The present study aims to report ovum pickup (OPU), in vitro embryo production (IVEP) and embryo transfer (ET) outcomes of fresh and vitrified buffalo embryos. For this purpose, 36 buffalo donors were submitted to 11 OPU sessions (n = 201). A total of 998 oocytes (5.0 +/- 0.5/donor/session) and 584 viable oocytes (2.9 +/- 0.3/donor/session) were recovered. Viable oocytes (grades 1, 2 and 3) were subjected to IVM, IVF (D0) and IVC. On D2, 54.5% of cleavage rate was obtained. Embryo yield on D7 was 44.9% (grade 1: 229 embryos, grade 2: 5 embryos and grade 3: 28 embryos). From this total, 115 fresh (grades 1 to 3) and 70 vitrified embryos (only grade 1) were transferred into recipients previously synchronized with fixed time embryo transfer (FTET) protocol. Vitrification was performed using the cryotop method. Pregnancy diagnosis in fresh and in vitrified groups were, respectively: 43.5% (50/115) and 37.1% (26/70) on 30 days after embryo transfer, and 41.7% (48/115) and 31.4% (22/70) on 60 days after embryo transfer. In conclusion, our results demonstrate the possibilities for commercial use of the techniques of OPU, IVEP and ET of fresh and vitrified embryos in buffaloes.

The aim of the present study was to evaluate the effects of season of the year (summer and winter) and parity (heifers and cows) on oocyte quality and number in buffaloes. For this purpose, 71 buffaloes had follicular wave emergence synchronized before OPU. OPU of all follicles >= 2mm was done 5 days after the beginning of the hormonal protocol, in 4 replicates (two for each season). Data were analyzed by ANOVA using PROC GLIMMIX, in a 2 x 2 factorial arrangement of treatments. No interactions were observed in following variables: number of follicles, number of total and viable oocytes, recovery rate, percentage of viable oocytes, grade I oocytes, grade II oocytes, grade III oocytes, denuded oocytes, expanded cumulus oocytes, and atretic/degenerated oocytes. Number of follicles visualized at OPU and recovery rate were not affected by parity or season. Relative to parity, number of total and viable oocytes were greater in heifers than in cows, respectively. Concerning season of the year, number of viable oocytes and viable oocyte rate were increased in winter. In conclusion, better oocyte quality can be obtained from heifers and during winter in buffaloes. However, the number of total oocytes seems to be more influenced by parity than by season of the year in this species.
Manipulation of follicle development to ensure optimal oocyte quality and conception rates in cattle

Over the last several decades, a number of therapies have been developed that manipulate ovarian follicle growth to improve oocyte quality and conception rates in cattle. Various strategies have been proposed to improve the responses to reproductive biotechnologies following timed artificial insemination (TAI), superovulation (SOV) or ovum pickup (OPU) programmes. During TAI protocols, final follicular growth and size of the ovulatory follicle are key factors that may significantly influence oocyte quality, ovulation, the uterine environment and consequently pregnancy outcomes. Progesterone concentrations during SOV protocols influence follicular growth, oocyte quality and embryo quality; therefore, several adjustments to SOV protocols have been proposed depending on the animal category and breed. In addition, the success of in vitro embryo production is directly related to the number and quality of cumulus oocyte complexes harvested by OPU. Control of follicle development has a significant impact on the OPU outcome. This article discusses a number of key points related to the manipulation of ovarian follicular growth to maximize oocyte quality and improve conception rates following TAI and embryo transfer of in vivo-and in vitro-derived embryos in cattle.

Gaze and movement behaviors of association football goalkeepers were compared under two video simulation conditions (i.e., verbal and joystick movement responses) and three in situ conditions (i.e., verbal, simplified body movement, and interceptive response). The results showed that the goalkeepers spent more time fixating on information from the penalty kick taker’s movements than ball location for all perceptual judgment conditions involving limited movement (i.e., verbal responses, joystick movement, and simplified body movement). In contrast, an equivalent amount of time was spent fixating on the penalty taker’s relative motions and the ball location for the in situ interception condition, which required the goalkeepers to attempt to make penalty saves. The data suggest that gaze and movement behaviors function differently, depending on the experimental task constraints selected for empirical investigations. These findings highlight the need for research on perceptual— motor behaviors to be conducted in representative experimental conditions to allow appropriate generalization of conclusions to performance environments.

