Parmi les différents problèmes récents animant les débats en éthique animale, celui de la prédation pose plusieurs défis. En effet, selon certaines théories, nous devrions intervenir dans la nature pour empêcher la prédation entre les espèces, parfois pour protéger le droit des proies (Regan) ou encore pour des raisons politiques (Donaldson et Kymlicka). Toutefois, personne ne semble vouloir accepter une telle implication puisque, comme le dirait Palmer, nous avons une forte intuition de « laisser-aller » en ce qui concerne la prédation. Dans cet article, je soutiendrai que le problème de la prédation n’en est pas vraiment un pour les approches conséquentialistes. Comme je le démontrerai au terme d’une révision des articles de Everett et Milburn, les approches conséquentialistes n’autorisent une telle intervention que sous des conditions extrêmement restreintes.

Das Beurteilen von Schülerleistungen, das Erfassen ihrer unterschiedlichen Lernvoraussetzungen und das Beobachten und Beurteilen von Schülerlernprozessen stellen wichtige Handlungsroutinen im Alltag von Lehrkräften dar. Dem gegenüber steht die Tatsache, dass sich noch bis vor einigen Jahren nur wenige Arbeiten und Projekte explizit mit der Diagnosekompetenz von angehenden und praktizierenden Lehrkräften beschäftigt haben. Die vorliegende Arbeit, die ein Teilprojekt innerhalb eines größeren interdisziplinären Projektes zur Diagnosekompetenz von Lehramtsstudierenden darstellt, möchte einen Beitrag leisten zur Debatte um die diagnostische Kompetenz. Ihr Hauptaugenmerk richtet sie dabei auf die diagnostische Kompetenz von Biologie-Lehramtsstudierenden im Bereich der naturwissenschaftlichen Erkenntnisgewinnung. Im Zuge der notwenigen theoretischen Konzeptionierung präsentiert die Arbeit einen im Gesamtprojekt erarbeiteten Vorschlag für eine Modellierung der (fachbezogenen) Diagnosekompetenz von Lehramtsstudierenden. Ergänzend dazu findet im Rahmen der Arbeit eine Klärung der in der Forschungsliteratur oftmals uneinheitlich benutzten Begrifflichkeiten zur diagnostischen Kompetenz statt. Weiterhin wird ein Vergleich von verschiedenen in der Literatur verwendeten Konzeptualisierungsansätzen vorgenommen. Da in der Forschungsliteratur keine geeigneten Instrumente zum Erfassen der hier im Speziellen betrachteten diagnostischen Kompetenz vorlagen, mussten diese im Rahmen der Arbeit neu entwickelt werden. Ein wesentlicher Ansatz dabei war eine Unterscheidung von status- und prozessdiagnostischen Kompetenzen und die Entwicklung entsprechend geeigneter Erhebungsinstrumente. Die neu entwickelten Testinstrumente wurden bei zwei Studierenden-Jahrgängen an verschiedenen Zeitpunkten im Verlauf ihres Studiums eingesetzt. Im Test zur Messung ihrer statusdiagnostischen Kompetenzen hatten die Studierenden Schülerlösungen aus dem Bereich der naturwissenschaftlichen Erkenntnisgewinnung zu analysieren und zu beurteilen. Dabei zeigte sich als ein zentraler Befund, dass die Probanden das Leistungsniveau der Schülerlösungen umso schlechter korrekt beurteilen konnten, je höher es war. Weiterhin wurde deutlich, dass die diagnostische Kompetenz der Lehramtsstudierenden für die typischen Teilschritte im Rahmen der Erkenntnisgewinnung unterschiedlich hoch ausgeprägt ist. So fiel es ihnen am leichtesten, die Schülerlösungen zum Teilschritt „Datenanalyse“ zu beurteilen, wohingegen sie deutlich größere Schwierigkeiten hatten, das Leistungsniveau der Schülerlösungen zum Teilschritt „Planen einer Untersuchung“ korrekt einzuschätzen. Im Test zum Erfassen der prozessdiagnostischen Fähigkeiten der Studierenden wurde ihnen ein Experimentierprozess zweier Schüler vorgelegt. Auffällig in Bezug auf ihre diagnostische Kompetenz waren die massiven Schwierigkeiten beim Beurteilen des Schülerumgangs mit den Variablen. Im Vergleich der status- und prozessdiagnostischen Fähigkeiten erwies sich die Prozessdiagnostik als die für die Studierenden schwierigere Tätigkeit. Ein wesentliches Plus in Bezug auf das Gewinnen von Informationen über die diagnostische Kompetenz brachte der Einsatz von Videoanalysen zusätzlich zu den beschriebenen paper-pencil basierten Testinstrumenten. Über eine Analyse der Testbearbeitungsprozesse der Studierenden wurde untersucht, aus welchen Beweggründen und auf Basis welcher Annahmen diese zu ihren diagnostischen Urteilen gekommen waren. Mit Hilfe der auf diese Weise gewonnenen prozessbezogenen Informationen war es möglich, die spezifischen Schwierigkeiten der Studierenden beim Diagnostizieren genauer herauszuarbeiten. Neben den genannten Untersuchungen zu den Ausprägungen von diagnostischer Kompetenz im Bereich Erkenntnisgewinnung wurden im Rahmen der Arbeit auch ihre postulierten Bedingungsfaktoren untersucht: das fachmethodische Vorwissen der Studierenden und ihr Wissenschaftsverständnis. Nur für ersteres konnte