Introducción: La evaluación de tecnologías en salud aplicadas a la selección de un módulo de proteína para uso hospitalario, tiene como finalidad servir de apoyo en la elección de productos costo efectivos y seguros, con el fin de favorecer la toma de decisiones a los diferentes agentes que participan en la elección de alternativas terapéuticas, recomendadas en pacientes con necesidades elevadas de proteínas, como es el caso de la presente investigación. Objetivo: Aplicar un método matemático - multicriterio que permita evaluar los módulos de proteína disponibles en el mercado para la terapia nutricional institucional. Métodos: Se establecieron dos fases, una revisión de la literatura para establecer y priorizar los criterios de evaluación técnica de las diferentes ofertas de módulos de proteína, y dos se realizó una aplicación de un modelo matemático con el fin de considerar el modulo proteico para uso dentro de las instituciones hospitalarias, el cual consistió en la asignación de un valor a cada una de las variables mediante una escala diferencial semántica establecida, que permitieron calcular el peso porcentual de cada una de las variables, cuya sumatoria arrojo la calificación porcentual de cada alternativa. Resultados: Respecto a la búsqueda de criterios de evaluación técnica para las diferentes ofertas de módulos de proteína, en la literatura se identificaron las siguientes variables para evaluación, la naturaleza o equivalencia, condiciones de administración y uso, seguridad, y eficacia. La naturaleza se evaluó mediante la calificación del cómputo químico de aminoácidos corregido por digestibilidad proteica (PDCAAS) con un peso en la evaluación del 39.05%, en referencia a las condiciones de administración y uso se tuvo en cuenta factores incluidos en los sistemas de distribución por dosis unitaria con un peso del 27.61%, la eficacia fue definida por la tasa de eficiencia proteica (PER) la cual impacta el 19.53% de la calificación y finalmente, el criterio de seguridad con un 13.81% referente al empaque y etiquetado. Conclusiones: Al realizar la evaluación de cuatro alternativas de módulos de proteína, ofertadas por las diferentes casas farmacéuticas, la mayor puntuación correspondiente a las alternativas con una calificación superior al 90%, la obtuvieron dos alternativas de módulos de proteína para uso hospitalario, las cuales contienen proteínas del suero (“Whey”) y aminoácidos en combinaciones.

La sang de porc és un subproducte comestible que es genera als escorxadors industrials durant el procés d'obtenció de la canal. Aquest subproducte es caracteritza per presentar una elevada càrrega contaminant i, degut a l'elevat volum que es genera, és necessari trobar estratègies que permetin la seva revaloració i aprofitament, a la vegada que disminuïm la contaminació ambiental i les despeses que es deriven del seu processament abans de l'abocament. La fracció cel·lular (FC) constitueix el 40 % de la sang de porc i conté principalment l'hemoglobina (Hb), que representa al voltant del 90 % del contingut en proteïna d'aquesta fracció (un 35 % aproximadament). L'elevat percentatge en proteïna i en ferro, i les seves bones propietats funcionals fan que l'aprofitament d'aquest subproducte com a primera matèria o ingredient de la indústria alimentària sigui una alternativa molt útil a l'hora de reduir les despeses de la indústria càrnia, sempre que es resolguin els problemes de l'enfosquiment i dels sabors estranys que pot conferir la FC quan s'addiciona a productes alimentaris. Una altra possible utilització de la FC és aprofitar les propietats colorants de l'Hb o del grup hemo, com a colorant d'origen natural en diversos productes alimentaris. Els objectius del present treball eren, en primer lloc, determinar les millors condicions d'aplicació del procés de conservació de la FC mitjançant la deshidratació per atomització i caracteritzar físico-químicament i microbiològica el concentrat d'Hb en pols. En segon lloc, avaluar l'eficàcia de diferents additius