The LY12-cz aluminium alloy sheet specimens with a central hole were tested under constant amplitude loading, Rayleigh narrow band random loading and a typical fighter broad band random loading. The fatigue life was estimated by means of the nominal stress and the Miner's rule. The stress cycles were distinguished by the rainflow count, range count and peak value count, respectively. The comparison between the estimated results and the test results was made. The effects of random loading sequence and small load cycles on fatigue life were also studied.

The effects of stress on both microalgal and macroalgal communities are considered. On one hand the contrasting approaches of studies of these two communities reflect intrinsic differences in plant size, longevity and ease of handling. On the other hand they reveal that biological monitoring of the potentially deleterious effects of man's activities has focused largely on freshwater environments in which macroalgae only occasionally dominate. Large conspicuous plants can be readily investigated as individuals, whereas it is virtually impossible to trace effects of stress on an individual cell of a vegetatively-reproducing microalga; a population approach is almost inevitably necessary. However, rapid turnover rates, a spectrum of ecological characteristics distributed between many taxa, and the potential for statistical analysis, have facilitated the use of microalgae in environmental impact studies. Failure to extend such investigations into marine systems rests as much on man's ability to ignore environmental deterioration until it affects his quality of life as on the visual dominance of seaweeds around our coasts. However, large gaps remain in our knowledge of both large and small algae; some reported community changes over time are suspect, and the causes of even blatant changes are not always apparent.

The role of life-history theory in population and evolutionary analyses is outlined. In both cases general life histories can be analysed, but simpler life histories need fewer parameters for their description. The simplest case, of semelparous (breed-once-then-die) organisms, needs only three parameters: somatic growth rate, mortality rate and fecundity. This case is analysed in detail. If fecundity is fixed, population growth rate can be calculated direct from mortality rate and somatic growth rate, and isoclines on which population growth rate is constant can be drawn in a ”state space” with axes for mortality rate and somatic growth rate. In this space density-dependence is likely to result in a population trajectory from low density, when mortality rate is low and somatic growth rate is high and the population increases (positive population growth rate) to high density, after which the process reverses to return to low density. Possible effects of pollution on this system are discussed. The state-space approach allows direct population analysis of the twin effects of pollution and density on population growth rate. Evolutionary analysis uses related methods to identify likely evolutionary outcomes when an organism's genetic options are subject to trade-offs. The trade-off considered here is between somatic growth rate and mortality rate. Such a trade-off could arise because of an energy allocation trade-off if resources spent on personal defence (reducing mortality rate) are not available for somatic growth rate. The evolutionary implications of pollution acting on such a trade-off are outlined.

This paper reviews the effectiveness of Gammarus scope for growth (SfG) as an indicator of water quality. In addition, the link between physiological changes and effects at higher levels of biological organisation is addressed. Exposure to a range of toxicants resulted in decreases in Gammarus SfG which were qualitatively and quantitatively correlated with subsequent reductions in growth and reproduction. Reductions in SfG were due principally to a decrease in energy intake (i.e. feeding rate) rather than an increase in energy expenditure. Gammarus pulex is an important shredder in many stream communities and stressed-induced reductions in its feeding activity were correlated with reductions in the processing of leaf litter by a semi-natural stream community. Hence, changes in the physiological energetics of Gammarus provide a general and sensitive indicator of stress which can be linked to effects at higher levels of biological organisation. Under long-term stress and hence prolonged reductions in SfG, animals may adapt by modifying their life-history strategies and producing fewer, larger offspring.