ein wenngleich geringer, aber doch signifikanter Zusammenhang festgestellt werden. Ergänzend zum Blick auf die möglichen Bedingungsfaktoren wurden auch Zusammenhänge zwischen der diagnostischen Kompetenz und verschiedenen ausgewählten Personen- und Kontextvariablen untersucht (z.B. die von den Studierenden gewählte Lehramtsform, ihre Fächerkombination, ihr Geschlecht und ihre Abiturnote). Von allen untersuchten Variablen erwies sich die von den Studierenden gewählte Lehramtsform als derjenige Faktor, der den wenngleich ebenfalls nur schwachen, aber doch durchgängig vorhandensten Zusammenhang aufwies. Zusätzlich zu der (objektiven) Erfassung der diagnostischen Kompetenzen mit Hilfe der Messinstrumente präsentiert die Arbeit Ergebnisse zum Verlauf der subjektiven Selbsteinschätzungen der Studierenden hinsichtlich ihres diagnostischen Wissens. Es zeigte sich, dass die Studierenden das Studium für die Ausbildung ihrer diagnostischen Kompetenz als weniger hilfreich empfinden als für den Aufbau der anderen Bereiche ihres Professionswissens. Die Arbeit endet mit einem Fazit zu den Testinstrument-Entwicklungen und den in den verschiedenen Untersuchungen gewonnenen Befunden, verbunden mit einem Ausblick auf mögliche Implikationen für die Lehrerausbildung und sich anschließende Forschungsvorhaben.

Cette intervention théâtralisée permet ultimement de dénoncer les faux semblants et le désordre qui règnent autour de l’empereur. Elle a une dimension purgative, visant la restauration de l’ordre moral, politique et sexuel, tout en promouvant une démarche heuristique placée au cœur du processus de lecture et d’interprétation.

Oxidative stress is considered to be of major relevance for a variety of pathological processes. Thus, it is valuable to identify compounds, which might act as antioxidants, i.e. compounds that antagonize the deleterious action of reactive oxygen species (ROS) on biomolecules. The mode of action of these compounds could be either to scavenge ROS directly or to trigger protective mechanisms inside the cell, thereby resulting in improved defense against ROS. Sulforaphane (SF) (1-isothiocyanato-(4R)-(methylsulfinyl)butane) is a naturally occurring cancer chemopreventive agent found as a precursor glucosinolate in Cruciferous vegetables like broccoli. Although SF is not a direct-acting antioxidant, there is substantial evidence that SF acts indirectly to increase the antioxidant capacity of animal cells and their abilities to cope with oxidative stress. Induction of phase 2 enzymes is one means by which SF enhances the cellular antioxidant capacity. Enzymes induced by SF include Glutathione S-transferases (GST) and NAD[P]H:quinone oxidoreductase (NQO1) which can function as protectors against oxidative stress. To protect themselves from oxidative stress, cells are equipped with reducing buffer systems including the GSH and thioredoxin (Trx) reductase. GSH is an important tripeptide thiol which in addition to being the substrate for GSTs maintains the cellular oxidation– reduction balance and protects cells against free radical species. Aim of the first part of this thesis was to investigate the ability of SF to induce the expression and the activity of different phase 2 and antioxidant enzymes (such as GST, GR, GPx, NQO1, TR, SOD, CAT) in an in vitro model of rat cardiomyocytes, and also to define if SF treatment supprts cells in counteracting oxidative stress induced by H2O2 It is well known that acute exhaustive exercise causes significant reactive oxygen species generation that results in oxidative stress, which can induce negative effects on health and well being. In fact, increased oxidative stress and biomarkers (e.g., protein carbonyls, MDA, and 8- hydroxyguanosine) as well as muscle damage biomarkers (e.g. plasmatic Creatine cinase and Lactate dehydrogenase) have been observed after supramaximal sprint exercises, exhaustive longdistance cycling or running as well as resistance-type exercises, both in trained and untrained humans. Markers of oxidative stress also increase in rodents following exhaustive exercise. Moreover, antioxidant enzyme activities and expressions of antioxidant enzymes are known to increase in response to exhaustive exercise in both animal and human tissues. Aim of this project was to evaluate the effect of SF supplementation in counteracting oxidative stress induced by physical activity through its ability to induce phase 2, and antioxidant enzymes in rat muscle. The results show that SF is a nutraceutical compound able to induce the activity of different phase 2 and antioxidant enzymes in both cardiac muscle and skeletal muscle. Thanks to its actions SF is becoming a promising molecule able to prevent cardiovascular damages induced by oxidative stress and muscle damages induced by acute exhaustive exercise.