antioxidants i/o segrestants del ferro per prevenir l'enfosquiment que pateix la FC durant la deshidratació. En tercer lloc, aplicar tractaments d'altes pressions hidrostàtiques com a procés d'higienització i avaluar els efectes d'aquest tractament sobre la microbiota contaminant, el color i les propietats funcionals de la FC. Finalment, desenvolupar un procés d'obtenció d'hidrolitzats proteics descolorats a partir de l'Hb amb la finalitat d'utilitzar-los com a ingredients nutricionals i/o funcionals. La millor temperatura de deshidratació per atomització de la FC hemolitzada era 140ºC. La FC en pols presentava un contingut en humitat del 5,3 % i un percentatge de solubilitat proteica del 96 %. La deshidratació per atomització induïa canvis en l'estructura nativa de l'Hb i, per tant, un cert grau de desnaturalització que pot conduir a una disminució de les seves propietats funcionals. L'extracte sec de la FC en pols estava composat per un 94,6 % de proteïna, un 3 % de sals minerals i un 0,7 % de greix. Els valors CIE L*a*b* del color de la FC en pols eren força constants i reflectien el color vermell marró fosc d'aquesta, a causa de l'oxidació del ferro hèmic que es produeix durant la deshidratació. La càrrega contaminant de la FC fresca de la sang de porc era força elevada i el tractament d'hemòlisi amb ultrasons i la centrifugació posterior no produïen una reducció significativa de la microbiota contaminant, obtenint un producte amb uns recomptes microbiològics de l'ordre de 106 ufc·mL-1. La deshidratació per atomització produïa una disminució d'una unitat logarítmica dels recomptes totals de la FC hemolitzada. Tanmateix, el producte en pols encara reflectia l'elevada contaminació de la primera matèria, fet que condiciona negativament la seva utilització com a ingredient alimentari, a no ser que es millorin les condicions de recollida de la sang a l'escorxador o que aquesta o la FC es sotmeti a algun tractament d'higienització prèviament a la deshidratació. Les isotermes de sorció a 20ºC de la FC en pols tenien forma sigmoïdal i una histèresi estreta i llarga. L'equació GAB és un bon model matemàtic per ajustar les dades de sorció obtingudes experimentalment i determinar la isoterma d'adsorció de la FC deshidratada per atomització. El percentatge d'humitat de la FC deshidratada a 140ºC es corresponia a un valor d'aw a 20 ºC d'aproximadament el 0,16. Tenint en compte que estava per sota dels valors d'aw corresponents a la capa monomolecular, es pot garantir la conservació a temperatura ambient del producte, sempre que s'envasi en recipients tancats que no permetin l'entrada d'humitat de l'exterior. De l'estudi de la possible estabilització del color de la FC deshidratada per atomització mitjançant l'addició d'antioxidants i/o segrestants de ferro, es va observar que només l'àcid ascòrbic, la glucosa, l'àcid nicotínic i la nicotinamida, tenien efectes positius sobre el color del producte en pols. L'ascòrbic i la glucosa no milloraven la conservació del color de l'Hb però disminuïen l'enfosquiment que es produeix durant la deshidratació, amb la qual cosa es pot obtenir un producte en pols de color marró més clar. L'addició de dextrina o L-cisteïna no disminuïa l'enfosquiment ni evitava el canvi de color de l'Hb. L'àcid nicotínic i la nicotinamida protegien el color de l'Hb durant el procés de deshidratació i l'emmagatzematge de la FC en pols. Les millors condicions d'aplicació del tractament amb altes pressions hidrostàtiques (HHP) sobre la FC eren 400 MPa, a 20ºC, durant 15 minuts, perquè produïen una millora significativa de la qualitat microbiològica, no afectaven negativament al color, no comprometien gaire la solubilitat proteica l'Hb i, malgrat que produïen un augment de la viscositat, la FC romania fluida després del tractament. Aquest tractament permetia una reducció de la microbiota contaminant de la FC d'entre 2 i 3 unitats logarítmiques. L'aplicació de l'alta pressió i la posterior deshidratació per atomització permetien obtenir un producte en