A cDNA and corresponding promoter region for a naturally occurring, feedback-insensitive anthranilate synthase (AS) α-subunit gene, ASA2, has been isolated from an unselected, but 5-methyl-tryptophan-resistant (5MTr), tobacco (Nicotiana tabacum) cell line (AB15–12-1). The ASA2 cDNA contains a putative transit peptide sequence, and Southern hybridization shows that more than one closely related sequence is present in the tobacco genome. The ASA2 cDNA complemented a trpE nonsense mutant Escherichia coli strain, allowing growth on 300 μm 5MT-containing minimal medium without tryptophan, and cell extracts contained feedback-insensitive AS activity. The 5MTr was lost when the E. coli strain was transformed with an ASA2 site-directed mutant (phenylalanine-107-arginine-108 → serine-107-glutamine-108). Identical nucleotide sequences encoding the phenylalanine-107-arginine-108 region have been found in polymerase chain reaction-amplified 326-bp ASA2 genomic fragments of wild-type (5-methyl-tryptophan-sensitive [5MTs]) tobacco and a progenitor species. High-level ASA2 transcriptional expression was detected only in 5MTr-cultured cells, not in 5MTs cells or in plants. Promoter studies indicate that tissue specificity of ASA2 is controlled by the promoter region between −2252 and −607. Since the ASA2 promoter sequences are not substantially different in the 5MTr and 5MTs lines, the increased levels of ASA2 mRNA in the 5MTr lines are most likely due to changes in a regulatory gene affecting ASA2 expression.

Cancer is a disease characterized by defects in growth control, and tumor cells often display abnormal patterns of cellular differentiation. The combination of recombinant human fibroblast interferon and the antileukemic agent mezerein corrects these abnormalities in cultured human melanoma cells resulting in irreversible growth arrest and terminal differentiation. Subtraction hybridization identifies a melanoma differentiation associated gene (mda-7) with elevated expression in growth arrested and terminally differentiated human melanoma cells. Colony formation decreases when mda-7 is transfected into human tumor cells of diverse origin and with multiple genetic defects. In contrast, the effects of mda-7 on growth and colony formation in transient transfection assays with normal cells, including human mammary epithelial, human skin fibroblast, and rat embryo fibroblast, is quantitatively less than that found with cancer cells. Tumor cells expressing elevated mda-7 display suppression in monolayer growth and anchorage independence. Infection with a recombinant type 5 adenovirus expressing antisense mda-7 eliminates mda-7 suppression of the in vitro growth and transformed phenotype. The ability of mda-7 to suppress growth in cancer cells not expressing or containing defects in both the retinoblastoma (RB) and p53 genes indicates a lack of involvement of these critical tumor suppressor elements in mediating mda-7-induced growth inhibition. The lack of protein homology of mda-7 with previously described growth suppressing genes and the differential effect of this gene on normal versus cancer cells suggests that mda-7 may represent a new class of cancer growth suppressing genes with antitumor activity.

Vaccination with synthetic peptides representing cytotoxic T lymphocyte (CTL) epitopes can lead to a protective CTL-mediated immunity against tumors or viruses. We now report that vaccination with a CTL epitope derived from the human adenovirus type 5 E1A-region (Ad5E1A234-243), which can serve as a target for tumor-eradicating CTL, enhances rather than inhibits the growth of Ad5E1A-expressing tumors. This adverse effect of peptide vaccination was rapidly evoked, required low doses of peptide (10 micrograms), and was achieved by a mode of peptide delivery that induces protective T-cell-mediated immunity in other models. Ad5E1A-specific CTL activity could no longer be isolated from mice after injection of Ad5E1A-peptide, indicating that tolerization of Ad5E1A-specific CTL activity causes the enhanced tumor outgrowth. In contrast to peptide vaccination, immunization with adenovirus, expressing Ad5E1A, induced Ad5E1A-specific immunity and prevented the outgrowth of Ad5E1A-expressing tumors. These results show that immunization with synthetic peptides can lead to the elimination of anti-tumor CTL responses. These findings are important for the design of safe peptide-based vaccines against tumors, allogeneic organ transplants, and T-cell-mediated autoimmune diseases.