Introdução: o óxido nítrico (NO) é um gás inorgânico com uma meia-vida curta e tem um papel crítico na manutenção da homeostase vascular e fluidez sanguínea. O NO é sintetizado a partir do aminoácido L-arginina por uma família de enzimas NO sintases (NOS). Estudos têm mostrado que eritrócitos expressam NOS endotelial (eNOS) funcional, que serve como uma fonte de NO intraluminal. Além disso, eritrócitos participam da defesa antioxidante removendo os radicais livres e prevenindo o dano oxidativo às membranas biológicas e a destruição do NO. Dietas hiperlípidicas estão associadas a um risco aumentado de doença cardiovacular e síndrome metabólica, mas os exatos mecanismos não estão completamente esclarecidos. O objetivo deste estudo foi investigar os efeitos de diferentes dietas hiperlípidicas na via L-arginina-NO e o estresse oxidativo em eritrócitos de camundongos. Metodologia: camundongos machos C57BL/6 de três meses de idade receberam diferentes dietas por 10 semanas: dieta normolipídica ou dieta hiperlipídica contendo banha de porco (HB), óleo de oliva (HO), óleo de girassol (HG) ou óleo de canola (HC). Foram analisados o transporte de L-arginina mediado pelos transportadores catiônicos y+ e y+L, a atividade da NOS, a expressão da eNOS e da NOS induzível (iNOS), a formação de substâncias reativas ao ácido tiobarbitúrico (TBARS) e a atividade das enzimas antioxidantes catalase (CAT) e superóxido dismutase (SOD). Resultados: o transporte total de L-arginina estava aumentado no grupo HO em comparação aos controles e aos outros grupos com dieta hiperlipídica. Quando o transporte foi fracionado, o sistema y+ estava mais ativado no grupo HO em relação aos controles e outros grupos que receberam dieta hiperlipídica. O transporte de L-arginina via sistema y+L estava maior nos grupos HO, HG e HC comparados aos grupos controle e HB. Adicionalmente, a atividade basal da NOS e a expressão de eNOS estavam aumentadas em eritrócitos independente do tipo de dieta hiperlípidica insaturada. Observou-se uma maior expressão da iNOS no grupo HO comparado ao controle. Em contraste, o grupo HB apresentou uma inibição da via L-arginina-NO. A análise da peroxidação lipídica, através da formação de TBARS, e da atividade da enzima antioxidante CAT não revelou diferenças entre os grupos, ao contrário do grupo HO, que induziu uma ativação de outra enzima antioxidante, a SOD. Conclusões: o presente estudo proporciona a primeira evidência de que os sistemas y+ e y+L regulam o transporte aumentado de L-arginina em eritrócitos de camundongos do grupo HO. Além disso, todas as dietas hiperlipídicas insaturadas induzem um aumento da atividade basal da NOS associada a uma expressão elevada da eNOS. É possível que diferentes mudanças na composição lipídica da membrana plasmática induzidas pelas dietas possam afetar transportadores e enzimas nos eritrócitos. Além disso, a inibição da via L-arginina-NO no grupo HB pode contribuir para o desenvolvimento da aterosclerose, enquanto dietas hiperlipídicas insaturadas podem ter um efeito protetor via aumento da geração de NO.

Ichthyoplankton surveys have been used to provide an independent estimate of adult spawning biomass of commercially exploited species and to further our understanding of the recruitment processes in the early life stages. However, predicting recruitment has been difficult because of the complex interaction of physical and biological processes operating at different spatial and temporal scales that can occur at the different life stages. A model of first-year life-stage recruitment was applied to Georges Bank Atlantic cod (Gadus morhua) and haddock (Melanogrammus aeglefinus) stocks over the years 1977–2004 by using environmental and densitydependent relationships. The best lifestage mortality relationships for eggs, larvae, pelagic juveniles, and demersal juveniles were first determined by hindcasting recruitment estimates based on egg and larval abundance and mortality rates derived from two intensive sampling periods, 1977–87 and 1995–99. A wind-driven egg mortality relationship was used to estimate losses due to transport off the bank, and a wind-stress larval mortality relationship was derived from feeding and survival studies. A simple metric for the density-dependent effects of Atlantic cod was used for both Atlantic cod and haddock. These life stage proxies were then applied to the virtual population analysis (VPA) derived annual egg abundances to predict age-1 recruitment. Best models were determined from the correlation of predicted and VPA-derived age-1 abundance. The larval stage was the most quantifiable of any stage from surveys, whereas abundance estimates of the demersal juvenile stage were not available because of undersampling. Attempts to forecast recruitment from spawning stock biomass or egg abundance, however, will always be poor because of variable egg survival.