pols amb recomptes totals de l'ordre de 2,8 unitats logarítmiques. El color de la FC pressuritzada en pols era igual que el de la FC control deshidratada, perquè ambdues mostres presentaven la mateixa susceptibilitat a l'oxidació del grup hemo produïda per la deshidratació. L'alta pressió incrementava la susceptibilitat de l'Hb als efectes desnaturalitzants de la deshidratació, fonamentalment a pH 7 (PIE), ja que es va observar una disminució de la solubilitat proteica a pH neutre després dels 2 processos tecnològics. La FC en pols presentava una màxima capacitat escumant al PIE de l'Hb. L'aplicació del tractament HHP produïa una disminució de la capacitat escumant de la FC en pols, però no tenia efectes negatius sobre l'estabilitat de l'escuma formada. Tampoc es van observar efectes negatius del tractament HHP sobre l'activitat emulsionant de l'Hb. La màxima activitat emulsionant de l'Hb s'aconseguia amb una concentració de FC en pols de l'1,5 % a pH 7 i de l'1 % a pH 4,5. Les pastes obtingudes per escalfament de la FC presentaven característiques molt diferenciades depenent del pH. A pH neutre es formaven unes pastes dures i consistents, mentre que a pH àcid les pastes eren poc consistents, molt adhesives i més elàstiques que les anteriors. Aquestes tenien una capacitat de retenció d'aigua molt superior que les de pH 7, en les quals l'aigua quedava retinguda per capil·laritat. La textura i capacitat de retenció d'aigua de les pastes tampoc eren afectades pel tractament HHP. El tractament HHP incrementava l'activitat de la Tripsina sobre l'Hb quan el substrat i l'enzim es tractaven conjuntament i afavoria el procés d'obtenció d'hidrolitzats descolorats a partir de la FC, la qual cosa permetia assolir el mateix grau de descoloració amb una dosi d'enzim inferior. El tractament d'hidròlisi de la FC amb la utilització combinada de Tripsina seguida d'un tractament amb Pepsina permetia l'obtenció d'un hidrolitzat proteic d'Hb descolorat i hidrolitzava completament la globina, donant lloc a 2 pèptids de 10,8 i 7,4 KDa. Val a dir que també produïa un 60 80 % de nitrogen soluble en TCA, constituït fonamentalment per pèptids petits i aminoàcids lliures. Els hidrolitzats trípsics i pèpsics d'Hb, obtinguts a partir de FC no pressuritzada i deshidratats per atomització a 180ºC, eren de color blanc i tenien un contingut en humitat del 4,7 %, un 84,2 % de proteïna i 9,7 % de sals minerals. El procés d'hidròlisi permetia una reducció considerable de la contaminació de la FC, obtenint un producte en pols amb uns recomptes totals de l'ordre de 102-103 ufc·g-1. Pel que fa a la funcionalitat dels hidrolitzats d'Hb deshidratats per atomització, aquests presentaven una elevada solubilitat proteica a pH 5 i 7 i romanien solubles després d'un escalfament a 80ºC durant 30 min. Tanmateix, aquesta hidròlisi afectava molt negativament la capacitat de mantenir escumes estables i l'activitat emulsionant.

To establish its significance during commercial breadmaking, dityrosine formation was quantified in flours and doughs of six commercial wheat types at various stages of the Chorleywood Bread Process. Dityrosine was formed mainly during mixing and baking, at the levels of nmol/g dry weight. Good breadmaking flours tended to exhibit higher dityrosine content in the final bread than low quality ones, but no relationship was found for dityrosine as a proportion of flour protein content, indicating that the latter was still a dominant factor in the analysis. There was no correlation between gluten yield of the six wheat types and their typical dityrosine concentrations, suggesting that dityrosine crosslinks were not a determinant factor for gluten formation. Ascorbic acid was found to inhibit dityrosine formation during mixing and proving, and have no significant effect on dityrosine in the final bread. Hydrogen peroxide promoted dityrosine formation, which suggests a radical mechanism involving endogenous peroxidases might be the responsible for dityrosine formation during breadmaking.

