The effectiveness of using frozen sheep semen in artificial insemination programs requires the development of extenders and freezing protocols that will increase pregnancy rate of inseminated females. This work aimed to survey the main extenders, additives and external and internal cryoprotectants that have been employed to improve the maintenance levels of sperm parameters after the freezing-thawing process. Better rates of post-thawing sheep sperm viability may be obtained with changes in the freezing extender composition, whether by the adjustment of its components or by introducing additives that inhibit the occurrence of sperm changes during the cryopreservation process. Thus, the possible changes proposed must take into consideration the intrinsic characteristics of ram semen and the individual variability among animals. It is important to emphasize that the sperm cryopreservation effectiveness requires that all process steps are conducted in an integrated manner, to maximize the fertility rate of frozen ram semen.

This study evaluated the prevalence and counting parasitism of different species of helminths of sheep from the micro-region of Jaboticabal of Sao Paulo state. For this, 66 animals naturally infected, four to 36 months of age, raised in pasture, were selected. The results of necropsy revealed the presence of seven genera and 12 species with the following prevalence and mean count: Haemonchus contortus: 100.0% (2947.2); Trichostrongylus colubriformis: 90.9% (3048.8); Cooperia curticei: 56.0% (256.5); Oesophagostomum columbianum: 48.4% (36.0); Cooperia punctata: 30.3% (94.5); Trichostrongylus axei: 22.7% (26.5); Strongyloides papillosus: 19.6% (83.0), Haemonchus contortus (L4): 7.5% (17.2), Cooperia pectinata: 10.6% (12.9), Trichuris ovis: 10.6 % (0.6); Cooperia spatulata 4.5% (0.3); Capillaria bovis: 4.5% (0.1). The mean parasitism of helminthswas 6524.7 per animal. Haemonchus contortus (adults and L4) and Trichostrongylus colubriformis corresponded to 45.4% and 46.7% of the average worm burden totally, respectively. Based in the results obtained in this study, can be concluded that the two most abundant species of helminths and important, the micro-region of Jaboticabal are Trichostrongylus colubriformis and Haemonchus contortus, and these two species amounted to 92.1% of the distribution percentage of helminths collected from all animals. These results demonstrate the importance of conducting a counts of eggs per gram of feces (EPG) in the herds of this region when FAMACHA is used on a particular property, since this method control does not allow to diagnostic the damage/clinical signs in animals infected by T. colubriformis, because this specie does not have hematophagism habit on animals.

The sheep industry has become increasingly prominent in agribusiness, transforming the stage production of Brazil, and thus contributes more to the socio-economic development of the country. The work aimed to verify the occurrence of brucellosis and leptospirosis in sheep from northwestern São Paulo state. In addition to determining the prevalence of major Leptospira in the region and to trace the diagnosis of sheep breeding in this part of the country. All the 1222 sheep serum samples from 49 properties did not react serologically to evidentiary testing for brucellosis, compared to antigens of B. abortus and B. ovis used in the testing of 2-ME and IDGA, respectively. The Microscopic Agglutination Test (MAT) test revealed that 19.14% (232/1212) of samples were positive for one or more serovars, with titles ranging from 100 to 800. The most frequent serovar was hebdomadis in the region, with Sentot and Sherman (18.10%, 11.64% and 8.62%, respectively). By profiling the system of sheep farming in the region, we found that most herds are composed of more than one race being the main purpose is for the court. According to the scheme adopted immunoprophylactics there is a homogeneous set schedule. It adopts the use of anti-helminth, non-prescription veterinary antibiotic. There is also the presence of diarrhea and abortions and the lack of criteria for los ovinos on management, making it the need for emergency development of programs for disease control, schema immunoprophylactics adequate sanitation and hygienic measures in sheep breeding.

Orchiepididymitis is the inflammation of both, testis and epididymis and may be caused by microbial agents such as Brucella ovis. Thus, the aim of the present report is to present a case of orchiepididymitis in a Lacaune ram with unilateral testicular enlargement. Ultrasonography of the testis revealed the presence of a circular hypoechogenic structure adjacent to the testicular parenchyma. Corynebacterium sp was isolated from the surgically removed testis. Two months after removal of the affected testis, the animal returned to the Veterinary Hospital due to an enlargement of the remaining testis. A new ultrasonographic exam revealed the presence of abscesses in the testicular parenchyma and periorchitis. After orchiectomy, Proteus mirabilis was isolated from the material.

