage over classical methods, particularly when tissue samples are limiting, as is often the case with coral due to availability and collection permit restrictions.

Juvenile chinook salmon, Oncorhynchus tshawytscha, from natal streams in California’s Central Valley demonstrated little estuarine dependency but grew rapidly once in coastal waters. We collected juvenile chinook salmon at locations spanning the San Francisco Estuary from the western side of the freshwater delta—at the confluence of the Sacramento and San Joaquin Rivers—to the estuary exit at the Golden Gate and in the coastal waters of the Gulf of the Farallones. Juveniles spent about 40 d migrating through the estuary at an estimated rate of 1.6 km/d or faster during their migration season (May and June 1997) toward the ocean. Mean growth in length (0.18 mm/d) and weight (0.02 g/d) was insignificant in young chinook salmon while in the estuary, but estimated daily growth of 0.6 mm/d and 0.5 g/d in the ocean was rapid (P≤0.001). Condition (K factor) declined in the estuary, but improved markedly in ocean fish. Total body protein, total lipid, triacylglycerols (TAG), polar lipids, cholesterol, and nonesterified fatty acids concentrations did not change in juveniles in the estuary, but total lipid and TAG were depleted in ocean juveniles. As young chinook migrated from freshwater to the ocean, their prey changed progressively in importance from invertebrates to fish larvae. Once in coastal waters, juvenile salmon appear to employ a strategy of rapid growth at the expense of energy reserves to increase survival potential. In 1997, environmental conditions did not impede development: freshwater discharge was above average and water temperatures were only slightly elevated, within the species’ tolerance. Data suggest that chinook salmon from California’s Central Valley have evolved a strong ecological propensity for a ocean-type life history. But unlike populations in the Pacific Northwest, they show little estuarine dependency and proceed to the ocean to benefit from the upwelling-driven, biologically productive coastal waters.

植物与昆虫的互作关系是个长期进化的过程,虫害给农业生产带来巨大损失。本研究以甘蓝型油菜(Brassica napus)为例,研究了不同环境条件和遗传背景下外源基因的表达与效用,同时利用蛋白质组技术,研究了虫害损伤模拟条件下植物可能存在的内源抗性机制。甘蓝型油菜中转入了人工合成的Bt(Bacillus thuringiensis)杀虫基因,能使植物产生抗虫蛋白抵御虫害。我们在湖北湖南两个实验点进行了大田实验,按植株生长发育的4个不同时期从转基因植株的叶片上采样,研究抗虫蛋白在植物体内的表达动态。植株顶部第三片展开叶的Bt毒蛋白浓度在结荚期前随植物生长而不断增加,而在结荚期出现或增或减的现象。采样叶片的可溶性总蛋白浓度含量一直呈增加的趋势,直到结荚以后出现含量的明显降低。同时,收集了转基因油菜与湘油15号在田间自然杂交形成的杂交后代种子用于栽培,用GFP仪检测杂交后代的绿色荧光蛋白(green fluorescent protein),并用聚合酶链式反应(polymerase chain reaction, PCR)检测并确认带有转基因的杂交植株。为了检测带有转基因的杂交后代油菜中Bt毒蛋白的杀虫效率,用对Bt毒蛋白敏感的试虫品系——初孵棉铃虫幼虫(Helicoverpa armigera)进行杀虫活性检测实验。结果表明,携带Bt基因的杂交湘油及其转基因亲本对试虫的体重增长量均产生了负面影响,可以推断在调查取样的植株生长发育阶段,转基因杂交后代与其转基因亲本植株的杀虫效率没有显著差异。转基因植物及其杂交后代中抗虫蛋白的持续表达及田间带有转基因的自播植物的出现会使害虫产生耐受抗性的潜在可能性增加。 相对于人为增加的抗虫基因,植物在长期对抗昆虫的过程中也进化形成了自我防御机制,能够产生特异的抗性蛋白来应对昆虫的取食。本研究用机械损伤模拟害虫取食,对比了油菜受到物理损伤前后可溶性总蛋白的含量变化并试图通过蛋白质组学技术来检测可能发生变化的蛋白质。Bradford定量测定发现,同一植株同一叶片损伤前后可溶性总蛋白含量差异显著,损伤后蛋白表达量显著增高。蛋白质组双向凝胶电泳及其差异分析显示,损伤前后有8个蛋白质点发生明显的上调或下调。选择其中2个差异蛋白点经过MALDI-TOF质谱鉴定,它们分别是Rubisco小亚基前体以及果糖-1,6-二磷酸醛缩酶和粪卟啉-3-氧化酶的混合物,这些蛋白质在其他植物的抗逆研究中也有报道,它们可能在油菜叶片应答机械损伤过程中对维持植物的生理功能也有重要作用。

The sublethal exposure (0.24 ppm) of Nuvan on some biochemical compositions such as serum protein, blood glucose, glutamate oxaloacetate transaminase (AST) and glutamate pyruvate transaminase (ALT) and on some hematological parameters such as red blood corpuscles (RBC), white blood corpuscles (WBC), hemoglobin content (Hb), mean corpuscular concentration (MCV), mean corpuscular hemoglobin concentration (MCHC) of Catla catla fingerlings were studied. The hematological and biochemical parameters evoked a significant reduction (excepting MCV, ALT and AST which is significantly increased) with increasing days of Nuvan exposure.

The fishery sector in Uganda has seen important changes in the last two decades. Among the changes registered, is the expansion of fish markets locally, regionally and internationally. Upon which, remarkable benefits have been realized at local and national levels, for instance, it is estimated that an average of 40m$ is being earned annually as foreign exchange. Besides, presently fish accounts for over 50% of total animal protein in take. However, it is argued that sustaining these gains has become an up hill task due to failure to maintain fish quality as a result of the rudimentary and inappropriate sanitary, fish handling and artisanal fish processing practices that both directly and indirectly affect the quality of fish and fish products. Therefore, against this background, a study of 507 respondents was undertaken among the Lake Victoria Communities specifically in Wakiso, Mayuge and Mukono districts. The study examined the perceptions of fishers on the social cultural practices of sanitation, fish handling and artisanal fish processing and consequently identified factors that influenced these practices.