Grass-based diets are of increasing social-economic importance in dairy cattle farming, but their low supply of glucogenic nutrients may limit the production of milk. Current evaluation systems that assess the energy supply and requirements are based on metabolisable energy (ME) or net energy (NE). These systems do not consider the characteristics of the energy delivering nutrients. In contrast, mechanistic models take into account the site of digestion, the type of nutrient absorbed and the type of nutrient required for production of milk constituents, and may therefore give a better prediction of supply and requirement of nutrients. The objective of the present study is to compare the ability of three energy evaluation systems, viz. the Dutch NE system, the agricultural and food research council (AFRC) ME system, and the feed into milk (FIM) ME system, and of a mechanistic model based on Dijkstra et al. [Simulation of digestion in cattle fed sugar cane: prediction of nutrient supply for milk production with locally available supplements. J. Agric. Sci., Cambridge 127, 247-60] and Mills et al. [A mechanistic model of whole-tract digestion and methanogenesis in the lactating dairy cow: model development, evaluation and application. J. Anim. Sci. 79, 1584-97] to predict the feed value of grass-based diets for milk production. The dataset for evaluation consists of 41 treatments of grass-based diets (at least 0.75 g ryegrass/g diet on DM basis). For each model, the predicted energy or nutrient supply, based on observed intake, was compared with predicted requirement based on observed performance. Assessment of the error of energy or nutrient supply relative to requirement is made by calculation of mean square prediction error (MSPE) and by concordance correlation coefficient (CCC). All energy evaluation systems predicted energy requirement to be lower (6-11%) than energy supply. The root MSPE (expressed as a proportion of the supply) was lowest for the mechanistic model (0.061), followed by the Dutch NE system (0.082), FIM ME system (0.097) and AFRCME system(0.118). For the energy evaluation systems, the error due to overall bias of prediction dominated the MSPE, whereas for the mechanistic model, proportionally 0.76 of MSPE was due to random variation. CCC analysis confirmed the higher accuracy and precision of the mechanistic model compared with energy evaluation systems. The error of prediction was positively related to grass protein content for the Dutch NE system, and was also positively related to grass DMI level for all models. In conclusion, current energy evaluation systems overestimate energy supply relative to energy requirement on grass-based diets for dairy cattle. The mechanistic model predicted glucogenic nutrients to limit performance of dairy cattle on grass-based diets, and proved to be more accurate and precise than the energy systems. Field experiments were carried out to assess the effects of nitrogen fertilization and seed rate on the Hagberg falling number (HFN) of commercial wheat hybrids and their parents. Applying nitrogen (200 kg N ha(-1)) increased HFN in two successive years. The HFN of the hybrid Hyno Esta was lower than either of its parents (Estica and Audace), particularly when nitrogen was not applied. Treatment effects on HFN were negatively associated with a-amylase activity. Phadebas grain blotting suggested two populations of grains with different types of a-amylase activity: Estica appeared to have a high proportion of grains with low levels of late maturity endosperm a-amylase activity (LMEA); Audace had a few grains showing high levels of germination amylase; and the hybrid, Hyno Esta, combined the sources from both parents to show heterosis for a-amylase activity. Applying nitrogen reduced both apparent LMEA and germination amylase. The effects on LMEA were associated with the size and disruption of the grain cavity, which was greater in Hyno Esta and Estica and in zero-nitrogen treatments. External grain morphology failed to explain much of the variation in LMEA and cavity size, but there was a close negative correlation between cavity size and protein content. Applying nitrogen increased post-harvest dormancy of the grain. Dormancy was greatest in Estica and least in Audace. It is proposed that effects of seed rate, genotype and nitrogen fertilizer on HFN are mediated through factors affecting the size and disruption of the grain cavity and therefore LMEA, and through factors affecting dormancy and therefore germination amylase. The chemical composition and fractional distribution of protein isolates prepared from species of Mucuna bean were studied. Using six different extraction media, the yield of protein based on the Kjeldahl procedure varied from 8% to 34%, and the protein content varied from 