Em lavouras de feijoeiro (Phaseolus vulgaris) da cultivar Carioca Comum, no município de Londrina, Estado do Paraná, foram encontradas plantas com sintomas de necrose da haste, mosaico clorótico leve e porte reduzido, semelhantes aos sintomas causados por infecção viral. Exames de microscopia eletrônica revelaram a presença de partículas isométricas. em testes de imunodifusão dupla em gel de ágar os extratos foliares de plantas infetadas reagiram positivamente com anti-soro específico para o Southern bean mosaic virus (SBMV). O vírus foi purificado e a massa molecular de sua proteína capsidial foi estimada em 30 kDa, valor esperado para proteínas do capsídeo de vírus do gênero Sobemovirus. A gama de hospedeiras do SBMV isolado no Paraná foi restrita ao feijoeiro e a algumas cultivares de soja (Glycine max). A separação de dois vírus isométricos comuns em infecções mistas no feijoeiro foi possível através da reação de imunidade ao SBMV apresentada por Crotalaria sp, Chenopodium quinoa e Mucuna deeringiana, e da reação de susceptibilidade dessas mesmas hospedeiras ao Bean rugose mosaic virus (BRMV).

O objetivo deste trabalho foi determinar a herdabilidade e o ganho na seleção para teor de fibra alimentar (solúvel, insolúvel e total) e para rendimento de grãos, em populações resultantes de cinco diferentes cruzamentos, além de identificar a população mais adequada, considerando-se as diferentes frações da fibra. Os cruzamentos entre os genitores CNFP 8100, FT 96-1282, Varre-Sai e Valente foram realizados em casa de vegetação. As populações obtidas (genitores, F1 e F2) foram avaliadas em campo, durante a primavera/verão de 2003/2004, em delineamento de blocos ao acaso. Diferenças significativas foram obtidas para fibra alimentar total; não foi detectada variabilidade genética para as frações da fibra - solúvel e insolúvel - e para rendimento de grãos nas populações. Correlação fenotípica positiva foi encontrada entre fibra insolúvel e fibra alimentar total. A seleção de plantas F2, em populações segregantes desenvolvidas a partir da combinação Valente x Varre-Sai, pode ser efetiva no desenvolvimento de germoplasma de feijão com maior teor de fibra alimentar total, em virtude da alta herdabilidade e maior ganho por seleção obtidos.

