Les WNTs sont des glycoprotéines sécrétées impliquées dans plusieurs processus tels que la spécification cellulaire, la prolifération, la différenciation, et beaucoup d’autres. Pour transmettre leur signal, les WNTs se lient aux récepteurs Frizzled (FZD) et au co-récepteur « Low-density-lipoprotein receptor Related Protein » (LRP) 6 ou 5, activant ainsi l’une des trois principales voies de signalisation: la voie de signalisation WNT/β-caténine (voie canonique), la voie « Planar Cell Polarity » (PCP) et la voie WNT/Ca2+ (voies non-canoniques). Des antagonistes de cette voie, les « Secreted Frizzled Related Protein » (SFRPs), peuvent aussi se lier aux WNTs pour empêcher leur liaison aux récepteurs FZDs. Bien que des rôles importants aient été associés à plusieurs composants de la voie des WNTs lors de la régulation de l’ovaire adulte, le fonctionnement exact de cette signalisation reste nébuleux. L’objectif global de cette thèse visait donc à mieux comprendre la voie de signalisation des WNTs au niveau de l’ovaire adulte de souris, par la caractérisation de deux autres composants de cette voie, FZD1 et SFRP4. La création et l’analyse de souris Fzd1 et Sfrp4 KO ont démontré que FZD1 est nécessaire pour la régulation des gènes associés à l’expansion du cumulus, dans le complexe ovocyte-cumulus (COC). Nous avons aussi constaté que SFRP4 avait un rôle à jouer lors de la régulation des gènes associés à l’expansion du cumulus mais cette fois, au niveau des cellules de la granulosa. Finalement, les résultats in vivo et in vitro de cette étude ont suggéré que la voie PCP, contrairement à la voie canonique, pourrait être modulée dans les cellules de la granulosa des souris Sfrp4 KO, possiblement grâce au signal induit par WNT4 et WNT5a. Ces données ont permis de créer un modèle hypothétique représentant la régulation de la signalisation ovarienne par les WNTs. Ce modèle servira de base pour l’élaboration de futurs projets de recherche visant à comprendre davantage la signalisation ovarienne et les possibles effets de sa dérégulation lors de processus pathologiques. Ces connaissances pourront ensuite être appliquées chez l’humain afin de traiter plusieurs maladies ou dérèglements ovariens.

