Introduction: Inherent and acquired cisplatin resistance reduces the effectiveness of this agent in the management of non-small cell lung cancer (NSCLC). Understanding the molecular mechanisms underlying this process may result in the development of novel agents to enhance the sensitivity of cisplatin. Methods: An isogenic model of cisplatin resistance was generated in a panel of NSCLC cell lines (A549, SKMES-1, MOR, H460). Over a period of twelve months, cisplatin resistant (CisR) cell lines were derived from original, age-matched parent cells (PT) and subsequently characterized. Proliferation (MTT) and clonogenic survival assays (crystal violet) were carried out between PT and CisR cells. Cellular response to cisplatin-induced apoptosis and cell cycle distribution were examined by FACS analysis. A panel of cancer stem cell and pluripotent markers was examined in addition to the EMT proteins, c-Met and β-catenin. Cisplatin-DNA adduct formation, DNA damage (γH2AX) and cellular platinum uptake (ICP-MS) was also assessed. Results: Characterisation studies demonstrated a decreased proliferative capacity of lung tumour cells in response to cisplatin, increased resistance to cisplatin-induced cell death, accumulation of resistant cells in the G0/G1 phase of the cell cycle and enhanced clonogenic survival ability. Moreover, resistant cells displayed a putative stem-like signature with increased expression of CD133+/CD44+cells and increased ALDH activity relative to their corresponding parental cells. The stem cell markers, Nanog, Oct-4 and SOX-2, were significantly upregulated as were the EMT markers, c-Met and β-catenin. While resistant sublines demonstrated decreased uptake of cisplatin in response to treatment, reduced cisplatin-GpG DNA adduct formation and significantly decreased γH2AX foci were observed compared to parental cell lines. Conclusion: Our results identified cisplatin resistant subpopulations of NSCLC cells with a putative stem-like signature, providing a further understanding of the cellular events associated with the cisplatin resistance phenotype in lung cancer. © 2013 Barr et al.

miR-126 has been implicated in the processes of inflammation and angiogenesis. Through these processes, miR-126 is implicated in cancer biology, but its role there has not been well reviewed. The aim of this review is to examine the molecular mechanisms and clinicopathological significance of miR-126 in human cancers. miR-126 was shown to have roles in cancers of the gastrointestinal tract, genital tracts, breast, thyroid, lung and some other cancers. Its expression was suppressed in most of the cancers studied. The molecular mechanisms that are known to cause aberrant expression of miR-126 include alterations in gene sequence, epigenetic modification and alteration of dicer abundance. miR-126 can inhibit progression of some cancers via negative control of proliferation, migration, invasion, and cell survival. In some instances, however, miR-126 supports cancer progression via promotion of blood vessel formation. Downregulation of miR-126 induces cancer cell proliferation, migration, and invasion via targeting specific oncogenes. Also, reduced levels of miR-126 are a significant predictor of poor survival of patients in many cancers. In addition, miR-126 can alter a multitude of cellular mechanisms in cancer pathogenesis via suppressing gene translation of numerous validated targets such as PI3K, KRAS, EGFL7, CRK, ADAM9, HOXA9, IRS-1, SOX-2, SLC7A5 and VEGF. To conclude, miR-126 is commonly down-regulated in cancer, most likely due to its ability to inhibit cancer cell growth, adhesion, migration, and invasion through suppressing a range of important gene targets. Understanding these mechanisms by which miR-126 is involved with cancer pathogenesis will be useful in the development of therapeutic targets for the management of patients with cancer.

