Resumen La Rinoneumonitis Equina (RE) es un término que describe un conjunto de enfermedades en caballos que pueden incluir cuadros clínicos respiratorios, aborto, neumonitis neonatal de los potros, o mieloencefalopatía, y está causada por los virus EHV-1 y EHV-4. La distribución de estos agentes se considera mundialmente enzoótica provocando importantes pérdidas económicas especialmente en países con buen desarrollo de la industria hípico-deportiva. En el Uruguay se considera enzoótica desde hace muchos años. El objetivo de este trabajo es estimar la seroprevalencia de EHV-1 en equinos de deporte en el Uruguay y determinar si está asociada con factores como la edad, raza, sexo, y los antecedentes de vacunación. Se colectaron 1517 sueros equinos en el período diciembre 2013 a marzo 2014, reseñando la raza, edad, sexo y registro de vacunación, la totalidad de las muestras fueron procesadas por la prueba de seroneutralización viral, y los resultados fueron estudiados mediante modelo de regresión logística. Se comprueba que el virus EHV-1 se encuentra ampliamente distribuido en la población de equinos de deporte en nuestro país, con una prevalencia estimadadel 29% (IC 95% = 27%-31%). Se observa que la presencia de anticuerpos para EHV-1 está asociada a factores como la raza y la vacunación contra RE (p<0,05)

A multiplex real-time PCR was designed to detect and differentiate equid herpesvirus 1 (EHV-1) and equid herpesvirus 4 (EHV-4). The PCR targets the glycoprotein B gene of EHV-1 and EHV-4. Primers and probes were specific to each equine herpesvirus type and can be used in monoplex or multiplex PCRs, allowing the differentiation of these two closely related members of the Alphaherpesvirinae. The two probes were minor-groove binding probes (MGB?) labelled with 6-carboxy-fluorescein (FAM?) and VIC® for detection of EHV-1 and EHV-4, respectively. Ten EHV-1 isolates, six EHV-1 positive clinical samples, one EHV-1 reference strain (EHV-1.438/77), three EHV-4 positive clinical samples, two EHV-4 isolates and one EHV-4 reference strain (EHV-4 405/76) were included in this study. EHV-1 isolates, clinical samples and the reference strain reacted in the EHV-1 real-time PCR but not in the EHV-4 real-time PCR and similarly EHV-4 clinical samples, isolates and the reference strain were positive in the EHV-4 real-time PCR but not in the EHV-1 real-time PCR. Other herpesviruses, such as EHV-2, EHV-3 and EHV-5 were all negative when tested using the multiplex real-time PCR. When bacterial pathogens and opportunistic pathogens were tested in the multiplex real-time PCR they did not react with either system. The multiplex PCR was shown to be sensitive and specific and is a useful tool for detection and differentiation of EHV-1 and EHV-4 in a single reaction. A comprehensive equine herpesvirus disease investigation procedure used in our laboratory is also outlined. This procedure describes the combination of alphaherpesvirus multiplex real-time PCR along with existing gel-based PCRs described by other authors.

Background Research in humans has demonstrated that high serum iron (sFe) concentration can predispose to infection, and many infections subsequently result in alterations of host sFe. A decrease in sFe concentration is an early and sensitive indicator of systemic inflammation caused by tissue necrosis, bacterial infections, or endotoxemia in horses. Serum iron parameters in acute equine herpesvirus type 1 (EHV-1) infection have not been evaluated previously. Objectives To document the sFe response to EHV-1 infection and to determine whether or not significant differences in sFe concentration exist between EHV-1 infected horses that develop neurologic disease and those that do not. Animals A total of 14 horses experimentally infected with EHV-1. Methods Data were collected as an ancillary data set during a blinded experimental EHV-1 infection. Horses were infected with the rAb4 strain of EHV-1. Temperature, neurologic score, packed cell volume (PCV), and sFe parameters (sFe concentration, % saturation, and total iron-binding capacity) were recorded daily for 2weeks. Data were evaluated using Wilcoxon signed rank tests and Wilcoxon rank sum tests with Bonferroni corrections. Conclusions and Clinical Relevance Serum iron concentration decreases significantly in a biphasic pattern after EHV-1 infection. There was no significant difference in sFe concentration in horses that developed neurologic disease and those that did not in these experimentally infected animals. Serum iron parameters may be useful in monitoring the clinical course of viral infections such as EHV-1.

