Algal blooms, worsening marine ecosystems and causing great economic loss, have been paid much attention to for a long time. Such environmental factors as light penetration, water temperature, and nutrient concentration are crucial in blooms processes. Among them, only nutrients can be controlled. Therefore, the threshold of nutrients for algal blooms is of great concern. To begin with, a dynamic eutrophication model has been constructed to simulate the algal growth and phosphorus cycling. The model encapsulates the essential biological processes of algal growth and decay, and phosphorus regeneration due to algal decay. The nutrient limitation is based upon commonly used Monod's kinetics. The effects of temperature and phosphorus limitation are particularly addressed. Then, we have endeavored to elucidate the threshold of phosphorus at different temperature for algal blooms. Based on the numerical simulation, the isoquant contours of change rate of alga as shown in the figure are obtained, which obviously demonstrate the threshold of nutrient at an arbitrary reasonable temperature. The larger the change rate is, the more rapidly the alga grows. If the phosphorus concentration at a given temperature remains larger than the threshold the algal biomass may increase monotonically, leading to the algal blooming. With the rising of temperature, the threshold is apparently reduced, which may explain why likely red tide disasters occur in a fine summer day. So, high temperature and sufficient phosphorus supply are the major factors which result in algal growth and blowout of red tide.

ENGLISH: An average of 78 m. of water was vertically displaced by wind-driven upwelling during the dry season (January-April) in Panama Bay at the head of the Gulf of Panama. The standing crop of phytoplankton and its productivity were significantly greater during the months of upwelling than during the rainy season. Equivalent results were found by three different methods used to calculate photosynthesis: direct measurement with radiocarbon; increase of dissolved oxygen in the water column; and decrease of phosphate-phosphorus in the water column. About 90 g. of carbon per square meter of sea surface were fixed by the phytoplankton during each upwelling season (January-April) and about 90 g. of carbon were fixed during each rainy season (May-December) resulting in an annual production of about 180 g. of carbon per square meter of sea surface SPANISH: Un promedio de 78 m. de agua fué desplazado verticalmente por la fuerza de los vientos que ocasionan el fenómeno conocido por afloramiento durante la estación seca (enero a abril) en la Bahía de Panamá, a la cabeza del Golfo de Panamá. La cosecha estable de fitoplancton y su productividad fueron significativamente mayores durante los meses de afloramiento, que durante la estación lluviosa. Se obtuvieron resultados equivalentes por medio de tres diferentes métodos usados para calcular la fotosíntesis: medición directa con radiocarbono, aumento del oxígeno disuelto en la columna de agua, y disminución del fosfato-fósforo en la columna de agua. Por medio del fitoplancton durante cada estación de afloramiento (enero-abril), se fijaron alrededor de 90 g. de carbono por metro cuadrado de la superficie del mar, y durante cada estación lluviosa (mayo-diciembre) se fijaron 90 g. de carbono lo que resulta en una producción anual de aproximadamente 180 g. de carbono por metro cuadrado de la superficie del mar.

With the advent of well-defined ruthenium olefin metathesis catalysts that are highly active and stable to a variety of functional groups, the synthesis of complex organic molecules and polymers is now possible; this is reviewed in Chapter 1. The majority of the rest of this thesis describes the application of these catalysts towards the synthesis of novel polymers that may be useful in biological applications and investigations into their efficacy.

A method was developed to produce polyethers by metathesis, and this is described in Chapters 2 and 3. An unsaturated 12-crown-4 analog was made by template- directed ring-closing metathesis (RCM) and utilized as a monomer for the synthesis of unsaturated polyethers by ring-opening metathesis polymerization (ROMP). The yields were high and a range of molecular weights was accessible. In a similar manner, substituted polyethers with various backbones were synthesized: polymers with benzo groups along the backbone and various concentrations of amino acids were prepared. The results from in vitro toxicity tests of the unsubstituted polyethers are considered.

The conditions necessary to synthesize polynorbornenes with pendent bioactive peptides were explored as illustrated in Chapter 4. First, the polymerization of various norbornenyl monomers substituted with glycine, alanine or penta(ethylene glycol) is described. Then, the syntheses of polymers substituted with peptides GRGD and SRN, components of a cell binding domain of fibronectin, using newly developed ruthenium initiators are discussed.

