AIM: This study investigated the ability of an osteoconductive biphasic scaffold to simultaneously regenerate alveolar bone, periodontal ligament and cementum. MATERIALS AND METHODS: A biphasic scaffold was built by attaching a fused deposition modelled bone compartment to a melt electrospun periodontal compartment. The bone compartment was coated with a calcium phosphate (CaP) layer for increasing osteoconductivity, seeded with osteoblasts and cultured in vitro for 6 weeks. The resulting constructs were then complemented with the placement of PDL cell sheets on the periodontal compartment, attached to a dentin block and subcutaneously implanted into athymic rats for 8 weeks. Scanning electron microscopy, X-ray diffraction, alkaline phosphatase and DNA content quantification, confocal laser microscopy, micro computerized tomography and histological analysis were employed to evaluate the scaffold's performance. RESULTS: The in vitro study showed that alkaline phosphatase activity was significantly increased in the CaP-coated samples and they also displayed enhanced mineralization. In the in vivo study, significantly more bone formation was observed in the coated scaffolds. Histological analysis revealed that the large pore size of the periodontal compartment permitted vascularization of the cell sheets, and periodontal attachment was achieved at the dentin interface. CONCLUSIONS: This work demonstrates that the combination of cell sheet technology together with an osteoconductive biphasic scaffold could be utilized to address the limitations of current periodontal regeneration techniques.

Melt electrospinning and its additive manufacturing analogue, melt electrospinning writing (MEW), are two processes which can produce porous materials for applications where solvent toxicity and accumulation in solution electrospinning are problematic. This study explores the melt electrospinning of poly(ε-caprolactone) (PCL) scaffolds, specifically for applications in tissue engineering. The research described here aims to inform researchers interested in melt electrospinning about technical aspects of the process. This includes rapid fiber characterization using glass microscope slides, allowing influential processing parameters on fiber morphology to be assessed, as well as observed fiber collection phenomena on different collector substrates. The distribution and alignment of melt electrospun PCL fibers can be controlled to a certain degree using patterned collectors to create large numbers of scaffolds with shaped macroporous architectures. However, the buildup of residual charge in the collected fibers limits the achievable thickness of the porous template through such scaffolds. One challenge identified for MEW is the ability to control charge buildup so that fibers can be placed accurately in close proximity, and in many centimeter heights. The scale and size of scaffolds produced using MEW, however, indicate that this emerging process will fill a technological niche in biofabrication.

Topographical cues can be exploited to regulate stem cell attachment, proliferation, differentiation and function in vitro and in vivo. In this study, we aimed to investigate the influence of different nanofibrous topographies on the chondrogenic differentiation potential of nasal septum derived progenitors (NSP) in vitro. Aligned and randomly oriented Ploy (L-lactide) (PLLA)/Polycaprolactone (PCL) hybrid scaffolds were fabricated via electrospinning. First, scaffolds were fully characterized, and then NSP were seeded on them to study their capacity to support stem cell attachment, proliferation and chondrogenic differentiation. Compared to randomly oriented nanofibers, aligned scaffolds showed a high degree of nanofiber alignment with much better tensile strength properties. Both scaffolds supported NSP adhesion, proliferation and chondrogenic differentiation. Despite the higher rate of cell proliferation on random scaffolds, a better chondrogenic differentiation was observed on aligned nanofibers as deduced from higher expression of chondrogenic markers such as collagen type II and aggrecan on aligned scaffolds. These findings demonstrate that electrospun constructs maintain NSP proliferation and differentiation, and that the aligned nanofibrous scaffolds can significantly enhance chondrogenic differentiation of nasal septum derived progenitors

