麻疯树(Jatropha curcas L.)属大戟科麻疯树属多年生亚乔木,耐干旱、高温和贫瘠等,具很强的抗逆性,在干热河谷等边际土地上生长良好。其种子富含油脂,是制备生物柴油的理想材料,为重要的能源植物之一。油体(oil body)是种子细胞中重要的细胞器, 脂肪酸以三脂酰甘油(triacylglyeerols,TAG)的形式储存其内,是种子萌发和幼苗生长时所需碳骨架和能量的主要来源。种子萌发为生命萌动并构建成自养个体的过程,是高等植物生长发育中的重要事件。 本论文运用高通量的蛋白质组学研究手段,结合电镜技术和生理学分析,对麻疯树种子油体以及种子萌发过程中蛋白质表达、生理学响应和细胞结构变化进行了研究。 从麻疯树种子胚乳中分离油体,再从油体中提取蛋白,经双向凝胶电泳后,得到油体蛋白质组的二维表达谱,这些蛋白质主要分布在等电点5 ~ 10、分子量12 ~ 66 kDa的范围内;图像分析表明,油体蛋白质组至少有141个蛋白点,其中酸性蛋白74个,碱性蛋白67个,表达丰度较高的多为低分子量碱性蛋白。对其中36个重要蛋白点进行LC-MS/MS质谱分析,得到鉴定的蛋白分别为30个基因的表达产物,主要包括油体重要的结构蛋白油质蛋白(oleosin)和caleosin,麻疯树种子毒蛋白curcin,以及新鉴定得到的另一种可能的麻疯树种子毒蛋白,人体过敏反应蛋白橡胶延伸因子(REF)。还有四个与脂肪酸代谢相关的酶,其中3-羟酰-酰基载体蛋白(ACP)脱水/催化酶和醇酰基转移酶与脂肪酸合成有关,而脂氧合酶和磷脂酶D在脂肪酸降解中发挥作用,显示部分脂肪酸代谢相关的酶在油体储存状态就已附着在油体上,为种子萌发时动员油脂做好了准备。 麻疯树种子胚乳发达,在32℃湿润土壤中很快就会萌动,胚轴伸长露出胚根,长出新根,约4天后形成出土子叶幼苗。种子萌发过程中胚乳主要成分含量测定表明,含水量在前24小时迅速上升,至48小时增加缓慢,此后开始较快上升,可分为三个阶段,呈现“S”型的变化;粗脂肪和粗蛋白在前两个阶段变化不大,进入第三阶段后其含量迅速下降,前者先于后者,分别在萌发后72小时和96小时后开始明显减少,说明被大量降解、转化,供萌发生长利用,其中主要组分亚油酸最为明显。细胞超微结构观察发现,排列整齐充满整个胚乳细胞的油体和嵌合在油体中的蛋白储存泡在种子萌发过程中,随着线粒体、乙醛酸循环体和液泡的出现增多或增大而被逐渐解体、减少或消失;同时,发现脂肪酸主要在乙醛酸循环体、蛋白颗粒主要在液泡中被降解或转化。 蛋白质组学分析表明,麻疯树种子在萌发72小时过程中变化量在两倍以上的差异蛋白点共有141个,所有的差异蛋白均通过LC-MS/MS分析和NCBI蛋白数据库搜索得到鉴定。其中包括多个参与降解储藏油脂的酶,如乙醛酸循环途径中的顺乌头酸酶,异柠檬酸裂解酶和苹果酸脱氢酶等,均从种子萌发48小时开始表达量明显上升;葡糖异生途径中的酶在种子萌发中的积累略晚于乙醛酸循环途径,如烯醇酶,磷酸甘油酸变位酶,磷酸甘油酸激酶,磷酸丙糖异构酶和醛缩酶大多在萌发约60小时后表达量开始上调。分析结果表明,乙醛酸循环途径在种子萌发48小时后被激活,与电镜观察胚乳细胞发现油脂在萌发48小时时开始被动员相一致,因而大规模的油脂动员开始于种子萌发的第三阶段。 同时,蛋白质组学的分析结果也得到了种子胚乳组分变化分析及电镜观察结果的印证。超微细胞结构观察显示种子储藏蛋白降解在萌发第二阶段启动,主要在液泡中进行降解。粗脂肪的含量在72小时时显著降低,而电镜观察显示此时胚乳细胞中出现中央大液泡,出现大量的线粒体和乙醛酸循环体,细胞结构发生重大变化,萌发96小时后仅有少量油体残留于胚乳细胞中,这些都为储藏油脂在麻疯树种子萌发过程中的降解方式提供了重要证据。许多其他的功能蛋白在种子萌发过程中也发生了变化,表明种子萌发过程中不仅发生储藏物质的动员,也发生抗逆反应以及植物形态的构建等众多其他生理生化反应。 本研究首次对麻疯树种子油体进行了蛋白组成分析,并结合电镜技术及生理分析深入探讨了种子储藏物质在萌发中的降解方式,为更好的理解油体结构、木本油料种子的萌发机制和对麻疯树进行品种的改良提供了参考。