75% to 95%. When the yields were high, the colour of the isolates generally tended to be dark and unsatisfactory. Hence, the use of chemical treatments and high pressure processing were explored. The solubility maxima for the protein isolates in water were found to occur at pH values of 2.0 and 11.0, while the pH corresponding to minimum solubility (i.e. isoelectric region) occurred at pH values of 4.0 and 5.0. The total essential amino acid in the isolates ranged from 495 to 557 mg g(-1) protein, which compares favourably with the recommended level for pre-school and school children. Methionine and cysteine were the limiting amino acids. A key nutritional attribute of the protein isolates was its high lysine content. The isolate can therefore complement cereal-based foods which are deficient in lysine. The proteins mainly consisted of albumins, glutelins and globulins. Prolamins were only present in trace concentration (< 0.3%). Gel filtration chromatograms of the isolates indicated the presence of major protein fractions with molecular weights of 40 and 15 kDa, while gel electrophoresis (SDS-PAGE) indicated a major broad zone with molecular weights of 36 +/- 7 and 17.3 +/- 13 kDa. A study was conducted to investigate the effects of wheat straw ammonisation and supplementation with a rumen undegradable protein (UDP) source on nutrient digestion and nitrogen balance by lambs while diets were supplemented with kibbled carob pods as energy source. Ammonisation increased the crude protein content of wheat straw by nearly 100% and decreased the contents of neutral detergent fibre and acid detergent fibre by 7% and 1.7% respectively. Treating the straw with ammonia resulted in significant (P<0.01) increase in nitrogen (N) intake and intakes of organic matter (OM) and dry matter (DM) tended toward significance (P<0.1). The UDP source had no effect (P>0.05) on DM and OM intakes but resulted in an increase (P<0.05) of N intakes. Both, ammonization and UDP supplementation increased (P<0.01) the DM, OM and N digestibility. In conclusion, the results of this study suggest that ammonisation and UDP supplementation is a practical dietary manipulation option to improve the nutritional status of ruminants fed on roughage-based diets.

Background: Seed storage proteins are a major source of dietary protein, and the content of such proteins determines both the quantity and quality of crop yield. Significantly, examination of the protein content in the seeds of crop plants shows a distinct difference between monocots and dicots. Thus, it is expected that there are different evolutionary patterns in the genes underlying protein synthesis in the seeds of these two groups of plants. Results: Gene duplication, evolutionary rate and positive selection of a major gene family of seed storage proteins (the 11S globulin genes), were compared in dicots and monocots. The results, obtained from five species in each group, show more gene duplications, a higher evolutionary rate and positive selections of this gene family in dicots, which are rich in 11S globulins, but not in the monocots. Conclusion: Our findings provide evidence to support the suggestion that gene duplication and an accelerated evolutionary rate may be associated with higher protein synthesis in dicots as compared to monocots.

Wheat gluten proteins, gliadins and glutenins, are of great importance in determining the unique biomechanical properties of wheat. Studies have therefore been carried out to determine their pathways and mechanisms of synthesis, folding, and deposition in protein bodies. In the present work, a set of transgenic wheat lines has been studied with strongly suppressed levels of γ-gliadins and/or all groups of gliadins, using light and fluorescence microscopy combined with immunodetection using specific antibodies for γ-gliadins and HMW glutenin subunits. These lines represent a unique material to study the formation and fusion of protein bodies in developing seeds of wheat. Higher amounts of HMW subunits were present in most of the transgenic lines but only the lines with suppression of all gliadins showed differences in the formation and fusion of the protein bodies. Large rounded protein bodies were found in the wild-type lines and the transgenic lines with reduced levels of γ-gliadins, while the lines with all gliadins down-regulated had protein bodies of irregular shape and irregular formation. The size and number of inclusions, which have been reported to contain triticins, were also higher in the protein bodies in the lines with all the gliadins down-regulated. Changes in the protein composition and PB morphology reported in the transgenic lines with all gliadins down-regulated did not result in marked changes in the total protein content or instability of the different fractions.