in this contribution we present a soft matter solid electrolyte which was obtained by inclusion of a polymer (polyacrylonitrile, PAN) in LiClO4/LiTFSI-succinonitrile (SN), a semi-solid organic plastic electrolyte. Addition of the polymer resulted in considerable enhancement in ionic conductivity as well as mechanical strength of LiX-SN (X=ClO4, TFSI) plastic electrolyte. Ionic conductivity of 92.5%-[1 M LiClO4-SN]:7.5%-PAN (PAN amount as per SN weight) composite at 25 degrees C recorded a remarkably high value of 7 x 10(-3) Omega(-1) cm(-1), higher by few tens of order in magnitude compared to 1 M LiClO4-SN. Composite conductivity at sub-ambient temperature is also quite high. At -20 degrees C, the ionic conductivity of (100 -x)%-[1 M LiClO4-SN]:x%-PAN composites are in the range 3 x 10(-5)-4.5 x 10(-4) Omega(-1) cm(-1), approximately one to two orders of magnitude higher with respect to 1 M LiClO4-SN electrolyte conductivity. Addition of PAN resulted in an increase of the Young's modulus (Y) from Y -> 0 for LiClO4-SN to a maximum of 0.4MPa for the composites. Microstructural studies based on X-ray diffraction, differential scanning calorimetry and Fourier transform infrared spectroscopy suggest that enhancement in composite ionic conductivity is a combined effect of decrease in crystallinity and enhanced trans conformer concentration. Plasma polymerization was used to coat a melt electrospun polycaprolactone scaffold to improve cell attachment and organization. Plasma polymerization was performed using an amine containing monomer, allylamine, which then allowed for the subsequent immobilization of biomolecules i.e. heparin and fibroblast growth factor-2. The stability of the plasma polymerized amine-coating was demonstrated by X-ray photoelectron spectroscopy analysis and imaging time-of-flight secondary ion mass spectrometry revealed that a uniform plasma amine-coating was deposited throughout the scaffold. Based upon comparison with controls it was evident that the combination scaffold aided cell ingress and the formation of distinct fibroblast and keratinocyte layers.

Ion transport in a recently demonstrated promising soft matter solid plastic-polymer electrolyte is discussed here in the context of solvent dynamics and ion association. The plastic-polymer composite electrolytes display liquid-like ionic conductivity in the solid state,compliable mechanical strength (similar to 1 MPa), and wide electrochemical voltage stability (>= 5 V). Polyacrylonitrile (PAN) dispersed in lithium perchlorate (LiClO4)-succinonitrile (SN) was chosen as the model system for the study (abbreviated LiClO4-SN:PAN). Systematic observation of various mid-infrared isomer and ion association bands as a function of temperature and polyme concentration shows an effective increase in trans conformer concentration along with free Li+ ion concentration. This strongly supports the view that enhancement in LiClO4-SN:PAN ionic conductivity over the neat plastic electrolyte (LiClO4-SN) is due to both increase in charge mobility and concentration. The ionic conductivity and infrared spectroscopy studies are supported by Brillouin light scattering. For the LiClO4-SN:PAN composites, a peak at 17 GHz was observed in addition to the normal trans-gauche isomerism (as in neat SN) at 12 GHz. The fast process is attributed to increased dynamics of those SN molecules whose energy barrier of transition from gauche to trans has reduced under influences induced by the changes in temperature and polymer concentration. The observations from ionic conductivity, spectroscopy, and light scattering studies were further supplemented by temperature dependent nuclear magnetic resonance H-1 and Li-7 line width measurements.

