Using a direct binding assay based on photoaffinity labeling, we studied the interaction of T cell receptor (TCR) with a Kd-bound photoreactive peptide derivative on living cells. The Kd-restricted Plasmodium berghei circumsporozoite (PbCS) peptide 253-260 (YIPSAEKI) was reacted NH2-terminally with biotin and at the TCR contact residue Lys259 with photoreactive iodo, 4-azido salicylic acid (IASA) to make biotin-YIPSAEK(IASA)I. Cytotoxic T lymphocyte (CTL) clones derived from mice immunized with this derivative recognized this conjugate, but not a related one lacking the IASA group nor the parental PbCS peptide. The clones were Kd restricted. Recognition experiments with variant conjugates, lacking substituents from IASA, revealed a diverse fine specificity pattern and indicated that this group interacted directly with the TCR. The TCR of four clones could be photoaffinity labeled by biotin-YIPSAEK(125IASA)I. This labeling was dependent on the conjugates binding to the Kd molecule and was selective for the TCR alpha (2 clones) or beta chain (1 clone), or was common for both chains (1 clone). TCR sequence analysis showed a preferential usage of J alpha TA28 containing alpha chains that were paired with V beta 1 expressing beta chains. The TCR that were photoaffinity labeled at the alpha chain expressed these J alpha and V beta segments. The tryptophan encoded by the J alpha TA28 segment is rarely found in other J alpha segments. Moreover, we show that the IASA group interacts preferentially with tryptophan in aqueous solution. We thus propose that for these CTL clones, labeling of the alpha chain occurs via the J alpha-encoded tryptophan residue.

The project aims at advancing the state of the art in the use of context information for classification of image and video data. The use of context in the classification of images has been showed of great importance to improve the performance of actual object recognition systems. In our project we proposed the concept of Multi-scale Feature Labels as a general and compact method to exploit the local and global context. The feature extraction from the discriminative probability or classification confidence label field is of great novelty. Moreover the use of a multi-scale representation of the feature labels lead to a compact and efficient description of the context. The goal of the project has been also to provide a general-purpose method and prove its suitability in different image/video analysis problem. The two-year project generated 5 journal publications (plus 2 under submission), 10 conference publications (plus 2 under submission) and one patent (plus 1 pending). Of these publications, a relevant number make use of the main result of this project to improve the results in detection and/or segmentation of objects.

We address the problem of comparing and characterizing the promoter regions of genes with similar expression patterns. This remains a challenging problem in sequence analysis, because often the promoter regions of co-expressed genes do not show discernible sequence conservation. In our approach, thus, we have not directly compared the nucleotide sequence of promoters. Instead, we have obtained predictions of transcription factor binding sites, annotated the predicted sites with the labels of the corresponding binding factors, and aligned the resulting sequences of labels—to which we refer here as transcription factor maps (TF-maps). To obtain the global pairwise alignment of two TF-maps, we have adapted an algorithm initially developed to align restriction enzyme maps. We have optimized the parameters of the algorithm in a small, but well-curated, collection of human–mouse orthologous gene pairs. Results in this dataset, as well as in an independent much larger dataset from the CISRED database, indicate that TF-map alignments are able to uncover conserved regulatory elements, which cannot be detected by the typical sequence alignments.