Stress hormones in Rocky Mountain bighorn sheep (Ovis canadensis canadensis), produced in response to environmental changes, road development, or high population density, may impact their immune systems to a threshold level that predisposes them to periodic, large-scale mortality. We compared the stress response to a novel environmental situation and repeated handling between bighorn sheep born and raised in captivity (CR) and bighorn sheep born in the wild (WC) and brought into captivity. We measured plasma epinephrine, norepinephrine, cortisol, and fecal glucocorticoid metabolites (FGM). Three weeks after each group’s arrival we used a one-time drop-net event to elicit an acute stress response, and we collected blood samples from each sheep over 35 minutes, as well as one fecal sample. We collected blood and fecal samples from both groups on 7 other occasions over the subsequent 6 months. We also collected fecal samples from the pen at approximately 24-hour intervals for 3 days following every handling event to monitor the stress response to handling. We found that CR sheep had a stronger autonomic nervous system response than WC sheep, as measured by epinephrine and norepinephrine levels, but we found a very similar hypothalamic–pituitary–adrenal axis (HPA) response, measured by cortisol levels, to the acute stress event of a drop-net restraint. We also found that once the WC sheep had acclimated, as indicated by the return to the initial baseline FGM levels within 12 weeks, the CR and WC groups’ HPA responses to sampling events were not significantly different from one another. Fecal samples can provide a noninvasive mechanism for managers to monitor baseline FGM for a given herd. Using long-term monitoring of FGM rather than values from a single point in time may allow managers to correlate these levels to outside influences on the herd and better understand the impacts of management changes, population density, or increased human developments on the health of the sheep population.

We report the development of a colourimetric PCR/dot blot assay targeting the mitochondrial gene NADH dehydrogenase subunit 1 (nad1) for differential diagnosis of taeniid eggs. Partial sequences of the cestode nad1 gene were aligned and new primers were designed based on conserved regions. Species-specific oligonucleotide probes (S-SONP) for canine taeniid cestodes were then designed manually based on the variable region between the conserved primers. Specifically, S-SONP were designed for the Taenia crassiceps, T. hydatigena, T. multiceps, T. ovis, T. taeniaeformis, Echinococcus granulosus (genotype 1), E. multilocularis and E. vogeli. Each probe showed high specificity as no cross-hybridisation with any amplified nad1 fragment was observed. We evaluated the assay using 49 taeniid egg-positive samples collected from dogs in Zambia. DNA from 5 to 10 eggs was extracted in each sample. Using the PCR/dot blot assay, the probes successfully detected PCR products from T. hydatigena in 42 samples, T. multiceps in 3 samples, and both species (mixed infection) in the remaining 4 samples. The results indicate that the PCR/dot blot assay is a reliable alternative for differential diagnosis of taeniid eggs in faecal samples.

To date, investigations of genetic diversity and the origins of domestication in sheep have utilised autosomal microsatellites and variation in the mitochondrial genome. We present the first analysis of both domestic and wild sheep using genetic markers residing on the ovine Y chromosome. Analysis of a single nucleotide polymorphism (oY1) in the SRY promoter region revealed that allele A-oY1 was present in all wild bighorn sheep (Ovis canadensis), two subspecies of thinhorn sheep (Ovis dalli), European Mouflon (Ovis musimon) and the Barbary (Ammontragis lervia). A-oY1 also had the highest frequency (71.4%) within 458 domestic sheep drawn from 65 breeds sampled from Africa, Asia, Australia, the Caribbean, Europe, the Middle East and Central Asia. Sequence analysis of a second locus, microsatellite SRYM18, revealed a compound repeat array displaying fixed differences, which identified bighorn and thinhorn sheep as distinct from the European Mouflon and domestic animals. Combined genotypic data identified 11 male-specific haplotypes that represented at least two separate lineages. Investigation of the geographical distribution of each haplotype revealed that one (H6) was both very common and widespread in the global sample of domestic breeds. The remaining haplotypes each displayed more restricted and informative distributions. For example, H5 was likely founded following the domestication of European breeds and was used to trace the recent transportation of animals to both the Caribbean and Australia. A high rate of Y chromosomal dispersal appears to have taken place during the development of domestic sheep as only 12.9% of the total observed variation was partitioned between major geographical regions.

The incidence of human brucellosis in Kyrgyzstan has been increasing in the last years and was identified as a priority disease needing most urgent control measures in the livestock population. The latest species identification of Brucella isolates in Kyrgyzstan was carried out in the 1960s and investigated the circulation of Brucella abortus, B. melitensis, B. ovis, and B. suis. However, supporting data and documentation of that experience are lacking. Therefore, typing of Brucella spp. and identification of the most important host species are necessary for the understanding of the main transmission routes and to adopt an effective brucellosis control policy in Kyrgyzstan. Overall, 17 B. melitensis strains from aborted fetuses of sheep and cattle isolated in the province of Naryn were studied. All strains were susceptible to trimethoprim-sulfamethoxazole, gentamicin, rifampin, ofloxacin, streptomycin, doxycycline, and ciprofloxacin. Multilocus variable number tandem repeat analysis showed low genetic diversity. Kyrgyz strains seem to be genetically associated with the Eastern Mediterranean group of the Brucella global phylogeny. We identified and confirmed transmission of B. melitensis to cattle and a close genetic relationship between B. melitensis strains isolated from sheep sharing the same pasture.