Cryogel matrices composed of different polymeric blends were synthesized, yielding a unique combination of hydrophilicity and hydrophobicity with the presence or absence of charged surface. Four such cryogel matrices composed of polyacrylamide-chitosan (PAAC), poly(N-isopropylacrylamide)-chitosan, polyacrylonitrile (PAN), and poly(N-isopropylacrylamide) were tested for growth of different hybridoma cell lines and production of antibody in static culture. All the matrices were capable for the adherence of hybridoma cell lines 6A4D7, B7B10, and H9E10 to the polymeric surfaces as well as for the efficient monoclonal antibody (mAb) production. PAAC proved to be relatively better in terms of both mAb production and cell growth. Further, PAAC cryogel was designed into three different formats, monolith, disks, and beads, and used as packing material for packed-bed bioreactor. Longterm cultivation of 6A4D7 cell line on PAAC cryogel scaffold in all the three formats could be successfully done for a period of 6 weeks under static conditions. Continuous packed-bed bioreactor was setup using 6A4D7 hybridoma cell line in the three reactor formats. The reactors ran continuously for a period of 60 days during which mAb production and metabolism of cells in the bioreactors were monitored periodically. The monolith bioreactor performed most efficiently over a period of 60 days and produced a total of 57.5 mg of antibody in the first 30 days (in 500 mL) with a highest concentration of 115 mu g mL(-1), which is fourfold higher than t-flask culture. The results demonstrate that appropriate chemistry and geometry of the bioreactor matrix for cell growth and immobilization can enhance the reactor productivity. In this study, fibers of barbed wire structure were obtained by electrospinning blend of organic conducting crystalline material and polyethylene oxide. Thermal and structural characterization of the blend fibers has been carried out to study the fiber characteristics. An increase in crystallinity in the electrospun fibers was observed and was attributed to both electrospinning process as well as addition of organic conducting crystalline material. The development of scaffolds for neural tissue engineering application requires an understanding of cell adhesion, proliferation, and migration of neuronal cells. Considering the potential application of carbon as scaffold materials and the lack of understanding of compatibility of amorphous carbon with neuronal cells, the carbon-based materials in the forms of carbon films and continuous electrospun carbon nanofibers having average diameter of approximate to 200 nm are being investigated with or without ultraviolet (UV) and oxy-plasma (OP) treatments for cytocompatibility property using mouse Neuroblastoma (N2a) and rat Schwann cells (RT4-D6P2T). The use of Raman spectroscopy in combination with Fourier transform infrared (FTIR) and X-ray diffraction establishes the amorphous nature and surface-bonding characteristics of the studied carbon materials. Although both UV and OP treatments make carbon surfaces more hydrophilic, the cell viability of N2a cells is statistically more significant on OP treated fibers/films compared to UV fiber/film substrates after 4 days in culture. The electrospun carbon fibrous substrate provides the physical guidance to the cultured Schwann cells. Overall, the experimental results of this study demonstrate that the electrospun amorphous carbon nanofibrous scaffolds can be used as a suitable biomaterial substrate for supporting cell adhesion and proliferation of neuronal cells in the context of their applications as artificial nerve implants. The growth of neuroblastoma (N2a) and Schwann cells has been explored on polymer derived carbon substrates of varying micro and nanoscale geometries: resorcinol-formaldehyde (RE) gel derived carbon films and electrospun nanofibrous (similar to 200 nm diameter) mat and SU-8 (a negative photoresist) derived carbon micro-patterns. MTT assay and complementary lactate dehydrogenase (LDH) assay established cytocompatibility of RE derived carbon films and fibers over a period of 6 days in culture. The role of length scale of surface patterns in eliciting lineage-specific adaptive response along, across and on the interspacing between adjacent micropatterns (i.e., ``on'', ``across'' and ``off'') has been assayed. Textural features were found to affect 3',5'-cyclic AMP sodium salt-induced neurite outgrowth, over a wide range of length scales: from similar to 200 nm (carbon fibers) to similar to 60 mu m (carbon patterns). Despite their innate randomness, carbon nanofibers promoted preferential differentiation of N2a cells into neuronal lineage, similar to ordered micro-patterns. Our results, for the first time, conclusively demonstrate the potential of RE-gel and SU-8 derived carbon substrates as nerve tissue engineering platforms for guided proliferation and differentiation of neural cells in vitro. In the present work, electrospraying of an organic molecule is carried out using various solvents, obtaining fibril structures along with a range of distinct morphologies. Solvent characteristics play a major role in determining the morphology of the organic material. A thiophene derivative (7,9-di(thiophen-2-yl)-8H-cyclopentaa]acenaphthylen-8-one) (DTCPA) of donor-acceptor-donor (DAD) architecture is used to study this solvent effect. Seven solvents with decreasing vapour pressure are selected for experiments. Electrospraying is conducted at a solution concentration of 1.5 wt% and a constant applied voltage of 15 kV. Gradual transformation in morphology of the electrospun product from spiked-spheres to only spikes is observed. A mechanism describing this transformation is proposed based on electron micrograph analysis and XRD analysis. These data indicate that the morphological change is due to the synergistic effect of both vapour pressure and dielectric constant of the solvents. Through a reasonable control of the crystallite size and morphology along with the proposal of the transformation mechanism, this study elucidates electrospraying as a prospective method for designing architectures in organic electronics.

Ag nanoparticle embedded NaYF4:0.05Tb center dot chi Ce/ PVP (PVP stands for poly(vinyl pyrrolidone)) composite nanofibers have been prepared by electrospinning. A field emission scanning electron microscope and x-ray diffraction have been utilized to characterize the size, morphology and structure of the as-prepared electrospun nanofibers. Obvious photoluminescence (PL) of NaYF4:0.05Tb center dot 0.05Ce/PVP electrospun nanofibers due to the efficient energy transfer from Ce3+ to Tb3+ ions is observed. The PL intensity of the electrospun nanofibers decreases gradually with the addition of Ag nanoparticles. No obvious surface plasmon resonance enhanced luminescence is observed. The reasons for the weakening of the emission intensity with the addition of Ag nanoparticles have also been discussed in this work.