This report describes the first detection of an equine herpesvirus 1 (EHV-1) neuropathogenic variant (G 2254/D 752) in Brazil from a case of fatal equine herpesvirus myeloencephalopathy (EHM) in a mare. The results of nucleotide sequencing of the EHV-1 ORF30 gene showed that two other Brazilian EHV-1 isolates from EHM cases are representatives of the non-neuropathogenic variant (A 2254/N 752), suggesting that other unidentified factors are probably also involved in the neuropathogenicity of EHV-1 in horses. These findings will contribute to the epidemiological knowledge of EHV-1 infection in Brazil.

Intranasal inoculation of equid herpesvirus type-1 (EHV-1) Brazilian strains A4/72 and A9/92 induced an acute and lethal infection in four different inbred mouse strains. Clinical and neurological signs appeared between the 2nd and 3rd day post inoculation (dpi) and included weight loss, ruffled fur, a hunched posture, crouching in corners, nasal and ocular discharges, dyspnoea, dehydration and increased salivation. These signs were followed by increased reactivity to external stimulation, seizures, recumbency and death. The virus was recovered consistently from the brain and viscera of all mice with neurological signs. Histopathological changes consisted of leptomeningitis, focal haemorrhage, ventriculitis, neuronal degeneration and necrosis, neuronophagia, non-suppurative inflammation, multifocal gliosis and perivascular infiltration of polymorphonuclear and mononuclear cells. Immunohistochemical examination demonstrated that EHV-1 strains A4/72 and A9/92 replicated in neurons of the olfactory bulb, the cortex and the hippocampus. In contrast, mice inoculated with the EHV-1 Brazilian strain A3/97 showed neither weight loss nor apparent clinical or neurological signs; however, the virus was recovered consistently from their lungs at 3 dpi. These three EHV-1 strains showed distinct degrees of virulence and tissue tropism in mice. EHV-1 strains A4/72 and A9/92 exhibited a high degree of central nervous system tropism with neuroinvasion and neurovirulence. EHV-1 strain A3/97 was not neurovirulent despite being detected in the brains of infected BALB/c nude mice. These findings indicate that several inbred mouse strains are susceptible to neuropathogenic EHV-1 strains and should be useful models for studying the pathogenesis and mechanisms contributing to EHV-induced myeloencephalopathy in horses. Twelve nasal swabs were collected from yearling horses with respiratory distress and tested for equid herpesvirus 1 (EHV-1) and equid herpesvirus 4 (EHV-4) by real-time PCR targeting the glycoprotein B gene. All samples were negative for EHV-1; however, 3 were positive for EHV-4. When these samples were tested for EHV-2 and EHV-5 by PCR, all samples were negative for EHV-2 and 11 were positive for EHV-5. All three samples that were positive for EHV-4 were also positive for EHV-5. These three samples gave a limited CPE in ED cells reminiscent of EHV-4 CPE. EHV-4 CPE was obvious after 3 days and was characterised by syncytia. None of the samples produced cytopathic effect (CPE) on African green monkey kidney (Vero) cells or hamster kidney (BSR) cells. Four of the samples, which were positive in the EHV-5 PCR, produced CPE on rabbit kidney (RK13) cells and equine dermis (ED) cells. EHV-5 CPE on both cell lines was slow and was apparent after four 7-day passages. On RK13 cells, the CPE was characteristic of equid herpesvirus, with the formation of syncytia. However, in ED cells, the CPE was characterised by ring-shaped syncytia. For the first time, a case of equine respiratory disease involving dual infection with EHV-4 and EHV-5 has been reported in Queensland (Australia). This was shown by simultaneously isolating EHV-4 and EHV-5 from clinical samples. EHV5 was recovered from all samples except one, suggesting that EHV5 was more prevalent in young horses than EHV2.

Meningoencephalitis by Herpesvirus type 5 (BoHV-5) in cattle has some features that are similar to those of herpetic encephalitis in humans and other animal species. Human Herpesvirus 3 (commonly known as Varicella-zoster virus 1), herpes simplex viruses (HSV), and equid Herpesvirus 1 (EHV-1) induce an intense inflammatory, vascular and cellular response. In spite of the many reports describing the histological lesions associated with natural and experimental infections, the immunopathological mechanisms for the development of neurological disorder have not been established. A total of twenty calf brains were selected from the Veterinary School, University of São Paulo State, Araçatuba, Brazil, after confirmation of BoHV-5 infection by virus isolation as well as by a molecular approach. The first part of the study characterized the microscopic lesions associated with the brain areas in the central nervous system (CNS) that tested positive in a viral US9 gene hybridization assay. The frontal cortex (Fc), parietal cortex (Pc), thalamus (T) and mesencephalon (M) were studied. Secondly, distinct pathogenesis mechanisms that take place in acute cases were investigated by an immunohistochemistry assay. This study found the frontal cortex to be the main region where intense oxidative stress phenomena (AOP-1) and synaptic protein expression (SNAP-25) were closely related to inflammatory cuffs, satellitosis and gliosis, which represent the most frequently observed neurological lesions. Moreover, MMP-9 expression was shown to be localized in the leptomeninges, in the parenchyma and around mononuclear infiltrates (p < 0.0001). These data open a new perspective in understanding the role of the AOP-1, MMP-9 and SNAP-25 proteins in mediating BoHV-5 pathogenesis and the strategies of host-virus interaction in order to invade the CNS.