La definizione di “benessere animale” e le modalità di determinazione di tale parametro sono ancora ampiamente dibattute. C’è, però, una generale concordanza sul fatto che una condizione di malessere dia origine a variazioni fisiologiche e comportamentali che possono essere rilevate e misurate. Tra i parametri endocrini, il più studiato è, senza dubbio, il cortisolo, in quanto connesso con l’attivazione dell’asse ipotalamico-pituitario-surrenale in condizioni di stress e quindi ritenuto indicatore ideale di benessere, benché debba essere utilizzato con cautela in quanto un aumento dei livelli di questo ormone non si verifica con ogni tipo di stressor. Inoltre, si deve considerare che la raccolta del campione per effettuare le analisi, spesso implica il confinamento ed il contenimento degli animali e può essere, quindi, essa stessa un fattore stressante andando ad alterare i risultati. Alla luce delle suddette conoscenze gli obiettivi scientifici di questa ricerca, condotta sul gatto e sul cane, sono stati innanzitutto validare il metodo di dosaggio di cortisolo dal pelo e stabilire se tale dosaggio può rappresentare un indicatore, non invasivo, di benessere dell’animale (indice di “stress cronico”). In seguito, abbiamo voluto individuare i fattori di stress psico-sociale in gatti che vivono in gattile, in condizioni di alta densità, analizzando i correlati comportamentali ed ormonali dello stress e del benessere in questa condizione socio-ecologica, ricercando, in particolare, l’evidenza ormonale di uno stato di stress prolungato e la messa in atto di strategie comportamentali di contenimento dello stesso e il ruolo della marcatura visivo-feromonale, inoltre abbiamo effettuato un confronto tra oasi feline di diversa estensione spaziale per valutare come varia lo stress in rapporto allo spazio disponibile. Invece, nel cane abbiamo voluto evidenziare eventuali differenze dei livelli ormonali tra cani di proprietà e cani di canili, tra cani ospitati in diversi canili e tra cani che vivono in diverse realtà familiari; abbiamo voluto valutare gli effetti di alcuni arricchimenti sui cani di canile ed, infine, abbiamo analizzato cani sottoposti a specifici programmi si addestramento. Il primo importante ed originale risultato raggiunto, che risponde al primo obiettivo della ricerca, è stato la validazione del dosaggio radioimmunologico di cortisolo in campioni di pelo. Questo risultato, a nostro avviso, apre una nuova finestra sul campo della diagnostica endocrinologica metabolica. Attualmente, infatti, il monitoraggio ormonale viene effettuato su campioni ematici la cui raccolta prevede un elevato stress (stress da prelievo) per l’animale data l'invasività dell'operazione che modifica l’attività di ipotalamo-ipofisi-surrene e, dunque, provoca repentine alterazioni delle concentrazioni ormonali. Questa metodica offre, quindi, il vantaggio dell’estrema semplicità di raccolta del campione e, in più, il bassissimo costo del materiale utilizzato. Dalle ricerche condotte sui gatti di gattile sono scaturite preziose indicazioni per future indagini sullo stress e sul comportamento sociale felino. I risultati dell’analisi congiunta del comportamento e delle concentrazioni ormonali hanno evidenziato che la disponibilità di postazioni di marcatura visivo-feromonale ha un effetto positivo sia sugli indicatori comportamentali, sia su quelli ormonali di stress. I risultati dell’analisi delle concentrazioni di cortisolo, derivanti dal confronto tra sette oasi feline di diversa estensione spaziale hanno permesso di evidenziare un aumento dei livelli dell’ormone inversamente proporzionale allo spazio disponibile. Lo spazio disponibile, però, non è l’unico fattore da prendere in considerazione al fine di assicurare il benessere dell’animale infatti, nelle colonie che presentavano instabilità sociale e variabilità territoriale il cortisolo aveva valori elevati nonostante le notevoli disponibilità di spazio. Infine, si è potuto costatare come anche lo stare appartati, aumenti proporzionalmente con l’aumentare dello spazio. Questo comportamento risulta essere molto importante in quanto mitiga lo stress ed è da prendere in considerazione nell’allestimento di colonie feline. Infatti, nelle colonie di dimensioni ridotte dove lo stress è già alto, l’impossibilità dei soggetti di appartarsi può contribuire a peggiorare la situazione; ecco perché si dovrebbero creare luoghi artificiali per fornire ai gatti la possibilità di appartarsi, magari sfruttando gli spazi sopraelevati (tetti, alberi, ecc.). Per quanto riguarda il confronto tra cani di proprietà e cani di canile non sono state evidenziate differenze significative nei livelli di cortisolo nel pelo mentre abbiamo rilevato che quest’ultimi sono influenzati dalla disponibilità di spazio: infatti sia i cani di proprietà che vivevano