人胚胎干细胞(ESC)的成功分离培养,吸引大批人对干细胞生物学的关注,特 别是ESC 在再生医学及人类早期胚胎发育研究的潜在价值。然而在人ESC 临床应用 之前需要找到合适的动物模型进行大量的预实验研究,从而评价其应用的安全性、有 效性及存活效率。因此,从其它物种建立稳定而可用的ESC 系也是必不可少的。ESC 能无限地自我更新并保持多潜能性,但控制其自我更新的分子机制现在仍然知之甚少 且物种间存在差异,了解ESC 的自我更新有利于提高建系率、改善培养体系及定向 分化体系。本文一方面对ESC 分离培养及自我更新机制的研究进展进行了综述,另 一方面对以下几个方面的内容进行了研究:1)建立了4 株稳定的兔ESC 系,能在体 外进行长期的培养并保持ESC 的多潜能Markers 及正常的XY 或XX 核型,具有碱性 磷酸酶活性、表达Oct-4、SSEA-1、SSEA-3、SSEA-4、TRA-1-60 及TRA-1-81。与人 和小鼠ESC 相似,兔ESC 表达多潜能基因(Oct-4、Nanog、Sox-2 及UTF-1),并表 达了与ESC 自我更新相关的信号通路(FGF、TGFβ及WNT)的许多基因。从形态 来说,兔ESC 与灵长类ESC 相似,但兔ESC 具有较快的增殖能力,与小鼠ESC 相类 似。在体外及体内兔ESC 均能分化成代表原始三胚层的各种细胞类型及组织。2) 从 受体抑制实验及生长因子的联合加入可以得出结论FGF 及TGF 信号通路对维持兔 ESC 的多潜能性发挥着重要的作用,这样的结果与人ESC 相类似。也表明FGF、TGF β及WNT 信号通路在兔ESC 的自我更新中都起着作用,而且他们之间可能形成了信 号调控网络,相互之间有着正负反馈作用。FGF2+Activin A 或TGFβ1+Noggin 的无 饲养层无血清培养体系不仅能显著抑制兔ESC 的分化,且能维持其长期的自我更新。 但与小鼠不同,TGFβ信号通路能影响其增殖能力,而对其多潜能性的维持并没有作 用。这就更说明了兔比小鼠更适宜成为人类疾病临床治疗之前的模型动物。3)四种 猕猴细胞系(MOF、MESF、MFG 和CMESF)可作为饲养层比MEFs(小鼠饲养层 细胞)更好或同等好支持猕猴ESC 的生长,保持其自我更新能力和分化的多能性。 而卵泡颗粒上皮样细胞(MFGE)不能支持猕猴ESC 的自我更新。进一步的研究表明 饲养层支持ESC 生长能力的差异可能是由于基因表达种类以及表达量上的差异而导 致的。

The cancer stem cell hypothesis may explain why conventional chemotherapies are unable to fully eradicate cancers. In this study, we examined both the prognostic and predictive significance of putative cancer stem cell markers in colorectal cancer. In this study, immunohistochemistry for three candidate cancer stem cell markers (CD133, Oct-4 and Sox-2) and for six other postulated prognostic markers (CK7, CK20, Cox-2, Ki-67, p27 and p53) were performed using tissue microarrays containing 501 primary colorectal cancer cases. Receiver-operating characteristic analysis was used to determine cut-off scores for positive protein expression. Neural stem cells (NSCs) are potential sources for cell therapy of neurodegenerative diseases and for drug screening. Despite their potential benefits, ethical and practical considerations limit the application of NSCs derived from human embryonic stem cells (ES) or adult brain tissue. Thus, alternative sources are required to satisfy the criteria of ready accessibility, rapid expansion in chemically defined media and reliable induction to a neuronal fate. We isolated somatic stem cells from the human periodontium that were collected during minimally invasive periodontal access flap surgery as part of guided tissue regeneration therapy. These cells could be propagated as neurospheres in serum-free medium, which underscores their cranial neural crest cell origin. Culture in the presence of epidermal growth factor (EGF) and fibroblast growth factor-2 (FGF-2) under serum-free conditions resulted in large numbers of nestin-positive/Sox-2-positive NSCs. These periodontium-derived (pd) NSCs are highly proliferative and migrate in response to chemokines that have been described as inducing NSC migration. We used immunocytochemical techniques and RT-PCR analysis to assess neural differentiation after treatment of the expanded cells with a novel induction medium. Adherence to substrate, growth factor deprivation, and retinoic acid treatment led to the acquisition of neuronal morphology and stable expression of markers of neuronal differentiation by more than 90% of the cells. Thus, our novel method might provide nearly limitless numbers of neuronal precursors from a readily accessible autologous adult human source, which could be used as a platform for further experimental studies and has potential therapeutic implications.

