Current methods to characterize mesenchymal stem cells (MSCs) are limited to CD marker expression, plastic adherence and their ability to differentiate into adipogenic, osteogenic and chondrogenic precursors. It seems evident that stem cells undergoing differentiation should differ in many aspects, such as morphology and possibly also behaviour; however, such a correlation has not yet been exploited for fate prediction of MSCs. Primary human MSCs from bone marrow were expanded and pelleted to form high-density cultures and were then randomly divided into four groups to differentiate into adipogenic, osteogenic chondrogenic and myogenic progenitor cells. The cells were expanded as heterogeneous and tracked with time-lapse microscopy to record cell shape, using phase-contrast microscopy. The cells were segmented using a custom-made image-processing pipeline. Seven morphological features were extracted for each of the segmented cells. Statistical analysis was performed on the seven-dimensional feature vectors, using a tree-like classification method. Differentiation of cells was monitored with key marker genes and histology. Cells in differentiation media were expressing the key genes for each of the three pathways after 21 days, i.e. adipogenic, osteogenic and chondrogenic, which was also confirmed by histological staining. Time-lapse microscopy data were obtained and contained new evidence that two cell shape features, eccentricity and filopodia (= 'fingers') are highly informative to classify myogenic differentiation from all others. However, no robust classifiers could be identified for the other cell differentiation paths. The results suggest that non-invasive automated time-lapse microscopy could potentially be used to predict the stem cell fate of hMSCs for clinical application, based on morphology for earlier time-points. The classification is challenged by cell density, proliferation and possible unknown donor-specific factors, which affect the performance of morphology-based approaches. This article presents a feasibility study with the objective of investigating the potential of multi-detector computed tomography (MDCT) to estimate the bone age and sex of deceased persons. To obtain virtual skeletons, the bodies of 22 deceased persons with known age at death were scanned by MDCT using a special protocol that consisted of high-resolution imaging of the skull, shoulder girdle (including the upper half of the humeri), the symphysis pubis and the upper halves of the femora. Bone and soft-tissue reconstructions were performed in two and three dimensions. The resulting data were investigated by three anthropologists with different professional experience. Sex was determined by investigating three-dimensional models of the skull and pelvis. As a basic orientation for the age estimation, the complex method according to Nemeskéri and co-workers was applied. The final estimation was effected using additional parameters like the state of dentition, degeneration of the spine, etc., which where chosen individually by the three observers according to their experience. The results of the study show that the estimation of sex and age is possible by the use of MDCT. Virtual skeletons present an ideal collection for anthropological studies, because they are obtained in a non-invasive way and can be investigated ad infinitum.

Having to carry input devices can be inconvenient when interacting with wall-sized, high-resolution tiled displays. Such displays are typically driven by a cluster of computers. Running existing games on a cluster is non-trivial, and the performance attained using software solutions like Chromium is not good enough. This paper presents a touch-free, multi-user, humancomputer interface for wall-sized displays that enables completely device-free interaction. The interface is built using 16 cameras and a cluster of computers, and is integrated with the games Quake 3 Arena (Q3A) and Homeworld. The two games were parallelized using two different approaches in order to run on a 7x4 tile, 21 megapixel display wall with good performance. The touch-free interface enables interaction with a latency of 116 ms, where 81 ms are due to the camera hardware. The rendering performance of the games is compared to their sequential counterparts running on the display wall using Chromium. Parallel Q3A’s framerate is an order of magnitude higher compared to using Chromium. The parallel version of Homeworld performed on par with the sequential, which did not run at all using Chromium. Informal use of the touch-free interface indicates that it works better for controlling Q3A than Homeworld.

Amyotrophic lateral sclerosis (ALS) is a progressive motor neuron disease, fatal within 1 to 5 years after onset of symptoms. About 3 out of 100’000 persons are diagnosed with ALS and there is still no cure available [1, 2]. 95% of all cases occur sporadically and the aetiology remains largely unknown [XXXX]. However, up to now 16 genes were identified to play a role in the development of familial ALS. One of these genes is FUS that encodes for the protein fused in sarcoma/translocated in liposarcoma (FUS/TLS). Mutations in this gene are responsible for some cases of sporadic as well as of inherited ALS [3]. FUS belongs to the family of heterogeneous nuclear ribonucleoproteins and is predicted to be involved in several cellular functions like transcription regulation [4], RNA splicing [5, 6], mRNA transport in neurons [7] and microRNA processing [8]. Aberrant accumulation of mutated FUS has been found in the cytoplasm of motor neurons from ALS patients [9]. The mislocalization of FUS is based on a mutation in the nuclear localization signal of FUS [10]. However, it is still unclear if the cytoplasmic localization of FUS leads to a toxic gain of cytoplasmic function and/or a loss of nuclear function that might be crucial in the course of ALS. The goal of this project is to characterize the impact of ALS-associated FUS mutations on in vitro differentiated motor neurons. To this end, we edit the genome of induced pluripotent stem cells (iPSC) using transcription activator-like effector nucleases (TALENs) [11,12] to create three isogenic cell lines, each carrying an ALS-associated FUS mutation (G156E, R244C and P525L). These iPSC’s will then be differentiated to motor neurons according to a recently establishe protocol (Ref Wichterle) and serve to study alterations in the transcriptome, proteome and metabolome upon the expression of ALS-associated FUS. With this approach, we hope to unravel the molecular mechanism leading to FUS-associated ALS and to provide new insight into the emerging connection between misregulation of RNA metabolism and neurodegeneration, a connection that is currently implied in a variety of additional neurological diseases, including spinocerebellar ataxia 2 (SCA-2), spinal muscular atrophy (SMA), fragile X syndrome, and myotonic dystrophy.

