Työn tavoitteena oli Imatra Steel Oy Ab:n jatkojalostusosaston lämpökäsittelylaitosten energiatalouden parantaminen. Työssä etsittiin lämpökäsittelylaitoksen energiatalouteen haitallisesti vaikuttavia tekijöitä ja pyrittiin löytämään ratkaisuja energiatalouden parantamiseksi.Työtä varten on kerätty tietoa kirjallisista lähteistä, Imatra Steel Oy Ab:n henkilökunnalta, insinööritoimistoilta, lämpökäsittelyuunien valmistajilta ja muilta asiantuntijoilta, jotka ovat olleet tekemisissä lämpökäsittelylaitosten, lämpökäsittelyuunien tai niihin läheisesti liittyvien energiataloudellisien tekijöiden parissa.Imatra Steel Oy Ab:n lämpökäsittelylaitosten suurimmat energiankuluttajat ovat lämpökäsittelyuunit, jotka ovat rakennettu pääosin vuosina 1965 ja 1971, jolloin energiankulutukseen ja sen aiheuttamiin kustannuksiin ei tarvinnut kiinnittää samalla tavoin huomiota kuin nykyisin. Työssä keskitytäänkin pääasiassa uunien energiatalouteen.Lämpökäsittelylaitosten energiatalouden parantamisessa on ensin keskityttävä primäärienergiana käytettävän maakaasun kulutuksen vähentämiseen, jonka jälkeen voidaan keskittyä uuneista syntyvän hukkalämmön hyödyntämiseen. Jotta lämpökäsittelyuunit saataisiin toimimaan energiataloudellisesti tehokkaasti ja nykyisessä markkinatilanteessa kilpailukykyisesti, pitäisi uuneille laatia oma kehitysohjelma. Uunien energiatalouden seuranta ja kehittäminen edellyttää nykyisten jatkuvatoimisesti mitattavien prosessitietojen ja erillisillä mittauksilla kerättävän tiedon nykyistä tehokkaampaa hyödyntämistä sekä uusien ennen mittaamattomien prosessiparametrien hyödyntämistä. Tulevaisuudessa toteutettava prosessitietojen keräysjärjestelmä antaakin aivan uusia mahdollisuuksia uunien kehittämiseen ja seurantaan.

Tässä työssä tarkastellaan teollisuuden ylijäämälämmön hyödyntämistä kaukolämpöverkoissa liiketoimintamallin näkökulmasta. Työn tilaaja on YIT Teollisuus Oy, joka haluaa osaltaan olla mukana ratkaisemassa ilmastonmuutoksesta ja hiilidioksidipäästöjen vähentämistarpeista aiheutuvia yhteiskunnan kehitystarpeita. Energiatehokkuuden parantaminen on yksi nopeimmista keinoista vähentää päästöjä. Teollisuuden energiatehokkuutta voidaan parantaa ottamalla talteen sähköntuotannossa ja tuotantoprosesseissa syntyvää ylijäämälämpöä. Aikaisempien tutkimusten perusteella tiedetään, että Suomessa syntyy vuosittain noin 4–6 TWh ylijäämälämpöä, joka voitaisiin hyödyntää jo olemassa olevien kaukolämpöverkkojen välityksellä rakennusten lämmittämiseen. Kuitenkin vuonna 2008 teollisuus myi ylijäämälämpöä kaukolämpöverkkoihin yhteensä vain 770 GWh, mikä vastaa noin 2,5 prosenttia kokonaiskaukolämmön tarpeesta. Tämän työn tuloksena syntyi liiketoimintamalli, joka esittelee ne palvelut, jotka YIT tuottaa asiakkailleen tilanteissa, joissa teollisuudessa syntyvää ylijäämälämpöä hyödynnetään kaukolämpöverkoissa. Jotta liiketoimintamalli toimisi käytännössä, on siitä oltava hyötyä kaikille osapuolille. Asiakkaan on siis voitava kattaa palvelusta ja sen rahoituksesta syntyvät kustannukset myydyn ylijäämälämmön tuotolla (teollisuuslaitos) tai säästyneistä energian hankintakustannuksista (kaukolämpöyhtiö). Eniten ylijäämälämmön käytöstä voivat hyötyä kaukolämpöyhtiöt, joiden tuotannosta korkeintaan pieni osa tulee yhteistuotannosta ja joilla uusiutuvien energialähteiden osuus on vähäinen. Lisäksi kaukolämpöverkon koon vuotuisena kulutuksena mitattuna on oltava riittävän suuri ja kaukolämmön hinnan suhteellisen korkea. Myös alueen ennustettu väestönkasvu ja uudet suunnitteilla olevat asuinalueet saattavat parantaa ylijäämälämmön hyödyntämisen houkuttelevuutta. YIT:n näkökulmasta ylijäämälämmön talteenottoprojektit ovat hyvä lisä sen nykyiseen palvelutarjontaan. Myös yhteiskunnallisella tasolla aihe on merkittävä. Vaikka nykytietämyksen mukaan energian käytön tehostaminen ja päästöttömän tuotannon lisääminen ovat molemmat yhtä merkittäviä keinoja ilmastotavoitteiden saavuttamisen kannalta, panostetaan Suomessa tällä hetkellä lähinnä tuotannon tukemiseen. Lähivuosien poliittiset ratkaisut vaikuttavatkin vahvasti siihen, kuinka paljon tulevaisuudessa ylijäämälämpöä hyödynnetään rakennusten lämmittämisessä.

