Since 1989, researchers with the Department of Primary Industries and Fisheries (DPI&F) in Queensland, Australia, have successfully used controlled low-water exchange green-water cultures to rear the larvae of estuarine fishes and crustaceans through to metamorphosis. High survivals and excellent fry condition have been achieved for several commercially important endemic species produced for various projects. They include barramundi or sea bass, Lates calcarifer, Australian bass, Macquaria novemaculeata, dusky flathead, Platycephalus fuscus, sand whiting, Sillago ciliata, red sea bream or snapper, Pagrus auratus, banana prawn, Fenneropenaeus merguiensis, and others. The consistent success of our standardised and relatively simple approach at different localities has led to it being incorporated into general fingerling production practices at several establishments in Australia. Although post-metamorphosis rearing methods have differed for each species investigated, due to various biological and behavioural traits and project requirements, these larval rearing methods have been successful with few species-specific modifications. Initially modelled on the Taiwanese approach to rearing Penaeids in aerated low-water exchange cultures, the approach similarly appears to rely on a beneficial assemblage of micro-organisms. Conceptually, these micro-organisms may include a mixture of the air-borne primary invaders of pure phytoplankton cultures when exposed to outdoor conditions. Whilst this would vary with different sites, our experiences with these methods have consistently been favourable. Mass microalgal cultures with eco-physiological youth are used to regularly augment larval fish cultures so that rearing conditions simulate an exponential growth-phase microalgal bloom. Moderate to heavy aeration prevents settlement of particulate matter and encourages aerobic bacterial decomposition of wastes. The green-water larval rearing approach described herein has demonstrated high practical utility in research and commercial applications, and has greatly simplified marine finfish hatchery operations whilst generally lifting production capacities for metamorphosed fry in Australia. Its potential uses in areas of aquaculture other than larviculture are also discussed.

Herpesviral haematopoietic necrosis is a disease of goldfish, Carassius auratus, caused by Cyprinid herpesvirus-2 (CyHV-2) infection. Quantitative PCR was carried out on tissue homogenates from healthy goldfish fingerlings, broodfish, eggs and fry directly sampled from commercial farms, from moribund fish submitted to our laboratory for disease diagnosis, and on naturally-infected CyHV-2 carriers subjected to experimental stress treatments. Healthy fish from 14 of 18 farms were positive with copy numbers ranging from tens to 10(7) copies mu g(-1) DNA extracted from infected fish. Of 118 pools of broodfish tested, 42 were positive. The CyHV-2 was detected in one lot of fry produced from disinfected eggs. Testing of moribund goldfish, in which we could not detect any other pathogens, produced 12 of 30 cases with 10(6)-10(8) copies of CyHV-2 mu g(-1) DNA extracted. Subjecting healthy CyHV-2 carriers to cold shock (22-10 degrees C) but not heat, ammonia or high pH, increased viral copy numbers from mean copy number (+/- SE) of 7.3 +/- 11 to 394 +/- 55 mu g(-1) DNA extracted after 24 h. CyHV-2 is widespread on commercial goldfish farms and outbreaks apparently occur when healthy carriers are subjected to a sharp temperature drop followed by holding at the permissive temperature for the disease.

This study identified Gram-positive bacteria in three sub-tropical marine fish species: Pseudocaranx dentex (silver trevally), Pagrus auratus (snapper) and Mugil cephalus (sea mullet). It further elucidated the role played by fish habitat, fish body part and ambient storage on the composition of the Gram-positive bacteria. A total of 266 isolates of Gram-positive bacteria were identified by conventional biochemical methods, VITEK, PCR using genus- and species-specific primers and/or 16S rRNA gene sequencing. The isolates were found to fall into 13 genera and 30 species. In fresh fish, Staphylococcus epidermidis and Micrococcus luteus were the most frequent isolates. After ambient storage, S. epidermidis, S. xylosus and Bacillus megaterium were no longer present whereas S. warned, B. sphaericus, Brevibacillus borstelensis, Enterococcus faecium and Streptococcus uberis increased in frequency. Micrococcus luteus and S. warned were the most prevalent isolates from P. dentex, while E. faecium and Strep. uberis were the most frequent isolates from P. auratus and M. cephalus. With respect to different parts of the fish body. E. faecium, Strep. uberis and B. sphaericus were the most frequent isolates from the muscles, E. faecium, Strep. uberis from the gills and M. luteus from the gut. This study showed a diversity of Gram-positive bacteria in sub-tropical marine fish; however, their abundance was affected by fish habitat, fish body part and ambient storage.