Changes of porosity, permeability, and tortuosity due to physical and geochemical processes are of vital importance for a variety of hydrogeological systems, including passive treatment facilities for contaminated groundwater, engineered barrier systems (EBS), and host rocks for high-level nuclear waste (HLW) repositories. Due to the nonlinear nature and chemical complexity of the problem, in most cases, it is impossible to verify reactive transport codes analytically, and code intercomparisons are the most suitable method to assess code capabilities and model performance. This paper summarizes model intercomparisons for six hypothetical scenarios with generally increasing geochemical or physical complexity using the reactive transport codes CrunchFlow, HP1, MIN3P, PFlotran, and TOUGHREACT. Benchmark problems include the enhancement of porosity and permeability through mineral dissolution, as well as near complete clogging due to localized mineral precipitation, leading to reduction of permeability and tortuosity. Processes considered in the benchmark simulations are advective-dispersive transport in saturated media, kinetically controlled mineral dissolution-precipitation, and aqueous complexation. Porosity changes are induced by mineral dissolution-precipitation reactions, and the Carman-Kozeny relationship is used to describe changes in permeability as a function of porosity. Archie’s law is used to update the tortuosity and the pore diffusion coefficient as a function of porosity. Results demonstrate that, generally, good agreement is reached amongst the computer models despite significant differences in model formulations. Some differences are observed, in particular for the more complex scenarios involving clogging; however, these differences do not affect the interpretation of system behavior and evolution.

The concentrations of the long-lived nuclear reaction products 129I and 36Cl have been measured in samples from the MEGAPIE liquid metal spallation target. Samples from the bulk target material (lead-bismuth eutectic, LBE), from the interface of the metal free surface with the cover gas, from LBE/steel interfaces and from noble metal absorber foils installed in the cover gas system were analysed using Accelerator Mass Spectrometry at the Laboratory of Ion beam Physics at ETH Zürich. The major part of 129I and 36Cl was found accumulated on the interfaces, particularly at the interface of LBE and the steel walls of the target container, while bulk LBE samples contain only a minor fraction of these nuclides. Both nuclides were also detected on the absorber foils to a certain extent (≪ 1% of the total amount). The latter number is negligible concerning the radio-hazard of the irradiated target material; however it indicates a certain affinity of the absorber foils for halogens, thus proving the principle of using noble metal foils for catching these volatile radionuclides. The total amounts of 129I and 36Cl in the target were estimated from the analytical data by averaging within the different groups of samples and summing up these averages over the total target. This estimation could account for about half of the amount of 129I and 36Cl predicted to be produced using nuclear physics modelling codes for both nuclides. The significance of the results and the associated uncertainties are discussed.

AnewRelativisticScreenedHydrogenicModel has been developed to calculate atomic data needed to compute the optical and thermodynamic properties of high energy density plasmas. The model is based on anewset of universal screeningconstants, including nlj-splitting that has been obtained by fitting to a large database of ionization potentials and excitation energies. This database was built with energies compiled from the National Institute of Standards and Technology (NIST) database of experimental atomic energy levels, and energies calculated with the Flexible Atomic Code (FAC). The screeningconstants have been computed up to the 5p3/2 subshell using a Genetic Algorithm technique with an objective function designed to minimize both the relative error and the maximum error. To select the best set of screeningconstants some additional physical criteria has been applied, which are based on the reproduction of the filling order of the shells and on obtaining the best ground state configuration. A statistical error analysis has been performed to test the model, which indicated that approximately 88% of the data lie within a ±10% error interval. We validate the model by comparing the results with ionization energies, transition energies, and wave functions computed using sophisticated self-consistent codes and experimental data.