OBJECTIVE: Fibrotic changes are initiated early in acute respiratory distress syndrome. This may involve overproliferation of alveolar type II cells. In an animal model of acute respiratory distress syndrome, we have shown that the administration of an adenoviral vector overexpressing the 70-kd heat shock protein (AdHSP) limited pathophysiological changes. We hypothesized that this improvement may be modulated, in part, by an early AdHSP-induced attenuation of alveolar type II cell proliferation. DESIGN: Laboratory investigation. SETTING: Hadassah-Hebrew University and University of Pennsylvania animal laboratories. SUBJECTS: Sprague-Dawley Rats (250 g). INTERVENTIONS: Lung injury was induced in male Sprague-Dawley rats via cecal ligation and double puncture. At the time of cecal ligation and double puncture, we injected phosphate-buffered saline, AdHSP, or AdGFP (an adenoviral vector expressing the marker green fluorescent protein) into the trachea. Rats then received subcutaneous bromodeoxyuridine. In separate experiments, A549 cells were incubated with medium, AdHSP, or AdGFP. Some cells were also stimulated with tumor necrosis factor-alpha. After 48 hrs, cytosolic and nuclear proteins from rat lungs or cell cultures were isolated. These were subjected to immunoblotting, immunoprecipitation, electrophoretic mobility shift assay, fluorescent immunohistochemistry, and Northern blot analysis. MEASUREMENTS AND MAIN RESULTS: Alveolar type I cells were lost within 48 hrs of inducing acute respiratory distress syndrome. This was accompanied by alveolar type II cell proliferation. Treatment with AdHSP preserved alveolar type I cells and limited alveolar type II cell proliferation. Heat shock protein 70 prevented overexuberant cell division, in part, by inhibiting hyperphosphorylation of the regulatory retinoblastoma protein. This prevented retinoblastoma protein ubiquitination and degradation and, thus, stabilized the interaction of retinoblastoma protein with E2F1, a key cell division transcription factor. CONCLUSIONS: : Heat shock protein 70-induced attenuation of cell proliferation may be a useful strategy for limiting lung injury when treating acute respiratory distress syndrome if consistent in later time points.

