Os fungicidas sistêmicos do grupo dos benzimidazóis são amplamente utilizados no controle de muitas doenças de plantas, tanto no solo como na parte aérea, e isso tem sido causa de contaminação ambiental. A ação de alguns microorganismos contribui para degradação e perda da atividade biológica desses fungicidas, podendo reduzir, dessa forma, o risco de impactos negativos. O objetivo deste trabalho foi selecionar fungos com potencial para degradar benomil e seu produto de hidrólise, carbendazim, e quantificar o potencial de degradação. Fungos foram isolados de três diferentes solos da região de agricultura irrigada do município de Guaíra, SP, com histórico de aplicação intensiva de fungicidas benzimidazóis. Dentre os fungos isolados, Alternaria alternata foi a linhagem menos afetada pelo aumento na concentração de benomil no meio de cultura. Na concentração máxima (100 mig mL-1) a inibição da taxa de crescimento de A. alternata foi 22%, enquanto nas demais linhagens foi superior a 45%. Em meio de cultura líquido suplementado com carbendazim, a taxa de crescimento em biomassa de A. alternata foi 43% maior do que a no meio sem carbendazim, o que indica o consumo do fungicida como fonte de carbono. A. alternata degradou rapidamente o fungicida, chegando a 66,21% de desaparecimento do produto em dois dias. A meia-vida de carbendazim nessas condições foi de 1,16 dias.

The carcinogenic potential of carbendazim and its metabolites was analyzed using statistical treatment of electronic parameters obtained from DFT/ 6-311++G(d,p) and AM1 calculations. The carcinogen-DNA interaction is described in the framework of the theory of unsynchronized resonance of covalent bond as a process of electron transfer involving the HOMO and LUMO frontier orbitals. Through a Principal Component Analysis (PCA) of the electron affinity, carcinogen-DNA interaction energy, electrostatic attraction and cell membrane permeability (dipole moment m and partition coefficient LogP) evidence was obtained showing carbendazim displays carcinogenic activity. For the metabolites of carbendazim, no evidence was found in the literature of their carcinogenic activities. However, the electronic parameters for these metabolites exhibited similarity to known carcinogens, thereby showing the importance of the results obtained in this study for a policy based on the precautionary principle.

Soybean target leaf spot, caused by the fungus Corynespora cassiicola, is controlled especially by leaf application of fungicides. In the last seasons, in the central-west region of Brazil, the disease chemical control efficiency has been low. This led to the hypothesis that the control failure could be due to the reduction or loss of the fungus sensitivity to fungicides. To clarify this fact, in vitro experiments were conducted to determine mycelial sensitivity of five C. cassiicola isolates to fungicides. Mycelial growth was assessed based on the growth of the mycelium on the culture medium, in Petri dishes. The medium potato-dextrose-agar was supplemented with the concentrations 0; 0.01; 0.1; 1; 10; 20 and 40 mg/L of the active ingredients carbendazim, cyproconazole, epoxiconazole, flutriafol and tebuconazole. The experiment was conducted and repeated twice in a controlled environment, temperature of 25±2ºC and photoperiod of 12 hours. Data on the percentage of mycelial inhibition were subjected to logarithmic regression analysis and the concentration that inhibits 50% of the mycelial growth (IC50) was calculated. Loss of sensitivity to carbendazim was observed for three fungal isolates, IC50 > 40 mg/L. Considering all five isolates, the IC50 for tebuconazole ranged from 1.89 to 2.80 mg/L, for epoxiconazol from 2.25 to 2.91, for cyproconazole from 9.21 to 20.32 mg/L, and for flutriafol from 0.77 to 2.18 mg/L. In the absence of information on the reference IC50 determined for wild isolates, the lowest values generated in our study can be used as standard to monitor the fungus sensitivity.

Field populations of earthworms have shown a varied response in mortality to the fungicide carbendazim, the toxic reference substance used in agrochemical field trials. The aim of this study was to determine the influence of soil conditions as a potential cause of this variation. Laboratory acute toxicity tests were conducted using a range of artificial soils with varying soil components (organic matter, clay, pH and moisture). Batch adsorption/desorption studies were run to determine the influence of the soil properties on carbendazim behaviour. Adsorption was shown to be correlated with organic matter content and pH and this in turn could be linked to Eisenia fetida mortality, with lower mortality occurring with increased adsorption. Overall while E.fetida mortality did vary significantly between several of the soils the calculated LC50 values in the different soils did not cover a wide range (6.04-16.00 mg kg(-1)), showing that under these laboratory conditions soil components did not greatly influence carbendazim toxicity to E.fietida. Carbendazim-amended soil was placed above or below unamended soil. Control tests comprised two layers of unamended soil. Allolobophora chlorotica earthworms were added to either the upper or the unamended soil. After 72 h vertical distributions of earthworms were compared between control and carbendazim-amended experiments. Earthworm distributions in the carbendazim-amended test containers differed significantly from the ‘normal’ distribution observed in the control tests. In the majority of the experiments, earthworms significantly altered their burrowing behaviour to avoid carbendazim. However, when earthworms were added to an upper layer of carbendazim-amended soil they remained in this layer. This non-avoidance is attributed to (1) the earthworms’ inability to sense the lower layer of unamended soil and (2) the toxic effect of carbendazim inhibiting burrowing. Earthworms modified their burrowing behaviour in response to carbendazim in the soil. This may explain anomalous results observed in pesticide field trials when carbendazim is used as a control substance.