L’ostéoarthrose (OA) se caractérise par une perte du cartilage articulaire, une sclérose osseuse, et une inflammation de la membrane synoviale. Des études in vivo et in vitro indiquent que les modifications du tissu osseux sont responsables de la perte du cartilage articulaire. Les ostéoblastes (Ob) OA présentent une réduction de la réponse à l’hormone parathyroïdienne (PTH), un signal anabolique important pour le tissu osseux, versus les Ob normaux. Le récepteur à la PTH (PTH-R) interagit avec le LRP6, le récepteur des ligands Wnts, et les antagonistes Dickkopf (DKK) bloquent cette interaction. Puisque le niveau de DKK2 est élevé en réponse au TGF-β1 dans les OA Ob, nous proposons que DKK2 altère l’interaction LRP6/PTH-R, le recyclage de PTH-R, et inhibe la réponse à la PTH. Nous avons utilisé des Ob OA et normaux humains en culture primaire. L’expression de PTH-R, LPR6 et DKK2 est mesurée par RT-PCR. Les niveaux protéiques de LRP6, PTH-R et DKK2 ont été déterminés par immunobuvardage. L’inhibition de DKK2 s’est effectuée par siRNA et l’AMP cyclique (AMPc) a été mesurée par ELISA. L’effet de TGF-β1 sur l’expression de DKK2 et PTH-R a été testé sur des cellules d’ostéosarcome SaOS-2. L’expression de PTH-R et LRP6 est similaire entre Ob normaux et OA, mais le niveau protéique de PTH-R est réduit dans les Ob OA. Par contre, l’expression et la production de DKK2 sont plus élevées dans les Ob OA. L’inhibition de DKK2 ne modifia pas l’expression de LRP6 et PTH-R dans les Ob OA mais a augmenté le niveau protéique de PTH-R détecté par immunobuvardage alors que celui de LRP-6 demeura inchangé. L’inhibition de DKK2 dans les Ob OA entraîna une augmentation de PTH-R dans la fraction membranaire et une diminution de la fraction intracellulaire. Les résultats de l'inhibition de la voie de clathrine par le triflupromazine ont montré une expression accrue du récepteur PTH dans la fraction membranaire qui peut être due à l'inhibition de sa dégradation par la voie de clathrine. L’induction de DKK2 dans les cellules SaOS-2 par TGF-β1 entraîna l’inhibition de PTH-R mais non celle de LRP6. L’inhibition de DKK2 dans les Ob OA a stimulé la production d’AMPc en réponse à la PTH par les Ob OA. En outre, le traitement de TGF-β1 dans les cellules SaOS-2 réduit la production d'AMPc en réponse à la PTH.Ces résultats démontrent que les niveaux élevés de DKK2, via l’inhibition de la signalisation Wnt/bcaténine, sont aussi responsables de l’altération de la réponse à la PTH observée dans les Ob OA. Le niveau élevé de DKK2 diminue spécifiquement l’affichage membranaire de PTH-R dans ces cellules et non son expression. Ces résultats suggèrent donc une altération du cross-talk entre LRP6 et PTH-R dans les Ob OA en lien avec leur niveau de DKK2.

Low-density lipoprotein (LDL) receptors are overexpressed in most neoplastic cell lines and provide a mechanism for the internalization and concentration of drug-laden nanoemulsions that bind to these receptors. The aim of the present study was to determine whether the administration of standard chemotherapeutic schemes can alter the expression of LDL and LDL receptor-related protein 1 (LRP-1) receptors in breast carcinoma. Fragments of tumoral and normal breast tissue from 16 consecutive volunteer women with breast cancer in stage II or III were obtained from biopsies before the beginning of neoadjuvant chemotherapy and after chemotherapy, from fragments excised during mastectomy. Tissues were analyzed by immunohistochemistry for both receptors. Because complete response to treatment was achieved in 4 patients, only the tumors from 12 were analyzed. Before chemotherapy, there was overexpression of LDL receptor in the tumoral tissue compared to normal breast tissue in 8 of these patients. LRP-1 receptor overexpression was observed in tumors of 4 patients. After chemotherapy, expression of both receptors decreased in the tumors of 6 patients, increased in 4 and was unchanged in 2. Nonetheless, even when chemotherapy reduced receptors expression, the expression was still above normal. The fact that chemotherapy does not impair LDL receptors expression supports the use of drug carrier systems that target neoplastic cells by the LDL receptor endocytic pathway in patients on conventional chemotherapy.

Görgeyite, K2Ca5(SO4)6··H2O, is a very rare monoclinic double salt found in evaporites related to the slightly more common mineral syngenite. At 1 atmosphere with increasing external temperature from 25 to 150 °C, the following succession of minerals was formed: first gypsum and K2O, followed at 100 °C by görgeyite. Changes in concentration at 150 °C due to evaporation resulted in the formation of syngenite and finally arcanite. Under hydrothermal conditions, the succession is syngenite at 50 °C, followed by görgyeite at 100 and 150 °C. Increasing the synthesis time at 100 °C and 1 atmosphere showed that initially gypsum was formed, later being replaced by görgeyite. Finally görgeyite was replaced by syngenite, indicating that görgeyite is a metastable phase under these conditions. Under hydrothermal conditions, syngenite plus a small amount of gypsum was formed, after two days being replaced by görgeyite. No further changes were observed with increasing time. Pure görgeyite showed elongated crystals approximately 500 to 1000 µ m in length. The infrared and Raman spectra are mainly showing the vibrational modes of the sulfate groups and the crystal water (structural water). Water is characterized by OH-stretching modes at 3526 and 3577 cm–1 , OH-bending modes at 1615 and 1647 cm–1 , and an OH-libration mode at 876 cm–1 . The sulfate 1 mode is weak in the infrared but showed strong bands at 1005 and 1013 cm–1 in the Raman spectrum. The 2 mode also showed strong bands in the Raman spectrum at 433, 440, 457, and 480 cm–1 . The 3 mode is characterized by a complex set of bands in both infrared and Raman spectra around 1150 cm–1 , whereas 4 is found at 650 cm–1.