K+ channels, which have been linked to regulation of electrogenic solute transport as well as Ca2+ influx, represent a locus in hepatocytes for the concerted actions of hormones that employ Ca2+ and cAMP as intracellular messengers. Despite considerable study, the single-channel basis for synergistic effects of Ca2+ and cAMP on hepatocellular K+ conductance is not well understood. To address this question, patch-clamp recording techniques were applied to a model liver cell line, HTC hepatoma cells. Increasing the cytosolic Ca2+ concentration ([Ca2+]i) in HTC cells, either by activation of purinergic receptors with ATP or by inhibition of intracellular Ca2+ sequestration with thapsigargin, activated low-conductance (9-pS) K+ channels. Studies with excised membrane patches suggested that these channels were directly activated by Ca2+. Exposure of HTC cells to a permeant cAMP analog, 8-(4-chlorophenylthio)-cAMP, also activated 9-pS K+ channels but did not change [Ca2+]i. In excised membrane patches, cAMP-dependent protein kinase (the downstream effector of cAMP) activated K+ channels with conductance and selectivity identical to those of channels activated by Ca2+. In addition, cAMP-dependent protein kinase activated a distinct K+ channel type (5 pS). These data represent the differential regulation of low-conductance K+ channels by signaling pathways mediated by Ca2+ and cAMP. Moreover, since low-conductance Ca(2+)-activated K+ channels have been identified in a variety of cell types, these findings suggest that differential regulation of K+ channels by hormones with distinct signaling pathways may provide a mechanism for hormonal control of solute transport and Ca(2+)-dependent cellular functions in the liver as well as other nonexcitable tissues.

A new classification of microtidal sand and gravel beaches with very different morphologies is presented below. In 557 studied transects, 14 variables were used. Among the variables to be emphasized is the depth of the Posidonia oceanica. The classification was performed for 9 types of beaches: Type 1: Sand and gravel beaches, Type 2: Sand and gravel separated beaches, Type 3: Gravel and sand beaches, Type 4: Gravel and sand separated beaches, Type 5: Pure gravel beaches, Type 6: Open sand beaches, Type 7: Supported sand beaches, Type 8: Bisupported sand beaches and Type 9: Enclosed beaches. For the classification, several tools were used: discriminant analysis, neural networks and Support Vector Machines (SVM), the results were then compared. As there is no theory for deciding which is the most convenient neural network architecture to deal with a particular data set, an experimental study was performed with different numbers of neuron in the hidden layer. Finally, an architecture with 30 neurons was chosen. Different kernels were employed for SVM (Linear, Polynomial, Radial basis function and Sigmoid). The results obtained for the discriminant analysis were not as good as those obtained for the other two methods (ANN and SVM) which showed similar success.

