Suite à la rencontre d’un antigène (Ag) présenté à la surface des cellules présentatrice de l’Ag (CPA), les lymphocytes T naïfs, ayant un récepteur des cellules T (RCT) spécifique de l’Ag, vont proliférer et se différencier en LT effecteurs (1). Suite à l’élimination de l’Ag la majorité des LTe vont mourir par apoptose alors que les restants vont se différencier en LT mémoire (LTm) protégeant l’organisme à long terme. Les mécanismes qui permettent la différenciation des LTe en LTm sont encore inconnus. Pour comprendre comment les LTm CD8+ sont générés à partir des LTe, nous avons émis l’hypothèse que la densité de l’Ag présenté par les CPA peut avoir un impact sur la sélection des LT CD8+ répondant l’Ag à se différencier en LTm. De manière intéressante, nos résultats montrent qu’une immunisation avec des cellules dendritiques (DCs) exprimant un haut niveau de complexe CMH/peptide à sa surface permet le développement de LTm. À l’inverse, le développement des LTm est fortement réduit (10-20X) lorsque les souris sont immunisées avec des DCs exprimant un niveau faible de complexes CMH/peptide à leur surface. De plus, la quantité d’Ag n’a aucune influence ni sur l’expansion des LT CD8+ ni sur l’acquisition de leurs fonctions effectrices, mais affecte de manière critique la génération des LTm. Nos résultats suggèrent que le nombre de RCT engagé lors de la reconnaissance de l’Ag est important pour la formation des LTm. Pour cela nous avons observé par vidéo-microscopie le temps d’interaction entre des LTn et des DCs. Nos résultats montrent que le temps et la qualité de l’interaction sont dépendants de la densité d’Ag présenté par les DCs. Effectivement, nous observons une diminution dans le pourcentage de LT faisant une interaction prolongée avec les DCs quand le niveau d’Ag est faible. De plus, nous observons des variations de l’expression des facteurs de transcription clefs impliqués dans la différenciation des LTm tels qu’Eomes, Bcl-6 et Blimp-1. Par ailleurs, la densité d’Ag fait varier l’expression du Neuron-derived orphan nuclear receptor 1 (Nor-1). Nor-1 est impliqué dans la conversion de Bcl-2 en molécule pro-apoptotique et contribue à la mort par apoptose des LTe pendant la phase de contraction. Notre modèle propose que la densité de l’épitope contrôle la génération des CD8+ LTm. Une meilleure compréhension des mécanismes impliqués dans la génération des LTm permettra le développement de meilleures stratégies pour la génération de vaccin. Dans un second temps, nous avons évalué le rôle du signal RCT dans l’homéostasie des LTm. Pour ce faire, nous avons utilisé un modèle de souris transgénique pour le RCT dont son expression peut être modulée par un traitement à la tétracycline. Ce système nous a permis d’abolir l’expression du RCT à la surface des LTm. De manière intéressante, en absence de RCT exprimé, les LTm CD8+ peuvent survivre à long terme dans l’organisme et rester fonctionnels. De plus, une sous population des LTm CD4+ a la capacité de survivre sans RCT exprimé dans un hôte lymphopénique alors que l’autre sous population nécessite l’expression du RCT.