in giardino, sia i cani dei canili che praticavano lo sgambamento presentavano livelli di cortisolo nel pelo più bassi rispetto, rispettivamente, ai cani di proprietà che vivevano in appartamento o appartamento/giardino e a quelli di canile che non praticavano lo sgambamento. L’arricchimento ambientale fornito ai cani di canile ha esercitato un’influenza positiva riducendo i livelli di cortisolo e migliorando la docilità dei soggetti, favorendone un’eventuale adozione. Si è inoltre messo in luce che i programmi di addestramento, eseguiti con tecniche “gentili”, non comportano situazioni stressanti per l’animale e aiutano i cani ad esprimere doti di equilibrio che rimarrebbero altrimenti celate dagli aspetti più istintivi del carattere. D’altra parte, l’impegno agonistico prima di una competizione e il livello di addestramento raggiunto dai cani, influenzano le concentrazioni di cortisolo a riposo e durante l’esercizio fisico. Questi risultati possono sicuramente dare utili suggerimenti per la gestione e la cura di gatti e cani al fine di migliorarne le condizioni di benessere.

Background: Several lines of evidence showed that inflammation is associated with changes in the expression of tachykinins both in human and animal models. Tachykinins, including substance P (SP), are small peptides expressed in the extrinsic primary afferent nerve fibres and enteric neurons of the gut: they exert their action through three distinct receptors, termed NK1, NK2 and NK3. SP modulates intestinal motility and enteric secretion, acting preferentially through the NK1 receptor. SP neural network and NK1 receptor expression are increased in patients with inflammatory bowel disease, and similar changes were observed in experimental models of inflammation. The 2,4 Dinitrobenzene Sulphonic Acid (DNBS) model of colitis is useful to study innate immunity, non-specific inflammation and wound healing; it has been suggested that the transmural inflammation seen in this model resembles that found in Crohns disease and can therefore be used to study what cells and mediators are involved in this type of inflammation. Aim: To test the possible protective effect of the NK1 receptor antagonist SSR140333 on: 1) acute model of intestinal inflammation; 2) reactivation of DNBS-induced colitis in rats. Methods: Acute colitis was induced in male SD rats by intrarectal administration of DNBS (15 mg/rat in 50% ethanol). Reactivation of colitis was induced by intrarectal injections of DNBS on day 28 (7.5 mg/rat in 35% ethanol). Animals were sacrificed on day 6 (acute colitis) and 29 (reactivation of colitis). SSR140333 (10 mg/kg) was administered orally starting from the day before the induction of colitis for 7 days (acute colitis) or seven days before the reactivation of colitis. Colonic damage was assessed by means of macroscopic and microscopic scores, myeloperoxidase activity (MPO) and TNF-α tissue levels. Enzyme immunoassay was used to measure colonic substance P levels. Statistical analysis was performed using analysis of variance (one-way or two-way, as appropriate) with the Bonferronis correction for multiple comparisons. Results: DNBS administration impaired body weight gain and markedly increased all inflammatory parameters (p<0.01). Treatment with SSR140333 10 mg/kg significantly counteracted the impairment in body weight gain, decreased macroscopic and histological scores and reduced colonic myeloperoxidase activity (p<0.01). Drug treatment counteracted TNF-α tissue levels and colonic SP concentrations (acute model). Similar results were obtained administering the NK1 receptor antagonist SSR140333 (3 and 10 mg/kg) for 5 days, starting the day after the induction of colitis. Intrarectal administration of DNBS four weeks after the first DNBS administration resulted in reactivation of colitis, with increases in macroscopic and histological damage scores and increase in MPO activity. Preventive treatment with SSR140333 10 mg/kg decreased macroscopic damage score, significantly reduced microscopic damage score but did not affect MPO activity. Conclusions: Treatment with SSR140333 significantly reduced intestinal damage in acute model of intestinal inflammation in rats. The NK1 receptor antagonist SSR140333 was also able to prevent relapse in experimental colitis. These results support the hypothesis of SP involvement in intestinal inflammation and indicate that NK receptor antagonists may have a therapeutic potential in inflammatory bowel disease.