In this study we evaluated whether administration of stem cells of neural origin (neural precursor cells, NPCs) could be protective against renal ischemia-reperfusion injury (IRI). We hypothesized that stem cell outcomes are not tissue-specific and that NPCs can improve tissue damage through paracrine mechanisms, especially due to immunomodulation. To this end, Wistar rats (200-250 g) were submitted to 1-hour ischemia and treated with NPCs (4 x 10(6) cells/animal) at 4 h of reperfusion. To serve as controls, ischemic animals were treated with cerebellum homogenate harvested from adult rat brain. All groups were sacrificed at 24 h of reperfusion. NPCs were isolated from rat fetus telencephalon and cultured until neurosphere formation (7 days). Before administration, NPCs were labeled with carboxyfluorescein diacetate succinimydylester (CFSE). Kidneys were harvested for analysis of cytokine profile and macrophage infiltration. At 24 h, NPC treatment resulted in a significant reduction in serum creatinine (IRI + NPC 1.21 + 0.18 vs. IRI 3.33 + 0.14 and IRI + cerebellum 2.95 + 0.78mg/dl, p < 0.05) and acute tubular necrosis (IRI + NPC 46.0 + 2.4% vs. IRI 79.7 + 14.2%, p < 0.05). NPC-CFSE and glial fibrillary acidic protein (GFAP)-positive cells (astrocyte marker) were found exclusively in renal parenchyma, which also presented GFAP and SOX-2 (an embryonic neural stem cell marker) mRNA expression. NPC treatment resulted in lower renal proinflammatory IL1-beta and TNF-alpha expression and higher anti-inflammatory IL-4 and IL-10 transcription. NPC-treated animals also had less macrophage infiltration and decreased serum proinflammatory cytokines (IL-1 beta, TNF-alpha and INF-gamma). The e.m.f. of a concentration cell for SO x (x=2,3)-O2 incorporating Nasicon as the main solid electrolyte has been measured in the temperature range 720 to 1080 K. The cell arrangement can be represented as,$$Pt, O'_2 + SO'_2 + SO'_3 \left| {Na_2 SO_4 \left\| {\left. {Nasicon} \right\|} \right.} \right.\left. {Na_2 SO_4 } \right|SO''_3 + SO''_2 + O''_2 , Pt$$ The Na2SO4 acts both as an auxiliary electrode, converting chemical potentials of SO x and O2 to equivalent sodium potentials, and as an electrolyte. The presence of Na2SO4 provides partial protection of Nasicon from chemical reaction with gas mixtures containing SO x . The open circuit e.m.f. of the cell is in close agreement with values given by the Nernst equation. For certain fixed inlet gas compositions of SO2+O2, the e.m.f. varies non-linearly with temperature. The intrinsic response time of the cell to step changes in gas composition is estimated to vary from sim2.0 ksec at 723K to sim 0.2 ksec at 1077K. The cell functions well for large differences in partial pressures of SO3(pPrimeSO 3/pprimeSO 3ap104) at the electrodes.

A solid-state sensor for SOx (x = 2, 3) species has been designed using ?-alumina as the solid electrolyte and Na2SO4 as the auxiliary electrode. The measured e.m.f. of the cell Pt, O?2 + SO?2 + SO?3|Na2SO4short parallel?-aluminashort parallelNa2SO4|SO?3 + SO?2 + O?2, PT in the temperature range 700 K to 1150 K agrees well with values calculated using the Nernst equation. The sodium sulphate acts both as a protective covering, preventing direct access of the gaseous SOx species to the ?-alumina electrolyte, and as an auxiliary electrode, converting chemical potentials of SOx species and O2 into an equivalent sodium potential. The open-circuit e.m.f. varies non-linearly with temperature for fixed composition of inlet gas mixtures containing SO2, O2 and Ar. The response time (t0.99) of the cell varies between 1.9 ks at 750 K and 0.06 ks at 1100 K. The e.m.f. response is faster when the partial pressure of SOx at the electrode is increased than when it is decreased.