Experiments were conducted over two years to quantify the response of faba bean (Vicia faba L.) to heat stress. Potted winter faba bean plants (cv. Wizard) were exposed to temperature treatments (18/10; 22/14; 26/18; 30/22; 34/26°C day/night) for five days during floral development and anthesis. Developmental stages of all flowers were scored prior to stress, plants were grown in exclusion from insect pollinators to prevent pollen movement between flowers, and yield was harvested at an individual pod scale, enabling effects of heat stress to be investigated at a high resolution. Susceptibility to stress differed between floral stages, flowers were most affected during initial green-bud stages. Yield and pollen germination of flowers present before stress showed threshold relationships to stress, with lethal temperatures (t50) ~28°C and ~32°C, while whole plant yield showed a linear negative relationship to stress with high plasticity in yield allocation, such that yield lost at lower nodes was partially compensated at higher nodal positions. Faba bean has many beneficial attributes for sustainable modern cropping systems but these results suggest that yield will be limited by projected climate change, necessitating the development of heat tolerant cultivars, or improved resilience by other mechanisms such as earlier flowering times.

A model is presented for the respiratory heat loss in sheep, considering both the sensible heat lost by convection (C-R) and the latent heat eliminated by evaporation (E-R). A practical method is described for the estimation of the tidal volume as a function of the respiratory rate. Equations for C-R and E-R are developed and the relative importance of both heat transfer mechanisms is discussed. At air temperatures up to 30 degreesC sheep have the least respiratory heat loss at air vapour pressures above 1.6 kPa. At an ambient temperature of 40 degreesC respiratory loss of sensible heat can be nil; for higher temperatures the transfer by convection is negative and thus heat is gained. Convection is a mechanism of minor importance for the respiratory heat transfer in sheep at environmental temperatures above 30 degreesC. These observations show the importance of respiratory latent heat loss for thermoregulation of sheep in hot climates.

Heat stress has negative effects on pregnancy rates of lactating dairy cattle. There are genetic differences in tolerance to heat stress; Bos taurus indicus (B. t. indicus) cattle and embryos are more thermotolerant than Bos taurus taurus (B. t. taurus). In the present study, the effects of sire and sire breed on conception and embryonic/fetal loss rates of lactating Holstein cows during the Brazilian summer were determined. In Experiment 1, cows (n = 302) were AI after estrus detection or at a fixed-time with semen from one Gyr (B. t. indicus) or one Holstein sire (B. t. taurus). Pregnancy was diagnosed 80 days after AI. In Experiment 2, cows (n = 811) were AI with semen from three Gyr and two Holstein sires. Pregnancy was diagnosed at 30-40 and at 60-80 days after AI. Cows diagnosed pregnant at the first examination but non-pregnant at the second were considered as having lost their embryo or fetus. Data were analyzed by logistic regression. The model considered the effect of sire within breed, sire breed, days postpartum, period of lactation, and AI type (AI after estrus versus fixed-time). There was no effect of the AI type, days postpartum or milk production on conception or embryonic loss rates. The use of Gyr bulls increased pregnancy rate when compared to Holstein bulls [9.1% (60/657) versus 5.0% (23/456), respectively, P = 0.008; data from Experiments 1 and 2 combined]. Additionally, in Experiment 2, cows inseminated using semen from sire #4 (Gyr) had lower embryonic loss (10%) when compared with other B. t. indicus (35.3% and 40%) or B. t. taurus sires (18.2% and 38.5%, P = 0.03). In conclusion, the use of B. t. indicus sires may result in higher conception rates in lactating Holstein cows during summer heat stress. Moreover, sire can affect embryonic loss and selection of bulls according to this criterion may result in higher parturition rates in lactating Holstein cows. (c) 2006 Elsevier B.V. The general principles of the mechanisms of heat transfer are well known, but knowledge of the transition between evaporative and non-evaporative heat loss by Holstein cows in field conditions must be improved, especially for low-latitude environments. With this aim 15 Holstein cows managed in open pasture were observed in a tropical region. The latent heat loss from the body surface of the animals was measured by means of a ventilated capsule, while convective heat transfer was estimated by the theory of convection from a horizontal cylinder and by the long-wave radiation exchange based on the Stefan-Boltzmann law. When the air temperature was between 10 and 36 degrees C the sensible heat transfer varied from 160 to -30 W m(-2), while the latent heat loss by cutaneous evaporation increased from 30 to 350 W m(-2). Heat loss by cutaneous evaporation accounted for 20-30% of the total heat loss when air temperatures ranged from 10 to 20 degrees C. At air temperatures > 30 degrees C cutaneous evaporation becomes the main avenue of heat loss, accounting for approximately 85% of the total heat loss, while the rest is lost by respiratory evaporation.

