En la segunda reunión de los Puntos Focales designados por los Gobiernos de los Países Signatarios de la Declaración sobre la aplicación del principio 10 de la Declaración de Río sobre el Medio Ambiente y el Desarrollo, que tuvo lugar en Guadalajara (México) los días 16 y 17 de abril de 2013, se decidió la creación de grupos de trabajo para avanzar hacia la consecución de un instrumento regional. Así, se estableció un grupo de trabajo sobre derechos de acceso e instrumento regional, con el objetivo de profundizar en el conocimiento de los derechos de acceso con miras a proponer la naturaleza y contenidos del instrumento regional. En su primera reunión, el grupo de trabajo consideró que un estudio descriptivo acerca de la tipología de instrumentos internacionales sería útil para alcanzar su objetivo. En este informe se estudian los diferentes tipos de instrumentos que se utilizan en el derecho internacional público, con énfasis en aquellos relativos al Principio 10. El informe está estructurado en tres capítulos, de la siguiente manera: En el primer capítulo se analiza el término "instrumento internacional" y se distingue entre instrumentos vinculantes y no vinculantes, presentando ejemplos ilustrativos. El segundo capítulo describe la función de mecanismos de cumplimiento e implementación en un instrumento internacional, a la vez que aporta ejemplos de dichos mecanismos. En el tercer capítulo se presentan los instrumentos multilaterales y regionales pertinentes a los derechos de acceso a la información, participación y justicia en asuntos ambientales en América Latina y el Caribe.

At the second meeting of the focal points appointed by the Governments of the signatory countries of the Declaration on the application of Principle 10 of the Rio Declaration on Environment and Development in Latin America and the Caribbean, which was held in Guadalajara, Mexico, on 16 and 17 April 2013, a decision was made to form working groups to advance towards the creation of a regional instrument. Thus, a working group on access rights and the regional instrument was formed for the purpose of gaining more in-depth knowledge on access rights in order to make a proposal on the nature and scope of the application of a regional instrument. At its first meeting, the working group determined that a study describing the different types of international instruments would be useful in helping it achieve its objective. This report explores the different types of instruments that are used in public international law, with an emphasis on the instruments that are relevant to Principle 10. The report has three chapters, which are as follows. The first chapter analyses the term “international instrument” and discusses the distinction between binding and non-binding legal instruments, illustrated with examples. The second chapter describes the function of implementation and compliance mechanisms in an international instrument, providing examples of these mechanisms. The third chapter presents the multilateral and regional instruments relevant to access rights regarding information, participation and justice in environmental matters in Latin America and the Caribbean.

The work consists of the historical city of Guaratinguetá, as well as data on their quality of life, economic development and population, transportation in Brazil and the transportation system of the city serving as a basis to support the development of the urban network. The urban fabric of the city was raised with plants of the city in different years, documents obtained by the Museum and Hall of Frei Galvão Guaratinguetá well to check the form of land use and growth drivers of the city over time. We studied also the main access roads to the city by focusing on an approach that has been the subject of a proposed urban intervention. A direct interview was performed and recorded with one of those responsible for planning the urban fabric of the city in order to gather information on the historical development, the future possibilities of interventions in the urban as well as problems encountered and other curiosities. Through the study it was observed that the urban grid Guaratinguetá developing a planned since 1970 and that the highlights of most cities in the Paraíba Valley. The work is grounded in concepts of sustainable city and also in quantitative and qualitative data from sectors responsible