Tartrate-resistant acid phosphatase (TRAP) is highly expressed in osteoclasts and in a subset of tissue macrophages and dendritic cells. It is expressed at lower levels in the parenchymal cells of the liver, glomerular mesangial cells of the kidney and pancreatic acinar cells. We have identified novel TRAP mRNAs that differ in their 5-untranslated region (5'-UTR) sequence, but align with the known murine TRAP mRNA from the first base of Exon 2. The novel 5'-UTRs represent alternative first exons located upstream of the known 5'-UTR. A similar genomic structure exists for the human TRAP gene with partial conservation of the exon and promoter sequences. Expression of the most distal 5'-UTR (Exon 1A) is restricted to adult bone and spleen tissue. Exon 1B is expressed primarily in tissues containing TRAP-positive nonhaematopoietic cells. The known TRAP 5'-UTR (Exon 1) is expressed in tissues characteristic of myeloid cell expression. In addition the Exon 1C promoter sequence is shown to comprise distinct transcription start regions, with an osteoclast-specific transcription initiation site identified downstream of a TATA-like element. Macrophages are shown to initiate transcription of the Exon 1C transcript from a purine-rich region located upstream of the osteoclast-specific transcription start point. The distinct expression patterns for each of the TRAP 5'-UTRs suggest that TRAP mRNA expression is regulated by the use of four alternative tissue- and cell-restricted promoters. The tartrate-resistant acid phosphatase (TRAP) is present in multiple tissues, including kidney, liver, lung, spleen, and bone. Recent study of (TRAP) gene expression has provided evidence for distinct promoters within the (TRAP) gene, suggesting that the gene has alternative, tissue-preferred mRNA transcripts. Examination of endogenous (TRAP) exon 1B and 1C mRNA transcripts revealed tissue-preferred transcript abundance with increased exon 1B transcripts detected in liver and kidney and increased exon 1C transcripts detected in bone and spleen. In this investigation, we have made transgenic mice that express a marker gene driven by two candidate promoters, designated BC and C, within the (TRAP) gene. The BC and C promoters are 2.2 and 1.6 kb, respectively, measured from the translation initiation site. Evaluation of BC transgenic lines demonstrated robust expression in multiple tissues. In contrast, significant transgene expression was not detected in C transgenic lines. Evaluation of transgene mRNAs in BC transgenic lines revealed that virtually all expression was in the form of B transcripts, suggesting that the tissue-preferred pattern of endogenous (TRAP) was not replicated in the BC transgenic line. Likewise, osteoclastogenic cultures from BC, but not C, transgenic bone marrow cells expressed the transgene following receptor activator of NFkappaB ligand/macrophage colony-stimulating factor stimulation. In conclusion, when compared with the 2.2-kb BC portion of the (TRAP) promoter region, the 1.6-kb C portion does not account for significant gene expression in vivo or in vitro; production of the bone- and spleen-preferred (TRAP) C transcript must depend on regulatory elements outside of the 2.2-kb promoter. As the majority of currently investigated transcription factors that influence transcriptional regulation of osteoclast gene expression bind within the 1.6-kb C portion of the (TRAP) promoter, it is likely that transcription binding sites outside of the 2.2-kb region will have profound effects on regulation of the gene in vivo and in vitro.

The stratigraphic and biogeographic distribution of more than 170 species of deep-water agglutinated benthic foraminifers (DWAF) from the North Atlantic and adjacent marginal seas has been compared with paleoenvironmental data (e.g. paleobathymetry, oxygenation of the bottom waters, amount of terrigenous input and substrate disturbance). Six general types of assemblages, in which deep water agglutinated taxa occur, are defined from the Turonian to Maastrichtian times: 1. High latitude slope assemblages 2. Low to mid latitude slope assemblages 3. Flysch-type assemblages 4. Deep water limestone assemblages (,,Scaglia,,-type) 5. Abyssal mixed calcareous-agglutinated assemblages 6. Abyssal purely agglutinated assemblages Latitudinal differences in faunal composition are observed, the most important of which is the lack or extreme paucity of calcareous forms in high latitude assemblages. East-to-west differences appear to be of comparatively minor importance. Most DWAF species occur in all studied regions and are thus considered as cosmopolitan. Biostratigraphic turnovers in the taxonomic content of assemblages are observed in the lowermost Turonian, mid-Campanian and in the upper Maastrichtian to lowermost Paleocene. These datum levels correspond to inter-regional and time-constant paleooceanographic events, which probably also affected the deep-water benthic biota. This allows us to use deep-water agglutinated foraminifers for biostratigraphy in the North Atlantic sequences deposited below CCD and to geographically extend the currently used zonal schemes which have been established in the Carpathian and Alpine areas.

