Lake Kinneret (LK) is a relatively fresh water take situated in the Dead Sea Rift (DSR) Valley. The pore water (PW) in the sediments underlying LK pelagic zone have significantly higher salinity than that of the lake. The concentrations of major ion solutes (Cl, Br, Na, K, Mg) in PW from six 2.4 m to 5.1 m long sediment cores increase linearly with depth, indicating the occurrence of saline, deep seated brines. The upper part of the PW column is affected by the much fresher boundary with LK water and in most cores is characterized by gradually increasing Br/Cl and decreasing Na, Mg, K/Cl molar ratios, which tend to stabilize at about 2.0 m below the sediment surface. The 'stable' molar ratios in the deeper PW vary spatially and are supposed to represent the ratios in the deep underlying brines at each site. When plotted as Na/Cl vs. Br/Cl, the stable ratios of the northern and central part of the lake fall close to a straight line which characterizes many of the brines in the DSR Valley. However, the respective ratios in the southern part of the lake fall markedly off the DSR line. Moreover, Na/Cl and K/Cl molar ratios in the south are significantly higher than in the central and northern parts. delta Cl-37 measured in present LK water is ca. 0.0 parts per thousand. Along the PW column at the lake center, delta Cl-37 is becoming more positive with depth, reaching values of about +0.5 parts per thousand to +0.6 parts per thousand at 3 m depth. Even more positive values (+0.7 parts per thousand to +0.8 parts per thousand) are detected further north, in PW from deeper sediment layers. In contrast, in PW from the southeastern part of the lake, delta Cl-37 is becoming more negative with depth (-1.0 parts per thousand at similar to 2.6 m). It is suggested that these isotopic differences are also indicative of spatial variability in the PW brine sources. O-18 and D values in the PW of all 3 m long cores are similar and resemble the respective levels in LK. The source of H2O in 3 m deep, bed sediments is claimed to be the overlying lake water, and therefore water isotopes do not provide a clue regarding the original water isotopic composition in the underlying brines. PW from the southeast with higher K/Cl and Na/Cl but lower concentrations of these solutes, suggest leaching by meteoric water of sub-surface halite and post-halite salt formations, while the more saline PW from the northern and central parts, that have lower K/Cl and Na/Cl, and higher Br/Cl, are similar to DSR brines and represent underlying residual brines. The number of solute-binding protein-dependent transporters in rhizobia is dramatically increased compared with the majority of other bacteria so far sequenced. This increase may be due to the high affinity of solute-binding proteins for solutes, permitting the acquisition of a broad range of growth-limiting nutrients from soil and the rhizosphere. The transcriptional induction of these transporters was studied by creating a suite of plasmid and integrated fusions to nearly all ATP-binding cassette (ABC) and tripartite ATP-independent periplasmic (TRAP) transporters of Sinorhizobium meliloti. In total, specific inducers were identified for 76 transport systems, amounting to approximate to 47% of the ABC uptake systems and 53% of the TRAP transporters in S. meliloti. Of these transport systems, 64 are previously uncharacterized in Rhizobia and 24 were induced by solutes not known to be transported by ABC- or TRAP-uptake systems in any organism. This study provides a global expression map of one of the largest transporter families (transportome) and an invaluable tool to both understand their solute specificity and the relationships between members of large paralogous families.