Lors d’une infection par un pathogène, des lymphocytes T CD8+ naïfs (LTn) spécifiques de l’antigène sont activés, prolifèrent et se différencient en LT effecteurs (LTe). Les LTe produisent différentes cytokines et acquièrent une activité cytotoxique menant à l’élimination du pathogène. Seulement 5 à 10 % des LTe survivront et se différencieront en LT mémoires (LTm), qui sont capables de répondre plus rapidement lors d’une seconde infection par le même pathogène, contribuant au succès de la vaccination. Toutefois, la compréhension de l’ensemble des mécanismes régulant le développement des LTe et des LTm demeure incomplète. Afin de mieux comprendre les signaux requis pour la différenciation des LT CD8+ lors de la réponse immune, nous avons posé deux hypothèses. Nous avons d’abord proposé que différentes cellules présentatrices d’antigène (CPA) fournissent différents signaux au moment de la reconnaissance antigénique influençant ainsi le devenir des LT CD8+. Vu leur potentiel d’utilisation en immunothérapie, nous avons comparé la capacité d’activation des LT CD8+ par les lymphocytes B activés via le CD40 (CD40-B) et les cellules dendritiques (CD). Nous avons montré que l’immunisation avec des CD40-B induit une réponse effectrice mais, contrairement à l’immunisation avec des CD, pratiquement aucun LTm n’est généré. Les LTe générés sont fonctionnels puisqu’ils sécrètent des cytokines, ont une activité cytotoxique et contrôlent une infection avec Listeria monocytogenes (Lm). Nous proposons qu’une sécrétion plus faible de cytokines par les CD40 B ainsi qu’une interaction plus courte et moins intime avec les LT CD8+ comparativement aux CD contribuent au défaut de différenciation des LTm observé lors de la vaccination avec les CD40-B. Ensuite, nous posé l’hypothèse que, parmi les signaux fournis par les CPA au moment de la reconnaissance antigénique, la voie de signalisation Notch influence le développement des LTe, mais aussi des LTm CD8+ en instaurant un programme génétique particulier. D’abord, grâce à un système in vitro, le rôle de la signalisation Notch dans les moments précoces suivant l’activation du LT CD8+ a été étudié. Ce système nous a permis de démontrer que la voie de signalisation Notch régule directement l’expression de la molécule PD-1. Ensuite, grâce à des souris où il y a délétion des récepteurs Notch1 et Notch2 seulement chez les LT CD8+ matures, un rôle de la voie de signalisation Notch dans la réponse immune des LT CD8+ a été démontré. Nos résultats démontrent que suite à une infection avec Lm ou à une immunisation avec des CD, la signalisation Notch favorise le développement de LTe, exprimant fortement KLRG1 et faiblement CD127, destinés à mourir par apoptose. Toutefois, la signalisation Notch n’a pas influencé la génération de LTm. De façon très intéressante, l’expression des récepteurs Notch influence la production d’IFN- en fonction du contexte d’activation. En effet, suite à une infection avec Lm, l’absence des récepteurs Notch n’affecte pas la production d’IFN- par les LTe, alors qu’elle est diminuée suite à une immunisation avec des CD suggérant un rôle dépendant du contexte pour la voie de signalisation Notch. Nos résultats permettent une meilleure compréhension des signaux fournis par les différentes CPA et de la voie de signalisation Notch, donc des mécanismes moléculaires régulant la différenciation des LT CD8+ lors de la réponse immunitaire, ce qui pourrait ultimement permettre d’améliorer les stratégies de vaccination.