In the last decades, the increase of industrial activities and of the request for the world food requirement, the intensification of natural resources exploitation, directly connected to pollution, have aroused an increasing interest of the public opinion towards initiatives linked to the regulation of food production, as well to the institution of a modern legislation for the consumer guardianship. This work was planned taking into account some important thematics related to marine environment, collecting and showing the data obtained from the studies made on different marine species of commercial interest (Chamelea gallina, Mytilus edulis, Ostrea edulis, Crassostrea gigas, Salmo salar, Gadus morhua). These studies have evaluated the effects of important physic and chemical parameters variations (temperature, xenobiotics like drugs, hydrocarbons and pesticides) on cells involved in the immune defence (haemocytes) and on some important enzymatic systems involved in xenobiotic biotransformation processes (cytochrome P450 complex) and in the related antioxidant defence processes (Superoxide dismutase, Catalase, Heat Shock Protein), from a biochemical and bimolecular point of view. Oxygen is essential in the biological answer of a living organism. Its consume in the normal cellular breathing physiological processes and foreign substances biotransformation, leads to reactive oxygen species (ROS) formation, potentially toxic and responsible of biological macromolecules damages with consequent pathologies worsening. Such processes can bring to a qualitative alteration of the derived products, but also to a general state of suffering that in the most serious cases can provoke the death of the organism, with important repercussions in economic field, in the output of the breedings, of fishing and of aquaculture. In this study it seemed interesting to apply also alternative methodologies currently in use in the medical field (cytofluorimetry) and in proteomic studies (bidimensional electrophoresis, mass spectrometry) with the aim of identify new biomarkers to place beside the traditional methods for the control of the animal origin food quality. From the results it’s possible to point out some relevant aspects from each experiment: 1. The cytofluorimetric techniques applied to O. edulis and C. gigas could bring to important developments in the search of alternative methods that quickly allows to identify with precision the origin of a specific sample, contributing to oppose possible alimentary frauds, in this case for example related to presence of a different species, also under a qualitative profile, but morpholgically similar. A concrete perspective for the application in the inspective field of this method has to be confirmed by further laboratory tests that take also in account in vivo experiments to evaluate the effect in the whole organism of the factors evaluated only on haemocytes in vitro. These elements suggest therefore the possibility to suit the cytofluorimetric methods for the study of animal organisms of food interest, still before these enter the phase of industrial working processes, giving useful information about the possible presence of contaminants sources that can induce an increase of the immune defence and an alteration of normal cellular parameter values. 2. C. gallina immune system has shown an interesting answer to benzo[a]pyrene (B[a]P) exposure, dose and time dependent, with a significant decrease of the expression and of the activity of one of the most important enzymes involved in the antioxidant defence in haemocytes and haemolymph. The data obtained are confirmed by several measurements of physiological parameters, that together with the decrease of the activity of 7-etossi-resourifine-O-deetilase (EROD linked to xenobiotic biotransformation processes) during exposure, underline the major effects of B[a]P action. The identification of basal levels of EROD supports the possible presence of CYP1A subfamily in the invertebrates, still today controversial, never identified previously in C. gallina and never isolated in the immune cells, as confirmed instead in this study with the identification of CYP1A-immunopositive protein (CYP1A-IPP). This protein could reveal a good biomarker at the base of a simple and quick method that could give clear information about specific pollutants presence, even at low concentrations in the environment where usually these organisms are fished before being commercialized. 3. In this experiment it has been evaluated the effect of the antibiotic chloramphenicol (CA) in an important species of commercial interest, Chamelea gallina. Chloramphenicol is a drug still used in some developing countries, also in veterinary field. Controls to evaluate its presence in the alimentary products of animal origin, can reveal ineffective whereas the concentration results to be below the limit of sensitivity of the instruments usually used in this type of analysis. Negative effects of CA towards the CYP1A- IPP proteins, underlined in this work, seem to be due to the attack of free radicals resultant from the action of the antibiotic. This brings to a meaningful alteration of the biotransformation mechanisms through the free radicals. It seems particularly interesting to pay attention to the narrow relationships in C. gallina, between SOD/CAT and CYP450 system, actively involved in detoxification mechanism, especially if compared with the few similar works today present about mollusc, a group that is composed by numerous species that enter in the food field and on which constant controls are necessary to evaluate in a rapid and effective way the presence of possible contaminations. 