O presente trabalho de pesquisa envolve os casos de toxicomanias, como um novo sintoma, e a relao com a subjetividade de nossa poca, provocando um questionamento sobre a clnica e a direo do tratamento. A mudana operada na cultura destaca o discurso do mestre atual como aquele que leva em si a marca do declnio dos ideais e das identificaes no mais estabelecidos por um significante mestre e, sim, pelo mercado, provocando um imperativo feroz de Um gozo que promete a satisfao plena atravs do objeto, no caso, a droga. Vimos nos casos das toxicomanias e do alcoolismo, a ilusria completude capturada por um gozo auto- ertico que ludibria o sintoma com a droga e silencia o sujeito. O resultado do entrelaamento do discurso capitalista com o discurso da cincia recai sobre a droga inserindo-a como mercadoria, cuja produo cada vez mais desenfreada, empurra a uma intoxicao generalizada, de difcil desintoxicao. Com a orientao psicanaltica possvel estabelecer um dispositivo que promova essa desintoxicao, reinventado caso a caso. ofertar atravs do discurso do analista, um vazio de significaes, de uma no resposta, em que, atravs da fala do sujeito, seja possvel pinar a funo da droga na economia psquica e os significantes que marcam sua histria. O debate sobre a poltica pblica e a funo da psicanlise nos servios pblicos deve-se orientar pela singularidade e a dimenso tica do gozo no que concerne a proposta universalizante da poltica pblica.

Nesta pesquisa, elegemos Alma Infantil (1912) como fonte e objeto de estudo: obra escrita pela poetisa Francisca Jlia da Silva em parceria com seu irmo, o tambm poeta Jlio Csar da Silva. A pesquisa busca evocar o cenrio literrio escolar do entressculos (XIX-XX) e do incio do sculo XX, em suas particularidades, coordenando o projeto poltico e pedaggico que conformava as escolas no perodo com os temas, autores e textos que circulavam nas salas de aula, sobretudo no estado de So Paulo, onde se verifica, no perodo, a maior incidncia de planos e aes pedaggicas. Temos por finalidade exprimir as relaes entre Alma Infantil e alguns projetos de nao propostos pelo governo na Primeira Repblica (1889-1930). Alma Infantil, publicado pela Livraria Editora Magalhes no incio do sculo XX livro para uso escolar, mais especificamente, livro de leitura suplementar. Este material, assim como muitos outros, estava comprometido com a moral e a ordem cvica dos primeiros anos do novo regime, com a exaltao natureza, aos animais e s riquezas naturais do pas, conforme a anlise dos poemas indica. O livro dos irmos Silva, em seus 48 poemas e quatro hinos, apresenta linguagem acessvel e adaptada leitura da criana, de fcil compreenso, conformados por um conceito de moral notoriamente verificado. Em grande parcela dos poemas, encontramos referncias a uma linguagem leve, descompromissada com a rigidez e culto forma diferentemente da que o exaltava. Outrossim, a presena dos hinos em Alma Infantil demarca ainda mais esta unidade patritica a que o Brasil se propunha compor no entressculos. Os temas dos hinos estudo, escola, trabalho, ptria - alinham os ideais republicanos de ordem, trabalho e progresso que vigorava no Brasil naquele momento. Sendo assim, atravs da pesquisa histrica que resultou neste texto, podemos afirmar por meio de anlise textual e documental que Alma Infantil obra confeccionada para uso escolar, como sua prpria capa o diz, e mais, uma legtima amostra do maquinrio poltico educacional dos primeiros anos do regime republicano neste pas.

Dinoflagellates possess many physiological processes that appear to be under post-transcriptional control. However, the extent to which their genes are regulated post-transcriptionally remains unresolved. To gain insight into the roles of differential mRNA stability and de novo transcription in dinoflagellates, we biosynthetically labeled RNA with 4-thiouracil to isolate newly transcribed and pre-existing RNA pools in Karenia brevis. These isolated fractions were then used for analysis of global mRNA stability and de novo transcription by hybridization to a K. brevis microarray. Global K. brevis mRNA half-lives were calculated from the ratio of newly transcribed to pre-existing RNA for 7086 array features using the online software HALO (Half-life Organizer). Overall, mRNA half-lives were substantially longer than reported in other organisms studied at the global level, ranging from 42 minutes to greater than 144 h, with a median of 33 hours. Consistent with well-documented trends observed in other organisms, housekeeping processes, including energy metabolism and transport, were significantly enriched in the most highly stable messages. Shorter-lived transcripts included a higher proportion of transcriptional regulation, stress response, and other response/regulatory processes. One such family of proteins involved in post-transcriptional regulation in chloroplasts and mitochondria, the pentatricopeptide repeat (PPR) proteins, had dramatically shorter half-lives when compared to the arrayed transcriptome. As transcript abundances for PPR proteins were previously observed to rapidly increase in response to nutrient addition, we queried the newly synthesized RNA pools at 1 and 4 h following nitrate addition to N-depleted cultures. Transcriptome-wide there was little evidence of increases in the rate of de novo transcription during the first 4 h, relative to that in N-depleted cells, and no evidence for increased PPR protein transcription. These results lend support to the growing consensus of post-transcriptional control of gene expression in dinoflagellates.