The use of polycaprolactone (PCL) as a biomaterial, especially in the fields of drug delivery and tissue engineering, has enjoyed significant growth. Understanding how such a device or scaffold eventually degrades in vivo is paramount as the defect site regenerates and remodels. Degradation studies of three-dimensional PCL and PCL-based composite scaffolds were conducted in vitro (in phosphate buffered saline) and in vivo (rabbit model). Results up to 6 months are reported. All samples recorded virtually no molecular weight changes after 6 months, with a maximum mass loss of only about 7% from the PCL-composite scaffolds degraded in vivo, and a minimum of 1% from PCL scaffolds. Overall, crystallinity increased slightly because of the effects of polymer recrystallization. This was also a contributory factor for the observed stiffness increment in some of the samples, while only the PCL-composite scaffold registered a decrease. Histological examination of the in vivo samples revealed good biocompatibility, with no adverse host tissue reactions up to 6 months. Preliminary results of medical-grade PCL scaffolds, which were implanted for 2 years in a critical-sized rabbit calvarial defect site, are also reported here and support our scaffold design goal for gradual and late molecular weight decreases combined with excellent long-term biocompatibility and bone regeneration. Having flexible notions of the unit (e.g., 26 ones can be thought of as 2.6 tens, 1 ten 16 ones, 260 tenths, etc.) should be a major focus of elementary mathematics education. However, often these powerful notions are relegated to computations where the major emphasis is on "getting the right answer" thus procedural knowledge rather than conceptual knowledge becomes the primary focus. This paper reports on 22 high-performing students' reunitising processes ascertained from individual interviews on tasks requiring unitising, reunitising and regrouping; errors were categorised to depict particular thinking strategies. The results show that, even for high-performing students, regrouping is a cognitively complex task. This paper analyses this complexity and draws inferences for teaching.

The mixed anion mineral dixenite has been studied by Raman spectroscopy, complimented with infrared spectroscopy. The Raman spectrum of dixenite shows bands at 839 and 813 cm-1 assigned to the (AsO3)3- symmetric and antisymmetric stretching modes. The most intense Raman band of dixenite is the band at 526 cm-1 and is assigned to the ν2 AsO33- bending mode. DFT calculations enabled the position of AsO22- symmetric stretching mode at 839 cm-1, the antisymmetric stretching mode at 813 cm-1, and the deformation mode at 449 cm-1 to be calculated. Raman bands at 1026 and 1057 cm-1 are assigned to the SiO42- symmetric stretching vibrations and at 1349 and 1386 cm-1 to the SiO42- antisymmetric stretching vibrations. Both Raman and infrared spectra indicate the presence of water in the structure of dixenite. This brings into question the commonly accepted formula of dixenite as CuMn2+14Fe3+(AsO3)5(SiO4)2(AsO4)(OH)6. The formula may be better written as CuMn2+14Fe3+(AsO3)5(SiO4)2(AsO4)(OH)6•xH2O.