Objetivos: (1) Estudiar la frecuencia de eventos cardiovasculares (CV) que presentaron los pacientes de una gran cohorte española de lupus eritematoso sistémico (LES) tras el diagnóstico de la enfermedad, y (2) investigar los principales factores de riesgo implicados en la aterosclerosis. Material y métodos: Estudio transversal retrospectivo basado en RELESSER, un registro multicéntrico, nacional de pacientes diagnosticados de LES. Pacientes: sujetos con diagnóstico de LES (criterios ACR 1997), atendidos en 45 servicios de Reumatología españoles. Variables: demográficas, clínicas, factores de riesgo tradicionales y eventos CV. La variable de desenlace fue evento CV que se definió por la presencia de al menos uno de los siguientes tras el diagnóstico de LES: 1) Enfermedad cardiaca isquémica (infarto agudo de miocardio y/o angina por clínica y/o electrocardiograma y/o enzimas y/o coronariografía), 2) accidente cerebrovascular (ACV) basado en un diagnóstico previo o por clínica y/o imagen; y 3) enfermedad arterial periférica por diagnóstico previo o por imagen. Los pacientes con eventos CV previos al diagnóstico de LES se excluyeron del análisis. Protocolo: la información fue recogida a partir de las historias clínicas usando un protocolo específico en una plataforma web y con previo entrenamiento de los investigadores. Análisis estadístico: Descriptivo y comparaciones entre grupos utilizando χ2, T-Student o U Mann-Whitney. Análisis de regresión logística múltiple para analizar los posibles factores de riesgo CV asociados a aterosclerosis. Resultados: Desde el año 2.011 al 2.012, 3.658 pacientes con LES fueron reclutados en RELESSER y solo 9 se excluyeron por insuficiente información sobre eventos CV. De los 3.649 restantes, 374 (10,9%) presentaron al menos un evento CV. Doscientos sesenta y nueve (7,4%) pacientes tuvieron 318 eventos CV tras el diagnóstico de LES. El 86,2% de esos pacientes eran mujeres, mediana [RIQ] de edad de 54,9 años [43,2 ̶ 66,1] y una duración del LES de 212.0 meses [120,8 ̶ 289,0]. La media (DE) de edad al cuando sufrieron el primer evento CV fue 48,6 (17,1) años. Los ACV (5,7%), fueron los eventos más frecuentes seguidos de la cardiopatía isquémica (3,8%) y la enfermedad arterial periférica (2,2%). El análisis multivariante mostró una fuerte asociación (OR [IC 95%]) entre los eventos CV y la edad (1,03 [1,02 ̶ 1,04]), la hipertensión arterial (1,71 [1,20 ̶ 2,44]), el tabaquismo (1,48 [1,06 ̶ 2,07]), la diabetes (2,2 [1,32 ̶ 3,74]), la dislipemia (2.18 [1,54 ̶ 3,09], el lupus neuropsiquiátrico (2,42 [1,56 ̶ 3,75]), la presencia de valvulopatía (2,44[1,34 ̶ 4,26]), serositis (1,54 [1,09 ̶ 2.18]), anticuerpos antifosfolípidos (1,57 [1,13 ̶ 2,17]), disminución del complemento (1,81 [1,12 ̶ 2,93]) y la toma de azatioprina (1,47 [1,04 ̶ 2,07]). Conclusiones: Los pacientes con LES presentan una alta prevalencia de enfermedad CV prematura que se asocia con factores de riesgo CV tradicionales y relacionados con el LES. Aunque se necesitan futuros estudios que lo comprueben, nuestro trabajo demuestra, por primera vez, una asociación entre la diabetes y los eventos CV en pacientes con LES.