Sediment and water samples were collected from mangrove and estuarine biotopes at fortnightly intervals. The physico-chemical characters of the overlying water were studied. In the mangrove biotope maximum temperature (31.5C) and in the estuarine biotope maximum salinity (35.6) were recorded during the summer season, whereas in post-monsoon period the sulphate content was increased to 516 p.p.m. and the pH was reduced to 7.4. Invariably both in the enriched sediment and water samples four major peaks (at wavelengths 460, 705, 772 and 850 nm) and two minor peaks (at wavelengths 580 and 663 nm) of absorption spectra were noticed. A pure culture of Chromatium sp., isolated from mangroves sediment, showed three peaks of absorption spectra at wavelengths, 500, 580 and 850 nm. The effect of sodium chloride on the growth of Chromatium sp., was also studied and it was observed that maximum growth occurred in the range 1-3% sodium chloride concentration. This isolate was also capable of utilizing various sulphur and carbon compounds. Glycerol and glucose did not show any specific effect whereas pyruvate, malate and acetate increased the growth.

Recent transcription profiling studies have revealed an unexpectedly large proportion of antisense transcripts in eukaryotic genomes. These antisense genes seem to regulate gene expression by interacting with sense genes. Previous studies have focused on the non-coding antisense genes, but the possible regulatory role of the antisense protein is poorly understood. In this study, we found that a protein encoded by the antisense gene ADF1 acts as a transcription suppressor, regulating the expression of sense gene MDF1 in Saccharomyces cerevisiae. Based on the evolutionary, genetic, cytological and biochemical evidence, we show that the protein-coding sense gene MDF1 most likely originated de novo from a previously non-coding sequence and can significantly suppress the mating efficiency of baker's yeast in rich medium by binding MAT alpha 2 and thus promote vegetative growth. These results shed new light on several important issues, including a new sense-antisense interaction mechanism, the de novo origination of a functional gene, and the regulation of yeast mating pathway.

Using next-generation sequencing technology alone, we have successfully generated and assembled a draft sequence of the giant panda genome. The assembled contigs (2.25 gigabases (Gb)) cover approximately 94% of the whole genome, and the remaining gaps (0.05 Gb) seem to contain carnivore-specific repeats and tandem repeats. Comparisons with the dog and human showed that the panda genome has a lower divergence rate. The assessment of panda genes potentially underlying some of its unique traits indicated that its bamboo diet might be more dependent on its gut microbiome than its own genetic composition. We also identified more than 2.7 million heterozygous single nucleotide polymorphisms in the diploid genome. Our data and analyses provide a foundation for promoting mammalian genetic research, and demonstrate the feasibility for using next-generation sequencing technologies for accurate, cost-effective and rapid de novo assembly of large eukaryotic genomes.

Classical swine fever virus (CSFV) non-structural protein 5B (NS5B) encodes an RNA-dependent RNA polymerase (RdRp), a key enzyme which initiates RNA replication by a de novo mechanism without a primer and is a potential target for anti-virus therapy. We expressed the NS5B protein in Escherichia coli. The rGTP can stimulate de novo initiation of RNA synthesis and mutation of the GDD motif to Gly-Asp-Asp (GAA) abolishes the RNA synthesis. To better understand the mechanism of viral RNA synthesis in CSFV, a three-dimensional model was built by homology modeling based on the alignment with several virus RdRps. The model contains 605 residues folded in the characteristic fingers, palm and thumb domains. The fingers domain contains an N-terminal region that plays an important role in conformational change. We propose that the experimentally observed promotion of polymerase efficiency by rGTP is probably due to the conformational changes of the polymerase caused by binding the rGTP. Mutation of the GDD to GAA interferes with the interaction between the residues at the polymerase active site and metal ions, and thus renders the polymerase inactive. (c) 2005 Elsevier B.V.