Not all experiences are memorized equally well. Especially, some types of stress are unavoidable in daily life and the stress experience can be memorized for life. Previous evidence has showed that synaptic plasticity, such as long-term potentiation (LTP) that may be the major cellular model of the mechanism underlying learning and memory, is influenced by behavioral stress. However, the effect of behavioral stress on age-related synaptic plasticity in-vivo was primarily known. Here we found that the LTP induction in the hippocampal CA1 region of anesthetized rats obviously showed inverted-U shape related to ages (4, 10 and 74 weeks old rats), but low-frequency stimulation was unable to induce reliable long-term depression (LTD) in these animals. Furthermore, acute elevated platform (EP) stress enabled reliable LTD significantly and completely blocked LTP induction at these ages. Importantly, LTD after exposure to acute EP stress showed similar magnitude over these ages. The present results that stress enables LTD but impairs LTP induction at these three ages strengthen a view that stress experience-dependent LTD (SLTD) may underlie stress form of aberrant memories. Recurrence is a key characteristic in the development of epilepsy. It remains unclear whether seizure recurrence is sensitive to postseizure stress. Here, tonic-clonic seizures were induced with a convulsive dose of pentylenetetrazole (PTZ), and acute seizure recurrence was evoked with a subconvulsive dose of the drug. We found that stress inhibited seizure recurrence when applied 30 minutes or 2 hours, but not 4 hours, after the tonic-clonic seizure. The time-dependent anti-recurrence effect of stress was mimicked by the stress hormone corticosterone and blocked by co-administration of mineralocorticoid and glucocorticoid receptor antagonists. Furthermore, in a PTZ-induced epileptic kindling model, corticosterone administered 30 minutes after each seizure decreased the extent of seizures both during the kindling establishment and in the following challenge test. These results provide novel insights into both the mechanisms of and therapeutic strategies for epilepsy. (C) 2010 Elsevier Inc. All rights reserved.

Flare tips are essential for safety. Maintenance is difficult and costly. Flare tips are subjected to high combustion temperatures, thermal cycling, oxidation and marine corrosion. Following a number of flare tip failures an in depth study by Imperial College was carried out into the failure of a flare tip from a UK platform, looking for service life improvement. Materials selection and design solutions were considered. The study considered alternative materials and concluded that materials selection was the smaller part of the answer; design changes can double service life. This study used failure investigation, high temperature experimental and thermo-mechanical modelling analysis. The modelling process simulated two common flaring conditions and correctly predicted the observed failure of initiation and crack propagation from holes used to bolt on flame stabilizing plates to the top of the flare. The calculated thermal stress and strains enabled the low cycle fatigue life and minimum creep life to be predicted. It was concluded that service life could be improved by replacing Incoloy alloy 800HT (UNS N08800) with Inconel alloy 625 (UNS N06625), an alloy with attractive mechanical properties and improved high temperature corrosion resistance. Repositioning or eliminating bolt holes can double service life. Copyright 2008, Society of Petroleum Engineers.

Many unicellular green algae can become yellow or red in various natural habitats due to mass accumulation of a secondary carotenoid, such as lutein, or astaxanthin. The accumulation of secondary carotenoids is generally thought to be a survival strategy of the algae under photo-oxidative stress or other adverse environmental conditions. The physiological role of the carotenoids in stress response is less well understood at the subcellular or molecular level. In this study, a stable astaxanthin overproduction mutant (MT 2877) was isolated by chemical mutagenesis of a wild type (WT) of the green microalga Haematococcus pluvialis Flotow NIES-144. MT 2877 was identical to the WT with respect to morphology, pigment composition, and growth kinetics during the early vegetative stage of the life cycle. However, it had the ability to synthesize and accumulate about twice the astaxanthin content of the WT under high light, or under high light in the presence of excess amounts of ferrous sulphate and sodium acetate. Under stress, the mutant exhibited higher photosynthetic activities than the WT, based on considerably higher chlorophyll fluorescence induction, chlorophyll autofluorescence intensities, and oxygen evolution rates. Cell mortality caused by stress was reduced by half in the mutant culture compared with the WT. Enhanced protection of the mutant against stress is attributed to its accelerated carotenogenesis and accumulation of astaxanthin. Our results suggest that MT 2877, or other astaxanthin overproduction Haematococcus mutants, may offer dual benefits, as compared with the wild type, by increasing cellular astaxanthin content while reducing cell mortality during stress-induced carotenogenesis.