This study examined the influence of both optic flow characteristics and intention on postural control responses. Two groups of 10 adults each were exposed to the room's movement either at 0.6 cm/s (low velocity group) or 1.0 cm/s (high velocity group). All the participants stood in the upright stance inside of a moving room and were informed about the room movement only after the fourth trial as they were asked to resist to its influence. Results revealed that participants from both groups were influenced by the imposed visual stimulus in the first trials, but the coupling strength was weaker for the high velocity group. The request to resist the visual influences decreased visual influences oil body sway, but only for the low velocity group. These results indicate that intention might play a role in stimulus influences on body sway but it is stimulus dependent.

Background
Over the past ten years MRSA has become endemic in hospitals and is associated with increased healthcare costs. Critically ill patients are most at risk, in part because of the number of invasive therapies that they require in the intensive care unit (ICU). Washing with 5% tea tree oil (TTO) has been shown to be effective in removing MRSA on the skin. However, to date, no trials have evaluated the potential of TTO body wash to prevent MRSA colonization or infection. In addition, detecting MRSA by usual culture methods is slow. A faster method using a PCR assay has been developed in the laboratory, but requires evaluation in a large number of patients.

Methods/Design
This study protocol describes the design of a multicentre, phase II/III prospective open-label randomized controlled clinical trial to evaluate whether a concentration of 5% TTO is effective in preventing MRSA colonization in comparison with a standard body wash (Johnsons Baby Softwash) in the ICU. In addition we will evaluate the cost-effectiveness of TTO body wash and assess the effectiveness of the PCR assay in detecting MRSA in critically ill patients. On admission to intensive care, swabs from the nose and groin will be taken to screen for MRSA as per current practice. Patients will be randomly assigned to be washed with the standard body wash or TTO body wash. On discharge from the unit, swabs will be taken again to identify whether there is a difference in MRSA colonization between the two groups.

Discussion
If TTO body wash is found to be effective, widespread implementation of such a simple colonization prevention tool has the potential to impact on patient outcomes, healthcare resource use and patient confidence both nationally and internationally.

OBJECTIVES: To determine whether the daily use of 5% tea tree oil (TTO) body wash (Novabac 5% Skin Wash) compared with standard care [Johnson's Baby Softwash (JBS)] had a lower incidence of methicillin-resistant Staphylococcus aureus (MRSA) colonization.

PATIENTS: The study setting was two intensive care units (ICUs; mixed medical, surgical and trauma) in Northern Ireland between October 2007 and July 2009. The study population comprised 391 patients who were randomized to JBS or TTO body wash.

METHODS: This was a Phase 2/3, prospective, open-label, randomized, controlled trial. Trial registration: ISRCTN65190967. The primary outcome was new MRSA colonization during ICU stay. Secondary outcomes included the incidence of MRSA bacteraemia and maximum increase in sequential organ failure assessment score.