Depuis 1992, la surveillance de la santé des mollusques marins du littoral français est assurée par le réseau de Pathologie des Mollusques (Repamo). Ses activités s’inscrivent dans le cadre de la Directive Européenne 2006/88/CE. Depuis son évaluation par la plateforme nationale d’épidémiosurveillance en santé animale en 2012, l’objectif de surveillance est la détection précoce des infections dues à des organismes pathogènes exotiques et émergents affectant les mollusques marins sauvages et d’élevage. L’année 2015 est la première année de transition pour laquelle un début d’évolution des modalités de surveillance de la santé des mollusques marins animées par l’Ifremer a été amorcé. Un dispositif hybride de surveillance a été mis en place, s’appuyant sur l’existant et intégrant des débuts d’évolution. La surveillance événementielle a constitué l’activité principale du dispositif en 2015 et s’est appuyée sur des réseaux existants : (1) la surveillance des mortalités observées sur des animaux sentinelles déployés sur les sites ateliers des réseaux Ifremer RESCO 2 (12 sites) pour l’huître creuse Crassostrea gigas et MYTILOBS 2 (8 sites) pour la moule bleue Mytilus edulis. Pour l’huître creuse Crassostrea gigas, la mortalité cumulée moyenne était de 50,3% (écart-type 10,9%) pour le naissain standardisé Ifremer (NSI), de 11,0% (écart-type 9,1%) pour les huîtres de 18 mois et de 7,3% (écart-type 5,6%) pour les huîtres de 30 mois. Les mortalités ont été observées principalement entre le début du mois de mai et la mi-juillet. Lors de ces épisodes de mortalité, des prélèvements d’animaux ont été réalisés en vue d’analyses diagnostiques : 7 prélèvements pour le NSI, 2 pour les huîtres de 18 mois et 1 pour les huîtres de 30 mois. Aucun agent réglementé n’a été détecté dans les échantillons d’huîtres creuses prélevés et analysés. Le virus OsHV-1 a été détecté dans les 7 échantillons analysés de NSI, dans 2 échantillons analysés d’huîtres de 18 mois et dans 1 échantillon analysé d’huîtres de 30 mois. La bactérie Vibrio aestuarianus a été détectée dans 5 échantillons analysés de NSI, dans 1 échantillon d’huîtres de 18 mois et dans 1’échantillon d’huîtres de 30 mois. Pour la moule bleue Mytilus edulis, des mortalités cumulées variant de 9% sur le site du Vivier à 51% sur le site des filières du Pertuis Breton ont été estimées. Les mortalités ont été observées au printemps sur des moules âgées d’une année et en automne sur des moules plus jeunes. Lors de ces épisodes de mortalités, des prélèvements d’animaux ont été réalisés en vue d’analyses diagnostiques : 2 prélèvements pour les moules d’une année et 1 pour les jeunes moules. Ces prélèvements ont eu lieu dans le Pertuis Breton. Aucun agent réglementé n’a été détecté dans les échantillons de moules prélevés et analysés. Des bactéries du groupe Splendidus ont été détectées dans les 3 échantillons de moules analysés. (2) la surveillance s’appuyant sur les déclarations de mortalités de mollusques par les conchyliculteurs et pêcheurs à pied professionnels auprès des Directions départementales des territoires et de la mer (DDTM). Cette modalité s’applique aux huîtres creuses et aux moules bleues lorsqu’il n’existe pas de site atelier RESCO 2 ou MYTILOBS 2 dans la zone où des mortalités sont déclarées par les conchyliculteurs ou pêcheurs à pied. Le réseau REPAMO 2 a réalisé 22 interventions, dont 15 pour les moules Mytilus edulis, 4 pour les coques Cerastoderma edule, 2 pour les palourdes Ruditapes sp. et 1pour les coquilles saint Jacques Pecten maximus. La recherche d’agents infectieux dans ces espèces de mollusques prélevés lors de hausse de mortalité a permis de mettre en évidence les parasites réglementés Perkinsus olseni dans 1 lot de palourdes, et Marteilia refringens dans 4 lots de moules, ainsi que le virus OsHV-1 dans 1 lot de palourdes et 1 lot de coques, la bactérie Vibrio aestuarianus dans 3 lots de coques, et des bactéries du groupe Splendidus dans 3 lots de coques et dans 13 lots de moules. L’année 2015 a également permis la démonstration sur un site atelier d’un exercice de surveillance programmée, ciblée et fondée sur les risques d’introduction et d’installation d’un organisme pathogène exotique. Elle a concerné le parasite Mikrocytos mackini de l’huître creuse Crassostrea gigas, sur un site atelier de la Charente-Maritime, suivi par le réseau RESCO 2. Le parasite Mikrocytos mackini n’a pas été détecté. En revanche, le parasite Marteilia refringens a été détecté dans ¾ des prélèvements d’huîtres réalisés. Dans le cadre du soutien scientifique et technique de l’évolution de la surveillance événementielle, l’année 2015 a également permis de poursuivre la démarche relative aux développements méthodologiques en lien avec la surveillance événementielle des mortalités de mollusques marins. Une étude de faisabilité de la recherche prospective de regroupements spatio-temporels d’événements de mortalités d’huîtres creuses a été préparée en collaboration avec tous les acteurs de la santé des mollusques marins en Normandie. Un outil de collecte et d’analyse des données de signalements des mortalités, automatisé, simple d’utilisation et flexible, a été élaboré.

