Johtavana ideana on tutkia, kuinka uusi tietojärjestelmään vaikuttaa varastointiyritykseen. Lisäksi kappaleessa on ratkaisuehdotukset näihin pulmiin. Teoria osassa kerrotaan varastonhallinnan käytännön taustalla olevat teoriat.

ABSTRACT For decades, many organizations worldwide have been enduring heavy losses due to partial and total failures regarding their investments in information systems (IS), posing serious challenges to all management levels and IS practitioners. Alarming statistics in this regard and decades of practice in the IS area lead the author to place an emphasis on the end users (EU) who are appointed in representation of their peers (EUR) to IS development projects (ISDP), considering them as highly influential factors on the problem. Especially, focusing on certain EUR success factors, and their dimensions, deemed critical to any IS development and implementation, omitted or not thoroughly analyzed neither in the theory nor in the empirical research on the subject, so far. No studies were found in Latin America or elsewhere addressing the phenomenon of IS success/failure from the perspective presented herein. Hence, this empirical research has assessed to what degree such factors can influence the outcomes of an ISDP and their feasible impact on the EU´s satisfaction, being the latter accepted by several authors as the main measure of IS success. This study was performed in Latin America embracing four major industrial enterprises, which vertically integrate the aluminum sector of Venezuela, subjected to a macro ISDP to install the ERP-type package SAP/R3. The field work included surveying and interviewing experienced professionals such as IS executives, IS developers, IS project leaders and end-user project leaders. A triangulation methodological approach allowed combining quantitative and interpretive analyses, obtaining convergent and complementing results. A statistical analysis, using Partial Least Squares (PLS), was carried out followed by a hermeneutical/dialectical analysis. Results confirmed a major finding: in problematic cases, paradoxically, the origins of IS rejection reasons argued by the EU, at a high degree, were usually traceable to the EUR and themselves. The results also confirmed the prevalence of cognitive, behavioral and political factors in these organizations as well as the high risk of taking for granted the presence and quality of those factors demanded from the EUR. The statistical validation of the proposed model revealed the construct EUR knowledge as the main latent variable, with its items exerting a major influence on IS quality and success. Another finding, in contradiction with that of other studies, proved knowledge of information technology (IT) aspects to be irrelevant. The payroll and the human resources administration IS were the most problematic, as is usually the case in large companies. The main conclusions confirm the EU´s decisive role for IS success and their relationship with the problem, which continues, demanding more research and, from organizations, more attention and preparation. Neglecting human and social factors in organizations as well as their effective planning and management in preparation for ISDP poses serious risks. Despite the limitations of this work, the analyzed problem tends to influence ISDP in a wide range of organizations; regardless of their size or type of IS, thus showing a high degree of generalization. Therefore it is believed that the results, conclusions and suggestions of this research have a high degree of generalization. A detailed checklist comprising key measures is provided for preventive actions. Finally, the factors evaluated can be used to expand the well-known model of DeLone & McLean (2003), by connecting them as latent variables of its independent variables information quality and IS quality.

In this work the influence of four different ligands present in the xylem sap of Quercus ilex (histidine, citric, oxalic and aspartic acids) on Ni(II) adsorption by xylem was investigated. Grinded xylem was trapped in acrylic columns and solutions of Ni(II), in the absence and presence of the four ligands prepared in KNO(3) 0-1 mol L(-1) at pH 5.5, were percolated through the column. Aliquots of solutions were recovered in the column end for Ni determination by graphite furnace atomic absorption spectrometry (GFAAS). The experimental. data to describe Ni sorption by xylem in both the presence and absence of ligands was better explained by the Freundlich isotherm model. The decreasing affinity order of ligands for Ni was: oxalic acid > citric acid > histidine > aspartic acid. On the other hand, the Ni(II) adsorption by xylem increased following the inverse sequence of ligands. Potentiometric titrations of acidic groups were carried out to elucidate the sorption site groups available in Q. ilex xylem. The potentiometric titration has shown three sorption sites: pK(a) 2.6 (57.7% of the sorption sites), related to monobasic aliphatic carboxylic acids or nitrogen aromatic bases, pK(a) 8.1 (9.6%) and pK(a) 9.9 (32.7%), related to phenolic groups. Rms1 is one of the series of five ramosus loci in pea (Pisum sativum L.) in which recessive mutant alleles confer increased branching at basal and aerial vegetative nodes. Shoots of the nonallelic rms1 and rms2 mutants are phenotypically similar in most respects. However, we found an up to 40-fold difference in root-sap zeatin riboside ([9R]Z) concentration between rms1 and rms2 plants. Compared with wild-type (WT) plants, the concentration of [9R]Z in rms1 root sap was very low and the concentration in rms2 root sap was slightly elevated. To our knowledge, the rms1 mutant is therefore the second ramosus mutant (rms4 being the first) to be characterized with low root-sap [9R]Z content. Like rms2, the apical bud and upper nodes of rms1 plants contain elevated indole-3-acetic acid levels compared with WT shoots. Therefore, the rms1 mutant demonstrates that high shoot auxin levels and low root-sap cytokinin levels are not necessarily correlated with increased apical dominance in pea. A graft-transmissible basis of action has been demonstrated for both mutants from reciprocal grafts between mutant and WT plants. Branching was also largely inhibited in rms1 shoots when grafted to rms2 rootstocks, but was not inhibited in rms2 shoots grafted to rms1 rootstocks. These grafting results are discussed, along with the conclusion that hormone-like signals other than auxin and cytokinin are also involved.