RESUMÉ Objectifs de la recherche: Depuis quelques années, l'utilisation de l'écologie dans le marketing a connu un essor considérable, aussi bien dans le monde académique que dans la pratique. De nos jours, la notion de label suscite un vif intérêt auprès des entreprises désireuses de promouvoir des produits "verts". Le principe de l'écolabellisation consiste à fournir aux consommateurs, en plus du prix, un nouvel élément de comparaison des produits. Les écolabels sont considérés comme l'un des meilleurs outils pour informer le consommateur, d'une manière claire et compréhensible, de l'impact du produit sur l'environnement. Nous nous intéressons, dans le cadre de notre travail, à l'étude du comportement d'achat des produits écolabellisés. En dépit de leur popularité croissante, les études académiques portant sur les labels écologiques sont relativement rares et de nombreuses problématiques demeurent en suspens. L'étude du comportement d'achat - au sens large - mérite notamment d'être approfondie. Notre recherche a plusieurs volets. Premièrement, nous étudions l'impact des valeurs, de l'implication vis-à-vis de l'écologie, du scepticisme, de la compréhension du label et de la connaissance de l'écologie sur le comportement d'achat de produits écolabellisés. Ensuite, nous testons la cohérence entre comportements écologiques en introduisant un comportement post- achat (le triage des déchets ménagers). Théories sous-jacentes: Notre étude repose sur différents apports académiques relatifs au comportement du consommateur "vert" mais également sur des concepts issus de la psychologie et de la sociologie. Nous présentons d'abord la littérature portant sur la caractérisation du consommateur "vert" (Webster, 1975; Arbuthnot, 1977; Van Liere et Dunlap, 1981; Balderjahn, 1988; Antil, 1984; Grunert et Juhl, 1995; Roberts, 1996). Nous abordons ensuite les études portant sur les valeurs (Schwartz, 1992; 1994), l'implication (Zaichkowsy, 1994), le scepticisme (Gray-Lee, Scammon et Mayer, 1994; Mohr et al., 1998), la compréhension des écolabels (van Dam et Reuvekamp, 1995; Thogersen, 2000) et la connaissance de l'écologie (Maloney et al., 1973, 1975; Arbuthnot, 1977; Pickett et al., 1993). Ces variables nous semblent être les plus à même d'influencer le comportement d'achat de produits écolabellisés. Enfin, sur la base des travaux de Valette-Florence et Roehrich (1993), nous développons un modèle de causalité, centré sur la relation entre les valeurs, l'implication et le comportement. Hypothèses de recherche et opérationnalisation des variables: Nous développons 16 hypothèses de recherche dont 12 portent sur les rapports de causalité entre construits. Par ailleurs, pour mieux comprendre la personnalité du consommateur de produits écolabellisés, nous distinguons les variables reflétant un intérêt collectif (altruisme) de celles reflétant un intérêt individuel (égocentrisme). La mesure des différents construits reposent sur la liste de Schwartz (1992, 1994) pour les valeurs, l'échelle d'implication de Zaichkowsy (1994) pour la publicité, l'échelle développée par Mohs et al. (1998) pour mesurer le scepticisme et une liste de questions portant sur l'écologie (Diekmann, 1996) pour tester le niveau de connaissance. Les comportements d'achat et post-achat sont mesurés respectivement à l'aide de questions relatives à la fréquence d'achat de produits écolabellisés et du triage des déchets ménagers. Collecte des données: Le recueil des données s'effectue par sondage, en ayant recours à des questionnaires auto-administrés. L'échantillon comprend de 368 étudiants provenant de diverses facultés de sciences humaines de Suisse Romande. Analyse des données et interprétation des résultats: Notre étude porte sur le comportement d'achat de trois labels écologiques et sur le triage de 7 déchets ménagers. Les différentes analyses montrent que certaines valeurs ont un impact sur l'implication et que l'implication a une influence positive sur le comportement d'achat et post-achat. Par ailleurs, nous montrons que l'implication sert de variable médiatrice entre les valeurs et le comportement. De plus, la compréhension de l'écolabel influence de manière positive l'achat de produits écolabellisés, la connaissance de l'écologie a une influence positive sur le comportement post-achat et le scepticisme vis-à-vis de l'utilité du triage des déchets ménagers influence négativement le comportement de triage. Implications managériales: Les résultats obtenus suggèrent qu'outre la valeur "protection de l'environnement", d'autres valeurs comme "la stimulation" ou encore "l'accomplissement" influencent de manière significative l'implication vis-à-vis de l'écologie qui à son tour influence l'achat de produits écolabellisés. Le manager doit donc tenir compte du fait que le consommateur est impliqué dans l'écologie. Ensuite, la compréhension du label joue un rôle prépondérant dans l'achat. De plus, le consommateur ne semble pas être sceptique par rapport aux informations fournies par les écolabels et le niveau de connaissance de l'écologie n'affecte pas son comportement. Enfin, le consommateur semble agir de manière cohérente en achetant différents produits écolabellisés. Apports, limites et voies de recherche: Notre étude contribue à l'enrichissement de la recherche sur le comportement du consommateur dans un contexte écologique à plusieurs égards, notamment par l'étude de la relation entre les valeurs, l'implication et le comportement. Néanmoins, la portée de notre recherche est naturellement restreinte en raison du nombre limité de cas étudiés, soit 3 labels écologiques relativement peu connus de notre échantillon. Il serait utile de répéter l'étude en utilisant des labels plus populaires. Par ailleurs, nous avons eu recours à des échelles développées dans des contextes quelque peu différents. En particulier, une échelle d'implication devrait être développée spécifiquement pour le contexte écologique.

