El projecte consisteix en l'arquitectura d'una solució Web aplicada a la gestió d'un centre de formació basat en una arquitectura fàcil de modificar i ampliar, per tenir en consideració la comoditat en l'adaptació de l'aplicació a qualsevol centre de formació mitjançant els mínims canvis pertinents. El sistema que s'ha implementat rep el nom de SIGENET.

Contemporary web-based software solutions are usually composed of many interoperating applications. Classical approach is the different applications of the solution to be created inside one technology/platform, e.g. Java-technology, .NET-technology, etc. Wide spread technologies/platforms practically discourage (and sometime consciously make impossible) the cooperation with elements of the concurrent technologies/platforms. To make possible the usage of attractive features of one technology/platform in another technology/platform some “cross-technology” approach is necessary. In the paper is discussed the possibility to combine two existing instruments – interoperability protocols and “lifting” of procedures – in order to obtain such cross-technology approach.

The solution structure of robustoxin, the lethal neurotoxin from the Sydney funnel-web spider Atrax robustus, has been determined from 2D H-1 NMR data, Robustoxin is a polypeptide of 42 residues cross-linked by four disulphide bonds, the connectivities of which were determined from NMR data and trial structure calculations to be 1-15, 8-20, 14-31 and 16-42 (a 1-4/2-6/3-7/5-8 pattern), The structure consists of a small three-stranded, anti-parallel beta-sheet and a series of interlocking gamma-turns at the C-terminus. It also contains a cystine knot, thus placing it in the inhibitor cystine knot motif family of structures, which includes the omega-conotoxins and a number of plant and animal toxins and protease inhibitors. Robustoxin contains three distinct charged patches on its surface, and an extended loop that includes several aromatic and non-polar residues, Both of these structural features may play a role in its binding to the voltage-gated sodium channel. The University of São Paulo has been experiencing the increase in contents in electronic and digital formats, distributed by different suppliers and hosted remotely or in clouds, and is faced with the also increasing difficulties related to facilitating access to this digital collection by its users besides coexisting with the traditional world of physical collections. A possible solution was identified in the new generation of systems called Web Scale Discovery, which allow better management, data integration and agility of search. Aiming to identify if and how such a system would meet the USP demand and expectation and, in case it does, to identify what the analysis criteria of such a tool would be, an analytical study with an essentially documental base was structured, as from a revision of the literature and from data available in official websites and of libraries using this kind of resources. The conceptual base of the study was defined after the identification of software assessment methods already available, generating a standard with 40 analysis criteria, from details on the unique access interface to information contents, web 2.0 characteristics, intuitive interface, facet navigation, among others. The details of the studies conducted into four of the major systems currently available in this software category are presented, providing subsidies for the decision-making of other libraries interested in such systems.

Nowadays video and web conferencing systems have become effective tools for communication and collaboration inside organizations. However, although these systems have evolved and now provide very nice features (e.g. sharing multimedia and documents), they are still too focused on the moment the meeting takes place. The existing systems provide very few facilities to organize the meeting and they do not take advantage of the possibilities the generated content offers once the meeting is finished. In this paper, we analyze the life cycle of a web conference and how existing systems monitor these conferences. Finally we present our solution, based on our know-how in videoconference management and our experience with these existing systems.

A family of potent insecticidal toxins has recently been isolated from the venom of Australian funnel web spiders. Among these is the 37-residue peptide omega-atracotoxin-HV1 (omega-ACTX-HV1) from Hadronyche versuta. We have chemically synthesized and folded omega-ACTX-HV1, shown that it is neurotoxic, ascertained its disulphide bonding pattern, and determined its three-dimensional solution structure using NMR spectroscopy. The structure consists of a solvent-accessible beta-hairpin protruding from a disulphide-bonded globular core comprising four beta-turns. The three intramolecular disulphide bonds form a cystine knot motif similar to that seen in several other neurotoxic peptides. Despite limited sequence identity, omega-ACTX-HV1 displays significant structural homology with the omega-agatoxins and omega-conotoxins, both of which are vertebrate calcium channel antagonists; however, in contrast with these toxins, we show that omega-ACTX-HV1 inhibits insect, but not mammalian, voltage-gated calcium channel currents.

The primary sequence and three-dimensional structure of a novel peptide toxin isolated from the Australian funnel-web spider Hadronyche infensa sp. is reported. ACTX-HI:OB4219 contains 38 amino acids, including eight-cysteine residues that form four disulfide bonds. The connectivities of these disulfide bonds were previously unknown but have been unambiguously determined in this study. Three of these disulfide bonds are arranged in an inhibitor cystine-knot (ICK) motif, which is observed in a range of other disulfide-rich peptide toxins. The motif incorporates an embedded ring in the structure formed by two of the disulfides and their connecting backbone segments penetrated by a third disulfide bond. Using NMR spectroscopy, we determined that despite the isolation of a single native homologous product by RP-HPLC, ACTX-HI:OB4219 possesses two equally populated conformers in solution. These two conformers were determined to arise from cis/trans isomerization of the bond preceding Pro30. Full assignment of the NMR spectra for both conformers allowed for the calculation of their structures, revealing, the presence of a triple-stranded antiparallel sheet consistent with the inhibitor cystine-knot (ICK) motif.

Constrained and unconstrained Nonlinear Optimization Problems often appear in many engineering areas. In some of these cases it is not possible to use derivative based optimization methods because the objective function is not known or it is too complex or the objective function is non-smooth. In these cases derivative based methods cannot be used and Direct Search Methods might be the most suitable optimization methods. An Application Programming Interface (API) including some of these methods was implemented using Java Technology. This API can be accessed either by applications running in the same computer where it is installed or, it can be remotely accessed through a LAN or the Internet, using webservices. From the engineering point of view, the information needed from the API is the solution for the provided problem. On the other hand, from the optimization methods researchers’ point of view, not only the solution for the problem is needed. Also additional information about the iterative process is useful, such as: the number of iterations; the value of the solution at each iteration; the stopping criteria, etc. In this paper are presented the features added to the API to allow users to access to the iterative process data.