RESULTS: A total of 445 patients were randomized to the study. After randomization, 54 patients were withdrawn; 30 because of a positive MRSA screen at study entry, 11 due to lack of consent, 11 were inappropriately randomized and 2 had adverse reactions. Thirty-nine (10%) patients developed new MRSA colonization (JBS n?=?22, 11.2%; TTO body wash n?=?17, 8.7%). The difference in percentage colonized (2.5%, 95% CI -?8.95 to 3.94; P?=?0.50) was not significant. The mean maximum increase in sequential organ failure assessment score was not significant (JBS 1.44, SD 1.92; TTO body wash 1.28, SD 1.79; P?=?0.85) and no study patients developed MRSA bacteraemia.

CONCLUSIONS: Compared with JBS, TTO body wash cannot be recommended as an effective means of reducing MRSA colonization.

This study was carried out to evaluate the effect of the inclusion of different linseed oil levels (0.0, 3.3, 6.6, or 9.9%) in iso-energy diets fed during the period of 21 to 56 days of age on the performance, carcass traits, and body composition of broilers. A total of 1,600 broilers were distributed according to a completely randomized experimental design consisting of four treatments with eight replicates of 40 birds per experimental unit. In the period of 21-42 days of age, weight gain increased, feed and calorie conversion ratios quadratically improved, and feed and metabolizable energy intakes linearly increased as dietary linseed oil level increased. Considering the entire experimental period, dietary linseed oil linearly increased weight gain and feed and energy intakes, and feed and calorie conversion ratios in a quadratic manner. On days 42 and 56, abdominal fat percentage and carcass yield were quadratically influenced by dietary linseed oil. Total body fat content at 56 days of age was quadratically influenced by dietary linseed levels.

Ninety (90) hatchery bred fingerlings of Clarias gariepinus (mean weight: 0.96 ± 0.1g) were randomly placed in 15 plastic baths (25 litres each) at the Research laboratory and were exposed to different concentrations of oil products to determine their effects on the fish, to facilitate inferential deductions that will enhance effective aquatic environmental management. Three (3) replicate basins of 5 experimental treatments (crude oil, petrol oil, kerosene oil, engine oil and control) were used at a concentration of 1.25ml. L-1. The control experiment was devoid of oil treatment. Six (6) fingerlings were placed in each replicate basin, flooded with 20 litres of clean tap water and fed with nutrafin cichilid food, 2 times daily at 3% body weight. The results showed that the feeding behaviour and swimming performances of fish were reduced after 24 hours of the addition of the various oil pollutants. Mortality of fingerlings in the oiled basins increased as the hours of exposure increased (i.e. 24, 48, 72 and 96 hours). Recovery was not immediate in the treated basin while surviving fingerlings in the control basins grew up to post-fingerlings after 90 days (3 months). There were significant differences (P<0.01 and P<0.05) in the effect of crude oil and the petroleum products on the mortality rate of C. gariepinus when exposed to oil pollutants at 1.25ml. We investigated developmental changes in the body compositions and fatty acid (FA) profiles of embryos and preparturition larvae of the quillback rockfish (Sebastes maliger). Comparisons of proximate composition data from early-stage embryos with data from hatched preparturition larvae taken from wild-caught gravid females indicated that embryos gain over one-third their weight in moisture while consuming 20% of their dry tissue mass for energy as they develop into larvae. Lipid contributed 60% of the energy consumed and was depleted more rapidly than protein, indicating a protein-sparing effect. Oil globule volume was strongly correlated with lipid levels, affirming its utility as an indicator of energetic status. FA profiles of early embryos differed significantly from those of hatched larvae. Differences in the relative abundances of FAs between early embryos and hatched larvae indicated different FA depletion rates during embryonic development. We conclude that some metabolically important FAs may prove useful in assessing the condition of embryos and preparturition larvae, particularly 20:4n-6, which cannot be synthesized by many marine fish and which is conserved during embryogenesis. Variability in body composition and energy use among rockfish species should be considered when interpreting any measures of condition.