The bridge over the Mittellandkanal is located in the municipality of Braunschweig, a city of Low Saxony located in Mid-North Germany, between Berlin and Hannover, 50 km away from this last city. The city of Braunschwig has 248.867 inhabitants (December 2010). The orography of the zone is practically even, with some slopes in the surrounding area, with a no important variation of the ground elevation, characterized by a big uniformity and a great visual quality. In this zone the canal flows from west to east, from the Dortmundkanal until the Elbe river, so it has a length of 325,3 km. The normal flora from the zone is farmland with some little forests not very big and not very important. But the Mittellandkanal lies at the North of Brauschweig, that means that in the zone of the canal there is a change between the urban area and the rural area. For this reason in the zone of the bridge we start to find farmlands and forest, however the constructions and buildings are still present in this area. The railway line that is going to be built will unite the North-center of Germany with the North-East of Germany because some of the following factors: •Overstress of the only existing line in the moment that connects Berlin with the North-East extreme of Germany. •No direct connection with the North-Easth Germany with other important German cities from South and West of Germany. •Replacement of the old existing one way line in the area that has to be renewed. •Modernization of the old railway lines of Germany. Schedule order by the European Union and the German Government. The actual lines of the zone do not have the necessary conditions and characteristics to satisfy the demand of the travelers that want to travel all over Germany with the railway avoiding using other transports like plan or car. For these reasons the necessity of the replacement of the old railway line and the aim to create a union with the North-East Germany that has a deficit of transport infrastructures and connections with the rest of the country and Europe. Although the new railway line provokes some disadvantages when constructing it is indispensable to build this railway line. The pass of the railway line through Braunschweig has not been random. The zone Braunschweig-Wolfsburg is a very industrialized area because of some companies like Wolkswagen and too because this zone is an important pass of important line from Berlin, South and East Germany. For all of these reasons the railway line goes through Braunschweig and connects to the city of Wolfsburg first, and after the North-East Germany zone.

El proyecto realizado, trata de una aplicación desarrollada en la plataforma Android, orientada a personas con algún tipo de discapacidad sensorial o psíquica. Dicha aplicación fomenta el uso y la integración de dispositivos móviles en este tipo de sector de población. Está pensada, para que, un usuario con discapacidad, pueda interactuar con ella y de forma transparente a él, se ha creado un sistema mediante el cual, se registra todo el comportamiento que ese usuario ha tenido durante el tiempo de uso de la aplicación, con el fin de llevar un seguimiento del mismo; evaluar si existen cambios en él; determinar si son necesarios algunos cambios en la aplicación que favorezcan una mejoría en cuanto al uso y consecución de resultados en el paciente, etc. Se ha combinado el uso de una aplicación instalada sobre un sistema operativo de libre distribución, concretamente Android, con un juguete de código abierto, como es Sphero. Eso ha permito el desarrollo de una aplicación perfectamente ajustada a los requisitos funcionales definidos, con una robustez y eficiencia similar a una aplicación de sistemas operativos móviles cerrados. Es importarte remarcar, que la primera finalidad de este proyecto es ofrecer la posibilidad de usar un juguete, como Sphero, que está orientado a un sector de población sin discapacidad, haciendo uso de cualquier dispositivo móvil, a personas con diferentes grados de discapacidad sensorial, motora y psíquica. Siempre clarificando, que no existe la posibilidad de usar esta aplicación para cualquier tipo y grado de discapacidad, ya que, ello supondría, un proyecto de una envergadura enorme. El hecho de usar un dispositivo móvil, es un derecho, que todos tenemos. Y por ello, se espera que tras la lectura y comprensión de este proyecto, se motive a los lectores a seguir desarrollando aplicaciones para que cualquier usuario, con discapacidad o no, tenga las mismas oportunidades de interactuación con dispositivos móviles. The carried out project, is an application developed on the Android platform , aimed at people with some kind of sensory or mental disability . This application encourages the use and integration of mobile devices in this type of population sector. It is designed to be interacted by a disable person and transparently to that user, it has been created a system by which all behavior, that user has had during the time of use of the application, is recorded, that’s to keep track of it ; assess whether there are changes in it, determine if changes are needed in the application that favor an improvement in the use and achieving patient outcomes , etc. . It has combined the use of an application installed on an open source operating system, namely Android, an open source toy , as is Sphero . That has made it possible to develop an application perfectly adjusted to the functional requirements defined with a robustness and efficiency similar to a mobile OS application closed. It is important to note, that the first aim of this project is to offer the possibility of using a toy, like Sphero , which is geared to a sector of the population without disabilities , using any mobile device , for people with different degrees of sensory impairment , motor and mental . Always clarifying that there is no possibility to use this application for any type and degree of disability, because the magnitude of the project would have been infinitely greater. The fact of using a mobile device, is a right we all have. And so, it is expected that upon reading and understanding of this project, motivate readers to continue developing applications for any user , disabled or not, have the same opportunities for interaction with mobile devices .

Applications based on Wireless Sensor Networks for Internet of Things scenarios are on the rise. The multiple possibilities they offer have spread towards previously hard to imagine fields, like e-health or human physiological monitoring. An application has been developed for its usage in scenarios where data collection is applied to smart spaces, aiming at its usage in fire fighting and sports. This application has been tested in a gymnasium with real, non-simulated nodes and devices. A Graphic User Interface has been implemented to suggest a series of exercises to improve a sportsman/woman s condition, depending on the context and their profile. This system can be adapted to a wide variety of e-health applications with minimum changes, and the user will interact using different devices, like smart phones, smart watches and/or tablets.