4. The investigations on fishes (Gadus morhua, and Salmo salar) and on a bivalve mollusc (Mytilus edulis) have allowed to evaluate different aspects related to the possibility to identify a biomarker for the evaluation of the health of organisms of food interest and consequently for the quality of the final product through 2DE methodologies. In the seafood field these techniques are currently used with a discreet success only for vertebrates (fishes), while in the study of the invertebrates (molluscs) there are a lot of difficulties. The results obtained in this work have underline several problems in the correct identification of the isolated proteins in animal organisms of which doesn’t currently exist a complete genomic sequence. This brings to attribute some identities on the base of the comparison with similar proteins in other animal groups, incurring in the possibility to obtain inaccurate data and above all discordant with those obtained on the same animals by other authors. Nevertheless the data obtained in this work after MALDI-ToF analysis, result however objective and the spectra collected could be again analyzed in the future after the update of genomic database related to the species studied. 4-A. The investigation about the presence of HSP70 isoforms directly induced by different phenomena of stress like B[a]P presence, has used bidimensional electrophoresis methods in C. gallina, that have allowed to isolate numerous protein on 2DE gels, allowing the collection of several spots currently in phase of analysis with MALDI-ToF-MS. The present preliminary work has allowed therefore to acquire and to improve important methodologies in the study of cellular parameters and in the proteomic field, that is not only revealed of great potentiality in the application in medical and veterinary field, but also in the field of the inspection of the foods with connections to the toxicology and the environmental pollution. Such study contributes therefore to the search of rapid and new methodologies, that can increase the inspective strategies, integrating themselves with those existing, but improving at the same time the general background of information related to the state of health of the considered animal organism, with the possibility, still hypothetical, to replace in particular cases the employment of the traditional techniques in the alimentary field.

“Naturally occurring cancers in pet dogs and humans share many features, including histological appearance, tumour genetics, molecular targets, biological behaviour and response to conventional therapies. Studying dogs with cancer is likely to provide a valuable perspective that is distinct from that generated by the study of human or rodent cancers alone. The value of this opportunity has been increasingly recognized in the field of cancer research for the identification of cancer-associated genes, the study of environmental risk factors, understanding tumour biology and progression, and, perhaps most importantly, the evaluation and development of novel cancer therapeutics”.(Paoloni and Khanna, 2008) In last years, the author has investigated some molecular features of cancer in dogs. The Thesis is articulated in two main sections. In section 1, the preliminary results of a research project aimed at investigating the role of somatic mutations of Ataxia-Telangiectasia mutated (ATM) gene in predisposing to cancer in boxer dogs, are presented. The canine boxer breed may be considered an unique opportunity to disclose the role of ATM somatic mutation since boxer dogs are known to be dramatically susceptible to cancer and since they may be considered a closed gene pool. Furthermore, dogs share with human the some environment. Overall, the abovementioned features could be considered extremely useful for our purposes. In the section 2, the results of our studies aimed at setting up accurate and sensitive molecular assays for diagnosing and assessing minimal residual disease in lymphoproliferative disorders of dogs, are presented. The results of those molecular assay may be directly translated in the field of Veterinary practice as well as the may be used to improve our objective evaluation of new investigational drugs effectiveness in canine cancer trials.