Remote engineering (also known as online engineering) may be defined as a combination of control engineering and telematics. In this area, specific activities require computacional skills in order to develop projects where electrical devives are monitored and / or controlled, in an intercative way, through a distributed network (e.g. Intranet or Internet). In our specific case, we will be dealing with an industrial plant. Within the last few years, there has been an increase in the number of activities related to remote engineering, which may be connected to the phenomenon of the large extension experienced by the Internet (e.g. bandwith, number of users, development tools, etc.). This increase opens new and future possibilities to the implementation of advance teleworking (or e-working) positions. In this paper we present the architecture for a remote application, accessible through the Internet, able to monitor and control a roller hearth kiln, used in a ceramics industry for firing materials. The proposed architecture is based on a micro web server, whose main function is to monitor and control the firing process, by reading the data from a series of temperature sensors and by controlling a series of electronic valves and servo motors. This solution is also intended to be a low-cost alternative to other potential solutions. The temperature readings are obtained through K-type thermopairs and the gas flow is controlled through electrovalves. As the firing process should not be stopped before its complete end, the system is equipped with a safety device for that specific purpose. For better understanding the system to be automated and its operation we decided to develop a scale model (100:1) and experiment on it the devised solution, based on a Micro Web Server.

No panorama socioeconómico atual, a contenção de despesas e o corte no financiamento de serviços secundários consumidores de recursos conduzem à reformulação de processos e métodos das instituições públicas, que procuram manter a qualidade de vida dos seus cidadãos através de programas que se mostrem mais eficientes e económicos. O crescimento sustentado das tecnologias móveis, em conjunção com o aparecimento de novos paradigmas de interação pessoa-máquina com recurso a sensores e sistemas conscientes do contexto, criaram oportunidades de negócio na área do desenvolvimento de aplicações com vertente cívica para indivíduos e empresas, sensibilizando-os para a disponibilização de serviços orientados ao cidadão. Estas oportunidades de negócio incitaram a equipa do projeto a desenvolver uma plataforma de notificação de problemas urbanos baseada no seu sistema de informação geográfico para entidades municipais. O objetivo principal desta investigação foca a idealização, conceção e implementação de uma solução completa de notificação de problemas urbanos de caráter não urgente, distinta da concorrência pela facilidade com que os cidadãos são capazes de reportar situações que condicionam o seu dia-a-dia. Para alcançar esta distinção da restante oferta, foram realizados diversos estudos para determinar características inovadoras a implementar, assim como todas as funcionalidades base expectáveis neste tipo de sistemas. Esses estudos determinaram a implementação de técnicas de demarcação manual das zonas problemáticas e reconhecimento automático do tipo de problema reportado nas imagens, ambas desenvolvidas no âmbito deste projeto. Para a correta implementação dos módulos de demarcação e reconhecimento de imagem, foram feitos levantamentos do estado da arte destas áreas, fundamentando a escolha de métodos e tecnologias a integrar no projeto. Neste contexto, serão apresentadas em detalhe as várias fases que constituíram o processo de desenvolvimento da plataforma, desde a fase de estudo e comparação de ferramentas, metodologias, e técnicas para cada um dos conceitos abordados, passando pela proposta de um modelo de resolução, até à descrição pormenorizada dos algoritmos implementados. Por último, é realizada uma avaliação de desempenho ao par algoritmo/classificador desenvolvido, através da definição de métricas que estimam o sucesso ou insucesso do classificador de objetos. A avaliação é feita com base num conjunto de imagens de teste, recolhidas manualmente em plataformas públicas de notificação de problemas, confrontando os resultados obtidos pelo algoritmo com os resultados esperados.

A velocidade de difusão de conteúdos numa plataforma web, assume uma elevada relevância em serviços onde a informação se pretende atualizada e em tempo real. Este projeto de Mestrado, apresenta uma abordagem de um sistema distribuído de recolher e difundir resultados em tempo real entre várias plataformas, nomeadamente sistemas móveis. Neste contexto, tempo real entende-se como uma diferença de tempo nula entre a recolha e difusão, ignorando fatores que não podem ser controlados pelo sistema, como latência de comunicação e tempo de processamento. Este projeto tem como base uma arquitetura existente de processamento e publicação de resultados desportivos, que apresentava alguns problemas relacionados com escalabilidade, segurança, tempos de entrega de resultados longos e sem integração com outras plataformas. Ao longo deste trabalho procurou-se investigar fatores que condicionassem a escalabilidade de uma aplicação web dando ênfase à implementação de uma solução baseada em replicação e escalabilidade horizontal. Procurou-se também apresentar uma solução de interoperabilidade entre sistemas e plataformas heterogêneas, mantendo sempre elevados níveis de performance e promovendo a introdução de plataformas móveis no sistema. De várias abordagens existentes para comunicação em tempo real sobre uma plataforma web, adotou-se um implementação baseada em WebSocket que elimina o tempo desperdiçado entre a recolha de informação e sua difusão. Neste projeto é descrito o processo de implementação da API de recolha de dados (Collector), da biblioteca de comunicação com o Collector, da aplicação web (Publisher) e sua API, da biblioteca de comunicação com o Publisher e por fim a implementação da aplicação móvel multi-plataforma. Com os componentes criados, avaliaram-se os resultados obtidos com a nova arquitetura de forma a aferir a escalabilidade e performance da solução criada e sua adaptação ao sistema existente.