The isotopic (dD, d18O, d13C, and 87Sr/86Sr) and geochemical characteristics of hydrothermal solutions from the Mid-Atlantic Ridge and the material of brucite-carbonate chimneys at the Lost City hydrothermal field at 30°N, MAR, were examined to assay the role of the major factors controlling the genesis of the fluid and hydrothermal chimneys of the Lost City field. The values of dD and d18O in fluid samples indicates that solutions at the Lost City field were produced during the serpentinization of basement ultramafic rocks at temperatures higher than 200°C and at relatively low fluid/rock ratios (<1). The active role of serpentinization processes in the genesis of the Lost City fluid also follows from the results of the electron-microscopic studying of the material of hydrothermal chimneys at this field. The isotopic (d18O, d13C, and 87Sr/86Sr) and geochemical (Sr/Ca and REE) signatures indicate that, before its submarine discharging at the Lost City field, the fluid filtered through already cold altered outer zones of the Atlantis Massif and cooled via conductive heat loss. During this stage, the fluid could partly dissolve previously deposited carbonates in veins cutting serpentinite at the upper levels of the Atlantis Massif and the carbonate cement of sedimentary breccias underlying the hydrothermal chimneys. Because of this, the age of modern hydrothermal activity at the Lost City field can be much younger than 25 ka.

I-2 is an avirulent strain of Newcastle disease virus. During establishment of the I-2 strain master vaccine seed, a series of selection procedures was carried out at 56 degrees C in order to enhance heat resistance. This master seed is used to produce a working seed, which is then employed to produce the vaccine. These two passages are done without further heat selection; however, it is not known how rapidly and to what extent thermostable variants would be lost during further passage. The study was therefore conducted to determine the effect of passage on thermostability of strain I-2. The virus was serially passaged and at various passage levels samples were subjected to heat treatment at 56 degrees C for 120 min. The inactivation rates for infectivity and haemagglutinin (HA) titres were assayed by use of chicken embryonated eggs and HA test, respectively. Thermostability of HA and infectivity of I-2 virus were reduced after 10 and 5 passages, respectively, without heat selection at 56 degrees C. These results suggest that 5 more passages could be carried out between the working seed and vaccine levels without excessive loss of thermostability. This would result in increased vaccine production from a single batch of a working seed.

Hsp90 is a molecular chaperone essential for cell viability in eukaryotes that is associated with the maturation of proteins involved in important cell functions and implicated in the stabilization of the tumor phenotype of various cancers, making this chaperone a notably interesting therapeutic target. Celastrol is a plant-derived pentacyclic triterpenoid compound with potent antioxidant, anti-inflammatory and anticancer activities; however, celastrol's action mode is still elusive. In this work, we investigated the effect of celastrol on the conformational and functional aspects of Hsp90α. Interestingly, celastrol appeared to target Hsp90α directly as the compound induced the oligomerization of the chaperone via the C-terminal domain as demonstrated by experiments using a deletion mutant. The nature of the oligomers was investigated by biophysical tools demonstrating that a two-fold excess of celastrol induced the formation of a decameric Hsp90α bound throughout the C-terminal domain. When bound, celastrol destabilized the C-terminal domain. Surprisingly, standard chaperone functional investigations demonstrated that neither the in vitro chaperone activity of protecting against aggregation nor the ability to bind a TPR co-chaperone, which binds to the C-terminus of Hsp90α, were affected by celastrol. Celastrol interferes with specific biological functions of Hsp90α. Our results suggest a model in which celastrol binds directly to the C-terminal domain of Hsp90α causing oligomerization. However, the ability to protect against protein aggregation (supported by our results) and to bind to TPR co-chaperones are not affected by celastrol. Therefore celastrol may act primarily by inducing specific oligomerization that affects some, but not all, of the functions of Hsp90α. To the best of our knowledge, this study is the first work to use multiple probes to investigate the effect that celastrol has on the stability and oligomerization of Hsp90α and on the binding of this chaperone to Tom70. This work provides a novel mechanism by which celastrol binds Hsp90α.