Pyrimidine-5'-nucleotidase type I (P5'NI) deficiency is an autosomal recessive condition that causes nonspherocytic hemolytic anemia, characterized by marked basophilic stippling and pyrimidine nucleotide accumulation in erythrocytes. We herein present two African descendant patients, father and daughter, with P5'N deficiency, both born from first cousins. Investigation of the promoter polymorphism of the uridine diphospho glucuronosyl transferase 1A (UGT1A) gene revealed that the father was homozygous for the allele (TA7) and the daughter heterozygous (TA6/TA7). P5'NI gene (NT5C3) gene sequencing revealed a further change in homozygosity at amino acid position 56 (p.R56G), located in a highly conserved region. Both patients developed gallstones; however the father, who had undergone surgery for the removal of stones, had extremely severe intrahepatic cholestasis and, liver biopsy revealed fibrosis and siderosis grade III, leading us to believe that the homozygosity of the UGT1A polymorphism was responsible for the more severe clinical features in the father. Moreover, our results show how the clinical expression of hemolytic anemia is influenced by epistatic factors and we describe a new mutation in the P5'N gene associated with enzyme deficiency, iron overload, and severe gallstone formation. To our knowledge, this is the first description of P5'N deficiency in South Americans.

OBJECTIVES: We investigated the influence of sildenafil on cardiac contractility and diastolic relaxation and examined the distribution of phosphodiesterase-5 in the hearts of hypertensive rats that were treated with by NG-nitro-L-arginine methyl ester (L-NAME). METHODS: Male Wistar rats were treated with L-NAME and/or sildenafil for eight weeks. The Langendorff method was used to examine the effects of sildenafil on cardiac contractility and diastolic relaxation. The presence and location of phosphodiesterase-5 and phosphodiesterase-3 were assessed by immunohistochemistry, and cGMP plasma levels were measured by ELISA. RESULTS: In isolated hearts, sildenafil prevented the reduction of diastolic relaxation (dP/dt) that was induced by L-NAME. In addition, phosphodiesterase-5 immunoreactivity was localized in the intercalated discs between the myocardial cells. The staining intensity was reduced by L-NAME, and sildenafil treatment abolished this reduction. Consistent with these results, the plasma levels of cGMP were decreased in the L-NAME-treated rats but not in rats that were treated with L-NAME + sildenafil. CONCLUSION: The sildenafil-induced attenuation of the deleterious hemodynamic and cardiac morphological effects of L-NAME in cardiac myocytes is mediated (at least in part) by the inhibition of phosphodiesterase-5.

Background: Primary hyperparathyroidism occurs in only 10%-30% of patients with multiple endocrine neoplasia type 2A (MEN2A), rarely as the sole clinical manifestation, and is usually diagnosed after the third decade of life. Summary: A5-year-old girl was referred for prophylactic thyroidectomy as she carried the p.C634R RET mutation. She was clinically asymptomatic, with a normally palpable thyroid and with the cervical region free of lymphadenopathy or other nodules. Preoperative tests revealed hypercalcemia associated with elevation of parathyroid hormone (PTH) (calcium = 11.2mg/dL, calcium ion = 1.48mmol/L, phosphorus = 4.0 mg/dL, alkaline phosphatase = 625U/L, parathyroid hormone (PTH) PTH = 998 pg/mL). A thyroid ultrasound was normal and parathyroid scintigraphy with (99m)Tc-Sestamibi revealed an area of radioconcentration in the upper half of the left thyroid lobe suggesting hyperfunctioning parathyroid tissue. She underwent total thyroidectomy and parathyroidectomy and developed hypocalcemia. The anatomopathological examination showed no histopathological changes in the thyroid tissue and an adenoma of the parathyroid gland, confirming the diagnosis of hyperparathyroidism. Conclusions: Primary hyperparathyroidism can be a precocious manifestation of MEN2A. This case report highlights that asymptomatic hypercalcemia should be scrutinized in children related to patients with MEN2A who carry a mutation in the RET proto-oncogene, especially mutations in the codon 634, before the currently recommended age of 8 years.