Carbendazim is highly toxic to earthworms and is used as a standard control substance when running field-based trials of pesticides, but results using carbendazim are highly variable. In the present study, impacts of timing of rainfall events following carbendazim application on earthworms were investigated. Lumbricus terrestris were maintained in soil columns to which carbendazim and then deionized water (a rainfall substitute) were applied. Carbendazim was applied at 4 kg/ha, the rate recommended in pesticide field trials. Three rainfall regimes were investigated: initial and delayed heavy rainfall 24 h and 6 d after carbendazim application, and frequent rainfall every 48 h. Earthworm mortality and movement of carbendazim through the soil was assessed 14 d after carbendazim application. No detectable movement of carbendazim occurred through the soil in any of the treatments or controls. Mortality in the initial heavy and frequent rainfall was significantly higher (approximately 55%) than in the delayed rainfall treatment (approximately 25%). This was due to reduced bioavailability of carbendazim in the latter treatment due to a prolonged period of sorption of carbendazim to soil particles before rainfall events. The impact of carbendazim application on earthworm surface activity was assessed using video cameras. Carbendazim applications significantly reduced surface activity due to avoidance behavior of the earthworms. Surface activity reductions were least in the delayed rainfall treatment due to the reduced bioavailability of the carbendazim. The nature of rainfall events' impacts on the response of earthworms to carbendazim applications, and details of rainfall events preceding and following applications during field trials should be made at a higher level of resolution than is currently practiced according to standard International Organization for Standardization protocols.

Uma vez que as técnicas cromatográficas permitem a separação, quantificação e identificação dos compostos organicos parentais e dos produtos de degradação e, considerando-se que a maioria dos fungicidas benzimidazois absorvem fortemente a luz ultra-violeta, testou-se o método de Cromatografia Liquida de Alta Eficiência (CLAE), para quantificar a biodegradabilidade do fungicida carbendazim. Neste experimento foi utilizada uma linhagem de Alternaria alternata isolada de solos agrícolas suplementados com benomil. Os frascos com os microorganismos crescidos foram repicados para frascos contendo meios de cultura batata-dextrose (50%) + carbendazim (100mg/ml). Estes foram incubados sob agitacao a 28oC mais ou menos 1oC por períodos de 2,4,7,15 e 30 dias. Apos extração e purificação das amostras procedeu-se a analise cromatográfica. Para tanto utilizou-se uma coluna de troca cationica nas seguintes condições: temperatura da coluna: 40oC, fase móvel: fosfato de amônio 0,0125M com fluxo de 0,2ml por minuto e detector de absorbância operando a 280nm. Sob estas condições, o tempo de retenção de carbendazim e 2 amino-benzimidazol foi de aproximadamente 4,5 e 6,3 minutos, respectivamente. A quantificação foi feita utilizando-se regressão linear de curvas de calibração obtidas em soluções padrões de carbendazim versus a resposta (altura ou área do pico), obtida no cromatograma. A confirmação da identidade do pico com alto grau de confiança foi possível pela comparação do tempo de retenção e o espectro de absorbância em ultra-violeta do carbendazim. Este espectro, em diferentes condições de cultivo de A alternata, indicou índices de similaridade da molécula variando de 0,99 a 1 entre as diferentes amostras. Por este método foram obtidas recuperações acima de 70% da concentração inicial de carbendazim. A degradação de carbendazim foi de 66,4% aos dois dias de incubação.

Este estudo verificou a eficiência da aplicação de diferentes moléculas em reduzir o comprimento relativo da lesão (CRL) da queima das bainhas em arroz. Plantas dos cultivares BR-Irga 409 e Labelle foram cultivadas em solução nutritiva e inoculadas com Rhizoctonia solani, no estádio de máximo perfilhamento. Às 24 horas antes da inoculação, as bainhas das plantas foram pulverizadas com soluções de silicato de potássio (SP), silicato de potássio + fósforo (SP+F), Acibenzolar-S-Metil (ASM), fungicida Carbendazim, quitosana desacetilada (QD), etileno (ET) e fosfito de potássio (FP). Plantas cujas bainhas foram pulverizadas com água destilada serviram como testemunhas. O efeito das moléculas contidas nesses produtos no crescimento micelial de R. solani foi testado in vitro. Para BR-Irga 409, o CRL foi menor com a aplicação do FP, em relação aos demais tratamentos, exceto o Carbendazim. A aplicação do Carbendazim reduziu em 86,1% o CRL, em relação à testemunha. O CRL foi significativamente menor no cultivar BR-Irga 409 do que no 'Labelle', com aplicação do FP. O crescimento micelial de R. solani foi reduzido apenas pelo FP e Carbendazim, em comparação com os demais tratamentos. Não houve diferença significativa entre os tratamentos testemunha, SP e SP+F para a concentração de Si nas bainhas das plantas dos dois cultivares.