This paper examines the role of powerful entities and coalitions in shaping international accounting standards. Specifically, the focus is on the process by which the International Accounting Standards Board (IASB) developed IFRS 6, Exploration for and Evaluation of Mineral Resources. In its Issues Paper, the IASB recommended that the successful efforts method be mandated for pre-production costs, eliminating the choice previously available between full cost and successful efforts methods. In spite of the endorsement of this view by a majority of the constituents who responded to the Issues Paper, the final outcome changed nothing, with choice being retained. A compelling explanation of this disparity between the visible inputs and outputs of the standard setting process is the existence of a “black box”, in which powerful extractive industries entities and coalitions covertly influenced the IASB to secure their own ends and ensure that the status quo was maintained

The structures of the anhydrous 1:1 proton-transfer compounds of isonipecotamide (4-carbamoylpiperidine) with picric acid and 3,5-dinitrosalicylic acid, namely 4-carbamoylpiperidinium 2,4,6-trinitrophenolate, C6H13N2O8+ C6H2N3O7- (I) and 4-carbamoylpiperidinium 2-carboxy-4,6-dinitrophenolate, C6H13N2O8+ C7H3N2O7-: two forms, the monoclinic alpha-polymorph (II) and the triclinic beta-polymorph (III) have been determined at 200 K. All compounds form hydrogen-bonded structures, one-dimensional in (II), two-dimensional in (I) and three-dimensional in (III). In (I), the cations form centrosymmetric cyclic head-to-tail hydrogen-bonded homodimers [graph set R2/2(14)] through lateral duplex piperidinium N---H...O(amide) interactions. These dimers are extended into a two-dimensional network structure through further interactions with anion phenolate-O and nitro-O acceptors, including a direct symmetric piperidinium N-H...O(phenol),O(nitro) cation--anion association [graph set R2/1(6)]. The monoclinic polymorph (II) has a similar R2/1(6) cation-anion hydrogen-bonding interaction to (I) but with an additional conjoint symmetrical R1/2(4) interaction as well as head-to-tail piperidinium N-H...O(amide) O hydrogen bonds and amide N-H...O(carboxyl) hydrogen bonds, give a network structure which include large R3/4(20) rings. The hydrogen bonding in the triclinic polymorph (III) is markedly different from that of monoclinic (II). The asymmetric unit contains two independent cation-anion pairs which associate through cyclic piperidinium N-H...O,O'(carboxyl) interactions [graph set R2/1(4)]. The cations also show the zig-zag head-to-tail piperidinium N-H...O(amide) hydrogen-bonded chain substructures found in (II) but in addition feature amide N-H...O(nitro) and O(phenolate) and amide N-H...O(nitro) associations. As well there is a centrosymmetric double-amide N-H...O(carboxyl) bridged bis(cation-anion) ring system [graph set R2/4(8)] in the three-dimensional framework. The structures reported here demonstrate the utility of the isonipecotamide cation as a synthon with previously unrecognized potential for structure assembly applications. Furthermore, the structures of the two polymorphic 3,5-dinitrosalicylic acid salts show an unusual dissimilarity in hydrogen-bonding characteristics, considering that both were obtained from identical solvent systems.

Over the last three years, in our Early Algebra Thinking Project, we have been studying Years 3 to 5 students’ ability to generalise in a variety of situations, namely, compensation principles in computation, the balance principle in equivalence and equations, change and inverse change rules with function machines, and pattern rules with growing patterns. In these studies, we have attempted to involve a variety of models and representations and to build students’ abilities to switch between them (in line with the theories of Dreyfus, 1991, and Duval, 1999). The results have shown the negative effect of closure on generalisation in symbolic representations, the predominance of single variance generalisation over covariant generalisation in tabular representations, and the reduced ability to readily identify commonalities and relationships in enactive and iconic representations. This chapter uses the results to explore the interrelation between generalisation and verbal and visual comprehension of context. The studies evidence the importance of understanding and communicating aspects of representational forms which allowed commonalities to be seen across or between representations. Finally the chapter explores the implications of the studies for a theory that describes a growth in integration of models and representations that leads to generalisation.