La brucelosis es una enfermedad que se presenta principalmente en personal que tiene contacto cercano con el ganado o que consume derivados que no se han tratado adecuadamente. Métodos: se realizó un estudio de corte transversal para determinar la seroprevalencia de brucelosis en trabajadores de mataderos de algunos municipios del Departamento del Tolima (Colombia). Se evaluaron 186 trabajadores a quienes se aplicaron las pruebas serológicas indirectas (rosa de bengala, fijación de complemento y Elisa). Resultados: se detectó una prevalencia de anticuerpos antibrucella cercana al 4% en la población evaluada, con una llamativa correlación entre condiciones de higiene del trabajo y seropositividad. Conclusiones: los trabajadores de mataderos con condiciones técnico-higiénicas desfavorables se encuentran en mayor riesgo de contagio de brucelosis.

This paper describes an approach for the analysis and design of 765kV/400kV EHV transmission system which is a typical expansion in Indian power grid system, based on the analysis of steady state and transient over voltages. The approach for transmission system design is iterative in nature. The first step involves exhaustive power flow analysis, based on constraints such as right of way, power to be transmitted, power transfer capabilities of lines, existing interconnecting transformer capabilities etc. Acceptable bus voltage profiles and satisfactory equipment loadings during all foreseeable operating conditions for normal and contingency operation are the guiding criteria. Critical operating strategies are also evolved in this initial design phase. With the steady state over voltages obtained, comprehensive dynamic and transient studies are to be carried out including switching over voltages studies. This paper presents steady state and switching transient studies for alternative two typical configurations of 765kV/400 kV systems and the results are compared. Transient studies are carried out to obtain the peak values of 765 kV transmission systems and are compared with the alternative configurations of existing 400 kV systems.

The components of EHV/UHV lines and substations can produce significant corona. To limit the consequent Radio Interference and Audible Noise on these systems, suitable corona control rings are employed. The shapes of these rings could vary from circular to rectangular with smooth bends. Many manufacturers seem to adopt trial and error method for arriving at the final design. As such neither the present testing standard nor the final design adopted consider the practical scenario like corona produced by deposition of dirt, bird droppings, etc. The present work aims to make a first step in addressing this practically important problem. This requires an accurate evaluation of the electric field and a reliable method for the evaluation of corona inception. Based on a thorough survey of pertinent literature, the critical avalanche criteria as applicable to large electrodes, has been adopted. Taking the rain drop on the surface as the biggest protrusion, conducting protrusions modeled as semi-ellipsoid is considered as representative for deposition of dust or the boundary of bird droppings etc. Through examples of 4 00 kV and 765 kV class toroidal corona rings, the proposed method is demonstrated. This work is believed to be useful to corona ring manufacturers for EHV/UHV systems.

This paper presents comparative evaluation of the distance relay characteristics for UHV and EHV transmission lines. Distance protection relay characteristics for the EHV and UHV systems are developed using Electromagnetic Transients (EMT) program. The variation of ideal trip boundaries for both the systems are presented. Unlike the conventional distance protection relay which uses a lumped parameter model, this paper uses the distributed parameter model. The effect of larger shunt susceptance on the trip boundaries is highlighted. Performance of distance relay with ideal trip boundaries for EHV and UHV lines have been tested for various fault locations and fault resistances. Electromagnetic Transients (EMT) program has been developed considering distributed parameter line model for simulating the test systems. The voltage and current phasors are computed from the signals using an improved full cycle DFT algorithm taking 20 samples per cycle. Two practical transmission systems of Indian power grid, namely 765 kV UHV transmission line and SREB 24-bus 400kV EHV system are used to test the performance of the proposed approach.

This paper presents a fast and accurate relaying technique for a long 765kv UHV transmission line based on support vector machine. For a long EHV/UHV transmission line with large distributed capacitance, a traditional distance relay which uses a lumped parameter model of the transmission line can cause malfunction of the relay. With a frequency of 1kHz, 1/4th cycle of instantaneous values of currents and voltages of all phases at the relying end are fed to Support Vector Machine(SVM). The SVM detects fault type accurately using 3 milliseconds of post-fault data and reduces the fault clearing time which improves the system stability and power transfer capability. The performance of relaying scheme has been checked with a typical 765kV Indian transmission System which is simulated using the Electromagnetic Transients Program(EMTP) developed by authors in which the distributed parameter line model is used. More than 15,000 different short circuit fault cases are simulated by varying fault location, fault impedance, fault incidence angle and fault type to train the SVM for high speed accurate relaying. Simulation studies have shown that the proposed relay provides fast and accurate protection irrespective of fault location, fault impedance, incidence time of fault and fault type. And also the proposed scheme can be used as augmentation for the existing relaying, particularly for Zone-2, Zone-3 protection.
