Methods are described for working with Nosema apis and Nosema ceranae in the field and in the laboratory. For fieldwork, different sampling methods are described to determine colony level infections at a given point in time, but also for following the temporal infection dynamics. Suggestions are made for how to standardise field trials for evaluating treatments and disease impact. The laboratory methods described include different means for determining colony level and individual bee infection levels and methods for species determination, including light microscopy, electron microscopy, and molecular methods (PCR). Suggestions are made for how to standardise cage trials, and different inoculation methods for infecting bees are described, including control methods for spore viability. A cell culture system for in vitro rearing of Nosema spp. is described. Finally, how to conduct different types of experiments are described, including infectious dose, dose effects, course of infection and longevity tests

Adult honey bees are maintained in vitro in laboratory cages for a variety of purposes. For example, researchers may wish to perform experiments on honey bees caged individually or in groups to study aspects of parasitology, toxicology, or physiology under highly controlled conditions, or they may cage whole frames to obtain newly emerged workers of known age cohorts. Regardless of purpose, researchers must manage a number of variables, ranging from selection of study subjects (e.g. honey bee subspecies) to experimental environment (e.g. temperature and relative humidity). Although decisions made by researchers may not necessarily jeopardize the scientific rigour of an experiment, they may profoundly affect results, and may make comparisons with similar, but independent, studies difficult. Focusing primarily on workers, we provide recommendations for maintaining adults under in vitro laboratory conditions, whilst acknowledging gaps in our understanding that require further attention. We specifically describe how to properly obtain honey bees, and how to choose appropriate cages, incubator conditions, and food to obtain biologically relevant and comparable experimental results. Additionally, we provide broad recommendations for experimental design and statistical analyses of data that arises from experiments using caged honey bees. The ultimate goal of this, and of all COLOSS BEEBOOK papers, is not to stifle science with restrictions, but rather to provide researchers with the appropriate tools to generate comparable data that will build upon our current understanding of honey bees.

Cells must rapidly sense and respond to a wide variety of potentially cytotoxic external stressors to survive in a constantly changing environment. In a search for novel genes required for stress tolerance in Saccharomyces cerevisiae, we identified the uncharacterized open reading frame YER139C as a gene required for growth at 37 degrees C in the presence of the heat shock mimetic formamide. YER139C encodes the closest yeast homolog of the human RPAP2 protein, recently identified as a novel RNA polymerase II (RNAPII)-associated factor. Multiple lines of evidence support a role for this gene family in transcription, prompting us to rename YER139C RTR1 (regulator of transcription). The core RNAPII subunits RPB5, RPB7, and RPB9 were isolated as potent high-copy-number suppressors of the rtr1Delta temperature-sensitive growth phenotype, and deletion of the nonessential subunits RPB4 and RPB9 hypersensitized cells to RTR1 overexpression. Disruption of RTR1 resulted in mycophenolic acid sensitivity and synthetic genetic interactions with a number of genes involved in multiple phases of transcription. Consistently, rtr1Delta cells are defective in inducible transcription from the GAL1 promoter. Rtr1 constitutively shuttles between the cytoplasm and nucleus, where it physically associates with an active RNAPII transcriptional complex. Taken together, our data reveal a role for members of the RTR1/RPAP2 family as regulators of core RNAPII function.

Postpartum hemorrhage (PPH) is one of the main causes of maternal deaths even in industrialized countries. It represents an emergency situation which necessitates a rapid decision and in particular an exact diagnosis and root cause analysis in order to initiate the correct therapeutic measures in an interdisciplinary cooperation. In addition to established guidelines, the benefits of standardized therapy algorithms have been demonstrated. A therapy algorithm for the obstetric emergency of postpartum hemorrhage in the German language is not yet available. The establishment of an international (Germany, Austria and Switzerland D-A-CH) "treatment algorithm for postpartum hemorrhage" was an interdisciplinary project based on the guidelines of the corresponding specialist societies (anesthesia and intensive care medicine and obstetrics) in the three countries as well as comparable international algorithms for therapy of PPH.The obstetrics and anesthesiology personnel must possess sufficient expertise for emergency situations despite lower case numbers. The rarity of occurrence for individual patients and the life-threatening situation necessitate a structured approach according to predetermined treatment algorithms. This can then be carried out according to the established algorithm. Furthermore, this algorithm presents the opportunity to train for emergency situations in an interdisciplinary team.

The loci of the porcine tumour necrosis factor genes, alpha (TNFA) and beta (TNFB), have been chromosomally assigned by radioactive in situ hybridization. The genomic probes for TNFA and TNFB yielded signals above 7p11-q11, a region that has been shown earlier to carry the porcine major histocompatibility locus (SLA). These mapping data along with preliminary molecular studies suggest a genomic organization of the SLA that is similar to that of human and murine major histocompatibility complexes.