As plantas apresentam moléculas que detectam a luz, entre elas estão os fitocromos que detectam mudanças na qualidade da luz nas regiões de vermelho (V) e vermelho-extremo (VE). O fitocromo age no crescimento das plantas possibilitando-as perceber o sombreamento por outras plantas, o que pode ocasionar no processo de estiolamento, ou seja, alongamento do caule. Assim, conforme aumenta o sombreamento, a razão V:VE diminui. Nas plantas de sol, quanto maior a proporção de VE, maior a taxa de estiolamento, enquanto que as plantas de sombra mostram pouca ou nenhuma redução na taxa de alongamento dos caules. Na sucessão florestal as plantas podem ser classificadas como pioneiras, sendo aquelas dependentes de luz e intolerantes à sombra, e as de transição ou clímax, que são tolerantes à sombra. Assim, este trabalho teve o intuito de analisar o processo de estiolamento de plântulas de espécies arbóreas nativas selecionadas, mantidas em condições de luz diferentes e correlacionar com a classe seral correspondente citada na literatura. As seguintes espécies foram analisadas: Bixa orellana, Caesalpinia peltophoroides, Dalbergia nigra, Erythrina speciosa, Eugenia uniflora, Hevea brasiliensis, Hymenaea courbaril, Licania tomentosa, Pachira aquatica, Piptadenia gonoacantha, Psidium guajava, Schizolobium parahyba. Todos os experimentos foram desenvolvidos no jardim experimental da UNESP de Rio Claro. As espécies foram expostas a seis condições luminosas diferentes, com razão V:VE de: 0,1; 0,19; 0,21; 0,35; 1,15 e 1,17. E radiação solar incidente de: 3,16UM; 44,78UM; 27,7UM; 73,54UM; 1987,00UM; 1453,00UM. Foram tiradas medidas do comprimento dos hipocótilos e epicótilos. Posteriormente foram realizadas análises estatísticas com nível de significância de 5%. As espécies consideradas pioneiras foram D. nigra e S. parahyba. It has been shown that ouabain (OUA) can activate the Na,K-ATPase complex and mediate intracellular signaling in the central nervous system (CNS). Inflammatory stimulus increases glutamatergic transmission, especially at N-methyl-D-aspartate (NMDA) receptors, which are usually coupled to the activation of nitric oxide synthase (NOS). Nuclear factor-kappa B (NF-kappa B) activation modulates the expression of genes involved in development, plasticity, and inflammation. The present work investigated the effects of OUA on NF-kappa B binding activity in rat hippocampus and the influence of this OUA-Na,K-ATPase signaling cascade in NMDA-mediated NF-kappa B activation. The findings presented here are the first report indicating that intrahippocampal administration of OUA, in a concentration that did not alter Na,K-ATPase or NOS activity, induced an activation of NF-kappa B, leading to increases in brain-derived neurotrophic factor (Bdnf), inducible NOS (iNos), tumor necrosis factor-alpha (Tnf-alpha), and B-cell leukemia/lymphoma 2 (Bcl2) mRNA levels. This response was not linked to any significant signs of neurodegeneration as showed via Fluoro-Jade B and Nissl stain. Intrahippocampal administration of NMDA induced NF alpha B activation and increased NOS and alpha 2/3-Na,K-ATPase activities. NMDA treatment further increased OUA-induced NF-kappa B activation, which was partially blocked by MK-801, an antagonist of NMDA receptor. These results suggest that OUA-induced NF-kappa B activation is at least in part dependent on Na,K-ATPase modulatory action of NMDA receptor in hippocampus. The interaction of these signaling pathways could be associated with biological mechanisms that may underlie the basal homeostatic state linked to the inflammatory signaling cascade in the brain. Background. Rest myocardial perfusion imaging (MPI) is effective in managing patients with acute chest pain in developed countries. We aimed to define the role and feasibility of rest MPI in low-to-middle income countries. Methods and Results. Low-to-intermediate risk patients (n = 356) presenting with chest pain to ten centers in eight developing countries were injected with a Tc-99m-based tracer, and standard imaging was performed. The primary outcome was a composite of death, non-fatal myocardial infarction (MI), recurrent angina, and coronary revascularization at 30 days. Sixty-nine patients had a positive MPI (19.4%), and 52 patients (14.6%) had a primary outcome event. An abnormal rest-MPI result was the only variable which independently predicted the primary outcome [adjusted odds ratio (OR) 8.19, 95% confidence interval 4.10-16.40, P = .0001]. The association of MPI result and the primary outcome was stronger (adjusted OR 17.35) when only the patients injected during pain were considered. Rest-MPI had a negative predictive value of 92.7% for the primary outcome, improving to 99.3% for the hard event composite of death or MI. Conclusions. Our study demonstrates that rest-MPI is a reliable test for ruling out MI when applied to patients in developing countries. (J Nucl Cardiol 2012;19:1146-53.)

Background aims. Mesenchymal stromal cells (MSC) are being used to treat and prevent a variety of clinical conditions. To be readily available, MSC must be cryopreserved until infusion. However, the optimal cryopreservation methods, cryoprotector solutions and MSC sensitivity to dimethyl sulfoxide (DMSO) exposure are unknown. This study investigated these issues. Methods. MSC samples were obtained from human umbilical cord (n = 15), expanded with Minimal Essential Medium-alpha (alpha-MEM) 10% human serum (HS), resuspended in 25 mL solution (HS, 10% DMSO, 20% hydroxyethyl starch) and cryopreserved using the BioArchive (R) system. After a mean of 18 +/- 7 days, cell suspensions were thawed and diluted until a DMSO concentration of 2.5% was reached. Samples were tested for cell quantification and viability, immunophenotype and functional assays. Results. Post-thaw cell recovery: 114 +/- 2.90% (mean +/- SEM). Recovery of viable cells: 93.46 +/- 4.41%, 90.17 +/- 4.55% and 81.03 +/- 4.30% at 30 min, 120 min and 24 h post-thaw, respectively. Cell viability: 89.26 +/- 1.56%, 72.71 +/- 2.12%, 70.20 +/- 2.39% and 63.02 +/- 2.33% (P<0.0001) pre-cryopreservation and 30 min, 120 min and 24 h post-thaw, respectively. All post-thaw samples had cells that adhered to culture bottles. Post-thaw cell expansion was 4.18 +/- 0.17 X, with a doubling time of 38 +/- 1.69 h, and their capacity to inhibit peripheral blood mononuclear cells (PBMC) proliferation was similar to that observed before cryopreservation. Differentiation capacity, cell-surface marker profile and cytogenetics were not changed by the cryopreservation procedure. Conclusions. A method for cryopreservation of MSC in bags, in xenofree conditions, is described that facilitates their clinical use. The MSC functional and cytogenetic status and morphologic characteristics were not changed by cryopreservation. It was also demonstrated that MSC are relatively resistant to exposure to DMSO, but we recommend cell infusion as soon as possible.