High protein dairy beverages are considered to be mouth drying. The drying sensation may be due to the product protein content; however the mechanism of this mouth drying is uncertain. This study investigated the potential adhesion of milk proteins to porcine oral mucosal tissues and their resistance to wash out with simulated saliva was monitored using fluorescence microscopy. Cadein was found to be more adhesive to porcine mucosa then lactogloubulin. Some investigation into the reason for this difference in mucoadhesion was conducted by thiol-content analysis, rheology and zeta-potential measurements. The higher viscosity of casein solution and smaller zeta-potential is believed to be responsible for its better retention on mucosal surfaces. These findings suggest that casein and whey protein are both capable of binding and eliciting mouth drying in high protein dairy beverages.

Wheat Distillers’ Dried Grains with Solubles (DDGS) and in-process samples were used for protein extraction. Prolamins were the predominant protein components in the samples. The absence of extractable α- and γ-gliadins in DDGS indicated protein aggregation during the drum drying processing stage. Prolamin extraction was performed using 70% (v/v) ethanol or alkaline-ethanol solution in the presence of reducing agent. DDGS extracts had relatively low protein contents (14-44.9%, w/w), regardless of the condition applied. The wet solids were the most suitable raw material for protein extraction, with recovery yields of ~ 55% (w/w) and protein content of ~58% (w/w) in 70% (v/v) ethanol. Protein extracts from wet solids were significantly rich in glutamic acid and proline. Mass balance calculations demonstrated the high carbohydrate content (~ 50%, w/w) of solid residues. Overall, the feasibility of utilising in-process samples of DDGS for protein extraction with commercial potential was demonstrated.

The expression of protein kinase C (PKC) isoforms (PKC-alpha, PKC-beta 1, PKC-delta, PKC-epsilon, and PKC-zeta) was studied by immunoblotting in whole ventricles of rat hearts during postnatal development (1-26 days) and in the adult. PKC-alpha, PKC-beta 1, PKC-delta, PKC-epsilon, and PKC-zeta were detected in ventricles of 1-day-old rats, although PKC-alpha and PKC-beta 1 were only barely detectable. All isoforms were rapidly downregulated during development, with abundances relative to total protein declining in the adult to < 25% of 1-day-old values. PKC-beta 1 was not detectable in adult ventricles. The specific activity of PKC was also downregulated. The rat ventricular myocyte becomes amitotic soon after birth but continues to grow, increasing its protein content 40- to 50-fold between the neonate and the 300-g adult. An important question is thus whether the amount of PKC per myocyte is downregulated. With the use of isolated cells, immunoblotting showed that the contents per myocyte of PKC-alpha and PKC-epsilon increased approximately 10-fold between the neonatal and adult stages. In rat ventricles, the rank of association with the particulate fraction was PKC-delta > PKC-epsilon > PKC-zeta. Association of these isoforms with the particulate fraction was less in the adult than in the neonate. In primary cultures of ventricular myocytes prepared from neonatal rat hearts, 1 microM 12-O-tetradecanoylphorbol-13-acetate (TPA) elicited translocation of PKC-alpha, PKC-delta, and PKC-epsilon from the soluble to the particulate fraction in < 1 min, after which time no further translocation was observed. Prolonged exposure (16 h) of myocytes to 1 microM TPA caused essentially complete downregulation of these isoforms, although downregulation of PKC-epsilon was slower than for PKC-delta. In contrast, PKC-zeta was neither translocated nor downregulated by 1 microM TPA. Immunoblotting of human ventricular samples also revealed downregulation of PKC relative to total protein during fetal/postnatal development.