Pododermatitis is frequent in captive flamingos worldwide, but little is known about the associated histopathologic lesions. Involvement of a papillomavirus or herpesvirus has been suspected. Histopathologic evaluation and viral assessment of biopsies from 19 live and 10 dead captive greater flamingos were performed. Selected samples were further examined by transmission electron microscopy and immunohistochemistry. Feet from 10 dead free-ranging greater flamingos were also evaluated. The histologic appearance of lesions of flamingos of increasing age was interpreted as the progression of pododermatitis. Mild histologic lesions were seen in a 3-week-old flamingo chick with no macroscopic lesions, and these were characterized by Micrococcus-like bacteria in the stratum corneum associated with exocytosis of heterophils. The inflammation associated with these bacteria may lead to further histologic changes: irregular columnar proliferations, papillary squirting, and dyskeratosis. In more chronic lesions, hydropic degeneration of keratinocytes, epidermal hyperplasia, and dyskeratosis were seen at the epidermis, as well as proliferation of new blood vessels and increased intercellular matrix in the dermis. Papillomavirus DNA was not identified in any of the samples, while herpesvirus DNA was seen only in a few cases; therefore, these viruses were not thought to be the cause of the lesions. Poor skin health through suboptimal husbandry may weaken the epidermal barrier and predispose the skin to invasion of Micrococcus-like bacteria. Histologic lesions were identified in very young flamingos with no macroscopic lesions; this is likely to be an early stage lesion that may progress to macroscopic lesions.

Einleitung: Sport unterrichtende Lehrpersonen sollten in der Lage sein, Sportunterricht entsprechend seinem Bildungsauftrag (Fries, Baumberger & Egloff, 2009) zu gestalten. Dies verweist auf bestimmte Kompetenzen, welche eine Lehrperson ausweisen muss (vgl. Baumert & Kunter, 2006), um ebendiese Zielsetzungen im Unterricht anzusteuern. Aus wissenschaftlicher Perspektive herrscht jedoch Uneinigkeit darüber, was (Sport unterrichtende) Lehrpersonen wissen und können sollten oder welche Kompetenzen besonders wichtig sind (Bauer, 2002). Dieser Frage wird im Forschungsprojekt KopS (Kompetenzprofile für das Fach Sport auf Vorschul- und Primarschulstufe) nachgegangen. Methode: Im Rahmen einer zweistufigen Delphibefragung wurden in einer ersten Runde ausgewählte Lehrpersonen mit Expertenstatus der Vorschul-, Unter- und Mittelstufe in halbstrukturierten Interviews zu den aus ihrer Sicht notwendigen Fähigkeiten für unterrichtende Lehrpersonen des Faches Sport befragt (N1 = 16). Daraus wurden mittels qualitativer Inhaltsanalyse nach Mayring (2008) stufenspezifische Kompetenzen abgeleitet. In der zweiten Runde wurden die gewonnenen Kompetenzen Fachdidaktikerinnen und Fachdidaktiker verschiedener Pädagogischer Hochschulen der Deutschschweiz (N2 = 10) in einem strukturierten Interview vorgelegt und nach deren Kommentierung und Beurteilung der Kompetenzen bezüglich Wichtigkeit gefragt. Ergebnisse: Die drei schulstufenspezifischen Kompetenzraster aus der ersten Delphi-Runde weisen zwischen 30-62 Kompetenzen auf. Diese sind stufenübergreifend ähnlich, wobei sich die Kompetenzen der Vor-schule tendenziell von jener der Primarschule abgrenzen lassen: Im Bewegungs- und Sportunterricht im Kindergarten wird insbesondere das Arrangieren von motorisch herausfordernden Raum- und Materialerfahrungen als zentral erachtet. In der Schule werden dagegen sportpraktisches und organisatorisches Können von Lehrpersonen als bedeutungsvoll eingeschätzt. Allen drei Kompetenzrastern werden sportunterrichtsspezifische Kompetenzen zu den Bereichen Organisation, Unterrichtsstruktur, sport- und bewegungsbezogenes Fachwissen, Beurteilung, Interaktion, Kommunikation und Sicherheit genannt. Die von den Lehrpersonen beschriebenen Kompetenzen werden von den Fachdidaktikerinnen und Fachdidaktiker als wichtig erachtet und nur marginal ergänzt. Diskussion: Mittels des beschriebenen Verfahrens wurden erstmals Kompetenzen von Sport unterrichtenden Lehrpersonen auf Vorschul- und Primarschulstufe erfasst. Die Ergebnisse zeigen, dass die sportspezifische Tönung der Kompetenzen Unterschiede zu bisherigen Kompetenzdarstellungen für Primarlehrpersonen nahelegt und es erforderlich ist, Kompetenzen stufenspezifisch zu diskutieren. Insects provide crucial ecosystem services for human food security and maintenance of biodiversity. Therefore, major declines in wild insects combined with losses of managed bees have raised great concern. Recent data suggest that honey bees appear to be less susceptible to stressors compared to other species. Here, we argue that eusociality plays a key role for the susceptibility of insects to environmental stressors due to superorganism resilience, which can be defined as the ability to tolerate the loss of somatic cells (= workers) as long as the germ line (= reproduction) is maintained. Life history and colony size appear critical for such resilience. Future conservation efforts should take superorganism resilience into account to safeguard ecosystem services by insects.
