On stirring an equimolar mixture of 4-oxo-4H-chromene-3-carbaldehyde, ninhydrin and cyclohexyl isocyanide in CH(2)Cl(2)-MeOH (7: 1) at room temperature produces 3-cyclohexylimino-1-(2-hydroxy-1,3-dioxo-2,3-dihydro-1H-inden-2-yl)-1,3-dihydro-9H-furo[3,4-b]chromen-9-one which on hydrolysis produces 1-(2-hydroxy-1,3-dioxo-2,3-dihydro-1H-inden-2-yl)-1H-furo[3,4-b]chromene-3,9-dione. The structure of the latter compound was confirmed by single crystal X-ray diffraction

The curculionid beetle Naupactus bipes (Germar, 1824) (Coleoptera: Curculionidae: Brachycerinae) has shown feeding preference for leaves of Piper gaudichaudianum, demonstrating an unexpected specificity for an insect considered to be a generalist. The leaves of P. gaudichaudianum contain the prenylated chromenes gaudichaudianic acid (4, major compound) and its methyl ester (5) in addition to a chromene (3) lacking one prenyl residue. In addition to 4, roots contain the chromone methyl ester (1) and methyl taboganate (2, major compound). Feeding on roots, larvae of N. bipes sequester exclusively the root-specific compounds 1 and 2. Adult beetles sequester the leaf-specific chromenes 3 and 4, but were found to also contain compounds 1 and 2 that are absent in leaves. Therefore, it is suggested that 1 and 2 are sequestered by larvae and can be found in the body of adult insects after long-term storage. In addition, 3 and 4, the major compounds in leaves were found to be associated with the eggs.

Metabolic studies involving the incorporation of [1-13C]-D- glucose into intact leaves of Piper aduncum (Piperaceae) have indicated that both the mevalonate (MVA) and the pyruvate-triose (MEP) non-mevalonate pathways are implicated in the biosynthesis of isoprene moieties present in methyl 2,2-dimethyl-2H-1-chromene-6-carboxylate (1) and methyl 2,2-dimethyl-8- (3′-methyl-2′-butenyl)-2H-1-chromene-6-carboxylate (2). The pattern of incorporation of label from [1-13C]-D-glucose into these chromenes was determined by quantitative 13C NMR spectroscopy. The results confirmed that biosynthetic compartment of 1 and 2 could either be the plastid and/ or the cytosol or, possibly, an additional compartment such as the plastid inter-membrane space. ©2007 Sociedade Brasileira de Química.

The aim of the study was to investigate the anti-trypanocidal activities of natural chromene and chromene derivatives. Five chromenes were isolated from Piper gaudichaudianum and P. aduncum, and a further seven derivatives were prepared using standard reduction, methylation and acetylation procedures. These compounds were assayed in vitro against epimastigote forms of Trypanosoma cruzi, the causative agent of Chagas disease. The results showed that the most of the compounds, especially those possessing electron-donating groups as substituents on the aromatic ring, showed potent trypanocidal activity. The most active compound, [(2S)-methyl-2-methyl-8-(3″-methylbut-2″-enyl)-2- (4′-methylpent-3′-enyl)-2H-chromene-6-carboxylate], was almost four times more potent than benznidazole (the positive control) and showed an IC 50 of 2.82 μM. The results reveal that chromenes exhibit significant anti-trypanocidal activities and indicate that this class of natural product should be considered further in the development of new and more potent drugs for use in the treatment of Chagas disease. © 2008 Pharmaceutical Society of Japan.

Chemical analysis carried out in leaves of 18 specimens of Piper aduncum L. (Piperaceae) occurring at Ripasa Reserve, Araraquara, SP, Brazil indicated two distinct populations when investigated over a period of 14 months (January 2000 to February 2001) and then submitted to cluster analysis. The two groups were characterized by accumulation of prenylated benzoic acids, chromenes and dihydrochalcone, respectively. A total of seven compounds were identified by HPLC analysis and compared with standards including two prenylated benzoic acid [aduncumene (1) and 3-(3′-7′-dimethyl-2′-6′-octadienyl)- 4-methoxy-benzoic acid (5)], four chromenes [methyl 2,2-dimethyl-8-(3′- methyl-2′-butenyl)-2H-1-chromene-6-carboxylate (4), methyl 2,2-dimethyl-2H-1-chromene-6-carboxylate (2b), methyl 8-hydroxy-2,2-dimethyl-2H- 1-chromene-6-carboxylate (3) and 2,2-dimethyl-2H-1-chromene-6-carboxylic acid (2a)] and one dihydrochalcone [2′,6′-dihydroxy-4′-methoxy- dihydrochalcone (6)]. © 2009 Academic Journals.