Polarized epithelial cells are responsible for the vectorial transport of solutes and have a key role in maintaining body fluid and electrolyte homeostasis. Such cells contain structurally and functionally distinct plasma membrane domains. Brush border and basolateral membranes of renal and intestinal epithelial cells can be separated using a number of different separation techniques, which allow their different transport functions and receptor expressions to be studied. In this communication, we report a proteomic analysis of these two membrane segments, apical and basolateral, obtained from the rat renal cortex isolated by two different methods: differential centrifugation and free-flow electrophoresis. The study was aimed at assessing the nature of the major proteins isolated by these two separation techniques. Two analytical strategies were used: separation by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) at the protein level or by cation-exchange high-performance liquid chromatography (HPLC) after proteolysis (i.e., at the peptide level). Proteolytic peptides derived from the proteins present in gel pieces or from HPLC fractions after proteolysis were sequenced by on-line liquid chromatography-tandem mass spectrometry (LC-MS/MS). Several hundred proteins were identified in each membrane section. In addition to proteins known to be located at the apical and basolateral membranes, several novel proteins were also identified. In particular, a number of proteins with putative roles in signal transduction were identified in both membranes. To our knowledge, this is the first reported study to try and characterize the membrane proteome of polarized epithelial cells and to provide a data set of the most abundant proteins present in renal proximal tubule cell membranes.

The aim of this review paper is to present experimental methodologies and the mathematical approaches used to determine effective diffusivities of solutes in food materials. The paper commences by describing the diffusion phenomena related to solute mass transfer in foods and effective diffusivities. It then focuses on the mathematical formulation for the calculation of effective diffusivities considering different diffusion models based on Fick's second law of diffusion. Finally, experimental considerations for effective diffusivity determination are elucidated primarily based on the acquirement of a series of solute content versus time curves appropriate to the equation model chosen. Different factors contributing to the determination of the effective diffusivities such as the structure of food material, temperature, diffusion solvent, agitation, sampling, concentration and different techniques used are considered. Polymer-stabilised liquid crystals are systems in which a small amount of monomer is dissolved within a liquid crystalline host, and then polymerised in situ to produce a network. The progress of the polymerisation, performed within electro-optic cells, was studied by establishing an analytical method novel to these systems. Samples were prepared by photopolymerisation of the monomer under well-defined reaction conditions; subsequent immersion in acetone caused the host and any unreacted monomer to dissolve. High performance liquid chromatography was used to separate and detect the various solutes in the resulting solutions, enabling the amount of unreacted monomer for a given set of conditions to be quantified. Longer irradiations cause a decrease in the proportion of unreacted monomer since more network is formed, while a more uniform LC director alignment (achieved by decreasing the sample thickness) or a higher level of order (achieved by decreasing the polymerisation temperature) promotes faster reactions.

Solution calorimetry offers a reproducible technique for measuring the enthalpy of solution (ΔsolH) of a solute dissolving into a solvent. The ΔsolH of two solutes, propranolol HCl and mannitol were determined in simulated intestinal fluid (SIF) solutions designed to model the fed and fasted states within the gut, and in Hanks’ balanced salt solution (HBSS) of varying pH. The bile salt and lipid within the SIF solutions formed mixed micelles. Both solutes exhibited endothermic reactions in all solvents. The ΔsolH for propranolol HCl in the SIF solutions differed from those in the HBSS and was lower in the fed state than the fasted state SIF solution, revealing an interaction between propranolol and the micellar phase in both SIF solutions. In contrast, for mannitol the ΔsolH was constant in all solutions indicating minimal interaction between mannitol and the micellar phases of the SIF solutions. In this study, solution calorimetry proved to be a simple method for measuring the enthalpy associated with the dissolution of model drugs in complex biological media such as SIF solutions. In addition, the derived power–time curves allowed the time taken for the powdered solutes to form solutions to be estimated.