The rapid growth in high data rate communication systems has introduced new high spectral efficient modulation techniques and standards such as LTE-A (long term evolution-advanced) for 4G (4th generation) systems. These techniques have provided a broader bandwidth but introduced high peak-to-average power ratio (PAR) problem at the high power amplifier (HPA) level of the communication system base transceiver station (BTS). To avoid spectral spreading due to high PAR, stringent requirement on linearity is needed which brings the HPA to operate at large back-off power at the expense of power efficiency. Consequently, high power devices are fundamental in HPAs for high linearity and efficiency. Recent development in wide bandgap power devices, in particular AlGaN/GaN HEMT, has offered higher power level with superior linearity-efficiency trade-off in microwaves communication. For cost-effective HPA design to production cycle, rigorous computer aided design (CAD) AlGaN/GaN HEMT models are essential to reflect real response with increasing power level and channel temperature. Therefore, large-size AlGaN/GaN HEMT large-signal electrothermal modeling procedure is proposed. The HEMT structure analysis, characterization, data processing, model extraction and model implementation phases have been covered in this thesis including trapping and self-heating dispersion accounting for nonlinear drain current collapse. The small-signal model is extracted using the 22-element modeling procedure developed in our department. The intrinsic large-signal model is deeply investigated in conjunction with linearity prediction. The accuracy of the nonlinear drain current has been enhanced through several issues such as trapping and self-heating characterization. Also, the HEMT structure thermal profile has been investigated and corresponding thermal resistance has been extracted through thermal simulation and chuck-controlled temperature pulsed I(V) and static DC measurements. Higher-order equivalent thermal model is extracted and implemented in the HEMT large-signal model to accurately estimate instantaneous channel temperature. Moreover, trapping and self-heating transients has been characterized through transient measurements. The obtained time constants are represented by equivalent sub-circuits and integrated in the nonlinear drain current implementation to account for complex communication signals dynamic prediction. The obtained verification of this table-based large-size large-signal electrothermal model implementation has illustrated high accuracy in terms of output power, gain, efficiency and nonlinearity prediction with respect to standard large-signal test signals.

Este artigo apresenta os primeiros resultados do projecto de investigação “iDTV-Saúde: Serviços inclusivos de promoção da saúde e bem-estar via televisão digital interactiva”, cujo objectivo é avaliar a potencialidade da televisão digital interactiva (iDTV) para a promoção de serviços, formatos e conteúdos originais relevantes no suporte dos cuidados de saúde pessoais e bem-estar nas pessoas acima dos 55 anos em território português. A prática social da televisão está-se a alterar de forma altamente contingente (Green, 2008) e este projecto ocorre num contexto de profunda transformação deste meio em função do surgimento de novas plataformas tecnológicas, como televisão digital terrestre (TDT) e IPTV, assim como novas tecnologias de comunicação móvel como a WimAX e a Long Term Evolution (LTE). Por conseguinte, e num contexto de adopção de novas tecnologias para a produção e distribuição de conteúdo televisivo, este projecto procura avaliar a satisfação e o potencial gerado pelo serviço de iDTV como um meio de suporte à saúde e bem-estar ao mesmo tempo que facilita o acesso à informação e visualização de conteúdos, nomeadamente no que diz respeito à sua capacidade de aumentar os níveis de capital social entre o grupo alvo.

[EN]The purpose of this project is to study the performance and the efficiency of the LDM technology for the transmission of multicast services in a cellular network and vehicular communication environment. In order to achieve this goal, it will be done a comparison with LTE, the technology nowadays used for multicast-grouping techniques. Thus, the specific parameters of each technology are going to be analyzed and a simulation software is going to be developed. Finally, the results from the above mentioned comparison will be shown, as well as an implementation of LDM in multicast-grouping technique.

Durante los años del colapso de la monarquía española, el Virreinato del Perú estuvo gobernado por Fernando de Abascal, quien, en alianza con la élíte de Lima, aplicó una política de represión en términos ideológicos y militares ante cualquier intento separatista o revolucionario, tanto dentro de su jurisdicción virreinal como fuera de ella, entre cuyos territorios vecinos estaban Quito, Chile y Charcas. Asimismo, mediante esta estrategia, el Virreinato de Lima intentó recuperar parte de la influencia perdida en América del Sur como consecuencia de la aplicación de las reformas borbónicas, especialmente durante la segunda mitad del siglo XVIII. El presente artículo pone atención a la respuesta de Lima contra al movimiento insurgente desatado en Quito a partir de 1809.

With the emerging prevalence of smart phones and 4G LTE networks, the demand for faster-better-cheaper mobile services anytime and anywhere is ever growing. The Dynamic Network Optimization (DNO) concept emerged as a solution that optimally and continuously tunes the network settings, in response to varying network conditions and subscriber needs. Yet, the DNO realization is still at infancy, largely hindered by the bottleneck of the lengthy optimization runtime. This paper presents the design and prototype of a novel cloud based parallel solution that further enhances the scalability of our prior work on various parallel solutions that accelerate network optimization algorithms. The solution aims to satisfy the high performance required by DNO, preliminarily on a sub-hourly basis. The paper subsequently visualizes a design and a full cycle of a DNO system. A set of potential solutions to large network and real-time DNO are also proposed. Overall, this work creates a breakthrough towards the realization of DNO.