In Chapter 5, the syntheses of homopolymers and a copolymer containing GRGDS and PHSRN, the more active forms of the peptides, are described. The ability of the polymers to inhibit human dermal fibroblast cell adhesion to fibronectin was assayed using an in vitro competitive inhibition assay, and the results are discussed. It was discovered that the copoymer substituted with both GRGDS and PHSR peptides was more active than both the GRGDS-containing homopolymer and the GRGDS free peptide.

Historically, one of the drawbacks to using metathesis is the removal of the residual ruthenium at the completion of the reaction. Chapter 6 describes a method where the water soluble tris(hydroxymethyl)phosphine is utilized to facilitate the removal of residual ruthenium from RCM reaction products.

When dissolved in water, compounds of nitrogen and phosphorus ought to contain the basic assimilated food requirements for autotrophic plants and therefore autotrophic algae. This article summarises the occurrence of nitrogen in water, how species of algae utilize nitrogen and phosphorus forms for growth and the capacities of algae to adapt to environments of different nutrient wealth. This topic has unquestionable importance not only for the purpose of survival of a species but also in deciding indirectly about the stability of ecosystems.

Acetyltransferases and deacetylases catalyze the addition and removal, respectively, of acetyl groups to the epsilon-amino group of protein lysine residues. This modification can affect the function of a protein through several means, including the recruitment of specific binding partners called acetyl-lysine readers. Acetyltransferases, deacetylases, and acetyl-lysine readers have emerged as crucial regulators of biological processes and prominent targets for the treatment of human disease. This work describes a combination of structural, biochemical, biophysical, cell-biological, and organismal studies undertaken on a set of proteins that cumulatively include all steps of the acetylation process: the acetyltransferase MEC-17, the deacetylase SIRT1, and the acetyl-lysine reader DPF2. Tubulin acetylation by MEC-17 is associated with stable, long-lived microtubule structures. We determined the crystal structure of the catalytic domain of human MEC-17 in complex with the cofactor acetyl-CoA. The structure in combination with an extensive enzymatic analysis of MEC-17 mutants identified residues for cofactor and substrate recognition and activity. A large, evolutionarily conserved hydrophobic surface patch distal to the active site was shown to be necessary for catalysis, suggesting that specificity is achieved by interactions with the alpha-tubulin substrate that extend outside of the modified surface loop. Experiments in C. elegans showed that while MEC-17 is required for touch sensitivity, MEC-17 enzymatic activity is dispensible for this behavior. SIRT1 deacetylates a wide range of substrates, including p53, NF-kappaB, FOXO transcription factors, and PGC-1-alpha, with roles in cellular processes ranging from energy metabolism to cell survival. SIRT1 activity is uniquely controlled by a C-terminal regulatory segment (CTR). Here we present crystal structures of the catalytic domain of human SIRT1 in complex with the CTR in an apo form and in complex with a cofactor and a pseudo-substrate peptide. The catalytic domain adopts the canonical sirtuin fold. The CTR forms a beta-hairpin structure that complements the beta-sheet of the NAD^+-binding domain, covering an essentially invariant, hydrophobic surface. A comparison of the apo and cofactor bound structures revealed conformational changes throughout catalysis, including a rotation of a smaller subdomain with respect to the larger NAD^+-binding subdomain. A biochemical analysis identified key residues in the active site, an inhibitory role for the CTR, and distinct structural features of the CTR that mediate binding and inhibition of the SIRT1 catalytic domain. DPF2 represses myeloid differentiation in acute myelogenous leukemia. Finally, we solved the crystal structure of the tandem PHD domain of human DPF2. We showed that DPF2 preferentially binds H3 tail peptides acetylated at Lys14, and binds H4 tail peptides with no preference for acetylation state. Through a structural and mutational analysis we identify the molecular basis of histone recognition. We propose a model for the role of DPF2 in AML and identify the DPF2 tandem PHD finger domain as a promising novel target for anti-leukemia therapeutics.