Dial-a-ride problem (DARP) is an optimization problem which deals with the minimization of the cost of the provided service where the customers are provided a door-to-door service based on their requests. This optimization model presented in earlier studies, is considered in this study. Due to the non-linear nature of the objective function the traditional optimization methods are plagued with the problem of converging to a local minima. To overcome this pitfall we use metaheuristics namely Simulated Annealing (SA), Particle Swarm Optimization (PSO), Genetic Algorithm (GA) and Artificial Immune System (AIS). From the results obtained, we conclude that Artificial Immune System method effectively tackles this optimization problem by providing us with optimal solutions. Este experimento teve como objetivo avaliar a possibilidade da utilização de fêmeas zebuínas de elevada qualidade genética, com problemas de fertilidade, adquiridos após um programa de transferência convencional de embriões, como doadoras de ovócitos em um programa de Ovum Pick Up e Produção In Vitro de embriões. Fêmeas da raça Nelore (n=16) (Bos taurus indicus), com idade média de 11 anos e 7 meses, foram submetidas às mesmas condições de ambiente, alimentação e manejo. Estes animais foram retirados do programa de T.E. por apresentarem problemas de fertilidade (cistos ovarianos, infecções genitais, mucometra e alguns casos de repetições de cio sem causa aparente). Os animais foram divididos em dois grupos conforme o tipo de patologia: GRUPO I (n=08) composto por animais que apresentaram patologias ovarianas (cistos) e GRUPO II (n=08) formado por animais com patologias extra ovarianas, ambos os grupos foram submetidos à aspiração folicular (OPU) utilizando dois tipos de agulhas (21G ; 19G ) e pressões (55 e 70mmHg) divididos em quatro tratamentos (T1 = 55/21G; T2 = 55/19G; T3 = 70/21G e T4 = 70/19G), posteriormente os ovócitos submeteramse a PIV. Foram realizadas 149 aspirações e colhidos 879 ovócitos (5,9 4,88) onde GRAU I = 16%; GRAU II = 43%; GRAU III = 21% e GRAU IV = 20%. As médias de ovócitos colhidos no GRUPO I foram (T1 = 7,06 5,10; T2= 7,00 3,64; T3 = 10,60 6,51 e T4 = 2,78 2,59) para os animais pertencentes ao GRUPO II obteve-se (T1= 4,90 4,47; T2= 2,93 3,05; T3 = 6,11 4,11 e T4 = 2,70 2,00). Para as médiasde ovócitos viáveis os resultados foram os seguintes: (T1 = 4,35 3,01; T2 = 4,88 2,96; T3 = 6,87 3,91 e T4 = 1,89 1,90) para o GRUPO I e (T1 = 2,70 2,22; T2 =1,92 1,98; T3 = 3,47 2,27 e T4 = 2,00 1,05) para o GRUPO II. Neste experimento foram levados à PIV 538 ovócitos dos quais 161 (32,85%) clivaram e 65 (13,26%) resultaram em Mórula ou Blastocistos. A análise estatística dos resultados não demonstrou diferença significativa entre os grupos quanto a colheita ovocitária. Quando analisada somente a pressão de sucção, foi observado que o tipo de agulha está diretamente relacionado a pressão da bomba de vácuo. Os melhores resultados foram obtidos com o tratamento T3 = 70/21G, além disso pode-se observar que as melhores médias de ovócitos colhidos foram as obtidas no grupo de animais que apresentaram cistos ovarianos (GRUPO I). Desta forma, concluímos neste experimento que a utilização da técnica de OPU/PIV é uma alternativa de grande utilidade no prolongamento da vida reprodutiva de animais zebuínos com alterações na fertilidade.

This paper presents a comparative study of the critical pickup and saltation velocities of particles in horizontal pipelines for pneumatic conveying design. A comparative study is performed using different existing correlations in the literature for the determination of the minimum velocity of transport as a function of the particle and pipe diameter, particle density, solid mass flow rate and particle sphericity. Their limitations and difficulties in predicting those critical velocities are analyzed. For the pickup velocity, an experimental study was also carried out in order to support the analysis. Recommendations are presented on the use of such correlations.