In alveolar macrophages, leukotriene (IT) B(4) and cysteinyl LTs (LTC(4), LTD(4) and LTE(4)) both enhance Fc gamma receptor (Fc gamma R)-mediated phagocytosis. In the present study we investigated the role of specific PKC isoforms (PKC-alpha and -delta), the MAP kinases p38 and ERK 1/2, and PI3K in mediating the potentiation of Fc gamma R-mediated phagocytosis induced by addition of leukotrienes to the AMs. It was found that exogenously added LTB(4) and LTD(4) both enhanced PKC-delta and -alpha phosphorylation during Fc gamma R engagement. Studies with isoform-selective inhibitors indicated that exogenous LTB(4) effects were dependent on both PKC-alpha and -delta, while LTD(4) effects were exclusively due to PKC-delta activation. Although both exogenous LTB(4) and LTD(4) enhanced p38 and ERK 1/2 activation, LTB(4) required only ERK 1/2, while LTD(4) required only p38 activation. Activation by both LTs was dependent on PI3K activation. Effects of endogenous LTs on kinase activation were also investigated using selective LT receptor antagonists. Endogenous LTB(4) contributed to Fc gamma R-mediated activation of PKC-alpha, ERK 1/2 and PI3K, while endogenous cysLTs contributes to activation of PKC-delta, p38 and PI3K. Taken together, our data show that the capacities of LTB(4) and LTD(4) to enhance Fc gamma R-mediated phagocytosis reflect their differential activation of specific kinase programs. In this work the results of a spectroscopic study of the southern field narrow-line Be star HD 171054 are presented. High dispersion and signal-to-noise ratio spectra allowed the estimation of the fundamental photospheric parameters such as the projected rotational velocity, effective temperature and superficial gravity from non-LTE stellar atmosphere models. From these parameters and microturbulence, the abundances of He, C, N, O, Mg, Al and Si for this object are estimated. Results show that C is depleted whereas N is overabundant compared with the sun and OB stars in the solar vicinity. Oxygen and helium are close to the solar value. Magnesium is down by 0.43 dex and aluminium and silicon are overabundant. (C) 2010 Elsevier B.V. All rights reserved.

The introduction of a new technology High Speed Downlink Packet Access (HSDPA) in the Release 5 of the 3GPP specifications raises the question about its performance capabilities. HSDPA is a promising technology which gives theoretical rates up to 14.4 Mbits. The main objective of this thesis is to discuss the system level performance of HSDPAMainly the thesis exploration focuses on the Packet Scheduler because it is the central entity of the HSDPA design. Due to its function, the Packet Scheduler has a direct impact on the HSDPA system performance. Similarly, it also determines the end user performance, and more specifically the relative performance between the users in the cell.The thesis analyzes several Packet Scheduling algorithms that can optimize the trade-off between system capacity and end user performance for the traffic classes targeted in this thesis.The performance evaluation of the algorithms in the HSDPA system are carried out under computer aided simulations that are assessed under realistic conditions to predict the results as precise on the algorithms efficiency. The simulation of the HSDPA system and the algorithms are coded in C/C++ language