Egg parasitism of Trichogramma pretiosum strain RV when presented with eggs of Anticarsia gemmatalis and Pseudoplusia includens was investigated at 18, 20, 22, 25, 28, 30 and 32 degrees C. The number of eggs parasitized per day decreased for both hosts as a function of the age of parasitoids, reaching 80% of lifetime parasitism more rapidly as temperature increased; on the 4th day at 32 degrees C and on the 12th day at 18 degrees C. The lifetime number of parasitized P. includens eggs achieved by the parasitoid maintained at 20 degrees C (44.95 +/- 3.94) differed from the results recorded at 32 degrees C (28.5 +/- 1.33). Differently, the lifetime number of A. gemmatalis parasitized eggs did not differ among the temperatures. When T. pretiosum reached 100% of lifetime parasitism, each adult female had parasitized from 28.5 +/- 1.33 to 44.95 +/- 3.94 and from 29.58 +/- 2.80 to 45.36 +/- 4.50 P. includens and A. gemmatalis eggs, respectively. Also, the longevity of these adult T. pretiosum females, for which P. includens or A. gemmatalis eggs were offered, was inversely correlated with temperature. Not only were the survival curves of those adult T. pretiosum females of type I when they were presented with eggs of A. gemmatalis but also with eggs of P. includens, i.e., there was an increase in the mortality rate with time as the temperature increased. In conclusion, T. pretiosum strain RV parasitism was impacted by temperature when on both host eggs; however, the parasitoid still exhibited high survival and, more importantly, high number of parasitized A. gemmatalis and P. includens eggs even at the extremes tested temperatures of 18 and 32 degrees C. Those results indicate that T. pretiosum strain RV might be well adapted to this studied temperature range and, thus, be potentially suitable for use in biological control programs of P. includens and A. gemmatalis in different geographical areas that fits in this range. It is important to emphasize the results here presented are from laboratory studies and, therefore, field trials still need to be carried out in the future with this strain in order to support the full development of the technology intend to use this egg parasitoid in soybean fields worldwide. O objetivo do trabalho foi avaliar a pegada hídrica das principais cultivares de soja como indicador de eficiência de uso da água em cultivos de grãos no município de Paragominas. Foi realizado trabalho de campo para acompanhar experimentos com soja visando obtenção de dados durante a safra agrícola 2013/2014. Considerou-se nas estimativas de pegada hídrica cinza o valor de 22 kg ha-1 de nitrogênio, com base na literatura nacional. As cultivares de soja BRS Candeia; BRS Sambaíba; BRS Tracajá; BRS Seridó RCH; BRS Babaçu foram consideradas nas avaliações da pegada hídrica de acordo com o ciclo médio, disponível na literatura. Considerou-se os padrões limites de lançamento de fertilizante nitrogenado em corpos hídricos, adotando-se valores conforme a legislação nº 357/2005 do Conselho Nacional de Meio Ambiente (CONAMA). Os resultados apontaram que a maior pegada hídrica foi na BRS Babaçu correspondendo a 1.306 m³ ton-1 e a menor foi na BRS Candeia com 1.015 m³ ton-1. Conclui-se que a pegada hídrica verde é um excelente indicador de eficiência de uso da água pela cultivar de soja utilizada em cada ano safra. Também, a quantidade de fertilizantes nitrogenados e o rendimento das cultivares são fatores determinantes na contabilidade de uso eficiente de água em polos grãos na Amazônia.

The expansion of cultivated areas with genetically modified crops (GM) is a worldwide phenomenon, stimulating regulatory authorities to implement strict procedures to monitor and verify the presence of GM varieties in agricultural crops. With the constant growing of plant cultivating areas all over the world, consumption of aflatoxin-contaminated food also increased. Aflatoxins correspond to a class of highly toxic contaminants found in agricultural products that can have harmful effects on human and animal health. Therefore, the safety and quality evaluation of agricultural products are important issues for consumers. Lateral flow tests (strip tests) is a promising method for the detection both proteins expressed in GM crops and aflatoxins-contaminated food samples. The advantages of this technique include its simplicity, rapidity and cost-effective when compared to the conventional methods. In this study, two novel and sensitive strip tests assay were developed for the identification of: (i) Cry1Ac and Cry8Ka5 proteins expressed in GM cotton crops and; (ii) aflatoxins from agricultural products. The first strip test was developed using a sandwhich format, while the second one was developed using a competitive format. Gold colloidal nanoparticles were used as detector reagent when coated with monoclonal antibodies. An anti-species specific antibody was sprayed at the nitrocellulose membrane to be used as a control line. To validate the first strip test, GM (Bollgard I® e Planta 50- EMBRAPA) and non-GM cotton leaf (Cooker 312) were used. The results showed that the strip containing antibodies for the identification of Cry1Ac and Cry8Ka5 proteins was capable of correctly distinguishing between GM samples (positive result) and non-GM samples (negative result), in a high sensitivity manner. To validate the second strip test, artificially contaminated soybean with Aspergillus flavus (aflatoxin-producing fungus) was employed. Food samples, such as milk and soybean, were also evaluated for the presence of aflatoxins. The strip test was capable to distinguish between samples with and without aflatoxins samples, at a sensitivity concentration of 0,5 μg/Kg. Therefore, these results suggest that the strip tests developed in this study can be a potential tool as a rapid and cost-effective method for detection of insect resistant GM crops expressing Cry1Ac and Cry8Ka5 and aflatoxins from food samples.