The reaction of various oxygen-containing benzo-fused cycloalkenes were studied with the hypervalent iodine reagent hydroxy(tosyloxy) iodo]benzene [PhI(OH)OTs, HTIB]. 2H-Chromene and 4-methyl-2H-chromene resulted in substituted 4H-chromene and cis-3,4-dialkoxy-4-methyl-3,4-dihydro-2H-chromenes, respectively. Ring contraction to chromanes and benzofurans was observed for dihydrobenzoxepines and 2,2-dimethyl-2H-chromenes, respectively.

Esta dissertação está dividida em duas partes. Na primeira parte reportam-se métodos de síntese de (E)-3-estirilflavonas e (E)/(Z)-2-aril- 4-cloro-3-estiril-2H-cromenos e estudos de ciclização das (E)-3- estirilflavonas em 5-arilbenzo[c]xantonas. Na segunda parte desenvolveram-se novas rotas de síntese de (E)-2-aril-3-estiril-4- quinolonas e posterior transformação em 5-fenil-12- metilbenzo[c]acridonas e 2,4-diarilfuro[3,2-c]quinolinas. Nesta parte estudou-se também a transformação de 2-aril-4-cloro-1,2-dihidroquinolina- 1,3-dicarbaldeídos em (E)/(Z)-2-aril-4-cloro-3-estiril-1,2- di-hidroquinolina-1-carbaldeídos. A síntese de novos derivados de (E)-3-estirilflavonas, abordada na primeira parte desta dissertação, envolveu estudos de otimização da reação de bromação seguida de ciclização de 3-aril-1-(2- hidroxiaril)propano-1,3-dionas/3-aril-3-hidroxi-1-(2-hidroxiaril)prop-2- en-1-onas em 3-bromoflavonas e o desenvolvimento de uma nova rota de síntese de 3-metilflavonas. As 3-metilflavonas foram sujeitas a bromação e seguidamente transformadas em sais de fosfónio antes de serem utilizadas na síntese de (E)-3-estirilflavonas via reação de Wittig. As 3-bromoflavonas foram também usadas na síntese de (E)-3- estirilflavonas via reação de Heck. A síntese de novos derivados de (E)/(Z)-2-aril-4-cloro-3-estiril-2H-cromenos, via reação de Wittig, envolveu a síntese e formilação de flavanonas. A última transformação reportada na primeira parte desta dissertação é a síntese de 5-arilbenzo[c]xantonas por reação de eletrociclização seguida de oxidação de (E)-3-estirilflavonas. Na segunda parte desta dissertação são estudadas duas vias de síntese de 2-aril-1-metil-4-quinolonas. A primeira via de síntese envolve a síntese de N-(2-acetilfenil)benzamidas, sua ciclização em 4-quinolonas seguida de metilação destas. A segunda via envolve a metilação e ciclização de N-(2-acetilfenil)benzamidas obtendo-se, num só passo, as 2-aril-1-metil-4-quinolonas. Posterior iodação das 2-aril-1-metil-4- quinolonas e subsequente reação de Heck das 2-aril-3-iodo-1-metil-4- quinolonas com estirenos comerciais possibilitaram a síntese de (E)-2- aril-3-estiril-1-metil-4-quinolonas. Estudos de eletrociclização seguidos de oxidação das (E)-2-aril-3-estiril-1-metil-4-quinolonas utilizando uma lâmpada de UV de mercúrio de alta pressão possibilitou a síntese de 5- fenil-12-metilbenzo[c]acridonas, enquanto que o aquecimento em refluxo de 1,2,4-triclorobenzeno deu origem a 2,4-diarilfuro[3,2- c]quinolinas. Nesta segunda parte aborda-se também a síntese de 2-aril-4-cloro-1,2- di-hidroquinolina-1,3-dicarbaldeídos, a partir da formilação de 2-aril- 2,3-di-hidro-4-quinolonas e a sua transformação em (E)/(Z)-2-aril-4- cloro-3-estiril-1,2-di-hidroquinolina-1-carbaldeídos por reação de Wittig. Todos os compostos sintetizados foram caracterizados por espectroscopia de ressonância magnética nuclear de protão e carbono 13C, espectros bidimensionais de correlação heteronuclear (HMBC e HSQC) e, nalguns casos espectros de efeito nuclear Overhauser (NOESY). Os novos produtos foram igualmente caracterizados por espectrometria de massa e sempre que possível análise elementar ou espectrometria de massa de alta resolução.