The interaction of the T cell antigen receptor with a photoreactive antigenic peptide derivative bound covalently to the H-2Kd (Kd) molecule was studied by photoaffinity labeling on cloned, CD8 positive cytotoxic T lymphocytes. The Kd-restricted Plasmodium berghei circumsporozoite peptide 253-260 (YIPS-AEKI) was conjugated with iodo-4-azidosalicylic acid at the N terminus and with 4-azidobenzoic acid at the T cell receptor residue Lys-259. Cell-associated or soluble Kd molecules were photoaffinity-labeled with the peptide derivative by selective photoactivation of the N-terminal photoreactive group. Incubation of cell-associated or soluble covalent Kd-peptide derivative complexes (ligands) with cytotoxic T lymphocytes that recognized this peptide derivative and activation of the orthogonal photoreactive group resulted in specific photoaffinity labeling of the T cell receptor. The labeling was inhibitable by an anti-Kd antibody and was absent on Kd-restricted cytotoxic T lymphocytes of different specificity. The binding of the soluble ligand reached a maximum after 2-4 min at 37 degrees C, after 30 min at 18 degrees C, and after 3 h at 4 degrees C. In contrast, binding of the cell-associated ligand reached a transient maxima after 50 and 110 min at 37 and 18 degrees C, respectively. The degree of binding at 37 degrees C was approximately 30% lower than that at 18 degrees C. No binding took place at 4 degrees C. Inhibition studies with antibodies and drugs indicated that the binding of the cell-associated, but not the soluble ligand, was highly dependent on T cell-target cell conjugate formation, whereas the binding of the soluble ligand was greatly dependent on CD8.

User generated content shared in online communities is often described using collaborative tagging systems where users assign labels to content resources. As a result, a folksonomy emerges that relates a number of tags with the resources they label and the users that have used them. In this paper we analyze the folksonomy of Freesound, an online audio clip sharing site which contains more than two million users and 150,000 user-contributed sound samplescovering a wide variety of sounds. By following methodologies taken from similar studies, we compute some metrics that characterize the folksonomy both at the globallevel and at the tag level. In this manner, we are able to betterunderstand the behavior of the folksonomy as a whole, and also obtain some indicators that can be used as metadata for describing tags themselves. We expect that such a methodology for characterizing folksonomies can be useful to support processes such as tag recommendation or automatic annotation of online resources.

Malignant melanoma, the deadliest form of skin cancer, is characterized by a predominant mutation in the BRAF gene. Drugs that target tumours carrying this mutation have recently entered the clinic. Accordingly, patients are routinely screened for mutations in this gene to determine whether they can benefit from this type of treatment. The current gold standard for mutation screening uses real-time polymerase chain reaction and sequencing methods. Here we show that an assay based on microcantilever arrays can detect the mutation nanomechanically without amplification in total RNA samples isolated from melanoma cells. The assay is based on a BRAF-specific oligonucleotide probe. We detected mutant BRAF at a concentration of 500 pM in a 50-fold excess of the wild-type sequence. The method was able to distinguish melanoma cells carrying the mutation from wild-type cells using as little as 20 ng µl(-1) of RNA material, without prior PCR amplification and use of labels.

T lymphocytes recognize antigen in the form of peptides that associate with specific alleles of class I or class II major histocompatibility (MHC) molecules. By contrast with the clear MHC allele-specific binding of peptides to purified class II molecules purified solubilized class I molecules either bind relatively poorly or show degenerate specificity. Using photo-affinity labelling, we demonstrate here the specific interaction of peptides with cell-associated MHC class I molecules and show that this involves metabolically active processes.