Survey of Fungal infestation of some species of fish in Tagwai Dam Minna was carried out from March to June 2002. Fungi were isolated from the scale/skin, gills and fins. Twenty-one fungi species were identified from 18 species of fish microbial growth was measured by direct cell count using Stuart colony counter. Most of the fungi encountered were of the mould group and infestation occurred among all the species sampled. The infestation was predominantly by Aspergillus species and the scale/skin was most widely affected. The study showed the identified fungi in order of frequency to be as follows. Aspergillius niger. Rrhizopus spp. Mmucor spp Aspergilus flavum. Aspergillus parasitous. Aspergillus fumigatus, Microsporum canis. Penicillum virridicalumand Fusarium spp respectively. Aspergillus niger occurred on all the species of fish sampled. Barrilius spa and Chrysichrhy auratus longifilis had significantly higher (P<0.05) mean fungal load on their fins and gills. There was no significant difference (P>0.05) in the mean fungal load in different parts of the body of other fish species sampled

Parameters of the von Bertalanffy growth function are presented for 42 fish stocks belonging to 16 families, 22 genera and 27 species. The growth performance index, Phi '(= log K + 2logL sub( infinity )), was computed for each stock and was found to be highest in male Gymnarchus niloticus (Gymnarchidae) from Lake Chad and lowest in Chrysichthys auratus (Bagridae) from the Cross River. Mean Phi ' for major fish genera and families are also presented and was highest in brackishwater fishes, closely followed by freshwater and inshore marine water fishes.

Quando as esterases acetilcolinesterase (AChE), butirilcolinesterase (BChE) e carboxilesterase (CarbE) hidrolisam ésteres de fosfato seus sítios ativos sofrem fosfatação inibitória. Por isto, tal fosfatação pode proteger seres vivos contra o espalhamento de xenobióticos organofosforados dentro de seus corpos, já que estas enzimas têm a capacidade de captar moléculas de pesticidas organofosforados estequiometricamente. Os organismos terrestres vivem em um ambiente com mais oxigênio do que os organismos aquáticos. Na água, quando o nível de oxigênio atinge aproximadamente 2,6 mg/L o ambiente está em hipoxia. Este fenômeno afeta ecossistemas aquáticos, uma vez que muitos organismos não conseguem se adaptar à baixa do oxigênio. Estudamos peixes em hipoxia e hiperoxia para entender melhor a bioquímica do funcionamento de suas enzimas captadoras de organofosforados quando eles estão expostos às variações físico-químicas de seus habitats. Dois grupos de no mínimo seis pacus (Piaractus mesopotamicus), seis peixes dourados (Carassius auratus auratus), seis tilápias (Oreochromis niloticus niloticus), seis piavussus (Leporinus macrocephalus), seis apaiaris (Astronotus ocellatus), ou seis carpas (Cyprinus carpio carpio) foram aclimatados à temperatura ambiente em dois aquários de 250 L. No primeiro aquário, pelo menos três animais ensaio de cada espécie sofreram hipoxia por diminuição da concentração de oxigênio até 0,5 mg/L através de borbulhamento de nitrogênio na água. Quando estes animais atingiram a hipoxia foram mantidos a 0,5 mg/L de oxigênio por 6, 8, 24 ou, no máximo, por 42 horas. Três peixes controle de cada espécie foram mantidos em normoxia (4,5 até 7,0 mg/L de oxigênio). Após estes tempos houve a retirada de cerca de 3,5 mL de sangue e dos fígados. Depois de coagular, o sangue foi centrifugado para retirada do soro sobrenadante, que foi usado como amostra para ensaios das esterases. Os fígados foram armazenados em freezer a -70 C e, no momento do ensaio, homogeneizados e centrifugados para obter as frações citosólica e microssomal. As atividades das esterases foram ensaiadas em espectrofotômetro com os substratos acetiltiocolina, butiriltiocolina ou p-nitrofenilacetato. As atividades sobre p-nitrofenilacetato (CarbE) do soro e do fígado sofreram queda em todos os exemplares das espécies submetidos à hipoxia. Tipicamente, esta atividade caiu cerca de 50% nos soros de pacus mantidos por 42 h sob concentrações de oxigênio abaixo de 1,0 mg/L. O tempo para que ocorresse a queda desta atividade enzimática variou de espécie para espécie.