The present study was performed to validate a spatial working memory task using pharmacological manipulations. The water escape T-maze, which combines the advantages of the Morris water maze and the T-maze while minimizes the disadvantages, was used. Scopolamine, a drug that affects cognitive function in spatial working memory tasks, significantly decreased the rat performance in the present delayed alternation task. Since glutamate neurotransmission plays an important role in the maintaining of working memory, we evaluated the effect of ionotropic and metabotropic glutamatergic receptors antagonists, administered alone or in combination, on rat behaviour. As the acquisition and performance of memory tasks has been linked to the expression of the immediately early gene cFos, a marker of neuronal activation, we also investigated the neurochemical correlates of the water escape T-maze after pharmacological treatment with glutamatergic antagonists, in various brain areas. Moreover, we focused our attention on the involvement of perirhinal cortex glutamatergic neurotransmission in the acquisition and/or consolidation of this particular task. The perirhinal cortex has strong and reciprocal connections with both specific cortical sensory areas and some memory-related structures, including the hippocampal formation and amygdala. For its peculiar position, perirhinal cortex has been recently regarded as a key region in working memory processes, in particular in providing temporary maintenance of information. The effect of perirhinal cortex lesions with ibotenic acid on the acquisition and consolidation of the water escape T-maze task was evaluated. In conclusion, our data suggest that the water escape T-maze could be considered a valid, simple and quite fast method to assess spatial working memory, sensible to pharmacological manipulations. Following execution of the task, we observed cFos expression in several brain regions. Furthermore, in accordance to literature, our results suggest that glutamatergic neurotransmission plays an important role in the acquisition and consolidation of working memory processes.

Mycotoxins are heterogeneous chemical compounds characterized by a low molecular weight and synthesized by the secondary metabolism of different molds. Fumonisins are water-soluble mycotoxins produced by Fusarium species spoiling corn and derived produc ts. These mycotoxins can be a health hazard when consuming contaminated cereals, but they can reach humans also indirectly through the consumption of food products derived from animals fed with contaminated feed. Fumonisins have been associated with several animal and human diseases: they are suspected risk factors for esophageal and liver cancers, neural tube defects and cardiovascular problems. Improved methods are needed to accurately assess fumonisins concentrations in food of vegetable and animal origin, in order to prevent acute and chronic human exposure. The aim of the present work was to evaluate the versatility and the performances of mass spectrometry, coupled with liquid chromatography, in fumonisins analysis from foods and matrices of animal origin. Different methods for the identification and quantification of fumonisins and related products have been developed and validated to determine fumonisin B1 in milk, fumonisin B1, fumonisin B2 and their complete hydrolyzed products (HFB1 and HFB2) in pig liver and fumonisins B1 and B2 in complete and complementary dry dog food. The experimental procedures have been carefully studied, considering matrices features, number and type of molecules to detect. Therefore, several extraction, clean up and separation techniques were tested in order to obtain the better conditions of sample processing. The fit for purpose sample preparation, matched with high mass spectrometry sensibility and specificity, have allowed to achieve good results in any tested animal matrices. Hence, the developed methods were validated and have shown a high accuracy, sensibility and precision, fulfilling performance requirements of Decision 2002/657/EC and of European Project Standard, Measuring and Testing (SMT). In any developed method, the analytes were identified and quantified even at very low concentrations : the limits of quantification resulted lower than other similar works, performed with different detectors. These methods were applied to some commercial samples and to some samples collected for research projects in the Department of Veterinary Public Health and Animal Pathology (DVPHAP) of University of Bologna. Although the disclosed data must be considered completely preliminary and without statistical significance, they emphasize the presence of mycotoxins in animal products. The outcomes obtained from the processed samples (bovine milk, pig liver and dry dog food) suggest the efficacy of these methods also on other food matrices, confirming the versatility and the performances of mass spectrometry, coupled with liquid chromatography, in fumonisins analysis. Moreover the results underline the need to set up a large scale monitoring in order to evaluate the presence of fumonisins in food of animal origin for human consumption.