The early life-history of Chinese rock carp Procypris rabaudi was investigated during a 56-day rearing period: 318 artificially propagated P. rabaudi larvae were reared throughout metamorphosis in a small-scale recirculation system (345 L water volume, 10 x 18 L rearing tanks, 150 L storage and filter compartment with bioballs, 20-30 larvae L-1) at the Institute of Hydrobiology, Wuhan, Hubei Province, China. The newly hatched larvae had an initial total length of 8.93 +/- 0.35 mm SD (n = 10) at 3 days post-hatch and reached an average total length of 33.29 mm (+/- 1.88 mm SD, n = 10) 56 days after hatching. Length increment averaged 0.45 mm day(-1), resulting in a mean growth of 24.4 mm within the 56-day period. High mortality rates of up to 92% derived from an introduced fungus infection and subsequent treatment stress with malachite green. Our results indicate that Chinese rock carp can be raised successfully from artificially fertilized eggs. We therefore assume this species to be a candidate for commercial aquaculture.

With the advancement in network bandwidth and computing power, multimedia systems have become a popular means for information delivery. However, general principles of system testing cannot be directly applied to testing of multimedia systems on account of their stringent temporal and synchronization requirements. In particular, few studies have been made on the stress testing of multimedia systems with respect to their temporal requirements under resource saturation. Stress testing is important because erroneous behavior is most likely to occur under resource saturation. This paper presents an automatable method of test case generation for the stress testing of multimedia systems. It adapts constraint solving techniques to generate test cases that lead to potential resource saturation in a multimedia system. Coverage of the test cases is defined upon the reachability graph of a multimedia system. The proposed stress testing technique is supported by tools and has been successfully applied to a real-life commercial multimedia system. Although our technique focuses on the stress testing of multimedia systems, the underlying issues and concepts are applicable to other types of real-time systems.

Polysaccharides isolated from Porphyra (porphyran) have been known to have diverse biological activities, including immunomodulatory and antioxidant activities. The molecular weight-antiaging activity relationship of degraded porphyrans was examined in this study. Natural porphyran was extracted from P. haitanensis, and then was degraded into different molecular weight fractions, P1 molecular weight 49 kDa, P2 molecular weight 30 kDa, P3 molecular weight 8.2 kDa, by free radical. The influence on life span and vitality of porphyrans were carried out on Drosophila melanogaster. We found that all the degraded porphyrans and natural porphyran (P), added daily to the diet, can significantly increase the life span of D. melanogaster, except for P3. Among them, P1 exhibited the most prolonging life span activity. Furthermore, vitality of middle-aged flies (assessed by measuring their mating capacity) receiving porphyrans was increased considerably in comparison with the controls. Finally, in the heat-stress test, we observed a remarkable increase in survival time, especially in P3-diet groups. These results suggest that porphyrans may be effective in reducing the rate of the aging process and molecular weight has important influence on the effects. It seems that P1 and P2, possessed higher molecular weight, may be more useful in normal metabolic condition and P3, possessed the lowest molecular weight, may be more beneficial for D. melanogaster in stress condition. Childhood sexual abuse is prevalent among people living with HIV, and the experience of shame is a common consequence of childhood sexual abuse and HIV infection. This study examined the role of shame in health-related quality of life among HIV-positive adults who have experienced childhood sexual abuse. Data from 247 HIV-infected adults with a history of childhood sexual abuse were analyzed. Hierarchical linear regression was conducted to assess the impact of shame regarding both sexual abuse and HIV infection, while controlling for demographic, clinical, and psychosocial factors. In bivariate analyses, shame regarding sexual abuse and HIV infection were each negatively associated with health-related quality of life and its components (physical well-being, function and global well-being, emotional and social well-being, and cognitive functioning). After controlling for demographic, clinical, and psychosocial factors, HIV-related, but not sexual abuse-related, shame remained a significant predictor of reduced health-related quality of life, explaining up to 10% of the variance in multivariable models for overall health-related quality of life, emotional, function and global, and social well-being and cognitive functioning over and above that of other variables entered into the model. Additionally, HIV symptoms, perceived stress, and perceived availability of social support were associated with health-related quality of life in multivariable models. Shame is an important and modifiable predictor of health-related quality of life in HIV-positive populations, and medical and mental health providers serving HIV-infected populations should be aware of the importance of shame and its impact on the well-being of their patients.