We present a method to integrate environmental time series into stock assessment models and to test the significance of correlations between population processes and the environmental time series. Parameters that relate the environmental time series to population processes are included in the stock assessment model, and likelihood ratio tests are used to determine if the parameters improve the fit to the data significantly. Two approaches are considered to integrate the environmental relationship. In the environmental model, the population dynamics process (e.g. recruitment) is proportional to the environmental variable, whereas in the environmental model with process error it is proportional to the environmental variable, but the model allows an additional temporal variation (process error) constrained by a log-normal distribution. The methods are tested by using simulation analysis and compared to the traditional method of correlating model estimates with environmental variables outside the estimation procedure. In the traditional method, the estimates of recruitment were provided by a model that allowed the recruitment only to have a temporal variation constrained by a log-normal distribution. We illustrate the methods by applying them to test the statistical significance of the correlation between sea-surface temperature (SST) and recruitment to the snapper (Pagrus auratus) stock in the Hauraki Gulf–Bay of Plenty, New Zealand. Simulation analyses indicated that the integrated approach with additional process error is superior to the traditional method of correlating model estimates with environmental variables outside the estimation procedure. The results suggest that, for the snapper stock, recruitment is positively correlated with SST at the time of spawning.

Bacterial flora associated with tail rot/fin rot of Carassius auratus, Xiphophorus helleri and hemorrhagic ulcers of Clarias spp were studied. Sensitivity pattern of 33 isolates comprising Aeromonas spp, Pseudomonas spp and Gram-positive rods from diseased C. auratus, X. helleri and Clarias spp were screened against six broad-spectrum antibiotics viz. ciprofloxacin, chloramphenicol, co-trimoxazole, gentamycin, nitro-furantoin and oxytetracycline. Ciprofloxacin was the most effective in inhibiting bacteria at 0.05-0.10 µg/ml level. About 44% of Pseudomonas spp. was resistant to nitrofurantoin. Resistance to oxytetracycline was seen in 27% of Aeromonas spp Gram-positive rods were comparatively more resistant to antibiotics. The multiple antibiotic resistances were seen in 21% of the bacterial isolates of diseased fish.

The effects of dietary supplementation of commercial human probiotic, Lactobacil and antibiotic, oxytetracycline on the growth, survival, disease resistance and content of intestinal microflora in two ornamental fishes, viz., goldfish, Carassius auratus and swordtail, Xiphophorus helleri were studied. The total wet weight gain, food conversion ratio and specific growth rate of C. auratus did not vary significantly (p>0.05) among treatments. While in X. helleri, significant differences existed in the total wet weight gain, survival, food conversion ratio and specific growth rate among treatment groups (p<0.05). The counts of antibiotic resistant bacteria in fish gut increased with days of culture in all the treatments and the increase was more in antibiotic fed fishes. A reduction in the development of antibiotic resistance among the bacterial flora of fish gut was noticed in probiotic fed groups of C auratus and X. helleri. The results of the present study revealed that the effects of human probiotic on the growth, survival and disease resistance of ornamental fishes are variable and difficult to reproduce the similar effect on different species.

A number of wide-ranging monitoring studies have been performed in order to estimate the degree of mercury (Hg) contamination in freshwater ecosystems. Knowledge regarding contamination of different levels of the food chain is necessary for estimation of total pollutant input fluxes and subsequent partitioning among different phases in the aquatic system. The growing international concern about this environmental data is closely related to the strongly developing ecological risk assessment activities. In addition,freshwater monitoring outputs hold a key position in the estimation of the Hg dose consumed by the human population as it is highly dependent on fish consumption. So monitoring of Hg in the tissue of edible fish is extremely important because of contaminated fish has caused serious neurological damage to new born babies and adults. Mercury tends to accumulate in fish tissue, particularly, in the form of methyl mercury, which is about 10 times more toxic than inorganic mercury. The Anzali lagoon is one of the biggest wetland of Guilan province, which joins to the Caspian sea. Many Chemical and industrial factories plus agricultural runoffs and urban and rural sewages are major polluting sources of the Anzali wetland. Since many of those polluting sources drain their wastes directly or indirectly into the Anzali wetland and their sewages may be polluted with Hg, this study was conducted to find out the bioaccumulation of Hg bioaccumulation in pike (Esox lucius) food chain from Anzali lagoon, Iran. Sampling were carried out from July 2004 to July 2005, in addition 318 speciments of 9 fish species were collected. T-Hg was measured by LECO AMA 254 Advanced Mercury Analyzer (USA) according to ASTM standard No D-6722. Each sample was analyzed 3 times. Accuracy of T-Hg analysis was checked by running three samples of Standard Reference Materials; SRM 1633b, SRM 2711 & Sra 2709. Detection limit was 0.001 mg/kg in dry weight. The Accuracy degree of analyzor equipment with RSD<%0.05 (N=7) was between %95.5 and %105. In overal eigth fish species were distingushed in the gut content of 87 speciments of pike with age 1-5 year and maximum length 550mm. The max. and min. concentration of T-Hg in dorsal muscle of pjke was 0.2ppm in one year and 1.2ppm in five year class. The mean of T-Hg significantly increased with age and length increased (P<0.05).Mercury accumulation pattern in pike was as well as muscle > liver > spleen (P<0.05). THg content in female was higher than male(P<0.05). In contrast the mean of THg concentration in dorsal muscle of eigth fish species as prey was 0.282, 0.261, 0.328, 0.254, 0.256, 0.286, 0.322 and 0.241 ppm for Carassius auratus gibelio, Hemiculter leucisculus, Blicca bjoerkna transcaucasica, Chalcalburnus mossulensis, Rhodeus sericeus amarus, Gambusia holbrooki, Alburnus charusini hohenackeri & Scardinius Erythrophthalmus respectively.Liner regresion indicated that high degree of relationship between age of pike and Uptak/Intake ratio (R2=%99.12) and indicated that the mercury bioaccumulation in the pike dorsal muscle increased with age increased. BFA was >1 and and indicating the mercury biomagnification in the pike food chain. Trophy level of pike in the Anzali lagoon was estimated as well as 3.5 and 4 . It is generally agreed that Hg concentration in carnivorous fish are higher than in noncarnivorous species.