With the rapid development of telecommunication industry, the IP multimedia Subsystem (IMS) could very well be the panacea for most telecom operators. It is originally defined as the core network for 3G mobile systems by the 3rd Generation Partnership Project (3GPP), the more recent development is merging between fixed line network and wireless networkd This report researchs the characteristic of the IMS data and proposes an IMS characterization analysis. We captured the IMS traffic data with 10 tousands users for about 41 hours. By analyzing the characteristics of the IMS, we know that the most important application in the IMS is VoIP call. Then we use the tool designed by Tsinghua University & Ericsson Company to recognize the data, and the results we got can be used to build the traffic models. From the results of the traffic models, I will get some reasons and conclusion. The traffic model gives out the types of session and types of VoIP call. I bring into a concept—busy hour. This concept is very important because it can help us to know which period is the peak of the VoIP call. The busy hour is from 10:00 to 11:00 in the morning. I also bring into another concept—connection ratio. This concept is significant because it can evaluate whether the VoIP call is good when it use IMS network. By comparing the traffic model with other one’s models, we found the different results from them, both the accuracy and the busy hour. From the contract, we got the advantages of our traffic models.

The introduction of new digital services in the cellular networks, in transmission rates each time more raised, has stimulated recent research that comes studying ways to increase the data communication capacity and to reduce the delays in forward and reverse links of third generation WCDMA systems. These studies have resulted in new standards, known as 3.5G, published by 3GPP group, for the evolution of the third generation of the cellular systems. In this Masters Thesis the performance of a 3G WCDMA system, with diverse base stations and thousand of users is developed with assists of the planning tool NPSW. Moreover the performance of the 3.5G techniques hybrid automatic retransmission and multi-user detection with interference cancellation, candidates for enhance the WCDMA uplink capacity, is verified by means of computational simulations in Matlab of the increase of the data communication capacity and the reduction of the delays in the retransmission of packages of information

Galactic stellar clusters have a great variety of physical properties that make valuable probes of stellar and galactic chemical evolution. Current studies show a discrepancy between the standard evolutionary models and observations, mainly considering the level of mixing and convective dilution of light elements, as well as to the evolution of the angular momentum. In order to better settle some of these properties, we present a detailed spectroscopic analysis of 28 evolved stars, from the turn-off to the RGB, belonging to the stellar open cluster M67. The observations were performed using UVES+FLAMES at VLT/UT2. We determined stellar parameters and metallicity from LTE analysis of Fe I and Fe II lines between 420 1100 nm. The Li abundance was obtained using the line at 6707.78 ˚A, for the whole sample of stars. The Li abundances of evolved stars of M67 present a gradual decreasing when decreasing the effective temperature. The Li dilution factor for giant stars of M67 with Teff ∼ 4350K is at least 2300 times greater than that predicted by standard theory for single field giant stars. The Li abundance as a function of rotation exhibits a good correlation for evolved stars of M67, with a much smaller dispersion than the field evolved stars. The mass and the age seem to be some of the parameters that influence this connection. We discovered a Li-rich subgiant star in M67 (S1242). It is member of a spectroscopic binary system with a high eccentricity. Its Li abundance is 2.7, the highest Li content ever measured for an evolved star in M67. Two possibilities could explain this anomalous Li content: (i) preservation of the Li at the post turn off stage due to tidal effects, or (ii) an efficient dredge-up of Li, hidden below the convective zone by atomic diffusion occurring in the post turn off stage. We also study the evolution of the angular momentum for the evolved stars in M67. The results are in agreement with previous studies dedicated to evolved stars of this cluster, where stars in the same region of the CM-diagram have quite similar rotations, but with values that indicate an extra breaking along the main sequence. Finally, we analize the distributions of the average rotational velocity and of the average Li abundance as a function of age. With relation to the average Li abundances, stars in clusters and field stars present the same type of exponencial decay law t−β. Such decay is observed for ages lesser than 2 Gyr. From this age, is observed that the average Li abundance remain constant, differently of the one observed in the rotation age connection, where the average rotational velocity decreases slowly with age