Wastewater treatment reduces environmental contamination by removing gross solids and mitigating the effects of pollution. Treatment also reduces the number of indicator organisms and pathogens. In this work, the fates of two coliform bacteria, Escherichia coli and Serratia marcescens, were analyzed in an activated sludge process to determine the main mechanisms involved in the reduction of pathogenic microorganisms during wastewater treatment. These bacteria, modified to express green fluorescent protein, were inoculated in an activated sludge unit and in batch systems containing wastewater. The results suggested that, among the different biological factors implied in bacterial removal, bacterivorous protozoa play a key role. Moreover, a representative number of bacteria persisted in the system as free-living or embedded cells, but their distribution into liquid or solid fractions varied depending on the bacterium tested, questioning the real value of bacterial indicators for the control of wastewater treatment process. Additionally, viable but nonculturable cells constituted an important part of the bacterial population adhered to solid fractions, what can be derived from the competition relationships with native bacteria, present in high densities in this environment. These facts, taken together, emphasize the need for reliable quantitative and qualitative analysis tools for the evaluation of pathogenic microbial composition in sludge, which could represent an undefined risk to public health and ecosystem functions when considering its recycling.

Os rios em todo o mundo estão ameaçados através da alteração do uso e cobertura do solo. Nesse contexto, o perifíton é sugerido como indicador biológico devido ao seu ciclo de vida curto, à comunidade autótrofa e à riqueza de espécies. O objetivo deste estudo foi identificar mudanças em parâmetros estruturais e funcionais da assembleia perifitica em relação aos diferentes tipos de cobertura do solo (pristino, perturbação intermediária e perturbado); em especial na limitação nutricional, crescimento e acúmulo, composição taxonômica e estequiometria da comunidade. O trabalho foi desenvolvido em três rios (Santa Maria, Itaperiti, Anil) da bacia Guapi-Macacu, RJ, Brasil. A limitação nutricional foi acessada através de Substratos Difusores de Nutrientes alocados in situ; estes substratos consistem em potes com agar-agar, enriquecidos com nutrientes (N, P, N+P), ou não (controle), que difundem até o filtro, servindo de substrato para o crescimento perifítico. Ainda, o crescimento foi acessado através da colonização de azulejos, amostrados em datas diferentes; na última data, a amostragem também foi feita para a verificação da composição taxonômica e estequiométrica da assembleia. Os resultados mostraram que a comunidade perifítica é limitada primariamente por luz nos pontos pristinos; já nos pontos perturbados, há limitação por nitrogênio ou fósforo, dependendo do uso do solo predominante agricultura com maior aporte de nitrogênio, causando limitação por fósforo, e pecuária com maior aporte de fósforo, causando limitação por nitrogênio. Os trechos perturbados apresentaram maior biomassa perifítica em termos de clorofila a; além disso, também mostraram as maiores taxas de crescimento da comunidade. Também, houve mudanças na composição taxonômica da comunidade no gradiente de perturbação, com o aumento de gêneros tolerantes à poluição e diminuição de gêneros não tolerantes e da diversidade e a equitabilidade. A intensidade luminosa foi determinante na presença dos táxons, e a concentração de fosfato, nas suas abundâncias. Por fim, as razões estequiométricas - C:N, C:P and N:P - diminuíram em função da concentração de nutrientes na água, indicando o potencial de remoção de nutrientes em córregos poluídos. O presente trabalho foi inovador na área limnológica devido à abordagem estequiométrica em conjunto com o grande número de outras variáveis biológicas. Em tempo, a dificuldade de separação dos efeitos das variáveis ambientais neste estudo in situ faz com que seja sugerido o uso de experimentos controlados em mesocosmos posteriormente