NOAA’s National Status and Trends Program (NS&T) collected oyster tissue and sediments for quantification of polycyclic aromatic hydrocarbons (PAHs) and petroleum associated metals before and after the landfall of oil from the Deepwater Horizon incident of 2010. These new pre- and post- landfall measurements were put into a historical context by comparing them to data collected in the region over three decades during Mussel Watch monitoring. Overall, the levels of PAHs in both sediment and oysters both pre- and post-landfall were within the range of historically observed values for the Gulf of Mexico. Some specific sites did have elevated PAH levels. While those locations generally correspond to areas in which oil reached coastal areas, it cannot be conclusively stated that the contamination is due to oiling from the Deepwater Horizon incident at these sites due to the survey nature of these sampling efforts. Instead, our data indicate locations along the coast where intensive investigation of hydrocarbon contamination should be undertaken. Post-spill concentrations of oil-related trace metals (V, Hg, Ni) were generally within historically observed ranges for a given site, however, nickel and vanadium were elevated at some sites including areas in Mississippi Sound and Galveston, Terrebonne, Mobile, Pensacola, and Apalachicola Bays. No oyster tissue metal body burden exceeded any of the United States Food and Drug Administration’s (FDA) shellfish permissible action levels for human consumption.

A laboratory trial was conducted to determine the digestible protein and energy value of fish meal, dextrin, fish oil and soybean oil for Thai sharpunti (Puntius gonionotus Bleeker). A reference diet containing 35% protein was formulated in which fish meal was the sole source of protein. Five test diets were formulated using reference diet and individual test ingredients (fish meal, dextrin, fish oil and soybean oil). Each treatment had three replicates with 15 fish per replicate. Fish were fed twice daily at the rate of 5% of their body weight. The result of the study indicated that the dietary protein in both reference and test diets were well digested and the apparent protein digestibility (APD) values of test diets ranged between 82.81 and 85.99%. The APD value of fish meal protein was 88.05%. The apparent digestible energy (ADE) value for the test ingredients ranged between 70.79 and 85.80% with soybean oil having the highest and fish meal the lowest value. The ADE values calculated in terms of Kcal/g of ingredients were 3.68, 3.22, 4.38 and 4.44 Kcal/g for fish meal, dextrin, fish oil and soybean oil respectively.

The effect of feeding 0, 4, 8 and 16% rapeseed oil from 12-42 days of age was studied in broiler chickens on performance, digestibility of nutrients, and development of gastrointestinal tract, protein and energy metabolism. Thirty six female chickens (Ross 208) with initial body weight average 246 g were allocated to the four groups and kept pair-wise in metabolism cages. The chickens were fed similar amounts of metabolisable energy (ME) per day and similar amounts of essential amino acids relative to ME by adjusting with crystalline amino acids. The chickens were subjected to four balance periods each of five days with two 24 h measurements of gas exchange in two open-air-circuit respiration chambers inserted on the second and third day of each period. The addition of rapeseed oil increased the amount of gutfill indicating a reduced rate of passage and causing a hypertrophy of the gastrointestinal tract. There was a positive effect on feed utilisation as well as on digestibility especially of dietary fat together with higher utilisation of protein with addition of rapeseed oil. The partial fat digestibility of rapeseed oil estimated by regression was 91.1% and the partial metabolisability (ME/GE) of the rapeseed oil was estimated to 85% yielding an apparent metabolisable energy value of 34.30 MJ/kg.

Individuals of Mytilus edulis L., collected from the Erme estuary (S.W. England) in 1978, were exposed to low concentrations (7 to 68 μg l-1) of the water-accommodated fraction (WAF) of North Sea crude oil. The pattern of accumulation of petroleum hydrocarbons in the body tissues was affected by the presence of algal food cells, the period of exposure, the hydrocarbon concentration in seawater, the type of body tissue and the nature of the hydrocarbon. Many physiological responses (e.g. rates of oxygen consumption, feeding, excretion, and scope for growth), cellular responses (e.g. lysosomal latency and digestive cell size) and biochemical responses (e.g. specific activities of several enzymes) were significantly altered by short-term (4 wk) and/or long-term (5 mo) exposure to WAF. Stress indices such as scope for growth and lysosomal latency were negatively correlated with tissue aromatic hydrocarbons.

