Cytogenetic studies were performed on two sympatric species of Characidium, C. gomesi and C. cf. zebra, from the Grande River basin, Minas Gerais State, Brazil. Although both species had a chromosome number of 50 with a karyotype exclusively consisting of meta- and submetacentric chromosomes, interspecific diversity was detected concerning the size of the two first chromosome pairs of the karyotypes. Active nucleolus organizer regions (NORs) were located at the terminal position on the long arm of the 17th pair of C. gomesi and at subterminal position on the long arm of the 23rd pair of C. cf. zebra. For both species the fluorochrome CMA3 stained only the NOR-bearing pair of chromosomes. The heterochromatin pattern also showed some differentiation between these species restricted to the centromeric or pericentromeric region of C. cf. zebra and practically absent in C. gomesi. These data are discussed concerning chromosome diversification in this fish group.

G- and C- banding patterns of seven species of the bat family Molossidae, Eumops glaucinus, E. perotis; Molossops abrasus, M. remminckii, Molossus ater, M. molossus, and Nyctinomops laticaudatus, were identified. Comparisons among the karyotypes of these species showed extensive homologies between E. perotis, M. ater, M. molossus, M. abrasus, and N. laticaudatus, demonstrating inter- and intrageneric conservatism, and a lesser degree of homologies in M. temminckii and glaucinus, reflecting intrageneric variation, Chromosomal variation was due to inversions, Robertsonian rearrangements, translocations, and variations in the location of constitutive heterochromatin and nucleolus organizer regions. The chromosome corresponding to No. 5 in the M. ater karyotype is discussed. We suggest that the Nyctinomops and Molossops karyotypes represent the primitive condition and that Molossus and Eumops have derived karyotypes.

Abnormalities in any component of the cell cycle regulatory machine may result in oral. cancer, and markers of cell proliferation have been used to determine the prognosis of tumor progression. The aim of this study was to determine whether silver-stained nucleolar organizer region (AgNOR) and Ki-67 measurements could improve the assessment of growth rates in oral lesions. Eighty-three oral biopsies were studied, 20 of which were classified as fibrous inflammatory hyperplasia (FIH), 40 as leukoplakia (LKP) and 23 as oral. squamous cell carcinoma (OSCC). Within the LKP group, 22 out of 29 biopsies were diagnosed as non-dysplastic leukoplakia (LK) and 18 as dysplastic teukoptakia (DLK), presenting discrete, moderate and severe dysplasia. Ki-67 immunotabeting of the lesions increased steadily in the following order: FIH, DLK, LK and OSCC, indicating that Ki-67 is a good marker for predicting the protiferative fraction among benign, premalignant and malignant oral lesions. The median values of AgNOR parameters indicate that the morphometric index gives better results regarding the proliferative rate than the numerical one. A series of linear regressions between AgNOR parameters and Ki-67 showed positive associations. We conclude that a combination of Ki-67 and morphometric AgNOR analyses could be used as an aid in the determination of the protiferative status of oral epithelial. cells in oral cancer. SYNAPTONEMAL COMPLEX-ANALYSIS IN SPERMATOCYTES OF TILAPIA, OREOCHROMIS-NILOTICUS (PISCES, CICHLIDAE)
Resumo:
Some adaptations of the synaptonemal complex (SC) whole-mounting technique first used in plants permitted its application to meiotic studies in tilapia, Oreochromis niloticus. Direct observation of the chromosome pairing process and bivalent structure during the meiotic prophase of this fish species by light and electron microscopy permitted the analysis of SCs in autosomes and the possible identification of sex chromosomes. The analysis of SCs in spermatocytes of 0. niloticus revealed that all 22 bivalent chromosomes completely paired, except for the occurrence of a size heteromorphism in the terminal region of the largest bivalent associated with the presence of an incompletely paired segment during the synapsis process, which may be the cytological visualization of an XX/XY sex chromosome system in this species.