O aumento da concentração de nutrientes nos corpos receptores, principalmente nitrogênio e fósforo oriundos de efluentes sanitários e industriais pode gerar o fenômeno da eutrofização. Para que isto não ocorra é necessário que este efluente passe por um tratamento adequado, no entanto, o papel desempenhado por diversos grupos de microrganismos encontrados nos sistemas de tratamento de efluentes não é completamente compreendido devido à complexidade das interações. Este trabalho teve como objetivo caracterizar a estrutura e dinâmica da comunidade microbiana (bactérias envolvidas no ciclo do nitrogênio e microfauna) e avaliar a atividade biológica dos reatores aeróbio e anaeróbio de uma indústria de alimentos. Os parâmetros físico-químicos da estação de tratamento foram monitorados, bem como foi feita a avaliação da estrutura e dinâmica da comunidade bacteriana envolvida no ciclo do nitrogênio por meio da técnica de Hibridização in situ Fluorescente. A microfauna do reator aeróbio foi caracterizada e classificada conforme o Índice Biótico do Lodo. A atividade biológica do lodo foi avaliada através do Teste de Respirometria e foram feitas correlações entre a microbiota encontrada no reator aeróbio e parâmetros físico-químicos. Os parâmetros físico-químicos analisados estiveram dentro dos limites permitidos pelas legislações federais e estaduais e os parâmetros Demanda Bioquímica de Oxigênio, Demanda Química de Oxigênio e Nitrogênio Kjeldahl foram reduzidos de 99,8%, 99,6% e 74,9%, respectivamente. Foi possível observar a presença tanto de bactérias oxidadoras de nitrito quanto de amônia em ambos os reatores analisados, bem como em cada ponto de coletas dentro dos reatores. A bactéria Pseudomonas fluorescens também ocorreu em todos os pontos de coleta dos dois reatores. Dentre os grupos que compõem a microfauna do lodo ativado, os ciliados rastejantes foram os mais frequentes, seguido pelas tecamebas, rotíferos, ciliados sésseis, ciliados livre natantes, flagelados e outros invertebrados. Além disso, não houve diferença entre as densidades dos grupos encontradas nos Pontos 1 e 2 do reator aeróbio e o Índice Biótico do Lodo encontrado foi igual a 8 (classe I). A semelhança apresentada entre a Taxa de Consumo de Oxigênio dos pontos 1 e 2, bem como a Taxa de Consumo de Oxigênio específica entre os pontos 1 e 2 sugere que o oxigênio é distribuído de forma homogênea dentro do tanque de aeração, fazendo com que os microrganismos tenham condições semelhantes de crescimento. Os ciliados livre natantes apresentaram correlação positiva com a DQO e DBO5 e os ciliados sésseis apresentaram correlação negativa com a DQO e com a DBO5. Os rotíferos apresentaram correlação negativa com Sólidos Suspensos Voláteis do reator aeróbio. Os ciliados rastejantes, tecamebas e rotíferos apresentaram correlação positiva com a microfauna total encontrada no reator aeróbio. Os ciliados livre natantes apresentaram correlação negativa com os ciliados sésseis, bactérias totais, Nitrobacter e outras bactérias; e correlação positiva com outros invertebrados. Os flagelados apresentaram correlação negativa com as bactérias totais, enquanto as outras bactérias apresentaram correlação positiva. Os outros invertebrados apresentaram correlação negativa com Nitrobacter.

The pigments (melanoidins) in molasses wastewater are refractory to conventional biological treatment. Ferric chloride was used as coagulant to remove color and chemical oxygen demand (COD) from molasses effluent. Using jar test procedure, main operating conditions such as pH and coagulant dosage were investigated. Under the optimum conditions, up to 86% and 96% of COD and color removal efficiencies were achieved. Residual turbidity in supernatant was less than 5 NTU and Fe3+ concentration was negligible because of effective destabilization and subsequent sedimentation. The results of high performance size exclusion chromatography (HPSEC) show that low molecular weight (MW) fraction of melanoidins is more reactive than high MW fraction and increase in the concentration of the lowest MW organic group is related to the capacity of charge neutralization. Aggregate size measurement reveals the size effect on the settleability of flocs formed, with larger flocs settling more rapidly. Charge neutralization and co-precipitation are proposed as predominant coagulation mechanism under the optimum conditions. (C) 2009 Elsevier B.V. All rights reserved.

Nutrient-rich effluents caused rising concern due to eutrophication of aquatic environment by utilization of a large amount of formula feed. Nutrient removal and water quality were investigated by planting aquatic vegetable on artificial beds in 36-m(2) concrete fishponds. After treatment of 120 days, 30.6% of total nitrogen (TN) and 18.2% of total phosphorus (TP) were removed from the total input nutrients by 6-m(2) aquatic vegetable Ipomoea aquatica. The concentrations of TN, TP, chemical oxygen demand (COD) and chlorophyll a in planted ponds were significantly lower than those in non-planted ponds (P<0.05). Transparency of water in planted ponds was much higher than that of control ponds. No significant differences in the concentration of total ammonia nitrogen (TAN), nitrate nitrogen (NO3-N) and nitrite nitrogen (NO2--N) were found between planted and non-planted ponds. These results suggested that planting aquatic vegetable with one-sixth covered area of the fishponds could efficiently remove nutrient and improve water quality.