In the present study, ectoparasite infestation of common freshwater ornamental fish species imported into Iran including: Dwarf gourami (Colisa lalia), Angelfish (Pterophyllum scalare), Oscar (Astronatus ocellatus), Red eye Tetra ( Moenkhansia Sanctaefilomenae), Barb( Capoeta tetrazona), Arowana (Osteoglossum bicirrhosum), Goldfish(Carassius auratus), Red fin Shark (Lebeo elythrurus, Catfish (Hypostomus plecostomus) and Pangasius sutchi ( Pangasius hypophthalmus) from May until April was investigated.A total of 6 specimens sample of each fish species) randomly were obtained and taken alive to the lab. After observing gill and skin wet smear under the microscope , gill was dissected and examined carefully .Photo and films were taken from the isolated parasites and parasite identification was performed according to Yamaguti AP1), Bychowsky (■Y).From a total of examined fishes 1\ ► sample (YY.V.) were parasitized .Parasite groups that were observed consisting ciliated protozoan myxosporidian , monogenean Digenean metacercaria and crustacean .The only non-parasitized fish was Barb. Monogenea Trianchoratus sp. , Cleidodiscus sp. , Ancylodiscoides sp. , Thaparocleidus sp. , Centrocestus formosanus metacercaria and Myxobolus longisporus report for the first time in ornamental fish which imported from Southeast of Asia to Iran. According to the results, it seems that severe quarantine and sanitary rules are necessary.

银鲫(Carassius auratus gibelio Bloch)由于其独特的遗传背景和繁殖方式而成为研究进化遗传学和选择育种的一个独特的模式生物。到目前为止,我们对新疆额尔齐斯河水系银鲫群体的多样性状况一直知之甚少。为了更好地了解额尔齐斯河水系银鲫群体的克隆多样性状况,本研究中,我们采集了来自新疆额尔齐斯河水系的4个鲫鱼群体。通过流式细胞术分析血细胞样品,结果证实这些鲫鱼均为三倍体银鲫。通过血清转铁蛋白电泳表型分析,我们从这些银鲫群体中鉴定出总共8个不同的克隆。在这些鉴定的克隆中,有4个克隆(克隆A

用细胞色素b(Cyt b)基因特异性引物,对萍乡肉红鲫(Carassius auratus var.pingxiangnensis)的线粒体Cytb基因进行PCR扩增和双向测序。在12个个体中均得到序列一致的Cytb基因全序列,长度为1 140 bp。其A、T、G、C含量分别为28.2%(321)、28.8%(328)、14.8%(170)和28.2%(321),A+T含量(57%)明显高于G+C含量(43%),与其他水生动物相同基因片段碱基含量相似。该基因中密码子第1位核苷酸中4种碱基组成较为均衡;第2

采用腹腔注射和混饲投喂两种方式研究中药甘草(Glycyrrhiza uralensis Fiseh)粗提物对鲫(Carassius auratus)的抗应激和免疫保护功效。将一部分粗提物按0.5%和2%的质量分数制成药饵投喂实验鲫,分别在第14 d、28 d、35 d、42 d、56 d采集血样;将另一部分粗提物用无菌生理盐水按0.5%和2%的浓度配成针剂腹腔注射鲫,分别在第3 d、7 d、14 d、21 d、28 d采集血样。结果显示:投喂实验中药物组和对照组血清皮质醇水平均呈先升后降趋势,除第14 d