Opinnäytetyö selvittää musiikkiteollisuuden ansaintalogiikkaa ja riippumattoman musiikintekijän tulomuotoja, sekä teknologisen kehityksen ja Internetin aiheuttaman rakennemuutoksen vaikutusta niihin. Alan nykyisenä ongelmana nähdään musiikintekijälle päätyvän rahan pieni osuus teollisuudessa liikkuvasta tulovirrasta. Työn tavoitteena on löytää kustannustehokkaita väyliä musiikin saattamiseen kuulijoille, pohtien samalla erilaisten promootiokanavien vaikutusarvoa. Lopullisena tarkoituksena on löytää tapa elättää itseään musiikin tekemisellä. Lähdeaineistona on käytetty alan kirjallisuutta, tilastoja, Internet-artikkeleita, sekä kirjoittajan omaa kokemusta alalta. Työ osoittaa, kuinka hankalaa musiikin tekemisellä tienaaminen on Suomessa. Kustannusten pitäminen mahdollisimman alhaisina ja asioiden tekeminen omatoimisesti mahdollisimman pitkälle ovat perusedellytys voitolliselle liiketoiminnalle. Digitaaliteknologia yhdessä Internetin kanssa tarjoaa mahdollisuuden tähän. Musiikintekijän merkittävimmiksi tulomuodoiksi osoittautuvat esiintymis- ja tekijänoikeuskorvaukset. Äänitteillä ansaitseminen on todennäköisintä pienlevy-yhtiöiden kanssa toimiessa, isojen levy-yhtiöiden raskas kulurakenne takaa sen että itse musiikintekijälle tuloutuu vain murto-osa tuotoista. Äänitteiden digijakelu ei vielä tarjoa varteenotettavaa tulonlähdettä musiikintekijälle, mutta sen merkitys kasvaa lähitulevaisuudessa, samoin kuin erilaisista edelleenlisensioinneista ja oheistuotteista kertyvät tulot.

We have tested the hypothesis that smaller alpha1B-adrenoceptor labeling by [3H]tamsulosin compared to [3H]prazosin is related to differential recognition of agonist low affinity states. Paired saturation binding experiments with [3H]prazosin and [3H]tamsulosin were performed in membrane preparations from rat liver and Rat- fibroblasts stably transfected with wild-type hamster alpha1B-adrenoceptors or a constitutively active mutant thereof. In all three settings [3H]tamsulosin labeled significantly fewer alpha1B-adrenoceptors than [3H]prazosin. In noradrenaline competition binding experiments, the percentage of agonist low affinity sites was smallest for the constitutively active alpha1B-adrenoceptor but the percentage of agonist low affinity sites recognized by [3H]tamsulosin and [3H]prazosin did not differ significantly. We conclude that [3H]tamsulosin labels fewer alpha1B-adrenoceptors than [3H]prazosin but this is not fully explained by a poorer labeling of agonist low affinity sites.

As worldwide consumer demand for high-quality products and for information about these products increases, labels and geographical indications (GIs) can serve to signal quality traits to consumers. However, GI systems among countries are not homogeneous and can be used as trade barriers against competition. Philosophical differences between the European Union and the United States about how GIs should be registered and protected led to the formation of a WTO dispute settlement panel. In this paper we discuss the issues behind the dispute, the World Trade Organization (WTO) panel decision, and the EU response to the panel decision leading to the new Regulation 510/2006. Given the potential for GI labels to supply consumer information, context is provided for the discussion using recent literature on product labeling. Implications are drawn regarding the importance of the panel decision and the EU response relative to GI issues yet to be negotiated under the Doha Round.

The subject "Value and prices in Russian economic thought (1890--1920)" should evoke several names and debates in the reader's mind. For a long time, Western scholars have been aware that the Russian economists Tugan-Baranovsky and Bortkiewicz were active participants to the Marxian transformation problem, that the mathematical models of Dmitriev prefigured forthcoming neoricardian based models, and that many Russian economists were either supporting the Marxian labour theory of value or being revisionists. Moreover, these ideas were preparing the ground for Soviet planning. Russian scholars additionally knew that this period was the time of introduction of marginalism in Russia, and that, during this period, economists were active in thinking the relation of ethics with economic theory. All these issues are well covered in the existing literature. But there is a big gap that this dissertation intends to fill. The existing literature handles these pieces separately, although they are part of a single, more general, history. All these issues (the labour theory of value, marginalism, the Marxian transformation problem, planning, ethics, mathematical economics) were part of what this dissertation calls here "The Russian synthesis". The Russian synthesis (in the singular) designates here all the attempts at synthesis between classical political economy and marginalism, between labour theory of value and marginal utility, and between value and prices that occurred in Russian economic thought between 1890 and 1920, and that embraces the whole set of issues evoked above. This dissertation has the ambition of being the first comprehensive history of that Russian synthesis. In this, this contribution is unique. It has always surprised the author of the present dissertation that such a book has not yet been written. Several good reasons, both in terms of scarce availability of sources and of ideological restrictions, may accounted for a reasonable delay of several decades. But it is now urgent to remedy the situation before the protagonists of the Russian synthesis are definitely classified under the wrong labels in the pantheon of economic thought. To accomplish this task, it has seldom be sufficient to gather together the various existing studies on aspects of this story. It as been necessary to return to the primary sources in the Russian language. The most important part of the primary literature has never been translated, and in the last years only some of them have been republished in Russian. Therefore, most translations from the Russian have been made by the author of the present dissertation. The secondary literature has been surveyed in the languages that are familiar (Russian, English and French) or almost familiar (German) to the present author, and which are hopefully the most pertinent to the present investigation. Besides, and in order to increase the acquaintance with the text, which was the objective of all this, some archival sources were used. The analysis consists of careful chronological studies of the authors' writings and their evolution in their historical and intellectual context. As a consequence, the dissertation brings new authors to the foreground - Shaposhnikov and Yurovsky - who were traditionally confined to the substitutes' bench, because they only superficially touched the domains quoted above. In the Russian synthesis however, they played an important part of the story. As a side effect, some authors that used to play in the foreground - Dmitriev and Bortkiewicz - are relegated to the background, but are not forgotten. Besides, the dissertation refreshes the views on authors already known, such as Ziber and, especially, Tugan-Baranovsky. The ultimate objective of this dissertation is to change the opinion that one could have on "value and prices in Russian economic thought", by setting the Russian synthesis at the centre of the debates.