Using artificial systems to simulate natural lake environments with cyanobacterial blooms, we investigated plankton community succession by polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) fingerprinting and morphological method. With this approach, we explored potential ecological effects of a newly developed cyanobacterial blooms removal method using chitosan-modified soils. Results of PCR-DGGE and morphological identification showed that plankton communities in the four test systems were nearly identical at the beginning of the experiment. After applying the newly developed and standard removal methods, there was a shift in community composition, but neither chemical conditions nor plankton succession were significantly affected by the cyanobacteria removal process. The planted Vallisneria natans successfully recovered after cyanobacteria removal, whereas that in the box without removal process did not. Additionally, canonical correspondence analysis indicated that other than for zooplankton abundance, total phosphorus was the most important environmental predictor of planktonic composition. The present study and others suggest that dealing with cyanobacteria removal using chitosan-modified soils can play an important role in controlling cyanobacterial blooms in eutrophicated freshwater systems.

Micronutrients play a very important role in biological processes for wastewater treatment. Many industrial wastewaters lack in nutrients (macronutrients and micronutrients) required for microbial growth, and this is one of the main problems at many activated sludge plants treating industrial wastewater. The microbial community structure is one of the important factors controlling the pollutant-degrading capacity of biological wastewater treatment system. In this study, the concentrations of micronutrients of the textile wastewater discharged from a textile plant were determined, and the effects of micronutrients on treatment efficiency and microorganism community structure of the biological treatment system were studied. The results showed that the optimal concentrations of magnesium, molybdenum, zinc, thiamine and niacin in the textile wastewater were 5.0, 2.0, 1.0, 1.0 and 1.0mg/L, respectively. The COD removal rates when magnesium, molybdenum, zinc, thiamine and niacin were added individually to the wastewater in their optimal concentrations were 1.8, 1.4, 1.3, 1.6 and 2.2 times of that of the control, respectively. The improving effects of combinations of zinc and thiamine, zinc and niacin, thiamine and niacin were better than single micronutrient. The diversity of quinones (DQ) changed significantly after the micronutrient was added into the wastewater treatment system. This indicated that there was probably a feasibility of optimizing the biological treatment performances and microorganism community structure of textile wastewater treatment system through micronutrient supplement.

Micronutrients play an important role in biological processes for wastewater treatment. Many industrial wastewaters lack in nutrients required for microbial growth, and this is one of the problems at many activated sludge plants treating them. In this study, the effects of the micronutrient niacin on the COD removal rates of textile wastewater, together with the effect of Mixed Liquor Suspended Solids (MLSS) on niacin, were studied. Certain improvement effects were found on the removal rates of COD, when 0.5 similar to 2.0 mg/L niacin was added to the textile wastewater. The optimal concentration of niacin was 1.0 mg/L, which was continuously added during textile wastewater treatment, and removal rates were 1.31 times compared to those of the control system. The concentration of MLSS was probably one of the factors influencing treatment efficiency, and the biological performance of treatment system could be optimized through micronutrient niacin supplements.

Pollution resulting from increased human activities is threatening Lake Donghu, its effects being characterized by serious eutrophication. A steady increase of phosphorus loading is the most important factor of the lake eutrophication. Pollution external control projects are being implemented and will be accomplished before the year 2010. In order to predict the restoration rate by the lake's self-purification after the projects of external control, a model of predicting the removal rate of total phosphorus (TP) from lake water is developed, and a brief method of estimating the release and export rate of sediment phosphorus is suggested. Results show that, on the premise of external loading fully controlled. The restoration needs about 55 years or more. Obviously, the great P pool in the sediment will be a limiting factor of preventing the improvement of water quality after the external loading is under control. Based on the estimation we conclude that after the external control projects before 2010, in order to restore the lake in a few years, although highly cost, the first step must be the sediment dredging to remove internal loading. The second step is diverting water of River Changjiang into the lake to accelerate the improvement of lake water. Otherwise, removal of pollutant sources will become meaningless.