Experimental evidence suggests that nitric oxide (NO) and hydrogen sulfide (H2S) signaling pathways are intimately intertwined, with mutual attenuation or potentiation of biological responses in the cardiovascular system and elsewhere. The chemical basis of this interaction is elusive. Moreover, polysulfides recently emerged as potential mediators of H2S/sulfide signaling, but their biosynthesis and relationship to NO remain enigmatic. We sought to characterize the nature, chemical biology, and bioactivity of key reaction products formed in the NO/sulfide system. At physiological pH, we find that NO and sulfide form a network of cascading chemical reactions that generate radical intermediates as well as anionic and uncharged solutes, with accumulation of three major products: nitrosopersulfide (SSNO−), polysulfides, and dinitrososulfite N-nitrosohydroxylamine-N-sulfonate (SULFI/NO), each with a distinct chemical biology and in vitro and in vivo bioactivity. SSNO− is resistant to thiols and cyanolysis, efficiently donates both sulfane sulfur and NO, and potently lowers blood pressure. Polysulfides are both intermediates and products of SSNO− synthesis/decomposition, and they also decrease blood pressure and enhance arterial compliance. SULFI/NO is a weak combined NO/nitroxyl donor that releases mainly N2O on decomposition; although it affects blood pressure only mildly, it markedly increases cardiac contractility, and formation of its precursor sulfite likely contributes to NO scavenging. Our results unveil an unexpectedly rich network of coupled chemical reactions between NO and H2S/sulfide, suggesting that the bioactivity of either transmitter is governed by concomitant formation of polysulfides and anionic S/N-hybrid species. This conceptual framework would seem to offer ample opportunities for the modulation of fundamental biological processes governed by redox switching and sulfur trafficking.

The quadrupolar hyperfine interactions of in-diffused (111)In -> (111)Cd probes in polycrystalline isostructural Zr(4)Al(3) and Hf(4)Al(3) samples containing small admixtures of the phases (Zr/Hf)(3)Al(2) were investigated. A strong preference of (111)In solutes for the contaminant (Zr/Hf)(3)Al(2) minority phases was observed. Detailed calculations of the electric field gradient (EFG) at the Cd nucleus using the full-potential augmented plane wave + local orbital formalism allowed us to assign the observed EFG fractions to the various lattice sites in the (Zr/Hf)(3)Al(2) compounds and to understand the preferential site occupation of the minority phases by the (111)In atoms. The effects of the size of the supercell and relaxation around the oversized In and Cd probe atoms were investigated in detail.

The viscosity of ionic liquids based on quaternary ammonium cations is reduced when one of the alkyl chains is replaced by an alkoxy chain (Zhou et al. Chem. Eur. J. 2005, 11, 752.). A microscopic picture of the role played by the ether function in decreasing the viscosity of quaternary ammonium ionic liquids is provided here by molecular dynamics (MD) simulations. A model for the ionic liquid N-ethyl-N,N-dimethyl-N-(2-methoxyethyl)ammonium bis(trifluoromethanesulfonyl)imide, MOENM(2)E TFSI, is compared to the tetraalky-lammonium counterpart. The alkoxy derivative has lower viscosity, higher ionic diffusion coefficients, and higher conductivity than the tetraalkyl system at the same density and temperature. A clear signature of the ether function on the liquid structure is observed in cation-cation correlations, but not in anion-anion or anion-cation correlations. In both the alkyl and the alkoxy ionic liquids, there is aggregation of long chains of neighboring cations within micelle-like structures. The MD simulations indicate that the less effective assembly between the more flexible alkoxy chains, in comparison to alkyl chains, is the structural reason for higher ionic mobility in MOENM(2)E TFSI.

O objetivo deste trabalho foi avaliar os indicadores fisiológicos da interação entre deficit hídrico e acidez do solo em plantas jovens de cana-de-açúcar. As plantas foram submetidas a três tratamentos de disponibilidade hídrica, medidos em percentagem de capacidade de campo (CC) - sem estresse (70% CC), estresse moderado (55% CC) e estresse severo (40% CC); e três tratamentos de acidez no solo, medidos em termos de saturação por bases (V) - baixa acidez (V = 55%), média acidez (V = 33%) e alta acidez (V = 23%). O experimento foi realizado em casa de vegetação a 29,7±4,3ºC e 75±10% UR. O delineamento experimental utilizado foi o de blocos ao acaso, em esquema fatorial 3x3, com quatro repetições. Após 60 dias, foram determinados os teores de solutos compatíveis - trealose, glicina betaína e prolina - na folha diagnóstico e o crescimento inicial da parte aérea. Os solutos compatíveis trealose, glicina betaína e prolina são indicadores do efeito da interação dos estresses hídrico e ácido no solo. O acúmulo dos solutos compatíveis nos tecidos foliares das plantas não é capaz de impedir a redução na produção de matéria seca da cana-de-açúcar, resultante do agravamento nas condições de disponibilidade hídrica e de acidez no solo.