Specimens of Hyla nana and Hyla sanborni from a syntopic population were studied cytogenetically. These species are morphologically very similar and are frequently misidentified, confused with each other. Both species had a diploid chromosome number, 2n = 30. However, the karyotypes of H. nana and H. sanborni differed considerably from each other in the number of submetacentric and telocentric chromosomes. The two species also differed in their primary NOR-bearing chromosomes (metacentric pair 13 in H. nana and telocentric pair 12 in H. sanborni). Additional nucleolus organizer regions (NORs) were detected by Ag-NOR staining and FISH in chromosome pairs 1, 5, 6, 12, and 14 in seven specimens of H. nana. Thus, a total of six patterns of NOR were identified. These differences in karyotype and in NOR location allowed the unambiguous identification of syntopic individuals of the two species. However, the chromosomal morphology of both species differed from that reported for populations from other geographic regions, suggesting that a systematic reevaluation of this group of Hyla may be necessary.

The mitotic chromosomes, nucleolus organizer regions (NORs), C-banding pattern and nuclear DNA content of Diplomystes mesembrinus were studied. The karyotype, with 2n=56 chromosomes (22m+24sm+6st+4a), has a high chromosome arm number (NF = 102), one chromosome pair with NORs, and a very small amount of heterochromatin. The NOR-bearing arm is entirely heterochromatic and exhibits a marked size polymorphism. The diploid DNA content detected in erythrocyte nuclei of D. mesembrinus was 2.57 ± 0.15 pg/nucleus. The chromosome evolution in Siluriformes is discussed on the basis of available cytogenetic data and it is proposed that 2n=56 is synapomorphic for the order.

Cytogenetic studies were carried out on Rhamdella microcephala collected from the headwaters of the Machado river, a tributary of the Sapucaí river, Minas Gerais, Brazil. Sixteen specimens exhibited 2n=56 chromosomes, with 18 metacentrics, 30 submetacentrics and 8 subtelocentric-acrocentrics (FN=84). The nucleolar organizing regions (NORs) were identified in the interstitial position on the long arm of a submetacentric pair (pair 12). Chromomycin A3 staining evidenced only the NOR-bearing segments. Positive C-band segments were identified in a pericentromeric position in most of the chromosomes and the NOR-bearing segments were also C-band positive. Some aspects related to the chromosomal characteristics of Rhamdella microcephala are discussed.

In the present study the karyotypes and the number and position of nucleolus organizer regions (NORs) of four species of Characiformes are described. The analyses showed that Acestrorhynchus lacustris had 2n=50 chromosomes (8M+34SM+6ST+2A), Oligosarcus longirostris had 2n=50 chromosomes (2M+20SM+10ST+18A), Oligosarcus cf. paranensis from Keller and Mourão rivers had 2n=50 chromosomes (2M+26SM+8ST+14A), and Rhaphiodon vulpinus had 2n=54 chromosomes (32M+12SM+8ST+2A). The number of NOR-bearing chromosome pairs ranged from one to three among the species studied. The results available show that there is no variation in the diploid number within the genera analyzed. However, clear differences related to the karyotypic formulae and the number and position of Ag-NORs were found even in the comparison between populations. The possible relationships between the genera analyzed and other Characiformes are discussed.

This paper describes the karyotype of Odontesthes regia by means of Giemsa staining, C-banding, to reveal the distribution of the constitutive heterochromatin, and by Ag-staining and fluorescent in situ hybridization (FISH), to locate ribosomal genes (rDNA). The chromosome diploid modal count in the species was 2n = 48. The karyotype is composed of one submetacentric pair (pair 1), 16 subtelocentric pairs (pairs 2 to 17), and 7 acrocentric pairs (pairs 18 to 24). With the exception of pair 1 it was not possible to classify the homologous chromosomes accurately because differences in chromosome size were too slight between adjacent pairs. The distribution of C-banded heterochromatin allowed for a more accurate matching of the majority of chromosomes of the subtelocentric series. Silver staining of metaphase spreads allowed for the identification of Nucleolus Organizer Regions (Ag-NOR) on pair 1. FISH experiments showed that 18S rDNA sequences were located, as expected, in the same chromosome pair identified as the Ag-NOR-bearing one.