Bixin is the main carotenoid found in annatto seeds (Bixa orellana L.) and is responsible for their reddish-orange color. The antioxidant properties of this compound are associated with its ability to scavenge free radicals, which may reduce damage and protect tissues against toxicity caused by anticancer drugs such as cisplatin. In this study, the genotoxicity and antigenotoxicity of bixin on cisplatin-induced toxicity in PC12 cells was assessed. Cytotoxicity was evaluated using the mu assay, mutagenicity, genotoxicity, and protective effect of bixin were evaluated using the micronucleus test and comet assay. PC12 cells were treated with bixin (0.05, 0.08, and 0.10 mu g/mL), cisplatin (0.1 mu g/mL) or a combination of both bixin and cisplatin. Bixin was neither cytotoxic nor genotoxic compared to the controls. In the combined treatment bixin significantly reduced the percentage of DNA in tail and the frequency of micronuclei induced by cisplatin. This result suggests that bixin can function as a protective agent, reducing cisplatin-induced DNA damage in PC12 cells, and it is possible that this protection could also extend to neuronal cells. Further studies are being conducted to better understand the mechanisms involved in the activity of this protective agent prior to using it therapeutically. (C) 2011 Elsevier Ltd. Human mesenchymal stem cells (hMSCs) are adult multipotent cells that have high therapeutic potential due to their immunological properties. They can be isolated from several different tissues with bone marrow (BM) being the most common source. Because the isolation procedure is invasive, other tissues such as human umbilical cord vein (UCV) have been considered. However, their interchangeability remains unclear. In the present study, total protein extracts of BM-hMSCs and UCV-hMSCs were quantitatively compared using gel-LC-MS/MS. Previous SAGE analysis of the same cells was re-annotated to enable comparison and combination of these two data sets. We observed a more than 63% correlation between proteomic and transcriptomic data. In silico analysis of highly expressed genes in cells of both origins suggests that they can be modulated by microRNA, which can change protein abundance. Our results showed that MSCs from both tissues shared high similarity in metabolic and functional processes relevant to their therapeutic potential, especially in the immune system process, response to stimuli, and processes related to the delivery of the hMSCs to a given tissue, such as migration and adhesion. Hence, our results support the idea that the more accessible UCV could be a potentially less invasive source of MSCs.

Populations in the Amazon are exposed to organic mercury via consumption of contaminated foods. These ethnic groups consume a specific plant seed annatto which contains certain carotenoids. The aim of this study was to find out if these compounds (bixin, BIX and norbixin, NOR), protect against DNA-damage caused by the metal. Therefore, rats were treated orally with methylmercury (MeHg) and with the carotenoids under conditions that are relevant to humans. The animals were treated either with MeHg (30 mu g/kg/bw/day), BIX (0.110 mg/kg/bw/day), NOR (0.011.0 mg/kg/bw/day) or combinations of the metal compound and the carotenoids consecutively for 45 days. Subsequently, the glutathione levels (GSH) and the activity of catalase were determined, and DNA-damage was measured in hepatocytes and leukocytes using single cell gel electrophoresis assays. Treatment with the metal alone caused a decrease in the GSH levels (35%) and induced DNA damage, which resulted in increased DNA migration after electrophoresis in liver and blood cells, whereas no effects were seen with the carotenoids alone. When BIX or NOR were given in combination with organic mercury, the intermediate and the highest concentrations of the carotenoids (1.0 and 10.0 mg/kg/bw/day BIX and 0.1 and 1.0 mg/kg/bw/day NOR) protected against DNA-damage. Furthermore, we found with both carotenoids, a moderate increase in the GSH levels in both metal-treated and untreated animals, while the activities of catalase remained unchanged. Our results indicate that consumption of BIX and NOR may protect humans against the adverse health effects caused by exposure to organic mercury. Environ. Mol. Mutagen., 2012.