Anxiety is an emotional phenomenon, and normally it is interpreted as an adaptative behavior front to adversities. In its pathological form, anxiety can severely affect aspects related to the personal and professional life. Studies have shown a close relationship between anxiety disorders and aversive memory processing. Considering that the pharmacotherapy of anxiety disorders is still limited, innovative anxiolytic agents are needed. In this regard, neuropeptides systems are interesting therapeutic targets to the treatment of psychopathologies. Neuropeptide S (NPS), a 20-aminoacid peptide, is the endogenous ligand of a G-protein coupled receptor (NPSR), which has been reported to evoke hyperlocomotion, awakefull states, besides anxiolysis and memory improvements in rodents. This study aimed to investigate the effects of biperiden (BPR; an amnesic drug), diazepam (DZP; an anxiolytic drug) and NPS at three distinct times: pre-training, post-training, and pre-test, in order to assess anxiety and memory process in the same animal model. The elevated Tmaze (ETM) is an apparatus derived from the elevated plus-maze test, which consists of one enclosed and two open arms. The procedure is based on the avoidance of open spaces learned during training session, in which mice were exposed to the enclosed arm as many times as needed to stay 300 s. In the test session, memory is assessed by re-exposing the mouse to the enclosed arm and the latency to enter an open arm was recorded. When injected pre-training, BPR (1 mg/kg) impaired learning and memory processing; DZP (1 and 2 mg/kg) evoked anxiolysis, but only at the dose of 2 mg/kg impaired memory; and NPS 0.1 nmol induced anxiolysis without affecting memory. Post-training injection of DZP (2 mg/kg) or BPR (1 and 3 mg/kg) did not affect memory consolidation, while the post-trainning administration of NPS 1 nmol, but not 0.1 nmol, improved memory in mice. Indeed, pre-trainning administration of NPS 1 nmol did not prevent memory impairment elicited by BPR (2 mg/kg, injected before training). In the open field test, BPR 1 mg/kg and NPS 1 nmol induced hyperlocomotion in mice. In conclusion, the proposed ETM task is practical for the detection of the anxiolytic and amnesic effects of drugs. The anxiolytic and memory enhancement effects of NPS were detected in the ETM task, and reinforce the role of NPS system as an interesting therapeutic target to the treatment of anxiety disorders

Objective. We previously documented that abatacept was effective and safe in patients with juvenile idiopathic arthritis (JIA) who had not previously achieved a satisfactory clinical response with disease-modifying antirheumatic drugs or tumor necrosis factor blockade. Here, we report results from the long-term extension (LTE) phase of that study.Methods. This report describes the long-term, open-label extension phase of a double-blind, randomized, controlled withdrawal trial in 190 patients with JIA ages 6-17 years. Children were treated with 10 mg/kg abatacept administered intravenously every 4 weeks, with or without methotrexate. Efficacy results were based on data derived from the 153 patients who entered the open-label LTE phase and reflect >= 21 months (589 days) of treatment. Safety results include all available open-label data as of May 7, 2008.Results. of the 190 enrolled patients, 153 entered the LTE. By day 589, 90%, 88%, 75%, 57%, and 39% of patients treated with abatacept during the double-blind and LTE phases achieved responses according to the American College of Rheumatology (ACR) Pediatric 30 (Pedi 30), Pedi 50, Pedi 70, Pedi 90, and Pedi 100 criteria for improvement, respectively. Similar response rates were observed by day 589 among patients previously treated with placebo. Among patients who had not achieved an ACR Pedi 30 response at the end of the open-label lead-in phase and who proceeded directly into the LTE, 73%, 64%, 46%, 18%, and 5% achieved ACR Pedi 30, Pedi 50, Pedi 70, Pedi 90, and Pedi 100 responses, respectively, by day 589 of the LTE. No cases of tuberculosis and no malignancies were reported during the LTE. Pneumonia developed in 3 patients, and multiple sclerosis developed in 1 patient.Conclusion. Abatacept provided clinically significant and durable efficacy in patients with JIA, including those who did not initially achieve an ACR Pedi 30 response during the initial 4-month open-label lead-in phase.