Nos últimos anos, a irradiação por micro-ondas tem sido cada vez mais usada na síntese de varias moléculas orgânicas, devido a uma série de vantagens que essa nova tecnologia apresenta. Até o momento, existem poucos estudos sobre polimerizações assistidas por micro-ondas e menos ainda sobre a modificação de polímeros usando essa nova fonte de energia. Polímeros heterocíclicos contendo anéis azóicos, como o tetrazol possuem interesse acadêmico e comercial devido as suas várias aplicações. Nesta Dissertação foi estuda a modificação química da poliacrilonitrila pela incorporação em sua cadeia polimérica de 10% de grupos tetrazol empregando-se a irradiação de micro-ondas e o método convencional (térmico). Foram utilizados um forno de micro-ondas doméstico e um reator de micro-ondas monomodo nas reações assistidas por micro-ondas. As reações em reator foram realizadas em vaso aberto, em solução (DMF) ou em massa, sendo variado o tempo e a temperatura reacional, bem como a potência do equipamento. Os copolímeros produzidos foram caracterizados por espectroscopia na região do infravermelho (FTIR) e os teores de grupos tetrazol incorporados na PAN foram determinados por titulometria de neutralização. A poliacrilonitrila foi modificada quimicamente com sucesso tanto no forno doméstico quanto no reator de micro-ondas, como foi verificado pela sua análise de FTIR. Pela determinação do grau de incorporação de grupos tetrazol nos polímeros concluiu-se que, para a reação em solução, nas temperaturas mais elevas, bem como em uma potência maior, o teor de heterocíclico incorporado no polímero está muito próximo do valor obtido pelo método convencional. Já as reações conduzidas em massa apresentaram graus de incorporação inferiores as das feitas em solução.

Polímeros contendo anéis heterocíclicos, como o imidazol, possuem um grande interesse comercial e científico devido ao seu emprego em diversas áreas. Esses polímeros podem ser obtidos pela modificação química de polímeros comerciais como a poliacrilonitrila. Geralmente, esse tipo de reação é realizada por aquecimento convencional (térmico). Entretanto, mais recentemente, a irradiação por micro-ondas também começou a ser utilizada como fonte de energia para esse fim. Nesta Dissertação foi estudada, comparativamente, a modificação química da poliacrilonitrila comercial com etilenodiamina sob ativação de P2S5, empregando como fonte de aquecimento: o método convencional (térmico) e irradiação de micro-ondas, geradas em forno de micro-ondas doméstico e em reator de micro-ondas (Discover CEM), para produzir o copolímero de poliacrilonitrila e 2-vinil-imidazol. As reações foram efetuadas em diferentes tempos reacionais, em diferentes temperaturas no reator de micro-ondas e em diferentes potências do forno doméstico. Os copolímeros produzidos foram caracterizados por espectroscopia na região do infravermelho (FTIR) e por ressonância magnética de núcleo de hidrogênio (NMR-1H). A poliacrilonitrila foi modificada quimicamente com sucesso em todos os métodos empregados, como foi verificado pela sua análise de FTIR. Os melhores resultados obtidos foram: em 48 h de aquecimento térmico, 20 min de irradiação de micro-ondas em forno doméstico e 20 min a 80C em reator de micro-ondas, no entanto todas as análises indicaram uma derivatização abaixo de 10 %

Processos corrosivos geram perdas incalculáveis para a sociedade. Existe, portanto, uma incessante busca por métodos cada vez mais econômicos, eficientes e que não gerem impacto ao meio-ambiente. Polímeros heterocíclicos são macromoléculas de elevada performance que vêm sendo estudados como inibidores de corrosão por atenderem as premissas citadas acima. Neste trabalho realizou-se a modificação química da poliacrilonitrila com a incorporação de grupos 2-vinil-2-oxazolina no teor teórico de 20%. O copolímero obtido e a poliacrilonitrila não modificada foram caracterizados por espectroscopia na região do infravermelho e avaliados como inibidores de corrosão para alumínio e aço carbono em meio salino em temperatura de 60C. As concentrações utilizadas foram de 20, 50, 100 e 200 ppm e os períodos de checagem: 1, 2, 3 e 4 semanas. Os polímeros estudados apresentaram atividade inibitória de corrosão apenas para o aço carbono. Para o alumínio, o ensaio em branco apresentou a menor taxa de corrosão dentre todos os ensaios realizados