The study assessed the efficacy of fish oil supplementation in counteracting the classic dyslipidemia of the atherogenic lipoprotein phenotype (ALP). In addition, the impact of the common apolipoprotein E (apoE) polymorphism on the fasting and postprandial lipid profile and on responsiveness to the dietary intervention was established. Fifty-five ALP males (aged 34 to 69 years, body mass index 22 to 35 kg/m2, triglyceride [TG] levels 1.5 to 4.0 mmol/L, high density lipoprotein cholesterol [HDL-C] <1.1 mmol/l, and percent low density lipoprotein [LDL]-3 >40% total LDL) completed a randomized placebo-controlled crossover trial of fish oil (3.0 g eicosapentaenoic acid/docosahexaenoic acid per day) and placebo (olive oil) capsules with the 6-week treatment arms separated by a 12-week washout period. In addition to fasting blood samples, at the end of each intervention arm, a postprandial assessment of lipid metabolism was carried out. Fish oil supplementation resulted in a reduction in fasting TG level of 35% (P<0.001), in postprandial TG response of 26% (TG area under the curve, P<0.001), and in percent LDL-3 of 26% (P<0.05). However, no change in HDL-C levels was evident (P=0.752). ANCOVA showed that baseline HDL-C levels were significantly lower in apoE4 carriers (P=0.035). The apoE genotype also had a striking impact on lipid responses to fish oil intervention. Individuals with an apoE2 allele displayed a marked reduction in postprandial incremental TG response (TG incremental area under the curve, P=0.023) and a trend toward an increase in lipoprotein lipase activity relative to non-E2 carriers. In apoE4 individuals, a significant increase in total cholesterol and a trend toward a reduction in HDL-C relative to the common homozygous E3/E3 profile was evident. Our data demonstrate the efficacy of fish oil fatty acids in counteracting the proatherogenic lipid profile of the ALP but also that the apoE genotype influences responsiveness to this dietary treatment.

Our objective was to determine whether the endothelial nitric oxide synthase (eNOS) Glu298Asp polymorphism influences vascular response to raised NEFA enriched with saturated fatty acids (SFA) or long-chain (LC) n-3 polyunsaturated fatty acids (PUFA). Subjects were prospectively recruited for genotype (Glu298, n = 30 and Asp298, n = 29; balanced for age and gender) consumed SFA on two occasions, with and without the substitution of 0.07 g fat/kg body weight with LC n-3 PUFA, and with heparin infusion to elevate NEFA. Endothelial function was measured before and after NEFA elevation (240 min), with blood samples taken every 30 min. Flow-mediated dilation (FMD) decreased following SFA alone and increased following SFA+LC n-3 PUFA. There were 2-fold differences in the change in FMD response to the different fat loads between the Asp298 and Glu298 genotypes (P = 0.002) and between genders (P < 0.02). Sodium nitroprusside-induced reactivity, measured by laser Doppler imaging with iontophoresis, was significantly greater with SFA+LC n-3 PUFA in all female subjects (P < 0.001) but not in males. Elevated NEFA influences both endothelial-dependent and endothelial-independent vasodilation during the postprandial phase. Effects of fat composition appear to be genotype and gender dependent, with the greatest difference in vasodilatory response to the two fat loads seen in the Asp298 females.

