Covalent “click” cycloaddition was used to functionalize silica substrates with pH-sensitive nanoparticles, thus producing uniform and highly luminescent analytical devices usable in both commercial fluorimeters and fluorescence microscopes. Quantitative and spatially-resolved extracellular pH measurements were successfully achieved on live cardiac fibroblasts with these novel ion-sensitive surfaces.

Aerial surveys of narwhals (Monodon monoceros) were conducted in the Canadian High Arctic during the month of August from 2002 to 2004. The surveys covered the waters of Barrow Strait, Prince Regent Inlet, the Gulf of Boothia, Admiralty Inlet, Eclipse Sound, and the eastern coast of Baffin Island, using systematic sampling methods. Fiords were flown along a single transect down the middle. Near-surface population estimates increased by 1.9%-8.7% when corrected for perception bias. The estimates were further increased by a factor of approximately 3, to account for individuals not seen because they were diving when the survey plane flew over (availability bias). These corrections resulted in estimates of 27 656 (SE = 14 939) for the Prince Regent and Gulf of Boothia area, 20 225 (SE = 7285) for the Eclipse Sound area, and 10 073 (SE = 3123) for the East Baffin Island fiord area. The estimate for the Admiralty Inlet area was 5362 (SE = 2681) but is thought to be biased. Surveys could not be done in other known areas of occupation, such as the waters of the Cumberland Peninsula of East Baffin, and channels farther west of the areas surveyed (Peel Sound, Viscount Melville Sound, Smith Sound and Jones Sound, and other channels of the Canadian Arctic archipelago). Despite these probable biases and the incomplete coverage, results of these surveys show that the summering range of narwhals in the Canadian High Arctic is vast. If narwhals are philopatric to their summering areas, as they appear to be, the total population of that range could number more than 60 000 animals. The largest numbers are in the western portion of their summer range, around Somerset Island, and also in the Eclipse Sound area. However, these survey estimates have large variances due to narwhal aggregation in some parts of the surveyed areas.

An understanding of the adaptability of belugas (Delphinapterus leucas) to changing ice conditions is required to interpret and predict possible changes in habitat selection in response to projected loss of sea ice throughout the circumpolar Arctic. We analyzed beluga observations made during spring aerial surveys for ringed seals conducted from 1975 to 1979 in the eastern Beaufort Sea. Despite interannual variability in the extent and distribution of sea ice, belugas consistently selected areas with water depths of 200-500 m and heavy ice concentrations (8/10 to 10/10) while areas of open water to light ice concentrations (0/10 to 1/10) were not selected. Belugas were also found in proximity to regions with >0.5 degrees seafloor slope which include the continental slope and other areas with the potential for oceanographic upwellings. In most years (4 of 5), fast-ice edges and coastal areas were not selected. In the lightest ice year analyzed, belugas showed less specificity in habitat selection as their distribution expanded and shifted shoreward to fast-ice edges. The observed distribution is discussed in terms of predator-prey relationships particularly with reference to beluga feeding on polar cod (Boreogadus saida). More research is required to examine and compare possible changes in distribution since the late 1970s and to investigate the factors driving the patterns described.

Pathogenesis-related proteins from intercellular fluid washings of stressed barley (Hordeum vulgare L.) leaves were analyzed to determine their binding to various water-insoluble polysaccharides. Three proteins (19, 16, and 15 kD) bound specifically to several water-insoluble β-1,3-glucans. Binding of the barley proteins to pachyman occurred quickly at 22°C at pH 5.0, even in the presence of 0.5 m NaCl, 0.2 m urea, and 1% (v/v) Triton X-100. Bound barley proteins were released by acidic treatments or by boiling in sodium dodecyl sulfate. Acid-released barley proteins could bind again specifically and singly to pachyman. Water-soluble laminarin and carboxymethyl-pachyman competed for the binding of the barley proteins to pachyman. The N-terminal sequence of the 19-kD barley β-1,3-glucan-binding protein showed near identity to the barley seed protein BP-R and high homology to other thaumatin-like (TL) permatins. The 16-kD barley protein was also homologous to TL proteins, whereas the 15-kD barley protein N-terminal sequence was identical to the pathogenesis-related Hv-1 TL protein. Antifungal barley protein BP-R and corn (Zea mays) zeamatin were isolated by binding to pachyman. Two extracellular proteins from stressed pea (Pisum sativum L.) also bound to pachyman and were homologous to TL proteins.
