This thesis describes, for the first time, the forces involved in the Nordic hamstring exercise, its reliability and the biomechanical effects of extra loading during the movement. The results provide practitioners with valuable information to enhance hamstring injury prevention and rehabilitation programs.

Pumpkin plants (Cucurbita maxima and C. moschata) with pumpkin yellow leaf curl (PYLC) disease were observed at production fields in Queensland, Western Australia and the Northern Territory. Diseased samples were positive for a phytoplasma indistinguishable from Candidatus Phytoplasma australiense, the phytoplasma associated with papaya dieback and strawberry lethal yellows. This is the first time Candidatus Phytoplasma australiense has been detected in pumpkin.

Gemini viral assembly and transport of viral DNA into nucleus for replication, ssentially involve DNA-coat protein interactions. The kinetics of interaction of Cotton LeafCtirl Kokhran Virus-Dabawali recombinant coat protein (rCP) with DNA was studied by electrophoretic mobility shift assay (EMSA) and Surface plasmon resonance (SPR). The rCP interacted with ssDNA with a K-A, of 2.6 +/- 0.29 x 10(8) M-1 in a sequence non-specific manner. The CP has a conserved C2H2 type zinc finger motif composed of residues C68, C72, H81 and H85. Mutation of these residues to alanine resulted in reduced binding to DNA probes. The H85A mutant rCP showed the least binding with approximately 756 fold loss in the association rate and a three order magnitude decrease in the binding affinity as compared to rCP. The CP-DNA interactions via the zinc finger motif could play a Crucial role ill Virus assembly and in nuclear transport. (C) 2009 Elsevier Inc.

Cotton leaf curl disease (CLCuD) is a major biosecurity threat to the Australian cotton industry. This proposal seeks cross-industry investment from the cotton (CRDC) and horticulture (HAL) industries to address the threat of exotic whitefly-transmitted viruses. Testing of silverleaf whitefly, the vector of CLCuD, could provide an alternative, cheaper strategy for early warning disease surveillance compared to surveys for disease symptoms. Control of whitefly-transmitted viruses in Australia and overseas will be reviewed to produce an integrated management package for their control in Australia. This will also involve a workshop with key stakeholders and selected overseas participants, to develop a working party to help formulate this package.

Background: Cotton leaf curl Kokhran Virus-Dabawali (CLCuKV-Dab) is a monopartite begomovirus encoding two proteins V1 and V2 in the virion sense and four proteins Cl, C2, C3 and C4 in the complementary sense. The C4 protein of monopartite begomoviruses has been implicated to play a role in symptom determination and virus movement. The present work aims at the biochemical characterization of this protein. Methods: The C4 protein of CLCuKV-Dab was purified in fusion with GST and tested for the ability to hydrolyze ATP and other phosphate containing compounds. ATPase activity was assayed by using radiolabeled gamma-32P]-ATP and separating the product of reaction by thin layer chromatography. The hydrolysis of other compounds was monitored by the formation of a blue colored phosphomolybdate complex which was estimated by measuring the absorbance at 655 nm. Results: The purified GST-C4 protein exhibited metal ion dependent ATPase and inorganic pyrophosphatase activities. Deletion of a sequence resembling the catalytic motif present in phosphotyrosine phosphatases resulted in 70% reduction in both the activities. Mutational analysis suggested arginine 13 to be catalytically important for the ATPase and cysteine 8 for the pyrophosphatase activity of GST-C4. Interaction of V2 with GST-C4 resulted in an increase in both the enzymatic activities of GST-C4. Conclusions: The residues important for the enzymatic activities of GST-C4 are present in a motif different from the classical Walker motifs and the non-classical ATP binding motifs reported so far. General significance: The C4 protein of CLCuKV-Dab, a putative natively unfolded protein, exhibits enzymatic activities.