Changes in diet carbohydrate amount and type (i.e., starch vs. fiber) and dietary oil supplements can affect ruminant methane emissions. Our objectives were to measure methane emissions, whole-tract digestibility, and energy and nitrogen utilization from growing dairy cattle at 2 body weight (BW) ranges, fed diets containing either high maize silage (MS) or high grass silage (GS), without or with supplemental oil from extruded linseed (ELS). Four Holstein-Friesian heifers aged 13 mo (BW range from start to finish of 382 to 526 kg) were used in experiment 1, whereas 4 lighter heifers aged 12 mo (BW range from start to finish of 292 to 419 kg) were used in experiment 2. Diets were fed as total mixed rations with forage dry matter (DM) containing high MS or high GS and concentrates in proportions (forage:concentrate, DM basis) of either 75:25 (experiment 1) or 60:40 (experiment 2), respectively. Diets were supplemented without or with ELS (Lintec[AU1: Add manufacturer name and location.]; 260 g of oil/ kg of DM) at 6% of ration DM. Each experiment was a 4 × 4 Latin square design with 33-d periods, with measurements during d 29 to 33 while animals were housed in respiration chambers. Heifers fed MS at a heavier BW (experiment 1) emitted 20% less methane per unit of DM intake (yield) compared with GS (21.4 vs. 26.6, respectively). However, when repeated with heifers of a lower BW (experiment 2), methane yield did not differ between the 2 diets (26.6 g/kg of DM intake). Differences in heifer BW had no overall effect on methane emissions, except when expressed as grams per kilogram of digestible organic matter (OMD) intake (32.4 vs. 36.6, heavy vs. light heifers). Heavier heifers fed MS in experiment 1 had a greater DM intake (9.4 kg/d) and lower OMD (755 g/kg), but no difference in N utilization (31% of N intake) compared with heifers fed GS (7.9 kg/d and 799 g/kg, respectively). Tissue energy retention was nearly double for heifers fed MS compared with GS in experiment 1 (15 vs. 8% of energy intake, respectively). Heifers fed MS in experiment 2 had similar DM intake (7.2 kg/d) and retention of energy (5% of intake energy) and N (28% of N intake), compared with GS-fed heifers, but OMD was lower (741 vs. 765 g/kg, respectively). No effect of ELS was noted on any of the variables measured, irrespective of animal BW, and this was likely due to the relatively low amount of supplemental oil provided. Differences in heifer BW did not markedly influence dietary effects on methane emissions. Differences in methane yield were attributable to differences in dietary starch and fiber composition associated with forage type and source.

BACKGROUND: Carriers of the apolipoprotein E ɛ4 (APOE4) allele are lower responders to a docosahexaenoic acid (DHA) supplement than are noncarriers. This effect could be exacerbated in overweight individuals because DHA metabolism changes according to body mass index (BMI; in kg/m²). OBJECTIVES: We evaluated the plasma fatty acid (FA) response to a DHA-rich supplement in APOE4 carriers and noncarriers consuming a high-saturated fat diet (HSF diet) and, in addition, evaluated whether being overweight changed this response. DESIGN: This study was part of the SATgenɛ trial. Forty-one APOE4 carriers and 41 noncarriers were prospectively recruited and consumed an HSF diet for 8-wk followed by 8 wk of consumption of an HSF diet with the addition of DHA and eicosapentaenoic acid (EPA) (HSF + DHA diet; 3.45 g DHA/d and 0.5 g EPA/d). Fasting plasma samples were collected at the end of each intervention diet. Plasma total lipids (TLs) were separated into free FAs, neutral lipids (NLs), and phospholipids by using solid-phase extraction, and FA profiles in each lipid class were quantified by using gas chromatography. RESULTS: Because the plasma FA response to the HSF + DHA diet was correlated with BMI in APOE4 carriers but not in noncarriers, the following 2 groups were formed according to the BMI median: low BMI (<25.5) and high BMI (≥25.5). In response to the HSF + DHA diet, there were significant BMI × genotype interactions for changes in plasma concentrations of arachidonic acid and DHA in phospholipids and TLs and of EPA in NLs and TLs (P ≤ 0.05). APOE4 carriers were lower plasma responders to the DHA supplement than were noncarriers but only in the high-BMI group. CONCLUSIONS: Our findings indicate that apolipoprotein E genotype and BMI may be important variables that determine the plasma long-chain PUFA response to dietary fat manipulation. APOE4 carriers with BMI ≥25.5 may need higher intakes of DHA for cardiovascular or other health benefits than do noncarriers