Objectives Exposure assessment to a single pesticide does not capture the complexity of the occupational exposure. Recently, pesticide use patterns analysis has emerged as an alternative to study these exposures. The aim of this study is to identify the pesticide use pattern among flower growers in Mexico participating in the study on the endocrine and reproductive effects associated with pesticide exposure. Methods A cross-sectional study was carried out to gather retrospective information on pesticide use applying a questionnaire to the person in charge of the participating flower growing farms. Information about seasonal frequency of pesticide use (rainy and dry) for the years 2004 and 2005 was obtained. Principal components analysis was performed. Results Complete information was obtained for 88 farms and 23 pesticides were included in the analysis. Six principal components were selected, which explained more than 70% of the data variability. The identified pesticide use patterns during both years were: 1. fungicides benomyl, carbendazim, thiophanate and metalaxyl (both seasons), including triadimephon during the rainy season, chlorotalonyl and insecticide permethrin during the dry season; 2. insecticides oxamyl, biphenthrin and fungicide iprodione (both seasons), including insecticide methomyl during the dry season; 3. fungicide mancozeb and herbicide glyphosate (only during the rainy season); 4. insecticides metamidophos and parathion (both seasons); 5. insecticides omethoate and methomyl (only rainy season); and 6. insecticides abamectin and carbofuran (only dry season). Some pesticides do not show a clear pattern of seasonal use during the studied years. Conclusions The principal component analysis is useful to summarise a large set of exposure variables into smaller groups of exposure patterns, identifying the mixtures of pesticides in the occupational environment that may have an interactive effect on a particular health effect.

Biological N2 fixation is a major factor contributing to the increased competitiveness of Brazilian soybeans on the international market. However, the contribution of this process may be limited by adverse conditions to symbiotic bacteria, such as fungicide seed treatments. This study aimed to evaluate the effects of the fungicides carbendazim + thiram and carboxin + thiram on soybean nodulation, plant growth and grain yield. Two field experiments were carried out in the Cerrado region of the State of Roraima, in a soil with a low organic matter content and no soybean bradyrhizobia. In 2005, seeds were treated with fungicide carbendazim + thiram and commercial inoculants containing the Bradyrhizobium elkanii strains SEMIA 5019 and SEMIA 587 and B. japonicum strains SEMIA 5079 and SEMIA 5080. In 2006, soybean seeds were treated with the fungicides carbendazim + thiram or carboxin + thiram and inoculated separately with each one of the four strains. The plants were evaluated for number of nodules and dry weight, shoot dry weight and total N accumulated in shoots 35 days after plant emergence, while grain yield and N grain content were determined at harvest. Both fungicides reduced soybean nodulation, especially in the presence of B. elkanii strains. The fungicide carbendazim + thiram reduced nodulation by about 50 % and grain yield by more than 20 % (about 700 kg ha-1), in the treatment inoculated with of strain SEMIA 587.

A fixação biológica de N é indispensável à soja no Brasil, dispensando o uso de N mineral e reduzindo o custo de produção. O objetivo deste estudo foi comparar a inoculação da soja com Bradyrhizobium no sulco de semeadura com a inoculação tradicional nas sementes, em solo com baixo teor de matéria orgânica e desprovido de bactérias nodulantes dessa cultura. Foi conduzido um experimento de campo no cerrado de Roraima no ano de 2006 e repetido nas mesmas condições em 2007, sendo avaliados os tratamentos: controle sem inoculação e sem N mineral, 200 kg ha-1 de N na forma de ureia e os métodos de inoculação no sulco de semeadura e inoculação nas sementes, sendo cada método avaliado em sementes sem tratamentos com fungicidas e com a aplicação de carboxin+tiram e carbendazim+tiram. A inoculação da soja no sulco proporcionou desempenho da fixação biológica do N igual ao da inoculação realizada nas sementes, não havendo diferenças na produção de matéria seca, rendimento de grãos e acúmulo de N na parte aérea e nos grãos. Por sua vez, quando as sementes foram tratadas com fungicidas, especialmente carbendazim+tiram, houve menor nodulação das plantas, desenvolvimento da parte aérea e rendimento de grãos com a inoculação nas sementes, enquanto para a inoculação no sulco não houve interferência. Assim, a inoculação no sulco de semeadura mostrou-se uma alternativa viável para a soja quando as sementes foram tratadas com fungicidas.