Com o objetivo de avaliar os efeitos de thidiazuron e de ácido giberélico nas características dos cachos de uvas 'Rubi', foi conduzido um experimento, utilizando-se de thidiazuron a 5 e 10 mg.L-1 e ácido giberélico a 20mg.L-1, combinados ou não. As aplicações dos produtos foram realizadas aos 14; 21 ou 28 dias após o pleno florescimento, por meio de imersão dos cachos. Todos os tratamentos com reguladores de crescimento aumentaram a massa dos cachos. A massa dos bagos e dos engaços foi identicamente influenciada pela aplicação dos produtos, porém menos evidente, quando as aplicações foram realizadas aos 28 dias após o pleno florescimento. As aplicações de thidiazuron a 5mg.L-1, aos 14 ou 21 dias após o florescimento, não diferiram das aplicações de ácido giberélico para as variáveis estudadas. Não houve diferenças significativas para as variáveis teor de sólidos solúveis totais, acidez titulável, porém os tratamentos com thidiazuron retardaram a maturação em até 7 dias.

The objective of this work was to evaluate conditions the effectiveness of acetolactate synthase (ALS) and protoporphyrinogen oxidase (PROTOX) inhibitors in the Bidens pilosa control under two water deficit conditions, as well as to determine the influence of water deficit under the contents of soluble carbohydrates and protein of weed. The experimental design was randomized completely design with the treatments, with four replications, setup in a factorial scheme 4x2, with four herbicides (fomesafen lactofen, chlorimuron-ethyl and imazethapyr), and two water conditions (-0.5 MPa and -0.01MPa). At 7, 14, 21 and 28 days after application (DAA), was assessed visually control efficiency of herbicides. For the determination of organic solutes plants were collected at 24, 48, 72 and 96 hours after application (HAA). Herbicides controlled efficiently plants of B. pilosa, except lactofen, whereas the water deficit affected the efficiency of control of all the herbicides. The soluble carbohydrate content increased with the imposition of water deficit, however, herbicides and water deficit further reduction of soluble proteins in plants of B. pilosa.

O objetivo deste estudo foi avaliar a eficácia dos herbicidas inibidores da ALS e PROTOX no controle de Ipomoea grandifolia sob dois níveis de déficit hídrico, bem como determinar a influência desse déficit hídrico sobre o conteúdo de carboidratos solúveis, proteínas totais e aminoácidos livres da planta daninha. O trabalho foi conduzido em casa de vegetação, no delineamento experimental inteiramente casualizado, com os tratamentos dispostos em esquema fatorial 4 x 2, sendo quatro herbicidas (fomesafen, lactofen, chlorimuron-ethyl e imazethapyr) e dois níveis de déficit hídrico (com e sem déficit: -0,5 MPa e -0,01 MPa, respectivamente). Aos 7, 14, 21 e 28 dias após a aplicação, o controle foi avaliado de forma visual, e a determinação dos solutos orgânicos foi feita 24, 48, 72 e 96 horas após a aplicação dos herbicidas. À exceção de chlorimuron-ethyl, os demais herbicidas controlaram eficientemente I. grandifolia. A eficiência de controle dos herbicidas foi afetada pela condição de déficit hídrico. Os herbicidas elevaram o conteúdo de carboidratos solúveis, independentemente da condição hídrica, e proporcionaram redução de proteínas solúveis nas plantas de I. grandifolia, bem como aumentaram as concentrações de aminoácidos livres.