Os riscos de poluição ao meio ambiente envolvendo petróleo envolvem, não só o seu transporte, como também seu refino. O prejuízo causado por um derramamento de petróleo vai além de danos à fauna e flora, pois envolvem também questões sociais. A emissão de óxidos de enxofre, denominadas SOx, durante o refino de petróleo através do craqueamento catalítico em leito fluidizado (FCC) também é uma das preocupações ambientais, já que esses óxidos estão relacionados com o a formação de chuva ácida e problemas respiratórios. Os hidrocarbonetos provenientes de um derramamento podem ser degradados em produtos menos agressivos ao meio ambiente, por oxidação química, por exemplo. Já as emissões de enxofre na unidade de FCC podem ser minimizadas por diversos processos, como por exemplo, o uso de aditivos nas unidades de FCC. Nesse trabalho óxidos de manganês dos tipos OMS-1 e OMS-2 foram sintetizados em presença e ausência de biomassa e óxidos OMS-2 foram dopados com os metais cobre, vanádio e ferro. Possíveis alterações em suas propriedades, suas atividades catalíticas em oxidação de hidrocarbonetos e em testes de captura de enxofre em condições de temperatura similares à unidade de FCC foram investigadas. Constatou-se uma diminuição na área superficial, tamanho e volume de poros nos óxidos sintetizados em presença de biomassa, através de uma análise de adsorção e dessorção de N2 (ASAP), porém seus difratogramas em uma análise de difração de raio X de pó (DRX) revelaram a obtenção de estruturas do criptomelano em todos os OMS-2. Os óxidos OMS-2 testados na oxidação do cicloexano, não sofreram modificações em sua estrutura após seu uso como catalisador, mas a presença da biomassa na síntese não aumentou sua atividade catalítica. Nos testes DeSOx, o óxido dopado com ferro apresentou o melhor desempenho e testes em ciclos mostraram ser possível sua reutilização

The deactivation of a silver-based hydrocarbon selective catalytic reduction catalyst by SOx and the subsequent regeneration under various operating conditions has been investigated. Using a sulfur trap based on a silica-supported catalyst it was found that, for a Ag/SiO2 + Ag/Al2O3 combination, the negative effect of SO2 on the n-octane-SCR reaction can be eliminated under normal operating conditions. The trap can be regenerated by hydrogen at low temperatures or at higher temperatures using a hydrocarbon reductant.

This dissertation examined three issues associated with Section 404 of the Sarbanes-Oxley Act (SOX) that are of current interest to regulators and the accounting profession. For the first essay, I examined auditor changes from 2003 to 2005 for 2,454 non-financial firms that filed their initial SOX 404 opinions prior to July 1, 2005. My results showed that there is a significant association between the receipt of an adverse SOX 404 opinion and auditor resignations - both before and after the issuance of the SOX 404 opinion. The data related to auditor dismissals show that clients are not successful in opinion shopping, since clients dismissing the auditor before the SOX 404 report also are more likely to receive an adverse SOX 404 opinion than clients not changing auditors. My second essay examined the relation between audit committee characteristics and changes in material weaknesses in internal control over financial reporting under the new SOX regime. My results showed that improvements in internal control in the second year of SOX are associated with: (1) the number of audit committee members and (2) financial expertise of audit committee members. My third essay examined the relation between the appointment of the new executives and the subsequent receipt of initial section 404 of SOX opinions. My results showed that adverse SOX 404 reports will be more likely at firms that recently hired a new chief financial officer (CFO).