The ability to carry out geranylations on aromatic substrates using enzymatic extracts from the leaves of Piper crassinervium (Piperaceae) was evaluated. A literature analysis pointed out its importance as a source of prenylated bioactive molecules. The screening performed on aromatic acceptors (benzoic acids, phenols and phenylpropanoids) including geranyl diphosphate as prenyl donor, showed the biotransformation of the 3,4-dihydroxybenzoic acid by the crude extract, and the p-hydroxybenzoic acid by both the microsomal fraction and the crude extract, after treating leaves with glucose. The analysis of the products allowed the identification of C- and O-geranylated derivatives, and the protease (subtilisin and pepsin) inhibition performed on the O-geranylated compounds showed weak inhibition. Electrophoretic profiles indicated the presence of bands/spots among 56-58 kDa and pI 6-7, which are compatible with prenyltransferases. These findings show that P. crassinervium could be considered as a source of extracts with geranyltransferase activity to perform biotransformations on aromatic substrates. (C) 2010 Elsevier Ltd. The bioactivity-guided fractionation of the crude extracts from leaves of Brazilian species Piper aduncum and Piper hostmannianum by means of bioautography using the fungi Cladosporium cladosporioides and C. sphaerospermum afforded prenylated methyl benzoate, chromenes, and dihydrobenzopyran derivatives as antifungal compounds. The isolation and structural elucidation of a new compound methyl 4-hydroxy-3-(2`-hydroperoxy-3`-methyl-3`-butenyl) benzoate were performed by application of chromatographic techniques and spectroscopic analyses. The methanol extract from aerial parts of the Peperomia blanda (Piperaceae) yielded two C-glycosyl-flavones. Their structures were elucidated on the basis of extensive spectroscopic analysis, including 1D and 2D NMR, chemical transformation and comparison with the related known compounds. The structure of the new flavonoids were established as 4`-methoxy-vitexin 7-O-beta-D-xylopyranoside (1) (7-O-beta-D-xylopyranosyl-8-C-beta-D-glucopyranosyl-4`-methoxy-apigenin) and vicenin-2 (2). The antioxidant activity of both compounds was investigated using the DPPH assay. Both compounds showed only modest activity, with IC50 values of 357.2 mu M for 1, and 90.5 mu M for 2. Five tetrahydrofuran lignans and two known flavones were isolated from the aerial parts of Peperomia blanda. The structures of the isolated lignans were elucidated by interpretation of their spectroscopic data, including by gHMQC and gHMBC. The relative and absolute configurations of the isolates were determined from NOESY interactions and optical properties, respectively. Four of the lignans were diastercomeric whilst one was of mixed biosynthetic origin. All but one of the lignans exhibited high in vitro trypanocidal activity when assayed against epimastigotes of Trypanosoma cruzi strain Y. (C) 2007 Elsevier Ltd. All rights reserved.

Gaudichaudianic acid, a prenylated chromene isolated from Piper gaudichaudianum, has been described as a potent trypanocidal compound against the Y-strain of Trypanosoma cruzi. We herein describe its isolation as a racemic mixture followed by enantiomeric resolution using chiral HPLC and determination of the absolute configuration of the enantiomers as (+)-S and (-)-R by means of a combination of electronic and vibrational circular dichroism using density functional theory calculations. Investigation of the EtOAc extract of the roots, stems, and leaves from both adult specimens and seedlings of P. gaudichaudianum revealed that gaudichaudianic acid is biosynthesized as a racemic mixture from the seedling stage onward. Moreover, gaudichaudianic acid was found exclusively in the roots of seedlings, while it is present in all organs of the adult plant. Trypanocidal assays indicated that the (+)-enantiomer was more active than its antipode. Interestingly, mixtures of enantiomers stowed a synergistic effect, with the racemic mixture being the most active.