This study describes a form of partial agonism for a CD8+ CTL clone, S15, in which perforin-dependent killing and IFN-gamma production were lost but Fas (APO1 or CD95)-dependent cytotoxicity preserved. Cloned S15 CTL are H-2Kd restricted and specific for a photoreactive derivative of the Plasmodium berghei circumsporozoite peptide PbCS 252-260 (SYIPSAEKI). The presence of a photoactivatable group in the epitope permitted assessment of TCR-ligand binding by TCR photoaffinity labeling. Selective activation of Fas-dependent killing was observed for a peptide-derivative variant containing a modified photoreactive group. A similar functional response was obtained after binding of the wild-type peptide derivative upon blocking of CD8 participation in TCR-ligand binding. The epitope modification or blocking of CD8 resulted in an > or = 8-fold decrease in TCR-ligand binding. In both cases, phosphorylation of zeta-chain and ZAP-70, as well as calcium mobilization were reduced close to background levels, indicating that activation of Fas-dependent cytotoxicity required weaker TCR signaling than activation of perforin-dependent killing or IFN-gamma production. Consistent with this, we observed that depletion of the protein tyrosine kinase p56(lck) by preincubation of S15 CTL with herbimycin A severely impaired perforin- but not Fas-dependent cytotoxicity. Together with the observation that S15 CTL constitutively express Fas ligand, these results indicate that TCR signaling too weak to elicit perforin-dependent cytotoxicity or cytokine production can induce Fas-dependent cytotoxicity, possibly by translocation of preformed Fas ligand to the cell surface.

The interaction of a parasite and a host cell is a complex process, which involves several steps: (1) attachment to the plasma membrane, (2) entry inside the host cell, and (3) hijacking of the metabolism of the host. In biochemical experiments, only an event averaged over the whole cell population can be analyzed. The power of microscopy, however, is to investigate individual events in individual cells. Therefore, parasitologists frequently perform experiments with fluorescence microscopy using different dyes to label structures of the parasite or the host cell. Though the resolution of light microscopy has greatly improved, it is not sufficient to reveal interactions at the ultrastructural level. Furthermore, only specifically labeled structures can be seen and related to each other. Here, we want to demonstrate the additional value of electron microscopy in this area of research. Investigation of the different steps of parasite-host cell interaction by electron microscopy, however, is often hampered by the fact that there are only a few cells infected, and therefore it is difficult to find enough cells to study. A solution is to profit from low magnification, hence large overview, and specific location of the players by fluorescence labels in a light microscope with the high power resolution and structural information provided by an electron microscope, in short by correlative light and electron microscopy.

The purpose of this study was to analyze the composition of 103 dietary supplements bought on the internet. The supplements were dispatched in four different categories according to their announced contents [creatine, prohormones, "mental enhancers" and branched chain amino acids (BCAA)]. All the supplements were screened for the presence of stimulants and main anabolic steroids parent compounds. At the same time, the research was focused on the precursors and metabolites of testosterone and nandrolone. The study pointed out three products containing an anabolic steroid, metandienone, in a very high amount. The ingestion of such products induced a high quantity of metandienone metabolites in urines that would be considered as a positive antidoping test. The results have also shown that one creatine product and three "mental enhancers" contained traces of hormones or prohormones not claimed on the labels and 14 prohormone products contained substances other than those indicated by the manufacturer. The oral intake of the creatine product revealed the presence of the two main nandrolone metabolites (19-norandrosterone and 19-noretiocholanolone) in urine.