The high affinity receptor for human granulocyte-macrophage colony-stimulating factor (GM-CSF) consists of a cytokine-specific alpha-subunit (hGMR alpha) and a common signal-transducing beta-subunit (hpc) that is shared with the interleukin-3 and -5 receptors, We have previously identified a constitutively active extracellular point mutant of hpc, I374N, that can confer factor independence on murine FDC-P1 cells but not BAF-B03 or CTLL-2 cells (Jenkins, B. J., D'Andrea, R. J., and Gonda, T. J. (1995) EMBO J. 14, 4276-4287), This restricted activity suggested the involvement of cell type-specific signaling molecules in the activation of this mutant. We report here that one such molecule is the mouse GMR alpha (mGMR alpha) subunit, since introduction of mGMR alpha, but not hGMR alpha, into BAF-B03 or CTLL-2 cells expressing the I374N mutant conferred factor independence, Experiments utilizing mouse/human chimeric GMR alpha subunits indicated that the species specificity lies in the extracellular domain of GMRa. Importantly, the requirement for mGMR alpha correlated with the ability of I374N (but not wild-type hpc) to constitutively associate with mGMRa. Expression of I374N in human factor-dependent UT7 cells also led to factor-independent proliferation, with concomitant up-regulation of hGMR alpha surface expression. Taken together, these findings suggest a critical role for association with GMR alpha in the constitutive activity of I374N.

An examination has been carried out of the secondary passive film on Type 304 stainless steel in 0.5 M H2SO4. The characterization techniques used were electrochemical (potentiodynamic; potentiostatic, and film reduction experiments) and surface analytical. A bilayer model for the secondary passive film is proposed. It appears that next to the metal, there is a modified passive film which controls the electrochemical response; i.e., governs the current for any applied potential. On top of this modified passive film, the experimental data are consistent with a ''porous'' corrosion-product film which adds to the total film thickness but has little influence on the electrochemical response. The composition of the secondary passive film corresponds most probably to a mixed Fe/Cr oxide/hydroxide enriched in Cr3+, With a composition similar to a primary passive film.

In the present study, we have performed a comparative analysis of the effect of selective inhibitors of phosphodiesterase (PDE) type III, IV and V on eosinophil chemotaxis triggered by platelet activating factor (PAF) and leukotriene B4 (LTB4) in vitro. The effect of the analogues N6-2'-O-dibutyryladenosine 3':5' cyclic monophosphate (Bt2 cyclic AMP) and N2-2'-O- dibutyrylguanosine 3':5' cyclic monophosphate (Bt2 cyclic GMP) has also been determined. The eosinophils were obtained from the peritoneal cavity of naive Wistar rats and purified in discontinuous Percoll gradients to 85-95% purity. We observed that pre-incubation of eosinophils with the PDE type IV inhibitor rolipram suppressed the chemotactic response triggered by PAF and LTB4, in association with an increase in the intracellular levels of cyclic AMP. In contrast, neither zaprinast (type V inhibitor) nor type III inhibitors milrinone and SK&F 94836 affected the eosinophil migration. Only at the highest concentration tested did the analogue Bt2 cyclic AMP suppress the eosinophil chemotaxis, under conditions where Bt2 cyclic GMP was ineffective. We have concluded that inhibition of PDE IV, but not PDE III or V, was able to block the eosinophil chemotaxis in vitro, suggesting that the suppressive activity of selective PDE IV inhibitors on tissue eosinophil accumulation may, at least, be partially dependent on their ability to directly inhibit the eosinophil migration.