Expression of the mRNAs encoding the astrocytic (EAAT1, EAAT2) and neuronal (EAAT3, EAAT4) excitatory amino acid transporters and the AMPA-type glutamate receptor subunits GluR2 and GluR3 was investigated in postmortem cerebellar extracts from a patient with olivopontocerebellar atrophy (OPCA) and in material from three age-matched controls. Decreased expression in the steady state level of EAAT4 mRNA in the OPCA sample was correlated with the selective loss of Purkinje cells. Neuropathological evaluation revealed reactive gliosis and concomitantly increased expression of the mRNA encoding astrocytic glial fibrillary acidic protein (GFAP). Expression of the mRNAs encoding the AMPA receptor subunits GluR2 and GluR3 subunits was found to be decreased in OPCA suggesting that excitotoxic mechanism could play a role in the pathogenesis of the selective neuronal cell death in this disorder.

It is predicted that dryland salinity will affect up to 17 Mha of the Australian landscape by 2050, and therefore, monitoring the health of tree plantings and remnant native vegetation in saline areas is increasingly important. Casuarina glauca Sieber ex Spreng. has considerable salinity tolerance and is commonly planted in areas with a shallow, saline water table. To evaluate the potential of using the nitrogenous composition of xylem sap to assess salinity stress in C. glauca, the responses of trees grown with various soil salinities in a greenhouse were compared with those of trees growing in field plots with different water table depths and groundwater salinities. In the greenhouse, increasing soil salinity led to increased allocation of nitrogen (N) to proline and arginine in both stem and root xylem sap, with coincident decreases in citrulline and asparagine. Although the field plots were ranked as increasingly saline-based on ground water salinity and depth-only the allocation of N to citrulline differed significantly between the field plots. Within each plot, temporal variation in the composition of the xylem sap was related to rainfall, rainfall infiltration and soil salinity. Periods of low rainfall and infiltration and higher soil salinity corresponded with increased allocation of N to proline and arginine in the xylem sap. The allocation of N to citrulline and asparagine increased following rainfall events where rain was calculated to have infiltrated sufficiently to decrease soil salinity. The relationship between nitrogenous composition of the xylem sap of C. glauca and soil salinity indicates that the analysis of xylem sap is an effective method for assessing changes in salinity stress in trees at a particular site over time. However, the composition of the xylem sap proved less useful as a comparative index of salinity stress in trees growing at different sites.

Stomatal conductance (g(s)) of pepper (Capsicum annuum L.) plants decreased during the second photoperiod (day 2) after withholding nitrate (N). Stomatal closure of N-deprived plants was not associated with a decreased shoot water potential (Psi(shoot)); conversely Psi(shoot) was lower in N-supplied plants. N deprivation transiently (days 2 and 3) alkalized (0.2-0.3 pH units) xylem sap exuded from de-topped root systems under root pressure, and xylem sap expressed from excised shoots by pressurization. The ABA concentration of expressed sap increased 3-4-fold when measured on days 2 and 4. On day 2, leaves detached from N-deprived and N-supplied plants showed decreased transpiration rates when fed an alkaline (pH 7) artificial xylem (AX) solution, independent of the ABA concentration (10-100 nM) supplied. Thus changes in xylem sap composition following N deprivation can potentially close stomata. However, the lower transpiration rate of detached N-deprived leaves relative to N-supplied leaves shows that factors residing within N-deprived leaves also mediate stomatal closure, and that these factors assume greater importance as the duration of N deprivation increases.