The strength of the North Atlantic Current (NAC) (based on sea-surface elevation sloped derived from altimeter data) is correlated with westerly winds (based on North Atlantic Oscillation [NAO] Index data over a nine year period [1992-2002] with 108 monthly values). The data time window includes the major change in climate forcing over the last 100 years (1995 to 1996). It is shown that the NAO Index can be used for early earning of system failure for the NAC. The correlation response or early warning time scale for western Europe and south England is six months. The decay scale for the NAC and Subtropical Gyre circulation is estimated as three years. Longer period altimeter elevation/circulation changes are discussed. The sea-surface temperature (SST) response of the North Sea to negative and positive NAO conditions is examined. The overall temperature response for the central North Sea to NAO index forcing, reflecting wind induced inflow, shelf circulation and local climate forcing, is similar to 5 months. In years with strong North Atlantic winter wind induced inflow, under marked NAO positive conditions, mean temperatures ( similar to 10.5 degree C) are about 1 degree C warmer than under negative conditions. In 1996 under extreme negative winter NAO conditions, the North Sea circulation stopped, conditions near the Dogger Bank became more continentally influenced and the winter (March) temperature fell to 3.1 degree C whereas in 1995 under NAO positive winter conditions the minimum temperature was 6.4 degree C (February). Seasonal advance of North Atlantic and North Sea temperature is derived in relation to temperature change. Temperature change and monthly NAO Index are discussed with respect to phytoplankton blooms, chlorophyll-a measurements, ocean colour data and the anomalous north-eastern Atlantic 2002 spring/summer bloom SeaWiFS chlorophyll concentrations.

A lens antenna consisting of a double-sided 3 x 3 array of curl antennas that can convert an incident LH/RHCP signal into a transmitted LH/RHCP signal is shown to have an additional property that allows the angle of incidence of an incoming linear or circularly polarised signal to be determined. Here the angle of arrival information is obtained via a simple boresight power measurement made at the lens receive side. It is shown that an unambiguous angle of arrival information for signals arriving at up to +/- 45 degrees can be obtained.

Objetivo: evaluar la reproducibilidad prueba-reprueba y el nivel de acuerdo entre mediciones de las pruebas 30 seg (30-s) Arm Curl y 30-s Chair Stand en una muestra de adultos mayores de Bucaramanga. Materiales y métodos: se realizó un estudio de evaluación de tecnologías diagnósticas. Ambas pruebas fueron administradas en dos oportunidades por el mismo evaluador a 111 adultos mayores de 59 años (70,4 ± 7,3), con un intervalo de 4 a 8 días. En el análisis se determinó la reproducibilidad prueba-reprueba mediante el coeficiente de correlación intraclase, CCI= 2,1 con sus respectivos intervalos de confianza de 95% (IC 95%). El nivel de acuerdo se estableció mediante el método de Bland y Altman. Resultados: la reproducibilidad prueba-reprueba para el 30-s Arm Curl test fue muy buena, con un CCI= 0,88 y para el 30-s Chair Stand test fue buena,con un CCI= 0,78. Así, el acuerdo fue muy bueno para las dos pruebas de resistencia muscular. Los límites de acuerdo de 95% estuvieron entre -3,8 y 3,2 repeticiones para la prueba 30-s ChairStand y entre -3,1 y 2,8 repeticiones para la prueba 30-s Arm Curl. Conclusión: las pruebas 30-s Arm Curl y 30-s Chair Stand tienen buena reproducibilidad y nivel de acuerdo para evaluar resistencia muscular en adultos mayores funcionalmente independientes.

Steep orography can cause noisy solutions and instability in models of the atmosphere. A new technique for modelling flow over orography is introduced which guarantees curl free gradients on arbitrary grids, implying that the pressure gradient term is not a spurious source of vorticity. This mimetic property leads to better hydrostatic balance and better energy conservation on test cases using terrain following grids. Curl-free gradients are achieved by using the co-variant components of velocity over orography rather than the usual horizontal and vertical components. In addition, gravity and acoustic waves are treated implicitly without the need for mean and perturbation variables or a hydrostatic reference profile. This enables a straightforward description of the implicit treatment of gravity waves. Results are presented of a resting atmosphere over orography and the curl-free pressure gradient formulation is advantageous. Results of gravity waves over orography are insensitive to the placement of terrain-following layers. The model with implicit gravity waves is stable in strongly stratified conditions, with N∆t up to at least 10 (where N is the Brunt-V ̈ais ̈al ̈a frequency). A warm bubble rising over orography is simulated and the curl free pressure gradient formulation gives much more accurate results for this test case than a model without this mimetic property.