In this work, all publicly-accessible published findings on Alicyclobacillus acidoterrestris heat resistance in fruit beverages as affected by temperature and pH were compiled. Then, study characteristics (protocols, fruit and variety, °Brix, pH, temperature, heating medium, culture medium, inactivation method, strains, etc.) were extracted from the primary studies, and some of them incorporated to a meta-analysis mixed-effects linear model based on the basic Bigelow equation describing the heat resistance parameters of this bacterium. The model estimated mean D* values (time needed for one log reduction at a temperature of 95 °C and a pH of 3.5) of Alicyclobacillus in beverages of different fruits, two different concentration types, with and without bacteriocins, and with and without clarification. The zT (temperature change needed to cause one log reduction in D-values) estimated by the meta-analysis model were compared to those ('observed' zT values) reported in the primary studies, and in all cases they were within the confidence intervals of the model. The model was capable of predicting the heat resistance parameters of Alicyclobacillus in fruit beverages beyond the types available in the meta-analytical data. It is expected that the compilation of the thermal resistance of Alicyclobacillus in fruit beverages, carried out in this study, will be of utility to food quality managers in the determination or validation of the lethality of their current heat treatment processes.

The association between a toothbrush and a dentifrice is the most used denture cleaning method. The purpose of this study was to evaluate the abrasiveness of specific and non-specific denture cleaning dentifrices on different heat-polymerized acrylic resins. Sixteen specimens (90x30x3mm) of each acrylic resin (QC-20, Lucitone 550, Clássico, Vipi-Cril) were prepared and randomly assigned to 4 groups: 1: control (distilled water), 2: Colgate, 3: Bonyplus and 4: Dentu-Creme. The specimens were subjected to simulated toothbrushing in an automatic brushing machine using 35,600 brush strokes for each specimen. Brushing abrasion run at a 200-g load with the specimens immersed in 2:1 dentifrice/water slurry. Specimens were reconditioned to constant mass and the mass loss (mg) was evaluated. Data were analyzed by 2-way ANOVA and Tukey's test (a=0.05). Analysis of dentifrices' abrasive particles was made by scanning electron microscopy. Colgate produced the greatest mass reduction (42.44 mg, p<0.05), followed by Dentu-Creme (33.60 mg). Bonyplus was the less abrasive (19.91 mg), similar to the control group (19.69 mg) (p>0.05). The mass loss values indicated that QC-20 (33.13 mg) and Lucitone 550 (33.05 mg) resins were less (p<0.05) resistant to abrasion than Clássico (26.04 mg) and Vipi-Cril (23.43 mg). In conclusion, Colgate produced the greatest abrasion. Specific dentifrices for dentures tend to cause less damage to acrylic resins.

The purpose of this study was to evaluate the flexural strength of a direct composite, for indirect application, that received heat treatment, with or without investment. One indirect composite was used for comparison. For determination of the heat treatment temperature, thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC) were performed, considering the initial weight loss temperature and glass transition temperature (Tg). Then, after photoactivation (600 mW/cm² - 40 s), the specimens (10 x 2 x 2 mm) were heat-treated following these conditions: 170ºC for 5, 10 or 15 min, embedded or not embedded in investment. Flexural strength was assessed as a means to evaluate the influence of different heat treatment periods and investment embedding on mechanical properties. The data were analyzed by ANOVA and Tukey's test (α = 0.05). TGA showed an initial weight loss temperature of 180ºC and DSC showed a Tg value of 157°C. Heat treatment was conducted in an oven (Flli Manfredi, Italy), after 37°C storage for 48 h. Flexural strength was evaluated after 120 h at 37°C storage. The results showed that different periods and investment embedding presented similar statistical values. Nevertheless, the direct composite resin with treatments presented higher values (178.7 MPa) compared to the indirect composite resin (146.0 MPa) and the same direct composite submitted to photoactivation only (151.7 MPa). Within the limitations of this study, it could be concluded that the heat treatment increased the flexural strength of the direct composite studied, leading to higher mechanical strength compared to the indirect composite.