First published August 5, 2009; doi:10.1152/ajprenal.90256.2008.-Sirolimus, an antiproliferative immunosuppressant, induces hypomagnesemia and hypokalemia. Rosiglitazone activates renal sodiumand water-reabsorptive pathways. We evaluated whether sirolimus induces renal wasting of magnesium and potassium, attempting to identify the tubule segments in which this occurs. We tested the hypothesis that reduced expression of the cotransporter NKCC2 forms the molecular basis of this effect and evaluated the possible association between increased urinary excretion of magnesium and renal expression of the epithelial Mg(2+) channel TRPM6. We then analyzed whether rosiglitazone attenuates these sirolimus-induced tubular effects. Wistar rats were treated for 14 days with sirolimus (3 mg/kg body wt in drinking water), with or without rosiglitazone (92 mg/kg body wt in food). Protein abundance of NKCC2, aquaporin2 (AQP2), and TRPM6 was assessed using immunoblotting. Sirolimus-treated animals presented no change in glomerular filtration rate, although there were marked decreases in plasma potassium and magnesium. Sirolimus treatment reduced expression of NKCC2, and this was accompanied by greater urinary excretion of sodium, potassium, and magnesium. In sirolimus-treated animals, AQP2 expression was reduced. Expression of TRPM6 was increased, which might represent a direct stimulatory effect of sirolimus or a compensatory response. The finding that rosiglitazone prevented or attenuated all sirolimus-induced renal tubular defects has potential clinical implications.

This paper describes a proteome analysis and changes in endogenous abscisic acid (ABA) contents during seed development of Araucaria angustifolia (Bert.) O. Ktze. Megagametophytes and embryonic axis tissues exhibited a similar ABA variation pattern during seed development, reaching maximum values at the pre-cotyledonary stage. The embryonic axis protein content increased until the cotyledonary stage with following stabilization at mature seed. The two-dimensional electrophoresis at the torpedo developmental stage showed approximately 230 polypeptides against 340 in the mature stage. Peptide mass fingerprinting analyses identified three polypeptides, corresponding to an AtSAC4, a late embryogenesis abundant (LEA) and a storage protein, respectively.

Hypnea musciformis (Wulfen in Jacqu.) J.V. Lamour. is the main source for carrageenan production in Brazil and strains with selected characteristics could improve the production of raw material. The effects of kinetin on growth rates, morphology, protein content, and concentrations of pigments (chlorophyll a, phycoerythrin, phycocyanin, and allophycocyanin) were assessed in the wild strain (brown phenotype) and in the phycoerythrin-deficient strain (green phenotype) of H. musciformis. Concentrations of kinetin ranging from 0 to 50 mu M were tested in ASP 12-NTA synthetic medium with 10 mu M nitrate (N-limited) and 100 mu M nitrate (N-saturated). In N-limited condition, kinetin stimulated growth rates of the phycoerythrin-deficient strain and formation of lateral branches in both colour strains. Kinetin stimulated protein biosynthesis in both strains. However, differences between both nitrogen conditions were significant only in the phycoerythrin-deficient strain. In the wild strain, effects of kinetin on concentrations of phycobiliproteins were not significant in both nitrogen conditions, except for chlorophyll content. However, the phycoerythrin-deficient strain showed an opposite response, and kinetin stimulated the phycobiliprotein biosynthesis, with the highest concentrations of phycoerythrin in N-saturated medium, while the highest concentrations of allophycocyanin and phycocyanin were observed in N-limited medium. These results indicate that the effects of kinetin on growth, morphology, protein and phycobiliprotein contents are influenced by nitrogen availability, and the main nitrogen storage pools in phycoerythrin-deficient strain of H. musciformis submitted to N-limited conditions were phycocyanin and allophycocianin, the biosynthesis of which was enhanced by kinetin.
